        국내산 백태 품종의 지방산 및 유리아미노산 조성

        The fatty acid composition and free amino acid content of domestic soybean cultivars were analyzed to confirm the quality characteristics of protein and fat contained in soybeans. The saturated fatty acid content of soybeans included palmitic acid at 9.47~11.15%, followed by stearic acid and myristic acid. The total saturated fatty acid content in soybeans was 12.56~14.34%, with Taekwang having the lowest content, followed by Daewon, Seonyu, Cheonga, and Jinpung. The linoleic acid content, an unsaturated fatty acid, was 45.69~58.17%, with Taekwang showing the lowest composition and Jinpung showing the highest composition. Next was oleic acid at 14.69~33.86%. Jinpung had the highest linoleic acid composition, had the lowest and Taekwang which had the least linoleic acid, had the highest. The unsaturated fatty acid content was in the order of linolenic acid, eicosatrienoic acid, eicosadienoic acid, and eicosapentaenoic acid. The total free amino acid content was 217.28~456.66 mg%, with Daewon showing the highest free amino acid content, followed by Seonyu, Taekwang, Cheonga, and Jinpung. The free amino acid content varied depending on the cultivars, but in general, the free amino acids in the soybeans used in the experiment showed higher aspartic acid, glutamic acid, and proline contents than other amino acids.

      • Unsaturated fatty acids protect trophoblast cells from saturated fatty acid-induced autophagy defects

        <P><B>Abstract</B></P> <P>Dysregulated serum fatty acids are associated with a lipotoxic placental environment, which contributes to increased pregnancy complications via altered trophoblast invasion. However, the role of saturated and unsaturated fatty acids in trophoblastic autophagy has yet to be explored. Here, we demonstrated that prolonged exposure of saturated fatty acids interferes with the invasiveness of human extravillous trophoblasts. Saturated fatty acids (but not unsaturated fatty acids) inhibited the fusion of autophagosomes and lysosomes, resulting in the formation of intracellular protein aggregates. Furthermore, when the trophoblast cells were exposed to saturated fatty acids, unsaturated fatty acids counteracted the effects of saturated fatty acids by increasing degradation of autophagic vacuoles. Saturated fatty acids reduced the levels of the matrix metalloproteinases (MMP)-2 and MMP-9, while unsaturated fatty acids maintained their levels. In conclusion, saturated fatty acids induced decreased trophoblast invasion, of which autophagy dysfunction plays a major role.</P> <P><B>Highlights</B></P> <P> <UL> <LI> SFAs induce insoluble protein aggregation in trophoblasts. </LI> <LI> SFAs impair autophagosome-lysosome fusion in trophoblasts. </LI> <LI> UFAs attenuate SFA-induced protein aggregate formation. </LI> <LI> UFAs restore the invasive capabilities of trophoblasts. </LI> </UL> </P>

        택사(澤瀉)가 유리지방산으로 유발된 HepG2 cell의 lipoapoptosis에 미치는 영향

        Objectives : This study was designed to investigate the effect on lipoapoptosis of Alisma orientale extract against free fatty acid-induced cellular injury. Methods : HepG2 cells were used in an vitro model. HepG2 cells were treated with free fatty acids to generate a cellular model of nonalcoholic fatty liver disease (NAFLD). Using this cellular model, the anti-apoptotic effect and reducing steatosis of Alisma orientale extract against free fatty acid-induced cellular injury was evaluated by measuring steatosis and apoptosis. Results : Alisma orientale extract significantly attenuated free fatty acid-induced intracellular steatosis. Alisma orientale extract inhibited free fatty acid-mediated activation of pJNK, PUMA, BAX, caspase-3, and -9, and apoptotic kinases that are correlated with NAFLD. Alisma orientale extract also promoted Bcl-2, a anti-apoptotic protein. Conclusions : From the above, the Alisma orientale extract decreased the hepatocyte steatosis and showed the hepatocelluar protective effect by the regulation of apoptosis-related protein. It proposes the possibility of Alisma orientale extract to the treatment of nonalcoholic fatty liver disease in clinics.

        추출기법에 따른 흰점박이 꽃무지 유충의 유리아미노산 및 지방산 특성

        In order to increase the industrial applicability of health functional materials using edible insects, effective extraction processes were explored to improve the quality characteristics of the free amino acids and fatty acids of Protaetia brevitarsis seulensis larvae (PBSL). After extracting PBSL under four conditions, water extraction at 50℃ (MWE), water extraction at 95℃ (HWE), ultrasonic extraction (USE), and high pressure extraction (HPE), the free amino acid content and fatty acid compositions of the extracts were evaluated. Essential free amino acid content as well as total free amino acid content were the highest in the HPE extract, followed by USE, HWE, and MWE extracts. In the HPE extract, the content of some essential amino acids increased dramatically, and the types of major amino acids had also changed. Additionally, the proportion of unsaturated fatty acids in PBSL was 69.9%, which was higher than the reported value in meat, the main protein source for Koreans. This level has also been further enhanced by USE (78.6%) and HPE (75%). These results show that HPE and USE can enhance the nutritive value of PBSL as a health functional material by improving the composition of free amino acids and fatty acids.

