Immunohistochemical study on the expression of calcium binding proteins (calbindin-D28k, calretinin, and parvalbumin) in the cerebral cortex and in the hippocampal region of nNOS knock-out(−/−) mice

        Yu Jin Cho,Jae Chul Lee,Bong Gu Kang1,2,Jaeyeol An,Hyeon Suk Song1,2,Onju Son,Do-Hyun Nam,Choong Ik Cha,Kyeung Min Joo 대한해부학회 2011 Anatomy & Cell Biology Vol.44 No.2

Nitric oxide (NO) modulates the activities of various channels and receptors to participate in the regulation of neuronal intracellular Ca2+ levels. Ca2+ binding protein (CaBP) expression may also be altered by NO. Accordingly, we examined expression changes in calbindin-D28k, calretinin, and parvalbumin in the cerebral cortex and hippocampal region of neuronal NO synthase knockout(−/−) (nNOS−/−) mice using immunohistochemistry. For the first time, we demonstrate that the expression of CaBPs is specifically altered in the cerebral cortex and hippocampal region of nNOS−/− mice and that their expression changed according to neuronal type. As changes in CaBP expression can influence temporal and spatial intracellular Ca2+ levels, it appears that NO may be involved in various functions, such as modulating neuronal Ca2+ homeostasis, regulating synaptic transmission, and neuroprotection, by influencing the expression of CaBPs. Therefore, these results suggest another mechanism by which NO participates in the regulation of neuronal Ca2+ homeostasis. However, the exact mechanisms of this regulation and its functional significance require further investigation.

Kang, Yun Sik,Park, Taehyun,Jang, Segeun,Choi, Mansoo,Yoo, Sung Jong,Cha, Suk Won Elsevier 2017 Journal of industrial and engineering chemistry Vol.47 No.-

Abstract

Membrane electrode assemblies (MEAs) with carbon paper and cloth for bendable polymer electrolyte membrane fuel cell were characterized as it is subject to repetitive bending. The performance of the MEA with carbon paper was decreased significantly while the MEA with carbon cloth remained constant after repetitive bending. Electrochemical impedance spectroscopy revealed ohmic and charge transfer resistances of the MEA with carbon paper were increased by repetitive bending. Such performance degradation is due to physically observed damages in carbon paper and its detachment from the MEA, which was not in the MEA with carbon cloth due to its intrinsic flexibility.

Highlights

The MEAs with carbon paper and carbon cloth were characterized as it is subject to repetitive bending.

The MEA with carbon paper exhibited significant performance degradation after repetitive bending.

The performance degradation in the MEA with carbon paper resulted from damaged components by cracks and fold lines.

The MEA with carbon cloth showed stable performance with good bending durability.

Graphical abstract

[DISPLAY OMISSION]

        강석,김종필,차응남,김경철,조창성,김용민,황정수,정필현 東國大學校醫學硏究所 1994 東國醫學 Vol.2 No.-

There have been several diagnostic methods to evaluate the knee injury patient with guarded accuracy. Magnetic Resonance Imaging(MRI) is recently developed diagnostic tool of highly improved benefits especially in orthopaedic field. MRI not only shows fine osseus and soft tissue structure, but is not invasive with upgraded accuracy. Authors analysed 30 cases of knee injury of which were diagnosed by simple X-ray including stress view, MRI, and physical examination under anesthesia. All the cases were confirmed at final operative intervention from Jan.1994 to Aug.1994 at our hospital. The purpose of this paper is to appreciate the diagnostic value of MRI in knee injured patient compair with simple X-ray including stress view and physical examination. The sensitivity and specificity of MRI were 78%,100% for ligament injury, and 100%,100% for meniscus injury, respectively. We believe that MRI, when combined with physical examination and X-ray finding gives the most accurate non-invasive method of information to delineate pathologic findings in the knee injury patient.

        강명근,박종구,장세진,고상백,차봉석 大韓産業醫學會 1998 대한직업환경의학회지 Vol.10 No.1

This study was conducted to understand health status by general characteristic, and to find out relationship between social support and worker's health status. Health status was measured using SF-36(Medical Outcome Study Short Form 36), a 36 item self administered instrument. The finding of this study were as follow; Mean scores of health status by sex were higher in male. The younger worker reported good health on physical functioning and role limitation-physical than did the older worker, but the older worker reported good health on social functioning and mental health. Mean scores of health status were higher in high income and white worker. When the relationship between social support and health status, social functioning, role limitation-emotion, mental health, vitality, general health were significantly related. Therefore we reviewed the evidence that socio-economic condition are associated with health status in this study, and that the strength of the social support was a important to maintain health.

