      • D-Amphetamine이 니코틴성 흥분작용에 의한 카테콜아민 분비작용에 미치는 영향

        본 연구의 목적은 d-arphetamine이 흰쥐의 적출부신 관류모델에서 니코틴 수용체 흥분에 의한 카테콜아민(CA) 유리작용에 미치는 영향을 검색하여 그 작용기전을 규명하고자 하였다. D-amphetamine은 흰쥐 부신정맥내로 60분간 관류시 d-amphetamine자체는 약한 CA 분비작용을 나타내었으나, d-amphetamine (30 μM)을 비롯한 강력한 neuronal nicotine 수용체 작용제인 cytisine (50 μM) 및 epibatidine (30 μM)에 의한 CA 유리작용을 처음 4-10분 동안만 유의하게 증강시켰다. 또한, d-amphetamine (30 μM)은 60분간 부신정맥 내로 관류한 상태에서 dihydropyridine L-형 칼슘통로 개방약물인 Bay-K-8644 (10 μM)과 세포질내 칼슘저장고에서 Ca^(2+) ATPase 억제제인 cyclopiazonic acid (10 μM)의 CA 유리작용을 처음 4분간만 유의하게 증강시켰다. 그러나, 고농도의 d-amphetamine (500 μM)은 상기한 모든 분비촉진제의 CA분비작용을 오히려 억제하였다. 이와 같은 연구결과로 보면, 흰쥐 관류 부신수질에서 d-amphetamine은 낮은 농도에서는 콜린성 니코틴 수용체 흥분에 의한 카테콜아민 분비반응을 증강시키지만, 고농도에서는 오히려 억제적으로 작용함을 시사한다. 따라서, d-amphetamine은 용량에 따라서 흰쥐 적출 관류부신수질의 니코틴 수용체의 작용제 및 길항제로 이중 작용(dual action)을 나타내는 것으로 생각된다. 이러한 d-amphetamine의 작용은 흰쥐 부신수질 크롬친화세포의 dihydropyridine계 L-형 칼슘통로의 활성화 및 세포 내 칼슘저장고로부터 칼슘유리작용과 관련성이 있는 것으로 사료된다. The purpose of the present study was to examine the effect of d-amphetamine on CA release evoked by nocotinic receptor stimulation from the isolated perfused model of the rat adrenal gland, and to establish its mechanism of action. D-amphetamine(30 μM), when perfused into an adrenal vein of the rat adrenal gland for 60 min, enhanced the CA secretory responses evoked by ACh (5.32 mM), nicotine (30 μM), cytisine (50 μM, a selective neuronal nicotinic Nn-receptor agonist) and epibatidine (30 nM, a selective neuronal nicotinic Nn receptor agonist) only for the first period (4~10 min), although it alone has weak effect on CA secretion. Moreover, d-amphetamine (30 μM) in to an adrenal vein for 60 min also augmented the CA release evoked by BAY-K-8644, an activator of the dihydropyridine L-type Ca^(2+) channels, and cyclopiazonic acid, an inhibitor of cytoplasmic Ca^(2+) ATPase only for the first peroid (4 min). However, in the presence rather inhibited the CA secretory responses evoked by the above all of secretagogues. Taken together, these experimental results suggest that d-amphetamine at a low concentration enhances the CA secretion from the rat adrenal medulla evoked by stimulation of cholinergic nicotininc receptors, but at a high concentration it rather inhibits them. It semms that d-amphetamine has dual action acting as both agonist and antagonist at nicotinic receptors of the isolated perfused rat adrenal medulla, which are might be dependent on the concentration. It is also thought that these actions of d-amphetamine are probably relevant to the activation of the dihydropyridine L-type Ca^(2+) channels located on the rat adrenomedullary chromaffin cell membrane and release of Ca^(2+) from the cytoplasmic store.

        Production and Characterization of Enzymes Involved in Chitin Catabolic Cascade from a Bacterial Strain Isolated from Soil

