        Effect of Non-animal-Derived Nitrogen Sources on the Production of Hyaluronic Acid by Streptococcus sp. KL0188

        Hyaluronic acid (HA) is a linear high-molecular-weight polysaccharide with useful biomedical applications. Streptococcus zooepidemicus, a typical HA-producing bacterium, requires an animal-derived nitrogen source such as tryptone, peptone or sheep blood as a nutrient. Sixteen non-animal-derived (NAD) nitrogen sources were tested as a replacement for the expensive animal-derived nitrogen sources, which may have safety issues. Among the sixteen tested NAD nitrogen sources, a yeast-derived nitrogen source (YE 0251) showed the highest HA productivity, which was equivalent to the control HA production medium containing tryptone in a 5-L jar and in 3,000-L industrial fermentations. In the 3,000-L fermentation, YE 0251 increased cell mass (dry cell weight) and HA production by 11% and 8%, respectively, compared with the control HA production medium. The fmal specific volumetric productivity (0.41 g/L h) was improved by about 70% after reducing the fermentation time from 20 h to 12 h, compared to the conventional production medium.

        주차원단위 산정 모형 개발에 관한 연구 -광주광역시 공동 주택 아파트를 대상으로-

        The rapid expansion of cities led to the shortage of housing in urban areas. The government compensated for this shortage through large scale residential developments that increased the housing supply. The supply of condominium apartments remains above 83% of the entire housing supply, and the proportion of apartments are at a steady increase, at about 50%. Due to the increase, illegally parked cars resulting from the shortage of parking spaces within the apartment complex have become increasingly problematic as they block the transit of emergency vehicles, and heighten the tension among neighboring residents in obtaining a parking space. Especially, the future residents are considered to plan the parking based on the estimated demand for parking. However, the parking unit method utilized to estimate the parking demand accounts for the exclusive use of space, which is believed to be far from the parking demands in reality. The reason for this discrepancy is that, as the number of households decrease, and area of exclusive space is expanded, the planned parking increases. On the other hand, when the number of households increase, and the area of exclusive space is reduced, the planned parking decreases, thus methods to recalculate the parking units based on estimated parking demand is an urgent concern. To estimate the parking units based on condominium apartments, this study first examined the existing research literature, and appointed the field of investigation to collect the necessary data. In addition, field study data and surveys collected and analyzed, in order to identify the problems underlying parking units, and problems regarding the current traffic impact assessment parking unit calculation method were deduced. Through identifying the influential factors on parking demand estimates, and performing a factorial analysis based on the collected data, the variables were selected in relation to the parking demand estimates, to develop the parking unit estimate model. Finally, through comparing and verifying the existing traffic impact assessment parking unit estimate against the newly developed model using collected data, a far more realistic parking unite estimate was suggested, reflecting the characteristics of the residents. The parking unit estimate model developed in this study is anticipated to serve as the guidelines for future parking lot legislature, as wel as the basis to provide a more realistic estimate of parking demands based on the resident characteristics of an apartment complex.

        Current Status of Ginseng Cultivation and Soil Characteristics of Northeastern Three Provinces in China

        This study was conducted to improve the continuous techniques for international competitiveness of ginseng industry to Korea-China FTA negotiation and conclusion, and provide the basic information for ginseng industry development of Korea. It was carried out the visiting of the northeastern three provinces (Jilin, Liaoling and Heilongjang) in China for 3-year from 2014 to 2016 and observed the farmers' fields of ginseng cultivation with soil environmental status. The types of ginseng cultivation could be observed in small scales of 0.5~3.0 ha, in middle scales of 4.0~10.0 ha and in large scales of 30~700 ha with the kinds of imhasam, Chinese ginseng, Korean ginseng and western ginseng. Also ginseng was cultivated in newly reclaimed land of forest in two types of direct seeding and transplanting of ginseng seedlings. The field beds of ginseng growing were covered with vinyl films in arch design of 100~130 cm height and vinyl was painted in spraying with blue, green and yellow colours for shading. It was investigated in status of the physico-chemical properties of soils. The physical information on the field soils were silt loam, loam and sandy loam in soil textures, and some plain in low slope, some alluvial fan or local valley in forest of land topography. Soil pH ranged within 5.0~5.2, soil EC was $0.93{\sim}3.78dS\;m^{-1}$, organic matter was $37{\sim}35g\;kg^{-1}$, nitrate nitrogen $63{\sim}490mg\;kg^{-1}$, available $P_2O_5$ $55{\sim}163mg\;kg^{-1}$, and in exchangeable cations, K was 0.30~0.98, Ca was 6.5~14.0, Mg was $1.1{\sim}5.3cmol_c\;kg^{-1}$ in ranges. Farmers used the fertilizer for ginseng cultivation in 10~11 t of compost, $200{\sim}400kg\;ha^{-1}$ of complex fertilizer and $750kg\;ha^{-1}$ of oil cakes. The northeastern three provinces of China can use the newly lands with large areas of ginseng cultivation in soil sickness by continuous cropping. and the soil basic fertility is batter than that of Korean in standard guide of ginseng cultivation soil.