KSHV vPK inhibits Wnt signaling via preventing interactions between β-catenin and TCF4

Cha, Seho,Kang, Myung-Suk,Seo, Taegun Elsevier 2018 Biochemical and biophysical research communication Vol.497 No.1

Abstract

Viral factors interact with host cellular proteins, leading to dysregulation of signaling pathways. The Wnt pathway is known to participate in embryonic development and oncogenesis under dysregulation conditions. A downstream factor of the Wnt signaling pathway, β-catenin, activates T-cell factor (TCF)-dependent transcription, which contributes to cell proliferation and tumorigenesis. In this study, we demonstrated that viral protein kinase (vPK) encoded by Kaposi's sarcoma-associated herpesvirus inhibits the Wnt signaling pathway without affecting nuclear localization and expression of β-catenin. Coimmunoprecipitation and chromatin immunoprecipitation assays revealed that vPK interacts with β-catenin, reducing the binding affinity on TCF binding regions as well as interactions of β-catenin with TCF4. Overexpression of vPK led to reduced mRNA expression of cyclin D1, a well-known transcriptional product of Wnt signaling, suggesting that vPK effectively regulates the host signaling pathway through direct interactions with cellular proteins.

Highlights

vPK decreases LiCl-mediated activation of TCF-dependent transcription.

vPK does not disrupt relocalization of β-catenin from the cytosol to nucleus.

vPK disrupts interactions of β-catenin with the TCF4-binding region.

vPK inhibits Wnt signaling via interactions with β-catenin.

Angiotensin II stimulates the synthesis of vascular endothelial growth factor through the p38 mitogen activated protein kinase pathway in cultured mouse podocytes.

Kang, Young Sun,Park, Yun Gyu,Kim, Bo Kyung,Han, Sang Youb,Jee, Yi Hwa,Han, Kum Hyun,Lee, Mi Hwa,Song, Hye Kyoung,Cha, Dae Ryong,Kang, Shin Wook,Han, Dae Suk Journal of Endocrinology (Ltd. by Guarantee) 2006 Journal of molecular endocrinology Vol.36 No.2

Angiotensin II (Ang-II) and vascular endothelial growth factor (VEGF) have an important role in the pathogenesis of diabetic nephropathy, but the signaling cascade of VEGF regulation in response to Ang-II in podocytes is largely unknown. In these experiments, we looked at the effect of Ang-II on the production of VEGF, and investigated whether VEGF production depends on the p38 mitogen activated protein kinase (MAPK) pathway in cultured mouse podocytes. Incubation of podocytes with Ang-II induced a rapid increase in VEGF mRNA expression and protein synthesis as well as its transcriptional activity in an Ang-II dose-dependent manner. To further define the role of angiotensin type 1 (AT1) and type 2 (AT2) receptors involved in Ang-II-mediated VEGF synthesis, the effects of selective AT1 and AT2 receptor antagonists were evaluated. Prior treatment with losartan significantly inhibited VEGF mRNA and protein synthesis induced by Ang-II, which suggests that the AT1 receptor is involved in Ang-II-mediated VEGF synthesis. Furthermore, stimulation of the cells with Ang-II increased both phosphorylation of p38 MAPK and MAP kinase kinase 3/6 (MKK3/6). Additionally, Ang-II enhanced the DNA binding activity to cAMP response element binding protein (CREB) and phosphorylation of CREB. In addition, to investigate the role of p38 MAPK in Ang-II-induced VEGF synthesis, podocytes were pretreated with or without the p38 MAPK inhibitor, SB203580 for 24 h to observe whether Ang-II-mediated VEGF synthesis was inhibited by blocking p38 MAPK. The addition of SB203580 led to a marked inhibition of the increased VEGF mRNA and protein production induced by Ang-II in a dose-dependent manner. Taken together, these results suggest that Ang-II stimulates the synthesis of VEGF in podocytes and the production of VEGF induced by Ang-II is mediated, in part, through the activation of the p38 MAPK pathway.
