        Quantitative Assay for the Binding of Jun-Fos Dimer and Activator Protein-1 Site

        The Jun and Fos families of eukaryotic transcription factors form heterodimers capable of binding to their cognate DNA enhancer elements. We are interested in searching for inhibitors or antagonists of the binding of the Jun-Fos heterodimer to the activator protein-1 (AP-1) site. The basic-region leucine zipper (bZIP) domain of c-Fos was expressed as a fusion protein with glutathione S-transferase, and allowed to form a heterodimer with the bZIP domain of c-Jun. The heterodimer was bound to glutathione-agarose, to which were added radiolabeled AP-1 nucleotides. After thorough washing, the gel-bound radioactivity was counted. The assay is faster than the conventional electrophoretic mobility shift assay because the gel electrophoresis step and the autoradiography step are eliminated. Moreover, the assay is very sensitive, allowing the detection of picomolar quantities of nucleotides, and is not affected by up to 50% dimethylsulfoxide, a solvent for hydrophobic inhibitors. Curcumin and dihydroguaiaretic acid, recently known inhibitors of Jun-Fos-DNA complex formation, were applied to this Jun-GST-fused Fos system and revealed to decrease the dimer-DNA binding.

        개에서 이중 결찰법을 통한 동맥관 개존증의 완치 예

        A ten months old, female Yorkshire terrier weighing 2.88 kg referred to veterinary teaching hospital of college of veterinary medicine, Konkuk University because of syncope, cough and dyspnea. First hematological and serum chemical test revealed thrombocytopenia, mild anemia, and increase of concentration of ALP (195 U/L). On 57 days later, second hematological and serum chemical test revealed polycythemia, increase of concentration of ALP (211 U/L), and Tchol (387 mg/dl). Right atrium enlargement, main pulmonary artery bulge and cardiomegaly (VHS = 11.5) were observed in radiographic findings. Ultrasonographic images showed both right and left ventricular dilation and turbulent flow between the descending aorta and the main pulmonary artery in color Doppler imaging. ECG showed left ventricular enlargement, SA block, and electrical alternant. Thoracotomy was performed through left fourth intercostal incision under isoflurane anesthesia. Patent ductus arteriosus was double ligated with 1-0 silk. Cough and dyspnea disappeared on 5 days after operation. Turbulent flow was not found in color doppler imaging of ultrasonography on 10 days after operation. Ten months later after the operation, syncope could not exist any more.

        한국인 직무 스트레스 측정도구의 개발 및 표준화

        Background and Purposes: Over the past three decades, numerous studies performed in Korea have reported that job stress is a determinant risk factor for chronic diseases and work disability. Every society has its own culture and occupational climate particular to their organizations, and hence experiences different occupational stress. An occupational stress measurement tool therefore needs to be developed to estimate it objectively. The purpose of this study is to develop and standardize the Korean Occupational Stress Scale (KOSS) which is considered to be unique and specific occupational stressors in Korean employees. Subjects and Methods: Data were obtained from the National Study for Development and Standardization of Occupational Stress (NSDSOS Project: 2002-2004). A total of 12,631 employees from a nationwide sample proportional to the Korean Standard Industrial Classification and the Korean Standard Occupational Classification were administered. The KOSS was developed for 2 years (2002-2004). In the first year, we collected 255 items from the most popular job stress measurement tools such as JCQ, ERI, NIOSH and OSI, and 44 items derived from the a qualitative study (depth interview). Forty-three items of KOSS, in the second year, were retained for use in the final version of the KOSS by using Delphi and factor analysis. Items were scored using conventional 1-2-3-4 Likert scores for the response categories. Results: We developed eight subscales by using factor analysis and validation process: physical environment (3 items), job demand (8 items), insufficient job control (5 items), interpersonal conflict (4 items), job insecurity (6 items), organizational system (7 items), lack of reward (6 items), and occupational climate (4 items). Together they explained 50.0% of total variance. Internal consistency alpha scores were ranged from 0.51 to 0.82. Twenty-four items of the short form of the KOSS (KOSS-SF) were also developed to estimate job stress in the work setting. Because the levels of the subscales of occupational stress were gender dependent, gender-specific standard norms for both the 43-item full version and the 24-item short form using a quartile for the subscales of KOSS were presented. Conclusion: The results of this study suggest that KOSS might be an appropriate measurement scale to estimate occupational stress of Korean employees. Further and more detailed study needs to be conducted to improve the validity of this scale.

      • 인터넷뱅킹 서비스품질의 구성요인에 관한 탐색적 고찰

        본 연구는 인터넷뱅킹 금융상품과 서비스의 품질 요인을 구성하고 있는 주요 속성을 살펴보기 위해 문헌연구를 통해 인터넷 뱅킹 서비스품질 속성을 고찰하고자 하였다. 본 연구에서 문헌연구를 통해 인터넷뱅킹 서비스품질에 대한 속성을 살펴본 결과 고객 서비스품질, 온라인 시스템품질, 은행서비스 금융상품품질 등의 세 개의 분류를 통해 인터넷뱅킹 서비스품질의 주요 구성 요인이 모두 19개의 차원으로 구성되어져 있음을 제시하였다. 즉, 고개 서비스품질 차원은 ① 신뢰성 ② 응답성 ③ 능력 ④ 예절 ⑤ 신뢰 ⑥ 접근가능성 ⑦ 커뮤니케이션 ⑧ 고객에 대한 이해 ⑨ 협조 ⑩ 지속적 개선차원으로, 온라인 시스템품질 차원은 ① 컨텐츠 ② 정확성 ③ 사용용이성 ④ 적시성 ⑤ 심미성 ⑥안전성 ⑦ 편의성 ⑧ 비용절감차원으로, 은행 서비스 금융상품품질 차원은 상품 다양성과 상품 특성의 차원으로 구분하였다.

