<P>Within the endoplasmic reticulum (ER), mannoses and glucoses, donated from dolichol-phosphate-mannose and -glucose, are transferred to N-glycan and GPI-anchor precursors, and serine/threonine residues in many proteins. Glycosyltransferases that mediate these reactions are ER-resident multitransmembrane proteins with common characteristics, forming a superfamily of >10 enzymes. Here, we report an essential component of glycosylphosphatidylinositol-mannosyltransferase I (GPI-MT-I), which transfers the first of the four mannoses in the GPI-anchor precursors. We isolated a Chinese hamster ovary (CHO) cell mutant defective in GPI-MT-I but not its catalytic component PIG-M. The mutant gene, termed phosphatidylinositolglycan-class X (PIG-X), encoded a 252-amino acid ER-resident type I transmembrane protein with a large lumenal domain. PIG-X and PIG-M formed a complex, and PIG-M expression was <10% in the absence of PIG-X, indicating that PIG-X stabilizes PIG-M. We found that Saccharomyces cerevisiae Pbn1p/YCL052Cp, which was previously reported to be involved in autoprocessing of proproteinase B, is the functional homologue of PIG-X; Pbn1p is critical for Gpi14p/YJR013Wp function, the yeast homologue of PIG-M. This is the first report of an essential subcomponent of glycosyltransferases using dolichol-phosphate-monosaccharide.</P>

Over the past decade, the Japanese government and Japanese universities have increased student mobility, both inbound and outbound, to accelerate the internationalization of higher education. However, student mobility was halted in early 2020 because of the COVID-19 pandemic, and international students who had planned to engage in a traditional study abroad program could not enter Japan. The current study examined whether the unexpected implementation of online distance classes because of the pandemic affected the learning strategies of graduate students, including international students. In addition, we investigated whether the online courses functioned as an alternative to face-to-face classes. An analysis using structural equation modeling revealed that the period of enrollment, self-regulation, and country of residence were factors that influenced help-seeking behavior. Graduate students who had enrolled before the pandemic and already experienced face-to-face classes were more likely to actively seek help from instructors and classmates in online classes. Furthermore, graduate students who were unable to enter the country but were taking classes online also tended to actively engage in help-seeking from their instructors and classmates. Students’ experiences of the sudden change to distance learning suggest that, to ensure a sustainable teaching and learning environment in various contexts, instructors should use class designs that consider distance learning, particularly designs that enhance students’ help-seeking, even under normal circumstances. In addition, ensuring sufficient online/virtual spaces for communication among teachers and students is important.

In liquid-propellant rockets, POGO instability can occur, in which a fluctuation of propellant supply to the engine, a thrust fluctuation, and a structural vibration are coupled. For the prediction of this instability, it is required to provide dynamic characteristics of the pump represented as the transfer matrix correlating the upstream and downstream pressure and flow rate fluctuations. In the present study, the flow rate fluctuation is evaluated from the fluctuation of pressure difference at the different locations assuming that the fluctuation is caused by the inertia of the flow rate fluctuation. The experiments were performed in some flow conditions, and it was shown that the tendencies of dynamic characteristics are related to excitation frequencies, cavitation numbers and flow rate coefficients.

Synthesis of Supercapacitor Electrode by Thermal Decomposition of Manganese Formate Complex

The supercapacitor is attracting attention as a high-power, long-life energy storage device capable of rapid charging and discharging. However, their low energy density (Wh/cm³) is an issue. In order to solve this problem, it is necessary to form thin films with high specific capacitance by volume (F/cm³), i.e., three-dimensional thin films. In this study, we attempted to synthesize electrodes for supercapacitors with high energy density by coating and sintering an ink containing manganese formate complexed with amine (TETA) on a porous metal substrate (Ni foam). The sintering temperature was varied from 200 °C to 300 °C, and the mechanism of electrode synthesis was analyzed by examining the difference in the electrode shape and the results of electrochemical measurements.

There is probably no kindergarten without an athletic festival Undokai in Japan. Froebel, the father of kindergarten, wrote about Play Festivals that are quite similar to the athletic festivals of today. However, the Japanese athletic festival did not develop from Froebel‘s concept. Rather, they came out of the festival tradition in Japanese naval colleges and universities. These festivals, over time, filtered down to elementary education and then to kindergartens in Japan. Athletic festival is characterized by physical competition and group training and these characteristics philosophically are counter to the principles of kindergarten education, which focuses more on children‘s autonomy and spontaneity. The two reasons given for the common practice of the athletic festival are (1) that it encourages parents to participate and, (2) that both parents and children under-stand the entertaining value of sharing the joy of watching growing children. Although instruction on group behavior or observing rules tends to be teacher-centered, teachers today pay close attention to creating an atmosphere and exchanging ideas through discussion so that children can be actively and willingly involved. The potential problem is that the teachers‘ instructional abilities can divert their expectations away from the viewpoint of child-centeredness. Athletic festivals have become a necessity to kindergartens in Japan yet at the same time they continue to raise a potential problem for encouraging the development of individuality and the sense of individuals contributing and belonging to a group.
