Streptococcus mutans의 우식 활성에 미치는 Propolis의 효과

Propolis, a natural resinous product collected by honey bees, is known to exhibit various biological activities including anti-microbial, anti-inflammatory, and anti-tumor activities. The present study was designed to investigate the effects of ethanol extracted propolis(EEP) on the growth, glucosyltransferase(GTase) activity, and adherence to the saliva-coated hydroxyapatite(S-HA) beads of Streptococcus mutans JC-2(serotype c), S.mutans LMM-7(serotype e), and S. mutans OMZ-175(serotype f) in vitro. We found that the EEP had minimal inhibitory concentrations of 500-1000㎍/㎖ against S.mutans strains when these strains were cultured for 48 hours, and inhibited both water-insoluble and water-soluble glucan synthesizing GTase activities of S. mutans strains to the 93-100% at the concentration of 1600㎍/㎖, and also we found that the EEP inhibited the adherence of S. mutans strains to S-HA beads to the 76-96% at the concentrations of 200㎍/㎖. These results indicate that EEP has anti-cariogenic activity in vitro.

培養鷄胚筋細胸 分化에 미치는 促進物質의 영향을 알아보고자 2가양이온 (Ca2+, Mg2+) 胸胚抽出物 (EE), polyethylene glycol (PEG), 再生中인 도마뱀 (Lacertilia)꼬리 추출물 (BE)을 처리하여 細胸融合指數를 비교 조사하였다. 정상배양시의 細胸融合指數는 배양시작 70 시간에 최대에 달했으며 더이상 증가하지 않았고, Ca2+와 Mg2+ 처리할 경우에는 농도가 증가할수록 細胸融合指數는 저하되고, 근세포의 굵기도 정상세포보다 가늘고 핵이 모여있었다. Polyethylene Glycol (PEG)와 Blastemata Extract (BE) 처리군에서는 대조구와 큰 차이가 없는 것으로 나타났다. 鷄胚抽出物 (EE) 을 筋細胞分化에 필수적이나 과량으로 添加시에는 細胞融合은 억제되고 세포수는 증가되는것으로 나타났다. To study the effects of promoting factors on the differentiation of muscle cells, chick embryonic myoblasts were cultured with divalent cations (Ca2+, Mg2+) Chick embryo extract (EE), Polyethylene Glycol (PEG) and Blastema extract of Lacertilia (BE). Fusion index and Growth ratio were measured at various culture time. The fusion index of normal myoblast culture was reached maximum at 70 hours and the divalent cations (Ca2+, Mg2+) treated groups were decreased at higher concentration. The cellular formation of myoblasts treated with divalent cations was thiner and more aggregated between nuclei than that of normal myoblasts calture was. When polyethylene glycol (PEG) and Blastema extract of Lacertilia (BE) were treated to the myoblasts, tha fusion indices were similar to the control. Embryo Extract (EE) was seemed to be necessary for the fusion of chick embryonic myoblasts in culture, however, when excess of EE was added the fusion index was decreased and the number of cells was increased.
A New Selective Medium for Detecting Acidovorax avenae subsp. avenae in Rice Seeds

A selective agar medium was developed and tested for the isolation of Acidovorax avenae subsp. avenae, the causal bacterial pathogen of bacterial brown stripe, from rice seeds. The new selective agar medium, designated sorbitol pyroglutamic acid agar (SPA) medium, contained 0.5 g of $K_2$HPO$_4$, 3.0 g of Na$_2$HPO$_4$, 2.0 g of D-sorbitol, 0.2 g of L-pyroglutamic acid, 10.0 $m\ell$ of tween 80, 40.0 mg of victoria blue B, 15.0 g of agar, 150.0 mg of ampicillin and 25.0 mg of vancomycin per litter. Colonies of A. avenae subsp. avenae on SPA medium were smooth, round, convex, shiny, blue and 1.5-2.0 mm in diameter 4 days after incubation at 28$^{\circ}C$. Blue colored colony having dark blue zone was typical type of A. avenae subsp. avenae colonies on the medium. Mean recovery of 8 isolates of A. avenae subsp. avenae on the selective SPA medium was 95.8% in comparison to that on KB medium. The saprophytic bacteria were reduced to 97.9% on SPA medium compared to those on KB medium. Most of other rice seedborne bacteria as well as reported pathogenic bacteria were failed to grow on SPA medium. This medium was highly selective for recovering A. avenae subsp. avenae from rice seed samples, and it could be used to enhance the recovery of this bacterium from rice seed samples, which may be contaminated with large numbers of competing microorganisms.
Research Notes : Occurrence of Clubroot on Shepherd`s-purse Caused by Plasmodiophora brassicae

Clubroot symptoms were frequently observed on roots of shepherd`s-purse (Capsella bursa-pastoris) grown in a field in Nonsan, Chungnam province, Korea in March, 2009. Many resting spores were found in the cells of the root gall tissues collected from the field. The clubroot pathogen was identified as Plasmodiophora brassicae based on its morphological and pathological characteristics. This is the first report that P. brassicae causes clubroot of shepherd`s-purse in Korea.
