        가죽나무(A. altissima) 부위별 에탄을 추출물의 생리활성

        한방생약자원으로 사용되는 가죽나무(A. altissima)의 생리학적 활성을 조사하고자 뿌리와 줄기, 잎을 에탄올을 용매로 추출하여 농도에 따라 전자공여능, SOD유사활성, 아질산염 소거능을 측정하였으며, xanthine oxidase와 tyrosinase 저해 활성을 측정하였다. 전자공여능은 $1,000{\mu}g/mL$의 농도에서 뿌리와 줄기 추출물이 64.04%와 63.27%이었으며, 잎 추출물은 17.47%를 나타내었다. SOD 유사활성능은 $7.66%{\sim}50.00%$로 잎 추출물이 뿌리와 줄기에 비해 약 5배 이상 높았고, 아질산염 소거능에 있어서도 가죽나무 잎 추출물이 $1,000{\mu}g/mL$의 농도에서 pH 1.2와 3.0에서 98%이상의 높은 소거효과를 나타내었으며 $500{\mu}g/mL$의 농도에서도 90% 이상의 소거효과를 나타내었다. Xanthine oxidase저해 활성은 $1,000{\mu}g/mL$ 농도에서 뿌리와 줄기, 잎 추출물 모두에서 90% 이상의 저해율을 보였고, tyrosinase저해는 $2,000{\mu}g/mL$의 농도에서 뿌리 추출물이 62.01%로 가장 높은 저해효과를 나타내었으며, $100{\mu}g/mL$의 농도에서도 57.70%의 저해 활성을 나타내었다. 그러므로 가죽나무는 한방생약자원으로 이용되는 뿌리(저근백피) 이외에도 잎은 높은 SOD 유사활성능과 아질산염 소거능을 나타내며 줄기에서도 뿌리보다 높은 전자공여능과 아질산염 소거능 및 유사한 xanthine oxidase 저해 활성을 나타내므로 유용한 한방생약자원인 것으로 판단된다. The study was carried out to optimize the electron donating ability (EDA), superoxide dismutase (SOD)-like activity, nitrite scavenging ability, and the inhibitory activities of xanthine oxidase and tyrosinase of Ailanthus altissima ethanol extracts from roots, stems and leaves. The EDA of the roots and stem extracts were 64.04% and 63.27% at $1,000{\mu}g/mL$, respectively. The SOD-like activity of the leaves extract was the highest (50.00%) at $1,000{\mu}g/mL$. The nitrite scavenging ability of the leaves extracts was over 98% at pH 1.2 and 3.0. The xanthine oxidase inhibitory rates of the extracts were $93.62%{\sim}95.40%$ and the tyrosinase inhibitory rate of the roots was the highest (62.01%) at the concentration of $1,000{\mu}g/mL$. These results indicated that the roots extract showed the highest EDA and tyrosinase inhibition, while the leaves extract had tile highest SOD-like activity and nitrate scavenging ability.

        애기똥풀(Chelidonium majus var. asiaticum) 추출물의 생리활성 연구

        This research is to useful physiological activities data of the aerial part of the Chelidonium majus var asiaticum, analyzed contents of polyphenol and flavonoid compound, nitrite scavenging, superoxide dismutatse (SOD)-like activity, electron donating, tyrosinase inhibition and xanthine oxidase inhibition. The RW (Reflux water extract of C. majus var asiaticum) showed the highest value of nitrite scavenging ability of 73.13% at pH 1.2, 2.0 mg/mL. The RE (Reflux 70% ethanol extract of C. majus var asiaticum) showed the highest of SOD-like activity of 78.67% at 2.0 mg/mL and electron donating ability of 86.07% at 0.3 mg/mL. The tyrosinase inhibitory of RW was 38.71%, it was significantly 2.5 higher compared to the RE (15.18%). The xanthine oxidase inhibitory of RW and RE were over 97% at 2.0 mg/mL. The both extracts were higher than ascorbic acid (55.36%) at 0.05 mg/mL. This suggests that C. majus var. asiaticum can be useful materials as functional healthy product, medicine, and cosmetic products.