        한국인 직무 스트레스 측정도구의 개발 및 표준화

        Background and Purposes: Over the past three decades, numerous studies performed in Korea have reported that job stress is a determinant risk factor for chronic diseases and work disability. Every society has its own culture and occupational climate particular to their organizations, and hence experiences different occupational stress. An occupational stress measurement tool therefore needs to be developed to estimate it objectively. The purpose of this study is to develop and standardize the Korean Occupational Stress Scale (KOSS) which is considered to be unique and specific occupational stressors in Korean employees. Subjects and Methods: Data were obtained from the National Study for Development and Standardization of Occupational Stress (NSDSOS Project: 2002-2004). A total of 12,631 employees from a nationwide sample proportional to the Korean Standard Industrial Classification and the Korean Standard Occupational Classification were administered. The KOSS was developed for 2 years (2002-2004). In the first year, we collected 255 items from the most popular job stress measurement tools such as JCQ, ERI, NIOSH and OSI, and 44 items derived from the a qualitative study (depth interview). Forty-three items of KOSS, in the second year, were retained for use in the final version of the KOSS by using Delphi and factor analysis. Items were scored using conventional 1-2-3-4 Likert scores for the response categories. Results: We developed eight subscales by using factor analysis and validation process: physical environment (3 items), job demand (8 items), insufficient job control (5 items), interpersonal conflict (4 items), job insecurity (6 items), organizational system (7 items), lack of reward (6 items), and occupational climate (4 items). Together they explained 50.0% of total variance. Internal consistency alpha scores were ranged from 0.51 to 0.82. Twenty-four items of the short form of the KOSS (KOSS-SF) were also developed to estimate job stress in the work setting. Because the levels of the subscales of occupational stress were gender dependent, gender-specific standard norms for both the 43-item full version and the 24-item short form using a quartile for the subscales of KOSS were presented. Conclusion: The results of this study suggest that KOSS might be an appropriate measurement scale to estimate occupational stress of Korean employees. Further and more detailed study needs to be conducted to improve the validity of this scale.

        <P>Parkinson's disease (PD) is the second leading neurodegenerative disease, and is known to be induced by environmental factors or genetic mutations. Among the verified genetic mutations of PD, Parkin, isolated from the PARK2 locus, shows an autosomal recessive inheritance pattern and is known to be an E3 ligase. However, the physiological target of Parkin and the molecular mechanism of Parkin-deficiency-induced PD have not been clearly demonstrated until now. It has recently been proposed that inflammation, suggesting as a causal factor for PD, is enhanced by Parkin deficiency. Thus, we examined the relationship between inflammation-related factors and Parkin. Here, we provide the evidence that Parkin suppresses inflammation and cytokine-induced cell death by promoting the proteasomal degradation of TRAF2/6 (TNF-α receptor-associated factor 2/6). Overexpression of Parkin can reduce the half-lives of TRAF2 and TRAF6, whereas si-Parkin can extend them. However, mutant Parkins did not alter the expression of TRAF2/6. Thus, loss of Parkin enhances sensitivity to TNF-α- or IL-1β-induced JNK activation and NF-κB activation. Indeed, si-Parkin-induced apoptosis is suppressed by the knockdown of TRAF6 or TRAF2. We also observed elevated expression levels of TRAF6 and a reduction of IκB in an 1-methyl-4-phenyl-1,2,3,6-tetrahydropyridine-induced PD mouse model. Moreover, elevated expression levels or aggregation of TRAF6 were detected in approximately half of the human PD tissues (7/15 cases) and 2 cases, respectively. In addition, TRAF6 and Parkin expression levels show a reverse relationship in human PD tissues. Our results strongly suggest that the reduction of Parkin or overexpression of TRAF2/6 by chronic inflammation would be the reason for occurrence of PD.</P>

        지상라이다를 이용한 미기록 외래종 갯쥐꼬리풀(Spartina Alterniflora)의 분포특성과 관리방안 연구 -다도해 해상국립공원 진도 남동리 해안을 사례로-

        Spartina alterniflora is native to the Atlantic coast from Newfoundland, the south of Canada and northern Argentina. There it forms a dominant part of brackish coastal saltmarshes. Due to its strong growth, the Spartina alternifolra was introduced to the rider area and coast as a use for ecological engineering. The Spartina alterniflora community can crowd out native species, reduce the biodiversity of saltmarsh and alter the environment. As a result, the invertebrates living in mud flats disappeared due to the encroaching of Spartina alterniflora, and the birds which eat off invertebrates went along as well. The Spartina alterniflora was first introduced in South Korea in 2008 and is known to have flowed in the Dadohaehaesang National Park. Compared to 2008, in 2015 the distribution area of the Spartina alterniflora has increased 550 times. (11.54㎡ in 2008 to 6,394.22㎡ in 2015). The large increase of Spartina alterniflora calls for urgent management and additionary study on the subject. Thus, this study analyzed the geo-spatial information on the Spartina alterniflora such as its distribution area, the height of habitats and expected expansion area, and can act as the preliminary data for management.
