        Oral Ingestion of AP Collagen Peptide Leads to Systemic Absorption of Gly-Pro-Hyp, Alleviating H2O2-Induced Dermal Fibroblast Aging

        Collagen-derived dipeptides and tripeptides have various physiological activities. In this study, we compared the plasma kinetics of free Hyp, peptide-derived Hyp, Pro-Hyp, cyclo(Pro-Hyp), Hyp-Gly, Gly-Pro-Hyp, and Gly-Pro-Ala after ingestion of four different collagen samples: AP collagen peptide (APCP), general collagen peptide, collagen, and APCP and γ-aminobutyric acid (GABA) combination. Each peptide was measured by high-performance liquid chromatography and triple quadrupole mass spectrometer. We found that, among all the peptides that were analyzed, only Gly-Pro-Hyp was significantly increased after ingestion of APCP compared with that of general collagen peptides and collagen. In addition, ingestion of the APCP and GABA combination improved the absorption efficiency of Gly-Pro-Ala. Finally, we reveal that Gly-Pro-Hyp was effective for preventing H2O2-induced reduction in extracellular matrix (ECM)-related genes, COL1A, elastin, and fibronectin, in dermal fibroblasts. Taken together, APCP significantly enhances the absorption of Gly-Pro-Hyp, which might act as an ECM-associated signaling factor in dermal fibroblasts, and the APCP and GABA combination promotes Gly-Pro-Ala absorption.         UVB-dependent inhibition of lipin-1 protects against proinflammatory responses in human keratinocytes

        Lipin-1 is an Mg2+-dependent phosphatidate phosphatase (PAP1) that catalyzes a critical step in the synthesis of glycerophospholipids and is also a cotranscriptional regulator. The role of lipin-1 in the regulation of inflammatory responses has been extensively studied in various cell types but not in skin cells. In the present study, the function of lipin-1 in UVB-induced proinflammatory responses was assessed in normal human epidermal keratinocytes (NHEKs). UVB radiation downregulated lipin-1 expression. Lipin-1 inhibition was mediated by UVB-dependent sterol-response element binding protein-1 (SREBP-1) inhibition. The UVB-dependent inhibition of lipin-1 and SREBP-1 was mediated by AMPK activation. UVB-induced activation of JNK was dependent on AMPK activation and mediated lipin-1 inhibition. Prevention of UVB-mediated lipin-1 repression by introducing a lipin-1 expression vector stimulated IL-6 and IL-8 production, suggesting that lipin-1 inhibition attenuates UVB-induced IL-6 and IL-8 production. The downregulation of lipin-1 ameliorated UVB-induced NF-ĸB phosphorylation, which might be attributed to the suppression of UVB-induced accumulation of free fatty acids (FFAs). Pharmacological inhibition of PAP1 with propranolol suppressed UVB-induced production of IL-6 and IL-8 in NHEKs and reconstituted human skin models. Taken together, lipin-1 is downregulated by exposure to UVB radiation, which confers protection against UVB-induced proinflammatory responses; therefore, the inhibition of lipin-1 is a potential strategy for photoaging.