        우산나물(Syneilesis palmata) 추출물의 항산화 활성

        기능성식품 소재를 개발하기 위한 연구의 일환으로 야생산채류인 우산나물의 지상부와 뿌리 추출물에 대한 플라보노이드와 폴리페놀 화합물 함량 그리고 생리활성 효과를 측정하였다. 플라보노이드는 지상부 에탄올 추출물이 31.72 ㎎/g으로 가장 많았으며, 폴리페놀은 뿌리의 에탄올 추출물에서 68.11 ㎎/g으로 가장 높았다. Xanthine oxidase에 대한 저해 효과를 측정한 결과 1.0 ㎎/mL의 농도에서 지상부 물 추출물과 뿌리의 에탄올 추출물 모두 98% 이상의 저해율을 보였으며, tyrosinase 저해는 뿌리 에탄올 추출물에서만 9.33%의 저해효과를 나타내었다. 우산나물 뿌리의 에탄올 추출물은 pH 1.2의 조건에서 70.89%의 아질산염 소거능을 보였으며, SOD 유사활성능도 13.06%의 활성을 나타내었다. 또한 전자공여능에서도 뿌리 에탄올 추출물이 98.58%로 천연항산화제인 ascorbic acid보다 높은 활성을 보였다. 이상의 결과 우산나물 뿌리의 에탄올 추출물의 폴리페놀 함량이 가장 높았고, tyrosinase 저해, 아질산염 소거, SOD 유사활성 및 전자공여능이 가장 우수하였다. 우산나물 뿌리의 물 추출물은 에탄올 추출물보다는 활성이 낮았으나 우산나물 지상부 추출물보다 높게 분석되었으며, xanthine oxidase 저해율이 가장 높았다. 본 실험결과 우산나물은 우수한 항산화 활성을 지닌 기능성식품 소재임을 확인할 수 있었으며, 식용으로 사용하고 있는 어린순 이외에 뿌리에도 다량의 폴리페놀을 함유하며, 우수한 생리활성 효과를 나타내므로 이를 항산화 활성을 지닌 기능성 건강식품 등의 재료로 활용할 수 있을 것으로 생각된다. This study was conducted to examine the antioxidant activities of the extracts from aerial parts and roots of Syneilesis palmata. The ethanol extract of aerial parts showed the highest content of flavonoid compounds (31.72 ㎎/g), and the ethanol extract of roots has the highest content of total polyphenol compounds (68.11 ㎎/g). The water extract of S. palmata roots showed the highest xanthine oxidase inhibition of 99.29% and the ethanol extract of aerial parts showed 98.48% at 1.0 ㎎/mL. The ethanol extract of roots showed the highest value of nitrite scavenging ability of 70.89% at pH 1.2, SOD-like activity of 13.06% and electron donating ability of 98.58% at 1.0 ㎎/mL concentration. The effect of tyrosinase inhibition was found at only the ethanol extracts of roots (9.33%). We found that root extracts contain abundance polyphenol compounds and their antioxidant activities were greater than those of the extracts of aerial part.

        Monascus purpureus P-57 변이주가 생산하는 홍국색소의 항산화효과

        Monascus purpureue KCCM 60016으로부터 색소생성 이 우수한 변이주 P-57이 생성한 홍국색소의 항산화 활성을 조사하였다. 홍국색소는 chloroform에 아주 잘 추출되며, 특히 hexane에서는 황색색소가 특이적으로 높게 추출되었다. DPPH radical소거 효과는 hexane분획, chloroform 분획, ethyl acetate분획, butanol 분획, water 분획의 순으로 높게 나타났다. Xanthine oxidase저해효과는 hexane분획, chloroform분획, ethyl acetate 분획의 순으로 높게 나타났으며, hexane 분획물의 경우 5 ppm에서 $41.2\%$, 50 ppm에서 $82.4\%$로 높은 저해율을 보였으며, 저해기작은 비경쟁적 저해였다. Antioxidant activity of monascus pigment of Monascus purpureus P-57 mutant was studied. Methanol extract from monascus pigment was separated into five organic solvent fractions; hexane, chloroform, ethyl acetate, butanol and water fractions. Hexane fraction showed the highest free radical scavenging effect on 1,l-diphenyl-2-picryl hydrazyl(DPPH), and the strongest inhibitory effect against xanthine oxidase, followed by chloroform fraction. But butanol and water fraction did not show inhibitory effect against the enzyme. Lineweaver-Burk plot showed that hexane fraction inhibited xanthine oxidase by non-competitive mode.

        Flavonoids from the Flower of Rhododendron yedoense var. Poukhanense and Their Antioxidant Activities

        Antioxidant flavonoids have been isolated from the flower of Rhododendron yedoense var. poukhanense. One new flavonoid and three known flavonoids, quercetin-5-O-β-D-glucopyranoside (1), quercetin (3), and quercitrin (4), were isolated from the butanol and ethyl acetate extracts of the plant. The new flavonoid was identified as myricitrin-5-methyl ether (2). The isolation of these flavonoids from this plant, for the first time, is a valuable finding. The flavonoids were evaluated for their antioxidant activities using 1,1-diphenyl-2-picrylhydrazyl free radical (DPPH), TBARS (thiobarbituric acid reactive substance) and superoxide anion radical (O2 −) in the xanthine/ xanthine oxidase assay system. In the DPPH scavenging assay, the IC50 values were 4.5±0.48 µM for compound 2 and 9.7±0.29 µM for compound 3, which showed an antioxidant activity approximately 1.5~2 times higher than the antioxidant activity of α-tocopherol (9.8±0.94 µM). Additionally, the antioxidant activities of myricitrin-5-methyl ether (2) (IC50 = 1.7±0.22 µM) and quercetrin (4) (IC50 = 1.9±0.63 µM) were higher than that of L-ascorbic acid (IC50 = 7.4±0.63 µM) when evaluated using a TBARS assay. Compound 2 showed high activity in both the inhibition of xanthine oxidase (1.1±0.21 mM) and in the activation of superoxide scavenging.

