High Resolution Melting Analysis을 이용한 고추 유전자원의 Cucumber mosaic virus 저항성 평가

In this study, total number of 1941 Capsicum accessions conserved at RDA Genebank was evaluated for their response to Cucumber mosaic virus (CMV). These accessions were composed with 9 species originated from 89 countries, included 839 Capsicum annuum, 277 C. baccatum, 395 C. chinense, 343 C. frutescens, 49 C. pubescens, and other 38 wild pepper species (C. chacoense, C. galapagoense, etc.). Resistant to CMV was screened with the 240H02SP6 SNP marker related to the Cmr1 (Cucumber mosaic resistance 1). Eighty nine accessions of pepper germplasm were resistant to CMV based on the marker. One hundred sixty two accessions showed heterozygosity. One thousand two hundred seventy accessions were susceptible to CMV. Four hundred twenty accessions did not show distinction by 240H02SP6 marker. These 89 resistant pepper germplasm can be used in a pepper breeding program against CMV.

This study was conducted to clarify the characteristics of seed germination and to promote the germinationrate in Pollia japonicaThunb. that was pointed to protect by the Office of Forestry in 1997. It was better the germinationPollia japonica showed thehighest germination rate at 20℃ any other treated temperature. To increase the germination rate of Pollia japonica, itwas conducted priming treatments and storage treatments at 4℃. Priming treatments using 1% NaOH, 1% KOH for30min were effective compared to control, but soaking of 100mg/L GA3 for 24 hours, 1% NaOCl for 30min were not90-day treatment, which improved 55.3% than dry storage in 90-day.

본 실험은 희귀 자생식물인 백서향의 삽목 증식 특성을 구명하기 위해 수행되었다. 백서향은 남부지방 해변의 산기슭에 나는 상록관목으로 이른 봄에 향기가 아주 뛰어난 식물이다. 자웅이주식물로 국내에는 대부분 수나무가 많아 채종이 어려워 종자번식이 힘든 상황이므로 삽목 방법을 통해 증식을 시도하였다. 백서향의 삽목에 적합한 토양을 찾기 위해 펄라이트, 버미큘라이트, 펄라이트와 버미큘라이트 혼용(1 : 1), 모래로 4처리를 수행한 결과, 발근율은 각각 86.7%, 95%, 75%, 95%로 나타났다. 온도를 달리하여 3처리로 삽목을 하였더니 발근율과 뿌리길이의 생육을 살펴볼 때 27℃에서 생육이 우수하였다. 시기별로 6월 19일, 7월 19일, 8월 19일에 삽목한 결과 생존율, 발근율은 통계적으로 오차 범위 내에서 공통적으로 우수하였고, 그 중 7월 삽목의 뿌리생장에 가장 우수하였다. 백서향의 삽목증식은 펄라이트와 버미큘라이트를 혼용하여 7월에 삽수를 채취하고 생육기간 중 온도를 27℃로 관리하는 것이 우수한 것으로 나타났다. This study was conducted to find out the propagation method of a rare variety Daphne kiusiana Miq. in Korea through softwood cuttings. Daphne kiusiana Miq are the evergreen broad leaf tree which have lived in the southern part of Korea. The flowering time of Daphne kiusiana Miq is early spring and the flower has an attractive fragrance. Generally, plants are dioecious and only the male plants are found in Korea. Propagation by seed is very difficult in this tree. Softwood cuttings were taken to establish an efficient propagation method in Daphne kiusiana Miq Cuttings were grown in perlite:vermiculite (1 : 1), perlite, vermiculite, sand rooting media under shaded greenhouses during summer in 2007. The rooting rates of cuttings were 86.7%, 95%, 75% and 95% in perlite:vermiculite (1 : 1), perlite, vermiculite and sand media, respectively. Softwood cuttings taken on July and rooting at 27℃ were significantly improved the growth and rooting rates. Softwood cutting can be an effective means of propagation in Daphne kiusiana Miq.