        The present study was aimed to isolate the bacteria producing chitinolytic enzymes including chitin deacetylase (CDA) from soil origin. About 24 bacteria were screened for their chitinolytic enzymes producing ability on the basis of colloidal chitin. Amongst one of the most potent isolates designated to JI-02 was further selected as the most prominent chitinolytic and CDAlike enzyme (CDA-X) producer based on the ability of deacetylation of an artificial substrate pNP-acetamilide. The maximum production of CDA-X in crude enzyme from JI-02 was observed in the presence of 1% colloidal chitin at 37°C and pH 6.4 after 120 h of incubation. Potent enzyme activity of CDA-X was determined toward pNP-acetamilide, demonstrating that optimal pH, temperature and ionic strength of the CDA-X were assessed to be 7.0, 52°C and 125 mM, respectively. Unexpectedly, Nacetylglucosamine (GlcNAc) was not fully converted by the action of CDA-X to glucosamine (GlcN) under the established conditions. Further study on enzyme activity toward chitin-oligosaccharides consisting of multi-N-acetylglucosamine (GlcNAc)n, n=2-5 may be necessary to elucidate the mode of action which requires the minimum size of (GlcNAc)n. However, our data suggest that CDA-X can convert chitin to chitosan with the maximum yield of approximately 0.08g/L during the fermentation of the strain in the presence of chitin. Furthermore, we found that crude enzyme showed the significant chitosanolytic activity. Taken together, our data suggest that the strain JI-02 may be a potential candidate as a particular strain for better understating of chitin catabolic pathway.

      • 동거형태에 따른 젊은 남자성인의 식행동과 건강관련 행동의 차이

        A survey was performed to 250 male young men who were living alone and shared living with friends in Chungnam province. Their average age was 22.6 years old, height was 171.8 cm, weight was 65.6 kg and BMI was 22.2 Comparing the prevalence of drinking, smoking and exercise, there were significantly different between living alone subjects and shared-living ones. The main value toward eating/meals was for hungry soultion. And we found many significant differences by living with or without companion. When young men living without companions, they shouved to be more in regular mealtimes, no-skipped meals, breakfast eating, adequate food amounts, eating-outs, etc. and to be less in pleasant mealtimes, various foods, snacking and especially late snacks. The good food intake pattern were highly scored in the alone-living group than the shared-living group, whereas the bad food intake pattern were also highly scoued in the alone-living group than the other. The latter might be due to the more food exposures because of companions. These findings could conclude that dietary behaviors of the young living alone were evaluated as good enough.

        고대 금속상감에 관한 시론

        금속상감은 바탕 금속에 홈을 만들어 홈 속에 바탕 금속과는 다른 금속을 물리적인 힘으로 감입하는 금공장식 기법이다. 본고는 상감홈과 상감선 등 제작에 관련된 여러 요소를 바탕으로 삼국시대와 일본 고분시대 출토 유물을 분류하고 기법상 차이와 유사점 등을 비교함으로써 고대 한일 금속상감의 제작공방과 그 계보를 상정하기 위한 시론이다. 금속상감은 연마라는 최종 제작과정과 부식이라는 금속의 특성으로 인해 육안관찰만으로는 제작 기법의 특징 파악이 곤란하다. 따라서 필자는 마이크로스코프와 X선 장치를 이용하여 상감유물의 기술적 특징을 파악하고 제작실험 비교를 통해 고대 금속상감 기법의 복원을 시도하였다. 그 결과, 상감홈의 경우 기존의 금공 연구를 통해 지적되어 왔던 축조와 모조기법의 양자가 존재하는 한편, 이제까지 언급되지 않았던 상감선의 제작기법에 있어서 단조 이외에 감기나 꼬기, 접기와 같은 다양한 제작기법이 존재한다는 것을 확인할 수 있었다. 각 기법의 사례에서는 시기와 지역차가 확인된다. 상감홈과 상감선 제작기법의 기술적 특징은 유라시아 대륙에서 그 원류를 찾을 수 있다. 이는 백제, 신라, 가야, 그리고 일본열도에 이르는 세선세공 기법의 전파루트와 상응하고 금제 보요의 기원과도 관련된다. 근년 발굴 조사된 러시아 초기철기 시대 상감유물, 그리고 후한대 중국 상감명문대도와 비교하여 각 기법 조합의 시기차와 지역차를 통해 고대한일 상감에 이르는 계보가 모조기법의 홈과 감기나 꼬기기법의 선을 특징으로 하는 <북방루 트>, 그리고 축조기법의 홈과 단조선을 특징으로 하는 <중국루트>의 가설을 제시하고자 한다. The metal inlaying technique is to putting gold or silver into a carved wire of the based metal with physical power. In this paper, based on the various elements such as quality of metal, inlaid groove and inlaid wire, presume the manufacture of workshop of the metal inlaying and its system by classifying the artifacts of the Three-Kingdom Period of the Korean Peninsula and Kofun Period of Japan and comparing the similarities and differences between them. It is hard to apprehend the feature of its technique with the naked eye because of polishing and corrosion. Therefore, I grasp the technical character by observing them with macroscopy as well as microscope, X-ray and restore the metal inlaying technique with comparing it with production experiment. As a result, in case of inlaid groove, there are lined-carving and triangled-carving that is commonly indicated in bronze artifacts. In case of inlaid wire, there are various technique such as winding, twisting, folding except hammering that is not indicated so far. It has got time and regional differences in example of each techniques. The feature of the technique of inlaid groove and inlaid wire is found in Eurasia. It corresponds to the diffusion route of the filigree technique to Baekje, Silla and Japan. By comparing its route connected with the origin of the golden ornament with the inlaid artifacts of Russian Early Iron Age and Chinese inlaid inscription sword in West Han Dynasty, abstract time and regional differences of the combination of the technique. Based on this, I propose a hypothesis that there are two different routes to the ancient Korea and Japan, one in the Northern-route; groove of the lined-carving and winded or twisted wire, the other in Chinese route; groove of triangled-carving and hammering wire.