        Antioxidant and Xanthine Oxidase Inhibition Activities of Cynomorium songaricum Extracts

        In this study, we evaluated the antioxidant activities and xanthine oxidase inhibition effects of water and ethanol extracts of Cynomorium songaricum. The ethanol extract of C. songaricum (EE) contained more phenolic and flavonoid compounds than the water extract (WE). The antioxidant activities of the extracts were increased as the concentration of the extract increased. The WE has better effectiveness than the EE for DPPH free radical scavenging activity and nitrite scavenging ability. The nitrite scavenging abilities of WE were 90.02% ($EC_{50}$ 653.15 ${\mu}g$/mL) at conditions of pH 1.2 and 2,000 ${\mu}g$/mL, and 84.34% ($EC_{50}$ 817.17 ${\mu}g$/mL) at pH 3.0. The EE has more effective SOD-like activity and XO inhibition than WE. The SOD-like activity of EE was 81.47% at a concentration of 2,000 ${\mu}g$/mL, $EC_{50}$ was 951.70 ${\mu}g$/mL. The xanthine oxidase inhibition of the EE, with an $EC_{50}$of 112.47 ${\mu}g$/mL, is greater than that of ascorbic acid, which was 192.50 ${\mu}g$/mL (p<0.05). These results suggest that the C. songaricum is a potentially useful antioxidant source for the development of nutraceuticals and medicines.

        Antioxidant, anti-acetylcholinesterase and xanthine oxidase inhibitory activities of three extracts from Phellinus igniarius

        This study was initiated to investigate antioxidant, anti-acetylcholinesterase, and xanthine oxidase inhibitory activities and properties of fruiting bodies, mycelia, and fermentation culture filtrates from Phellinus igniarius. The contents of total phenols and flavonoid of fruit bodies, mycelia, and culture filtrate were 15.35-1.36 mg/g, 10.35-7.85 mg/g, and 8.25-5.36 mg/g. The 1,1- diphenyl-1-picrylhydrazyl (DPPH) radical scavenging abilities of the extracts from the fruiting bodies, mycelia, and culture filtrates were 90.25-95.60%, 78.82-85.24%, and 76.32-82.50% at 50-400 μg/mL, respectively. The chelating ability of fruiting body extract on ferrous ions was higher than those of mycelia and culture filtrates tested. The anti-acetylcholinesterase inhibitory activity of the fruiting body extract at 400 μg/mg exhibited 91.10% on AChE, which is lower than that of positive control, galanthamine (94.82%). The xanthine oxidase inhibitory activities of the fruiting bodies, mycelia, and culture extract were 85.47%, 78.13%, and 72.49% at 400 μg/mL, respectively. Overall, the fruiting body extract has better anti-acetylcholinesterase, antioxidant and xanthine oxidase inhibitory activities than those from mycelia and culture filtrate.

        In Vitro a-Amylase, a-Glucosidase, Pancreatic Lipase, Xanthine Oxidase Inhibiting Activity of Agaricus bisporus Extracts

        In this study, the a-amylase inhibitory activity, a-glucosidase inhibitory activity, pancreatic lipase inhibitory activity, and Xanthine Oxidase inhibitory activity of the fruiting body extracts of 5 varieties of Agaricus bisporus (AB) were confirmed. First, the a-amylase inhibi- tory activity of AB12, AB13, AB18, AB34, and AB40 methanol extracts was lower than that of acarbose, a positive control, in all concentration ranges. The a-glucosidase inhibitory activity of the AB40, AB13, and AB12 methanol extracts at the extract concentration of 1.0 mg/mL was 80.5%, 81.3%, and 78.5%, respectively, similar to that of acarbose, a positive control. The pancreatic lipase inhibitory activity of the methanol extract of Agaricus bisporus fruiting body was significantly lower than that of the positive control orlistat in the concentration range of 50 1.000 (mg/mL). The Xanthine Oxidase inhibitory activity was 0.5 8.0 mg/mL of each extract, which was significantly lower than that of the positive control allopurinol in the same concentration range. However, the Xanthine Oxidase inhibitory activity of AB13 and AB40 at 8.0 mg/mL was about 70%, which was higher than that of other mush- rooms. In conclusion, five kinds of Agaricus bisporus fruiting bodies seem to have inhibitory effects on enzymes such as a-amylase, a-glucosidase, pancreatic lipase, and Xanthine Oxidase that degrade starch and protein. In particular, it has an inhibitory effect and a reduction effect on xanthine oxidase that causes gout, so it is expected that it can be developed and used as a food or health supplement with health functional properties through future research.