        청소년의 지적능력과 인기에 대한 암묵적 신념, 학업적 및 사회적 성취목표지향성, 수업참여 및 또래관계 질의 구조적 관계

        본 연구는 청소년의 지적능력과 인기에 대한 암묵적 신념, 학업적 및 사회적 성취목표지향성, 수업참여 및 또래관계 질의 구조적 관계를 살펴보는데 목적이 있었다. 중학생 576명(남 353명)을 대상으로 연구한 결과, 첫째, 청소년의 지적능력에 대한 증진신념은 학업적 성취목표를 모두 정적으로 예측할 뿐만 아니라 사회적 숙달목표 및 수행접근목표도 정적으로 예측하였다. 둘째, 인기에 대한 암묵적 신념은 또래관계 질에 직접적 효과가 있었으나 사회적 성취목표를 유의하게 예측하지 못했다. 셋째, 지적능력에 대한 증진신념과 고정신념은 학업적 숙달목표 뿐만 아니라 사회적 숙달목표를 매개하여 수업참여 및 또래관계의 질을 예측하였으며 인기에 대한 증진신념과 고정신념은 각각 학업적 숙달목표를 매개하여 수업참여를 예측하였다. 넷째, 학업적 및 사회적 숙달목표는 해당 영역의 적응을 강하게 예측하였으며 학업적 숙달목표는 사회적 적응에도 유의한 효과가 있었다. 본 연구는 학업적 영역의 변인들과 사회적 영역의 변인들을 하나의 구조에 놓고 영역내 및 영역간 상호 관계를 탐구하고 학업적 및 사회적 숙달목표의 영역을 교차한 효과를 확인한 점에서 의의가 있다. The purpose of this study was to investigate the structural relationship among implicit self-theories of intelligence and popularity, academic and social achievement goal orientation, academic engagement, and quality of peer relationships. In particular, we tried to examine whether implicit theories directly or indirectly influence school adaptation through achievement goal orientation across the academic and social domains. The participants were 576 middle school students (353 males) recruited from five middle schools. The results were as follows. First, implicit self-theories of intelligence had significant effects not only on academic achievement goals, but also on social achievement goals. The incremental belief of intelligence had positive effects on academic mastery, academic performance-approach, social mastery, and social performance-approach goals. Second, the implicit self-theory of popularity directly affected quality of peer relationships, but did not predict social achievement goals. Third, incremental beliefs and entity beliefs of intelligence predicted academic engagement and quality of peer relationships by mediating not only academic mastery goals but also social mastery goals. Incremental beliefs and entity beliefs of popularity predicted academic engagement by mediating academic mastery goals. Fourth, mastery goals in the academic and social domain strongly predicted adaptation in the corresponding domain, and academic mastery goals had a significant effect on social adjustment as well. This study’s findings explore the interrelationships between implicit theories, achievement goals, and adjustment within academic and social domains, as well as across these domains.

        ATG5 expression induced by MDMA (ecstasy), interferes with neuronal differentiation of neuroblastoma cells

        <P>The amphetamine derivative 3, 4-methylenedioxymethamphetamine (MDMA) has become a popular recreational drug, and has also been shown to cause serotonergic neurotoxicity. This report shows that MDMA impairs brain development in a whole mouse embryo culture. The results of quantitative real-time PCR analysis showed that autophagy-related protein 5 (Atg5) expression is elevated in mouse embryo and neuroblastoma cells after MDMA treatment. This elevated Atg5 expression interferes with the neuronal differentiation of neuroblastoma cells such as SH-SY5Y and PC12 cells. Thus, our results suggest that the use of MDMA during pregnancy may impair neuronal development via an induction of Atg5 expression.</P>

