        Origin-related study of genetic diversity and heteroplasmy of Mongolian sheep (Ovis aries) using mitochondrial DNA

        Food and agricultural production sector, especially livestock production is vital for Mongolia’s economic and social development. Domestic sheep play key roles for Mongolians, providing food (meat, milk) and raw materials (wool, sheepskin), but genetic diversity, origin of sheep populations in Mongolia have not been well studied. Studies of population genetic diversity is important research field in conservation and restoration of animal breeds and genetic resources. Therefore, this study aimed to investigate genetic characteristics and estimate origin through the analysis of mitochondrial DNA control region D-loop and Cytochrome b of Mongolian indigenous sheep (Mongolian native, Orkhon and Altanbulag) and one Europe sheep (Suffolk). As a result of there were found, 220 SNPs (Single nucleotide polymorphism) in the D-loop region, 28 SNPs in the Cytochrome B region, furthermore, 77 Haplotypes. The nucleotide diversity was only found in D-loop region (n = 0.0184). Phylogenetic analysis showed that 3 (A, B, and C) of 5 haplogroups of sheep have been identified in our research. Haplogroup C was only found in Mongolian indigenous sheep. Haplogroup D and E were not observed. As a result of haplogroups, haplogroup A was dominant (n = 46 of 94 sheeps), followed by haplogroup B (n = 36) and haplogroup C (n = 12). Sequence analysis showed that T deletion, insertion and heteroplasmy in D-loop region occurred at a high rate in Mongolian indigenous sheep population (T insertion = 47, T deletion = 83). The heteroplasmy, which has never been found in Mongolian sheep, has been newly discovered in this study. As a result, the Mongolian sheep varieties, which mainly derived from Asia, were in hybridization with European sheep varieties.

        관행과 유기농 고추 재배지의 토양미생물 군집 비교

        Agricultural practices are known to have a crucial influence not only on soil physico-chemical properties but also on microbial communities. To investigate the effect of farming practices on soil microbial communities, a total of 10 soil samples were collected, including five conventional and five organic farming soils cultivated with peppers in plastic greenhouse. We conducted barcorded-pyrosequencing of V1-V3 regions of 16S rRNA genes to examine soil microbial communities of two different farming practices. Taxonomic classification of the microbial communities at the phylum level indicated that a total of 22 bacterial phyla were present across all samples. Among them, seven abundant phyla (>3%) including Proteobacteria, Actinobacteria, Firmicutes, Acidobacteria, Bacteroidetes, Chloroflexi, and Gemmatimonadetes were found, and Proteobacteria (33.0±5.7%), Actinobacteria (19.9±9.7%), and Firmicutes (13.6±5.0%) comprised more than 66% of the relative abundance of the microbial communities. Organic farming soils showed higher relative abundances of Proteobacteria and Firmicutes, while Actinobacteria and Chloroflexi were more abundant in conventional farming soils. Notably, the genera Bacillus (higher in organic farming soils) and Streptomyces (higher in conventional farming soils) exhibited significant variation in relative abundance between organic and conventional farming soils. Finally, correlation analysis identified significant relationships (p<0.05) between soil chemical properties, in particular, pH and organic matter content and microbial communities. Taken together, this study demonstrated that the changes of soil physico-chemical properties by agricultural farming practices effected significantly (p<0.05) on soil microbial communities.

        부모의 과보호적 양육태도와 관계중독과의 관계에서 내현적 자기애의 매개효과

        본 연구는 부모의 과보호적 양육태도와 관계중독과의 관계에서 내현적자기애의 매개효과를 살펴보았다. 이를 위해 대학에 재학 중인 500명에게 부모-자녀 결합 척도(Parental Bonding Instrument: PBI), 내현적 자기애 척도(Covert Narcissism Scale: CNS), 관계중독 척도를 실시하였고 한부모자녀, 이성관계 경험이 없는 대상을 제외한 384명의 자료를 분석하였다. 연구가설을 검증하기 위해 SPSS를 활용하여 기초통계분석을 실시하였으며 AMOS를 활용하여 구조방정식모형을 검증하고 분석을 실시하였다. 본 연구의 결과는 다음과 같다. 첫째, 부모의 과보호적 양육태도는 관계중독과 내현적 자기애에 정적인 상관이 있으며 내현적 자기애도 관계중독과 정적인 상관이 있는 것으로 나타났다. 둘째, 부모의 과보호적 양육태도와 관계중독과의 관계에서 내현적 자기애가 부분매개효과를 가지를 것으로 나타났다. 마지막으로 본 연구가 갖는 의의와 제한점 및 후속 연구에 대한 제언을 논의하였다.

