        자동혈구분석기 Hemavet 950FS를 이용한 돼지 혈액 내 혈구분석의 유효시간 평가

        본 연구는 자동혈구분석기 Hemavet 950FS를 이용한 돼지 혈액 내 혈구분석의 유효성을 검증하고, 정확한 분석을 위한 유효시간을 결정하기 위하여 실시하였다. 분석시료는 체중 60 kg (±3.5)의 Duroc 종 34두로부터 채취하였으며, 이들 중에서 분석치가 정상범위를 벗어나거나 용혈이 발견된 시료는 연구에서 제외하고 최종적으로 10개의 시료를 본 연구에 이용하였다. 임의의 하나의 시료에 대해 10회 연속분석을 통하여 일내변동을 조사한 결과, leukocytes계 혈구들 중에서 백혈구, 호중구, 림프구에서는 낮은 일내변동으로 분석의 유효성이 인정되었으나, 단핵구, 호산구, 호염구에 있어서는 큰 변동을 나타내었다. 이와 반면에 erythrocytes 및 thrombocytes계 혈구들의 모든 지표들은 안정적인 일내변동이 관찰되었다. 채혈 후 시간경과에 따른 혈구변화를 분석한 결과 leukocytes계 혈구 및 thrombocytes계 혈구는 시간경과에 따라 검출농도가 유의적으로 감소되는 것으로 관찰되었다. 반면, erythrocytes계 혈구들은 채혈 후 120시간까지는 안정적으로 분석이 가능한 것으로 나타났다. 본 연구의 결과를 종합하면, Hemavet 950FS는 돼지의 혈액 내에서 단핵구, 호산구, 호염구를 제외하고 일반 혈구분석에 유용한 것으로 판단된다. 또한, leukocytes 및 thrombocytes계 혈구들에 대한 정확한 분석을 위해서는 채혈 후 4시간 이내에 분석이 이루어져야하며, erythrocytes계 혈구들은 채혈 후 120시간까지는 안정적으로 검출이 가능한 것으로 사료된다. This study was conducted to determine the valid time for accurate detection of complete blood cell count (CBC) in pig blood using an automatic blood corpuscle analyzer (Hemavet 950FS). Blood samples were collected from 34 pigs (Duroc) with a 60 kg (±3.5) body weight. Ten specimens with CBC parameters in normal range and with no hemolysis were selected among 34 samples and used in this study. Regarding leukocytes parameters, white blood cell (WBC), neutrophil (NE), and lymphocyte (LY) counts showed a low daily variation (coefficient of variation, CV), whereas monocyte (MO), eosinophil (EO), and basophil (BA) CVs were significantly high (19.7, 56.9, and 53.3%, respectively). On the other hand, all parameters of erythrocytes and thrombocytes showed stable daily variation. All parameters of leukocytes and thrombocytes were significantly reduced as storage time passed (P<0.01 or 0.001), except for lymphocytes (P=0.535). However, no significant differences were observed in parameters of erythrocytes from blood up to 120 hours. From above results, we assert that Hemavet 950FS is useful in analyzing CBC, except for MO, EO, and BA. For accurate detection of leukocyte and thrombocyte parameters, analysis should be performed within 4 hours after blood collection when using Hemavet 950FS. On the other hand, parameters of erythrocytes could be stably detected for at least 120 hours after blood collection.

        쏘가리 (Siniperca scherzeri) 말초혈액 백혈구의 미세구조 분석

        광학현미경과 전자현미경을 이용하여 쏘가리(Siniperca scherzeri0의 말초혈액 세포의 형태적 특징을 조사하였다. 쏘가리 말초 혈구는 크게 적혈구, 트롬보사이트, 백혈구 등 3가지로 구분되었다. 적혈구는 성숙 적혈구, 미성숙 적혈구, 적혈 아세포 등 3가지로 구분되었다. 트롬보사이트는 둥근형에서 방추난형으로 구분되었고, 핵은 이질염색질이 풍부한 난형이었으며, 세포질에는 소포, 미세간질이 관찰되었다. 림프구는 소형 림프구(type Ⅰ), 중형 림프구(type Ⅱ), 대형 림프구(type Ⅲ)로 구분되었다. 과립백혈구는 핵과 과립의 형태에 따라 이질핵 과립백 혈구와 호산성 백혈구로 구분되었는데, 이질핵 과립백혈구는 다형립 핵과 세포질내 크고 작은 과립이 풍부했고, 호산성 과립백혈구는 둥근 핵과 크고 작은 둥근 과립이 풍부했다. 과립백혈구의 과립의 형태는 크고 작은 방추형과 원형, 침형 또는 난형으로 구분되었다. 쏘가리의 단핵구/대식세포는 형태적으로 포유동물과 유사하였다. The morphological characteristics of peripheral blood cells in the freshwater mandarin fish (Siniperca scherzeri) were investigated by light and electron microscopy. Erythrocytes, thrombocytes, lymphocytes, granulocytes and monocytes were identified as the peripheral blood cells. Erthrocytes were divided into three form types: mature, immature, and erthroblasts. Thrombocytes were round to long oval (fusiform), each containing vesicular and microtubular structures and an oval nucleus with abundant heterochromatins. Lymphocytes were divided into three types in size: small, medium, and large. Granulocytes were distinguished into three types (type Ⅰ, type Ⅱ, and type Ⅲ) according to the morphology of the granules and nucleus. Heterophilic granulocytes possessed lobulated nuclei and a large number of cytoplasmic granules, each of which was oval and contained electron-dense materials and a crystalloid. Acidophilic granulocytes possessed large and small granules, and round nuclei. The granules were filled with regularly arranged fibriform materials, some needle-like structures, and oval forms. Monocytes and lymphocytes in this fish species were morphologically similar to those of mammals.