      • ASMR MARKETING IN FASHION BRANDS USING EMOTIONAL COMMITMENT AND NOSTALGIA

        Recently, as the development of media contents accelerated, auditory-based contents, especially ASMR, has been taking center stage in its field than visual-based contents. They are meaningful in a way that they are alternative contents that suggest a new space and possibility through aural stimulation, getting out from the boundary of excessive visual stimulation of existing media. According to Jang, Park, & Lyou (2016), ASMR is originally the abbreviation of a medical term, ‘Autonomous Sensory Meridian Response’. However in 2010, Jennifer Allen for the first time presented the definition that ‘ASMR is a specific sound that gives pleasure', and recently it is used as a coined word among young people rather than medical terminology (Jang, Park,& Lyou, 2016). Consumers listen to ASMR for stress relief, psychological stabilization, and relaxation through repeated, constant sound triggers (Zhang, 2015). The triggers used in ASMR marketing draw a stronger commitment within the consumers, by accompanying visual stimuli consistent with auditory stimuli (Sim, 2014). As reported by Hong (2017), commitment is a state of sensorial arousal when a person is totally lost in something and among all affective commitment relieves stress and enhances enjoyment; therefore raises quality of life and helps to live a rich life (Hong, 2017). This shows that there is a close relationship between satisfaction and enjoyment about experiences of ASMR content that leads to commitment, and such phenomenon creates a state of immersion that collapses the distinction between consumers’ past, present and future. Throughout the process, consumers meet the chance to recall the familiar sound that they have experienced in the past, and therefore remind of nostalgia on it (Zhang, 2015). Belk (1990), within the view of marketing, referred to the conceptual definition of nostalgia as a kind of longing atmosphere promoted by sensory stimuli such as scene, smell, and music. Zhang (2015) says that one of the important factors that cause nostalgia is sound, and one calls of the past through a specific sound which is an external stimulus – ASMR is included in it. Consumers can acquire prior knowledge of the brand while enjoying ASMR contents, and they can even have indirect experiences without having direct experiences of actually purchasing or wearing products. This helps to inspire the expertise of the product, and also plays a role in shaping positive consumption emotion such as pleasure and sensibility (Yang, 2016). Fashion products, especially, are more sensible than general consumer goods and due to excessive diversity of products, rational and deliberate purchasing rarely happen. Thus, consumers can build positive brand equity through ASMR marketing by learning specialized knowledge about products that could not be obtained from other marketing. In this study, we propose the new compound word ‘ASMR marketing,’ which combines marketing with the definition of ASMR of the coined term. Electronic word of mouth(EWOM) by ASMR marketing is a process in which the consumers communicate about product information and usage in the online community without the intervention of the seller. Through the characteristics of electronic word of mouth and ASMR, it is easy to infiltrate into daily life of the consumers and easily imprint the individuality of the brand to them (Wang, 2015). Although ASMR appears in ads of HeatTech and AIRism of UNIQLO, a fashion brand, ASMR triggers are not used as a background music and narration. In fashion industry, the use of sounds from the production process of fashion products, or the sounds that occur when wearing them, highlights professionalism and stimulates nostalgia by attracting consumers' emotional commitment. In addition, when ASMR marketing is applied to a luxury brand rather than SPA brand, expertise can be emphasized more effectively. ASMR has been spreading like a fashion in recent years, and marketing of ASMR in fashion brands has not yet been researched actively. The purposes of this study are: first, to analyze significant meaning and value of ASMR in auditory sense field in modern days; second, to identify the characteristics of ASMR marketing through emotional commitment and nostalgia; third, to investigate the effect of ASMR marketing on consumers. Although ASMR, therefore, is used only as a mitigation tool in medical field, this study combines media and marketing with ASMR, suggests it as a tool to enhance the profitability and brand equity of fashion brand. This study would help shape the trend of new culture by collaborating ASMR marketing with fashion brand. Furthermore, a cornerstone of related research can be established by presenting a causal relationship that emotional commitment affects nostalgia and brand equity. This study will be able to offer fashion companies diverse ways to establish ASMR marketing strategy.