        가감청영탕(加減淸營湯)에 효과를 보인 열성 아토피 피부염 환자 6례 증례 보고

        Objective : The purpose of this study is to investigate the effect of Gagamchengyoung-tang($Ji\bar{a}ji\check{a}nq\bar{i}ngy\acute{i}ng-t\bar{a}ng$) for atopic dermatitis patients diagnosed as the febrile tendency. Method : This case study was done on 6 atopic patients with febrile tendency who have visited Korean Medicine Clinic from 2013.08.01. to 2014.04.30.. Gagamchengyoung-tang($Ji\bar{a}ji\check{a}nq\bar{i}ngy\acute{i}ng-t\bar{a}ng$) was prescribed to all 6 patients and evaluated the symptom change through photographs and questionnaire. Objective SCORAD Index(OSI) was used to evaluate the objective symptom and VAS was used to evaluate the subjective change. Results & Conclusion : The Objective SCORAD index and VAS were decreased in all patients. The average OSI reduction score was 26.3. Most symptoms of 6 patients were improved. Gagamchengyoung-tang ($Ji\bar{a}ji\check{a}nq\bar{i}ngy\acute{i}ng-t\bar{a}ng$) can be effective in patients who were diagnosed as febrile tendency.

      • Benson's Group Additivity Method를 이용한 염화다환방향족 화합물의 열역학적물성치 평가와 생성반응경로에 관한 연구

        도시폐기물 소각시설에서 다이옥신류는 chlorobenzene, chlorophenols, PCBs등의 염소성분이 포함된 방향족화합물과 함께 폐기물의 소각처리시 불완전 연소로 인해 주로 생성된다. 이들 다이옥신 생성에 대한 반응메카니즘의 예측에 필요한 다이옥신을 형성하는 전구물질인 분자 및 radical의 정확한 열역학적 물성치를 평가하기 위해 Benson's group additivity method에서는 고려되지 않는 Cl, F, OH, CH3등과 같은 2개 이상의 치환기가 결합된 방향족화합물에 대한 computer code인 THERM을 이용하여 엔탈피(Hf), 엔트로피(St), 열용량(Cp)을 평가하였다. 또한 이와 같은 열역학적 물성치 평가법의 신뢰성을 검토하기 위해 다양한 방향족화합물에 대한 실험적으로 측정된 문헌치와 평가치와의 절대오차는 para-cresol의 0.02kcal/mole에서 hexamethylbenzene의 3.46kcal/mole로 좁은 범위를 나타내었다. 또한 다이옥신을 생성하는 전구물질들의 열역학적 물성치를 THERM을 이용해 간접적으로 평가함으로써 다이옥신을 형성하는 주요 생성반응 경로에 대해 살펴본 결과는 다이옥신을 형성하는 기초반응(elementary reaction)중 욤소수가 많은 반응물질일수록 반응이 더 잘 일어남을 알수 있었다. Chlorinated dioxins are often observed in effluent from oxidation or incineration such as chlorobenzenes, chlorophenols and PCBs. It would be of significant value to have an accurate and fundamental understanding of the mechanism for dioxin formation, in addition to the knowledge of dioxin thermodynamic properties, in order to assist in the design of stategies to effectively control or reduce emissions of these compounds. Very little has been published on the thermodynamic properties and elementary reactions which lead to formation of dioxins. Benson group additivity method, when used for the estimation of thermodynamic properties of molecules, does not account for all interactions between functional groups or atoms on aromatics, such as chlorines, hydroxyl or ether linkages. Therefore thermodynamic properties of dioxins are estimated by THERM based on Benson's group additivity method corrected by interactions group. Absolute errors between our predictions and literature values range from a low of 0.02 kcal/mole for paracresol to a high 3.46kcal/mole for hexamethylbenzene. We use these thermodynamic calculations to show the feasibility of several elementary reaction pathways leading to dioxin formation, where the presence of chlorine acts to enhance or lower the Gibbs free energy of reactions.