      • 비혈연간 조혈모세포이식 후 재발한 만성골수성백혈병에서 Gleevec에 의한 완전관해 및 공여자 조혈 재건 1예

        저자들은 만성골수성백혈병으로 동종 골수이식 후 림프구성급성전환으로 재발한 환자에서, Gleevec 투약 후 DLI (donor lymphocyte infusion) 없이 공여자 조혈(donor hematopoiesis)의 재건이 이루어지고, 세포유전학적, 분자생물학적 기준에서 모두 완전관해에 이르고, 12개월째 관해를 유지하고 있는 환자를 경험하였기에 이를 보고하는 바이다. 본례의 환자에서 Gleevec은 동종 골수이식 후 급성전환기로 재발한 환자에서도 매우 효과적인 것으로 나타났다. Gleevec은 동종 골수이식 후 재발한 만성골수성백혈병에서 Interferon-α, DLI, 2차골수이식 등 기존의 치료의 문제점을 보완할 수 있는 유용한 치료방법으로 사용될 수 있을 것으로 생각되며, 이의 장기적 치료효과의 평가와 기존치료와의 병합요법의 유용성 등에 대한 향후 연구가 필요할 것으로 생각된다. We report a 32-year old woman with chronic myelongenous leukemia (CML) in lymphoid blast crisis, who underwent allogenic bone marrow transplantation (BMT) from HLA phenotype full mached unrelated donor. Her disease relapsed into full-blown blast crisis. Upon treatment with gleevec, she achieved immediate remission of leukemia as well as the donor type hematopoiesis without donor lymphocyte infusion (DLI). Our case suggests the high effectiveness of gleevec in CML relapsed after allogeneic BMT. Further studies are needed to define the role of gleevec (with or without DLI) in the management of CML relapsed after allogeneic BMT.

      • 단기배양을 통한 말초혈액 CD34 양성세포의 체외증폭

        Background: It is suggested that clinical practice in the areas of bone marrow transplantation and gene therapy might rely on the ex vivo expansion of hematopoietic stem/progenitor cells. However, the condition for ex vivo expansion of hematopoietic progenitor cells is not well established. The authors pursued a series of experiments to define the proper conditions for the expansion of hematopoietic cells in the short-term liquid suspension culture of mobilized peripheral blood CD34+ cells. Methods: 1.0ml cultures were initiated with 9×10^3 PB CD34+ cells, which were isolated from PB mononuclear cells (MNCs) by high-gradient cell sorting, in 12 well plates with the various combinations of hematopoietic growth factors(HGF). The following recombinant human HGFs were used: stem cell factor(SCF) 100ng/ml, granulocyte colony-stimulating factor(G-CSF) 100ng/ml, GM-CSF(granulocyte, macrophage colony-stimulating factor) 100ng/ml, interleukin-1 beta(IL-1B) 1ng/ml, interleukin-3(IL-3) 20ng/ml, interleukin-6 (IL-6) 100ng/ml. At the end of culture, colony-forming cells were evaluated by semisolid clonogenic assay. Results: 1) Using the high-gradient magnetic sorting system, CD34^+ cells were isolated with a yield of 40 3% 2) In 7 day culture of PB CD34^+ cells(9×10^3 cells), nucleated cells expanded mean 10×10^3(range, 9 to 20×10^3) with the addition of SCF alone, 35×10^3(range, 10 to 60×10^3) with SCF plus G-CSF plus GM-CSF, and 130×10^3(range, 40 to 300×10^3) with the combination of SCF, G-CSF, IL-1, IL-3, IL-6, GM-CSF. In 14 day culture, nucleated cells expanded 10×10^3 to 1,860×10^3 with combination of human hematopoietic growth factors. 3) In 10 day culture without medium change of PB CD34^+ cells, CFU-GM numbers expanded 16. 5 fold(range, 7 to 59 fold) with the addition of SCF plus G-CSF plus Il-1 plus IL-3, 31.3 fold(range, 20.5 to 101.1 fold) with the combination of SCF, G-CSF, IL-1, IL-3, GM-CSF. In 14 day culture with or without medium change of PB CD34^+ cells was inferior to 10 day culture for CFU-GM expansion. 4) There was no significant difference for CFU-GM expansion between five growth factors(SCF,G-CSF,IL-1,IL-3,GM-CSF) and six growth factors(five growth factors plus IL-6). Conclusion: The authors could confirm that short-term suspension culture of peripheral blood CD34+ cells could expand hematopoietic progenitor cells. Ten-day culture with medium change of CD34+ cells with the addition of five growth factors, i.e. SCF, G-CSF, IL-1B, IL-3, and GM-CSF, might be the most efficient in this system.

        The purpose of the research is to study the prediction of heat transfer on flat heat pipe applid to solar energy collector by the numerical study. Numerical analysis was performed by finite element method for the prediction of heat transfer on flat transfer on flat heat pipe. A comparison between the experimental and the numerical results show qualitatively agreement and it was able to predict the performence of flat heat pipe collector. The Biot number, (h_(ev)l)/Ks in order to know the effect of heat transfer on flat heat pipe was show to decrease by increas of heat transfer coefficient in vapor region, and with higher conductivity of container, Ks.