        닭의 혈액내 단핵세포 표면항원 특이 단클론성 항체 생산

        This study was performed to produce monoclonal antibodies (mAb) specifically reacting with chicken leukocyte surface antigens. Popliteal lymph node cells of BALB/c mice previously immunized through foot-pad with peripheral blood mononuclear cells (PBMC) of chickens separated by Ficoll-Histopaque method. They were fused with P3X63Ag14 mouse myeloma cells. A total of 34 hybridomas secreted antibodies specifically binding to the PBMC. According to the reactivity patterns with PBMC, the mAbs were divided into 4 groups. Group 1 mAbs (IIB3, IIB10, IIE10) specifically reacted with non-adherent lymphocytes but not with adherent cells which were mainly composed of thrombocytes and monocytes in PBMC culture. These mAbs were reactive with 25-59% of thymus cells and 42-64% of spleen cells of chickens. They did not show any significant reactivity with cells in the bursa of Fabricius, T-cell (MDCC-MSB1) and B-cell (LSCC-1104B1) lines. These results indicate that Group I mAbs specifically reacted with T-lymphocyte subpopulation. Monoclonal antibodies in Group II (IC6, IG2-2 and IID9) showed specific reactivity with monocytes but not with thrombocytes or non-adherent cells in PBMC culture. These mAbs, though not reacted with the chicken macrophage cell line, HD11, also bound to macrophages of the spleen and lung in immunohistochemical staining. Five mAbs in Group III showed characteristics of binding to lymphocytes and monocytes, but not to thrombocytes. Twenty-three mAbs in Group IV showed specific reactivity to lymphocytes, monocytes, and thrombocytes. Two mAbs (IC3 and IE9) in Group IV reacted with most of PBMC.

      • Prognostic Significance of Basic Laboratory Methods in Non-Small-Cell-Lung Cancer

        Background: In our study, the LDH, albumin, hemoglobin, neutrophile, thrombocyte, lymphocyte counts and prognostic significance of neutrophile-lymphocyte and thrombocyte-lymphocyte ratios in NSCLC derived from these counts obtained during regular examinations of patients were examined. Materials and Methods: Histopathologically diagnosed non-small-cell-lung cancer patients between 2008 and 2010 were included in the study. Before the treatment, full blood count including routine lymphocyte count, blood biochemistry examinations including liver (AST, ALT, total protein, Albumin), LDH and kidney (BUN, Cre) function tests were performed. Results: A total of 156 patients, 76 of whom (48.7%) were female and 80 of whom (51.3%) were male were included. Mean hemoglobin level was determined as 12. Overall survival was found to be significantly dependent on whether patients were anemic or not (p: 0.005). Mean LDH level was determined as 233.4. There was nosurvival difference between patients with and without high LDH (p: 0.532). In patients where NLR showed systemic inflammatory response, overall survival was 10.8 months whereas this duration was 19.6 months in patients where the systemic inflammatory response was negative (p: 0.012). In patients where TLR showed systemic inflammatory response, overall survival was 13.6 months whereas this duration was 21.9 months in patients where the systemic inflammatory response was negative (p: 0.04). Conclusions: Molecular methods have been changing rapidly in today's world and they manage the treatment besides defining the prognosis of patients. However, easily accessible and cheap laboratory parameters should be considered in the prognosis of patients besides these new methods.

        견(犬)에 있어서 비장적출(脾臟摘出)이 혈액(血液) 및 골수거대핵세포상(骨髓巨大核細胞像)에 미치는 영향(影響)

        Present experiments were undertaken in order to clarify the effect of splenectomy on the hematology and marrow megakaryocyte picture and to know the genesis of postsplenectomy thrombocytosis in dogs. Six mongrel dogs weighing 8.5~18㎏ were used, of which three were splenectomized and the other three were laparotomized for comparison. Erythrocyte count, total and differential leukocyte counts, thrombocyte count and packed cell volume measurement were made using the blood samples. In addition, bone marrow samples obtained from the femur at 7th and 23rd day of the operation were examined for the number per low-power field, the diameter, and the distribution frequency of the megakaryocyte. From these experiments, following results were obtained : Erythrocyte count and packed cell volume showed significant decrease beginning on the 15th day of splenectomy. Total and differential leukocyte counts showed marked increase for the first 2 days of postsplenectomy. The thrombocyte count of splenectomized dogs increased from the 2nd day of the operation, reached to the peak count on the 15th day, and returned to the preoperation count by the 28th day. The megakaryocyte count per low-power field of the biopsied preparation increased in according to the increase in thrombocyte count. The megakaryocyte diameter of splenectomized dog showed no increase on the 7th or 23rd day of the operation. However, the distribution frequency of the larger megakaryocyte was higher in the splenectomized dogs than in the laparotomized dogs. The total plasma protein concentration showed no significant change after splenectomy or laparotomy. From these results, it may be concluded that the postsplenectomy thrombocytosis results from the increased megakaryocytopoesis or the activated thrombocytopoesis of the marrow megakaryocytes.

        Cholestatic Hepatitis and Thrombocytosis in a Secondary Syphilis Patient

        The incidence of acute hepatitis in syphilis patient is rare. First of all, our patient presented with hepatitis comorbid with thrombocytosis. To our knowledge, this is only the second report of syphilitic hepatitis with thrombocytosis. The 42-yr-old male complained of flulike symptoms and skin eruptions on his palms and soles. Laboratory findings suggested an acute hepatitis and thrombocytosis. Serologic test results were positive for VDRL. He recovered from his symptoms and elevated liver related enzymes with treatment. Because syphilitic hepatitis can present without any typical signs of accompanying syphilis, syphilis should be considered as a possible cause in acute hepatitis patients.