        글로벌 금융위기 기간 금융제약과 기업의 성장에 관한 실증분석

        본 연구는 글로벌 금융위기 기간 동안 기업의 금융제약이 기업의 성장에 미치는 영향을 기업의 수출활동유무, 소유주체 등의 특징을 고려하여 비금융위기 기간과 비교 분석했다. 상장기업뿐 아니라 비상장기업을 포괄적으로 포함하는 통계청의 2006-2013년 기업활동조사 불균형 패널데이터를 이용해, 기업의 성장에 대한 현금흐름비율의 민감도를 기업진단별로 금융위기와 비금융위기 기간 각각 System GMM을 이용하여 추정하였다, 실증분석 결과, 기업의 금융제약이 성장에 미치는 영향이 통계적으로 유의하게 나타났고, 이러한 기업의 성장에 미치는 금융제약은 금융위기와 비금융위기 기간 모두 나타났다. 기업의 특성을 고려하여 기업집단별로 분석한 결과, 외국기업은 금융위기와 비금융위기 기간 모두 금융제약으로 인해 성장에 제한을 받지 않는 것으로 나타난 반면에, 국내기업은 금융위기와 비금융위기 기간 모두 내부금융이 기업의 성장에 영향을 미치는 것으로 나타났다. 외국기업은 외국 모회사로부터 다양한 외부지원 접근성 용이로 인한 비교우의로 인해 내부금융이 기업의 성장에 미치는 영향이 통계적으로 유의하지 않게 나타난 것으로 판단된다. 또한 수출활동 유무에 따라 기업집단을 구분한 분석결과, 내수기업이 직면한 금융제약이 수출기업보다 소폭 크게 나타났다. 또한, 내수기업은 금융위기기간에 기업의 금융제약이 기업의 성장에 미친 영향이 비금융위기 기간에 비교해 크게 증가한것으로 나타났다. 수출기업은 내부금융이 성장에 미친 영향이 금융위기 기간에는 통계적으로 유의하지 않게 나타난 반면에, 비금융위기 기간에는 통계적으로 유의하게 나타났다. 금융위기기간, 수출기업의 외부금융 및 정책자금에 대한 접근성 확대로 인한 결과로 판단된다. 실증분석결과 기업의 성장이 금융제약으로 제한을 받는 것으로 나타났고, 이러한 내부금융이 성장에 미치는 영향은 기업집단별 특징에 따라 상이하게 나타난 만큼, 차별화된 기업 금융 지원 정책으로 기업의 성장의 제한하는 금융제약을 완화시킬 필요가 있다. This paper analyzes the effect of financial constraints on firm’s growth during global financial crisis vs non-crisis periods, taking into account firm s characteristics of export participation activity as well as ownership structure. This paper investigates the effect of firm s cash-flow ratio, the measurement of financial constraints, on firm asset growth rate, using a manufacturing firm-level unbalanced panel dataset of South Korea for 2006–2013. The estimation results from system GMM show that the effects of financial constraints on firm’s growth vary according to firm’s characteristics. We do not find that foreign MNCs subsidiary’s growth is affected by their cash flow ratio during both crisis and non-crisis periods while we find that domestic firm’s growth is affected by their cash flow ratio during both crisis and non-crisis periods. This empirical result can be explained by the comparative advantage of foreign MNCs subsidiaries in terms of easier access to the external finance through their parents. In addition, non-exporting firms experience severer financial constraints on their growth during crisis than non-crisis period. However, the effect of financial constraints on firm’s growth for exporting firms is not statistically significant during the crisis period while it is significant during the non-crisis period.

        Coxsackie and Adenovirus Receptor Binding Ablation Reduces Adenovirus Liver Tropism and Toxicity

        <P>Human adenovirus-based vectors have emerged as a new promising vehicle for in vivo gene transfer-mediated therapy. However, the full potential of this methodology has not been fully realized because of the nonspecific tissue distribution of adenoviral vectors. Adenovirus infection is initiated by forming a complex between the fiber protein and a ubiquitously expressed host cell membrane protein, coxsackie B virus and adenovirus receptor (CAR). Therefore, ablating the adenovirus vector's ability to bind to the CAR is the first step in redirecting adenoviral tropism. To ablate CAR binding, we mutated the Bbeta sheet of the fiber knob, generating CAR-binding ablated replication-incompetent (dl-K420A-Z) and replication-competent (YKLK420A) adenoviral vectors. The in vitro transduction efficiency of dl-K420A-Z was significantly reduced in comparison to dl-LacZ carrying the wild-type fiber in CAR-positive cells but not in CAR-negative cells, suggesting that the mutation introduced in the Bbeta sheet of the fiber knob could disable the CAR-dependent transduction pathway. The in vivo transduction was also dramatically reduced in the liver and other organs for mice treated with dl-K420A-Z, compared with a cognate control vector, dl-LacZ. Concomitant with this attenuated gene transfer efficiency in vivo was a substantial reduction in the amount of general toxicity observed in the YKL-K420A-treated mice. Diminished toxicity was surmised from quantitative measurement of serum level of enzymes for liver and kidney function, hematologic chemistries, histopathology, and differences in lethality. Significant decrease in serum transaminases (alanine transferase [ALT] and aspartate transferase [AST]) was observed in mice treated with YKL-K420A. In addition, the lethality was lower in the YKLK420A- treated groups compared to the YKL-1-treated groups at all doses examined. Furthermore, the hepatopathologic analysis revealed that YKL-1 induced focal zonal necrosis and hepatocyte degeneration, while YKL-K420A induced mild spotty necrosis. In summary, this decreased vector tropism of CAR-binding ablated adenoviruses in normal tissues may increase the amount of virus available for infecting tumor cells and thus increase the antitumor efficacy with fewer unwanted side effects.</P>
