갈근(葛根)으로부터 분리된 puerarin의 항당뇨 효과

본 연구에서는 갈근 및 그의 주 이소플라본인 puerarin의 활성을 세포수준에서 분석하였다. 먼저 갈근에 함유된 이소플라본의 양을 분석한 결과, puerarin이 총 이소플라본의 90 % 차지하였다. 다음으로는 puerarin의 항당뇨 활성을 검정한 결과 먼저 탄수화물 및 지방소화효소저해활성에 대해서는 거의 미비한 것으로 나타났으나 인슐린 감수성 및 지방세포의 분화의 유도에 대해서는 농도의존적으로 작용하는 것으로 관찰되었다. 따라서 puerarin은 지방조직내로 포도당의흡수를 촉진함으로서 항당뇨 효능을 발휘하는 것으로 추정된다. In this study we evaluated the anti-diabetic potential of Puerariae Radix and its isoflavone (puerarin) by investigating their inhibitory activities against digestive enzymes, α-amylase, α-glucosidase and lipase and effect on glucose uptake and PPAR γ expression. The activities of carbohydrate digestive enzymes were not inhibited by puerarin. Glucose uptake in differentiated adipocytes was stimulated by puerarin. Furthermore, puerarin enhanced the differentiation of preadipocytes as evaluated by triacylglycerol (TG) accumulation, which is specific for differentiated adipocytes. The effect of puerarin on expression of peroxisome proliferator-activated receptor γ (PPAR-γ) gene, which is associated with obesity and dyslipidemia, was examined by both real-time PCR and reverse transcriptase PCR. The study demonstrated that puerarin increased the expression of PPAR-γ. In conclusion, puerarin showed potential to exert anti-diabetic action by enhancing cellular glucose uptake and thereby TG accumulation in adipocyte tissue.

토란의 기지 shoot를 암배양하여 절간이 신장한 황하경을 얻고 이황하경의 절편을 계대배양하여 기내증식을 시도하였다. 황하경은 NAA 0.2mg/ℓ가 첨가된 배지에서 가장 길게 신장하였다. 황하경은 자구형성형이 모구배대형보다 길게 신장하였다. 호르몬이 첨가된 거의 모든 배지에서 protocorm이 형성되었으나 0.2mg/ℓ NAA + 0.2mg/ℓ BA를 첨가한 배지에서 특히 많이 형성되었고 이 protocorm을 MS기본 배지에 옮겼을 때 8.5%의 재분화율을 보였다. Etiolated stems with elongated internodes were obtained from the in vitro culture of shoots of Colocasia esculenta schout under dark condition and plant differntiation was achived by subculturing the ethiolated stem segments. The etiolated stems elongates better on the media containing 0.2mg/ℓNAA than of those lacking it. Subculture of the etiolated stem segments produced protocorm-like structures in all the media examined. The medium containing 0.2mg/ℓNAA and 0.2mg/ℓBA produced the higest number of protocorm-like structure, which regenerated into plantets at 85% when transferred to phytohormone-free MS medium.

Health Risk Appraisal(HRA) is usually defined as a process by which we expect of individual's chances of death or acquiring specific diseases within a defined period of time. The concept of worksite as an area of health maintenance and promotion is newly developing. Our movement for health promotion of employees has been increasingly known in the worksite. The aim of this study is to develop health risk appraisal tools about health promotion at the worksite for employees. We performed this study by two steps; one step was to develop a HRA questionnaire of worksites in Korea, another was to evaluate the reliability of the questionnaire. For developing HRA questionnaire, we reviewed scientific examples at first, and then weighted the score by delphi. To evaluate the reliability of developed questionnaire, we carried out survey by test-retest method. A total of 131 employees completed HRA questionnaire on two times. The results are as follows. The total score of health risk in unhealthy workers was higher than that of healthy workers. The range of test-retest reliability of responses to the questionnaire was 0.57-0.94. Therefore it seems that this questionnaire was very suitable to assess the health behavior of workers. In conclusion, the developed HRA questionnaire can be used as a tool for evaluating health behavior and for providing health counseling materials.