        항산화제 만니톨의 히알루론산 가교체의 유변학적 성질 및 기능에 미치는 영향

        Purpose: This study aimed to evaluate the effect of mixing mannitol into crosslinked hyaluronic acid gels on their rheological properties. Methods: Cross-linked hyaluronic acid gels were prepared by mixing in mannitol to our own manufacturing process, and then the changes in their rheological properties due to chemical and thermal oxidative stress were investigated. The rheological factors evaluated were the storage modulus (G'), loss modulus (G"), complex viscosity (G * ) and phase angle (P/A). Results: Mixing mannitol into the cross-linked hyaluronic acid gels caused their elasticity (G') and viscosity (G") to resist both frequency increase and oxidative stress caused by chemical and thermal stress. Adding mannitol also resulted in improved urine secretion in an animal model, and we therefore expect that it will reduce edema due to osmotic pressure. Conclusion: Mannitol’s antioxidant effect contributed to the improving the cross-linked hyaluronic acid gels’ rheological properties. 목적: 본 본 연구는 라디칼 소거제로인 만니톨 혼합에 의한 가교 히알루론산 겔의 유변학적 물성의 개선효과를 평가하기 위함이다. 방법: 가교겔의 제조공정에 따라서 만니톨을 혼합하여 가교겔을 제조하였으며, 그 유변학적 성질을 화학적 및 가열 산화적 스트레 스를 가하여 탄성(G'), 점성(G"), 복소점도(G * ) 및 위상각(P/A)을 측정하여 관찰하였다. 결과: 만니톨 혼합 가교겔은 주파수의 증가및 화학적 또는 가열 산화적 스트레스에 대한 저항력을 나타내었다. 또한 동물실험에서 만니톨은 첨가농도에서 뇨 배출의 증진을 나타내었다. 이러한 결과는 삼투압에 의한 뇨 배출로 인한 부종의 감소를 기대할 수 있을 것으로 평가된다. 결론: 만니톨의 항산화 기능은 히알루론산 가교 겔의 유변학적 물성의 개선을 나타내었다.

        반응표면분석법 이용한 더덕 추출조건의 최적화

        본 연구에서는 더덕 열수 추출물의 이화학적 특성과 더덕의 주요성분의 함량 등을 측정하여 반응표면분석법으로 더덕의 최적 추출조건을 설정하였다. 실험계획은 중심합성 계획법으로 실시하여 3가지 독립변수로서 용매비(10~30 mL/g; X1), 추출온도(70~90°C; X2) 및 추출시간(1~5시간; X3)을 설정하고, 종속변수로는 추출물의 추출수율, 총당, 환원당, 조사포닌 및 lobetyolin 함량 등을 측정하여 추출조건을 최적화하였다. 그 결과 추출수율은 35.01~71.53%였고 R2값은 0.9574로 높은 신뢰도를 보였다. 추출물의 총당과 환원당 함량은 각각 3.19~6.04 mg/mL와 1.58~3.11 mg/mL의 범위였고 1% 이내의 유의성을 보였다. 더덕 추출물의 사포닌 함량은 0.09~0.17%였으며 R2값은 0.9373으로 1% 이내의 유의성을 보였다. 추출물의 lobetyolin 함량은 16.72~ 32.82 μg/mL였고 R2값은 0.9791로 1% 이내의 유의성을 보였다. 조사포닌 함량과 lobetyolin 함량의 반응표면을 superimposing 하여 최적조건을 설정한 결과 용매비 12 mL/g, 추출온도 75°C 및 추출시간 3시간으로 나타났다. The deodeok extraction conditions were optimized by a response surface methodology (RSM). The physicochemical properties and major components of the deodeok extracts were analyzed. A central composite design was applied to examine the effects of three independent variables: solvent to sample ratio (10∼30 mL/g; X1), extraction temperature (70∼90°C; X2), and extraction time (1∼5 hour; X3). The extraction conditions were optimized with the characteristics of the extracts as a dependent variable. As a result, the extraction yields were 35.01∼71.53% and the coefficient (R2) of the derived equation from the response surface regression for extraction yield was 0.9574. The total sugar and reducing sugar contents in all extracts were 3.19∼6.04 mg/mL and 1.58∼3.11 mg/mL, respectively, which were within the 1% significance level. The crude saponin contents in all extraction samples were 0.09∼0.17% with a R2 value of 0.9373 and were within the 1% significance level. The lobetyolin content as a major component in all extraction samples was 16.72∼32.82 μg/mL with a R2 value of 0.9791 that was within the 1% significance level. The optimal conditions based on the superimposing of the four-dimensional RSM on the crude saponin and lobetyolin contents under various conditions were established as follows: solvent to sample ratio, 12 mL/g; extraction temperature, 75°C; and extraction time, 3 hours.

        생체신호 측정기와 스마트 디바이스를 활용한 심전도 데이터 기반의 원격진료 시스템

        In this paper, we propose a telemedicine system based ECG data using a bio-signal meter and a smart device for treating faraway patients. This system is composed of a patch-shaped portable bio-signal meter, patient's smart device application, and doctor's PC software. Using these components, doctors and patients can do telemedicine. First, a patient measures his own ECG signal with a bio-signal meter and send the data to a doctor using a smart device application. Then, the doctor checks the ECG data, and make and send a diagnosis chart to web server. Likewise, doctors and patients can be offered a medical environment without time and space restraints. Applying this system to real medical system can improve the problem of low accessibility and efficiency and also can reduce medical expenses.