        It is the plan that storage gets energy done by in structure with what thermal storage of structure changes only supply air duct among the existing HVAC system, and a basic principle coordinates the damper that it was installed makeup air in supply air duct like former HVAC system to an inside at supply and a night by day, and do makeup air supply with in ceiling space of thermal storage of structure HVAC system. This thermal storage of structure HVAC system uses the existing utility system, and energy saving can get structure of building done with energy storage, before air conditioning and peak load cut getting day heat load done.

        중등학교 미디어교육을 위한 문화기호학적 방법론

        이 연구는 미디어 환경 변화에 따른 학교 미디어교육의 필요성을 인식하고, 중등학교 미디어교육을 위한 교수학습방법론을 제시 하는 데 목적을 두고 있다. 이를 위해 미디어교육의 패러다임 변화에 따른 미디어교육 개념의 변화에 대해 조사하고, 학교 미디어 교육으로서 특히 중등 국어과의 미디어교육적 요소를 검토하게 된다. 나아가 현행 국어과에서 적용되는 미디어교육의 방법론을 개괄하고 문제점을 도출하게 된다. 아직까지 미디어교육이 ‘미디어를 통한 교육’ 즉 미디어를 통해 특정 교육 요소를 이해시키는데 활용하는 도구적 입장을 취하고 있다. 이 연구에서는 이런 점을 비판적으로 성찰하고 ‘미디어에 대한 교육’이 중점적으로 이루어 질 수 있도록 교육적 대안을 마련하게 된다. 그것이 바로 문화기호학적 교수학습방법론이다. 일찍이 기호학은 구조주의를 바탕으로 한 질적 연구방법으로서 교육적 영역에 도입되었다. 이 연구에서는 그레마스에 의해 시도되었던 행동자 모형과 내러티브 분석 기법을 중등학교 미디어교육에 도입한다. 이 문화기호학적 방법론은 멀티 리더러시를 가능케 하는 데, 이는 미디어 수용자 혹은 잠재적인 미디어 학습자가 특정 미디어 텍스트를 통해 문화적 차원의 의미 해석에 기여한다. 뿐만 아니라 수용자나 학습자가 처한 사회문화적 콘텍스트를 비판적으로 성찰 하는데 도움을 줄 것이다. The research explores acceptance of media Education in Korean Language Education. The concept of media education extends to teach communication skills as 'new language'. Using internet is not only the technology but the process of communication. For the new generation, media education value the process of communication like the constructivism paradigm. Media education have to be get through via the idea of literacy. For going forward expansion of communication ability, the purpose and contents of media education need to be re-established. Results of this article were as follow: First, the idea of media education was reflected on the 7th Korean curriculum. And some Korean teachers group developed series of textbooks for media education in secondary schools. Second, the teaching methodologies based cultural semiotics, multi-literacy methodology and narrative scheme methodology play an important role for secondary Korean class.

        RAW 264.7 세포에 대한 중국산 천연 광물성 섬유 TAFMAG의 독성효과

        Objectives : This study was designed to evaluate cytotoxicity of TAFMAG, which is a trade name of natural mineral fiber mined and produced in China. Methods : The cytotoxicity of TAFMAG was evaluated by measuring iron content, lipid peroxidation, erythrocyte hemolysis, and cytotoxicity in vitro. These results were compared with the data of chrystotile and wollastonite as a positive and negative control, respectively. Results : There was significant increase of Fenton activity in TAFMAG and chrysotile with dose-response pattern. The iron chelating agent, desferrioxamine, significantly decreased Fenton activity of the particulates except wollastonite. TAFMAG and chrysotile fibers significantly increased malondialdehyde concentration from lipid peroxidation of the red blood cell membrane. In erythrocyte hemolysis test, TAFMAG & chrysotile had stronger effect on erythrocyte hemolysis than wollastonite with the concentration of 1,000g/ml. Furthermore, TAFMAG was more hemolytic than chrysotile with the concentration of 5,000g/ml. There was a significant cytotoxic effect in TAFMAG and chrysotile on RAW cell compared with wollastonite. Conclusions : In vitro study suggested that TAFMAG may have a similar health hazard as usual asbestos.