Background and Objectives: Benign tumors of the nose and paranasal sinues are rare in comparison with malignant growths. Authors aimed to investigate the incidence and characteristics of benign sino-nasal tumors. Materials and Methods: From 1994 through 2004, 25 cases of benign sino-nasal tumors (17 men and 8 women) were diagnosed and cared at the Department of Otorhinolaryngology-Head and Neck Surgery, College of Medicine, Konkuk University. We reviewed medical records of patients to find out clinical features, radiologic findings, operative findings, therapeutic options, and outcomes. Postoperative observation was given for fifteen months on the average. Results: The most common symptom was nasal obstruction, followed by nasal mass, frequent nasal bleeding, headache, and rhinorrhea, and the mass mainly placed in the lateral nasal wall. There were 12 cases (48%) of inverted papilloma, 4 (16%) cases of squamous papilloma, 4 (16%) cases of hemangioma, 1 (4%) case of pleomorphic adenoma, 1 (4%) case of leiomyoma, 1 (4%) case of lipoma. All the tumors were surgically removed. For the treatment of inverted papilloma, medial maxillectomy had been performed before 1997(2 cases); however, most cases have been managed with endonasal endoscopic technique since 1997. Recurrence was observed in two patients with inverted papilloma and one patients with squamous papilloma. Conclusion: Inverted papilloma is the most common benign tumor in the nose and paranasal sinuses, followed by squamous papilloma and hemangioma. The optimal management of benign tumors is complete excision of tumor and endonasal endoscopic surgery is an effective treatment for an inverted papilloma.
Genistein Induces Glucose-Regulated Protein 78 in Mammary Tumor Cells

<P>Preliminary studies have shown that genistein modulates the expression of some heat shock proteins in mammary tumor cells. In this study, we investigated the effect of genistein pretreatment on the expression of glucose-regulated protein 78 (GRP78) in both estrogen receptor-positive (MCF-7) and -negative (MDA-MB-231) cells. Genistein increased the expression of GRP78 in a dose- and time-dependent manner and suppressed glucose uptake in both cell lines. However, induction of GRP78 by genistein appears not to be directly associated with inhibition of glucose uptake. Genistein treatment also made MDA-MB-231 cells more sensitive to doxorubicin, probably via increased GRP78 expression, but had no effect or even decreased drug sensitivity in MCF-7 cells. These results suggest that genistein may be exploited as an enhancer of chemotherapeutic agents in certain types of breast cancer.</P>
PPARγ activation by baicalin suppresses NF-κB-mediated inflammation in aged rat kidney.

<P>Baicalin, a herb-derived flavonoid compound, has beneficial activities, including the modulation of oxidative stress and inflammation. Nuclear receptor peroxisome proliferator-activated receptor-γ (PPARγ) is a ligand-activated transcription factor that plays an important role in regulating nuclear factor-κB (NF-κB)-induced age-related inflammation. We investigated the anti-inflammatory action of baicalin, which depends on its ability to activate PPARγ, and subsequently to suppress NF-κB. We examined baicalin-treated kidney tissue from 24-month-old Fischer 344 aged rats (10 or 20 mg/kg/day for 10 days) and baicalin-fed mice (10 mg/kg/day for 3 days) for in vivo investigations, and used endothelial YPEN-1 cells for in vitro studies. In the baicalin-fed aged rats, there was a marked enhancement of both nuclear protein levels and DNA binding activity of PPARγ, and a decreased expression of NF-κB target genes (VCAM-1, IL-1β, and IL-6) compared with non-baicalin-fed aged rats. Furthermore, to confirm the anti-inflammatory action of PPARγ activated by baicalin, we used lipopolysaccharide (LPS)-treated cells and mice. The results showed that baicalin induced PPARγ-selective activation in YPEN-1 cells, and that the effects of baicalin were blocked by the PPARγ receptor antagonist, GW9662. In addition, baicalin treatment prevented RS generation, NF-κB activation and the expression of pro-inflammatory genes, whereas it increased PPARγ expression in LPS-treated cells and mouse kidney. Our data suggest that baicalin-induced PPARγ expression reduced age-related inflammation through blocking pro-inflammatory NF-κB activation. These results indicate that baicalin is a novel PPARγ activator and that this agent may have the potential to minimize inflammation.</P>