        This study was carried out to investigate the changes of free sugars, non-volatile organic and fatty acids in flue-cured leaf tobacco during aging. The threshed leaf tobacco(B1O and C1L) produced in 2002 crop year was aged for 21 month in warehouse of Oc-Cheon Leaf Tobacco Processing Factory. The leaf tobacco were sampled at three month intervals for analysis of free sugars, non-volatile organic and fatty acids. The major free sugars of flue-cured were glucose and fructose regardless of tobacco grades. After aging period of 21 months, the contents of glucose and fructose showed a tendency to slightly decrease, and there was no appreciable change in the contents of sucrose in B1O and C1L grades. The major compound of non-volatile organic acid and fatty acid were malic, oxalic and citric, linolenic, linoleic and palmitic acid regardless of tobacco grade. After aging period of 21 months, the contents of malic, malonic, oxalic and fumaric acid showed a tendency to slightly decrease, whereas succinic acid was slightly increased, and maleic acid was not changed in B1O and C1L grades. The decreases in fatty acid contents in B1O and C1L grade tobacco loaves after 21 month aging were 16.5% and 9.8%, respectively. The decreases in linolenic acid contents in two grades were the highest, showing 22.1% and 12.0% reduction after 21 month aging.

        야생 돌복숭아 씨와 과육의 아미노산 및 지방산 조성에 관한 연구

        우리나라 전역에서 자생하는 야생 돌복숭아(Prunus persica Batsch var, davidiana Max.)의 씨와 과육 중의 각종 생리활성 물질을 규명하여 바이오헬스 기능성 소재 등의 자원으로서 이용 가능성을 검토하기 위한 자료로, 돌복숭아 씨와 과육 중의 일반성분, 구성 총 아미노산, 유리 아미노산 및 지질 구성 지방산 조성을 분석한 결과는 다음과 같다. 돌복숭아 씨 및 과육 중의 일반성분 함량은 탄수화물이 63.92% 와 75.11%로 대부분을 차지하였으며, 조단백질 27.85%, 12.77%, 수분 3.61%, 4.69%, 조지 방이 1.21%, 4.80% 그리고 조회분이 3.41%와 2.63% 함유되어 있는 것으로 나타났다. 돌복숭아 씨 중의 구성 총 아미노산의 함량은 3,444.35 mg%로 이 중 aspartic acid가 681.10 mg%, glutamic acid 495.48 mg%, alanine 283.66 mg%, serine 251.36 mg%, proline 229.80 mg%, lysine 192.31 mg% 및 leucine이 191.34 mg%의 순으로 조성되어 있었으며, 특히 aspartic acid 및 glutamic acid가 대부분을 차지하였다. 돌복숭아 과육의 단백질 구성 아미노산 조성의 총 함량은 1,064.02 mg%로, aspartic acid 250.15 mg%, glutamic acid는 129.63 mg%로 과육의 주요 총 아미노산이었고 lysine, proline, leucine, alanine, serine 등의 순으로 함유되어 있었다. 돌복숭아 씨 중 유리 아미노산 조성의 총 함량은 6,215.34 mg%로, 이 중 glutamic acid가 827.25 mg%로 가장 많았으며, threonine, valine, $\beta-aminobutyric$ acid 등의 순으로 함유되어 있었다. 과육 중의 유리 아미노산 조성의 총 함량은 683.82 mg%로, glutamic acid(339.49 mg%)가 대부분 이었으며, serine, proline, alanine, $\gamma-amino-n-butyric$ acid의 순으로 함유되어 있었다. 지질 구성 지방산 조성은 포화지방산이 12.49%, 단불포화지방산 13.11% 및 다불포화지방산이 74.36%의 함유비율을 보였으며, 필수지방산인 n-6계 linoleic acid$(C_{18:2})$와 n-3계 linolenic acid$(C_{18:3})$가 대부분을 차지하였다. 야생 돌복숭아 과육 중의 지방산 조성은 포화지방산이 16.74%, 단불포화지방산 17.51% 및 다불포화지방산이 65.73%의 함유 비율을 보였는데, 이 중 다불포화지방산인 n-6계 linoleic acid$(C_{18:2})$와 n-3계 linolenic acid$(C_{18:3})$가 지질 구성 총 지방산의 대부분을 차지하는 함유 비율을 나타내었다. Amino acid and fatty acid compositions of the physiological activity substance in the seed and pulpy substance of feral peach (Prunus persica Batsch var. davidiana Max.) were analyzed for the use as an biohealth functional processed products. The proximate compositions in the vacuum freeze dried seed and pulpy substance of feral peach were carbohydrate 63.92% and 75.11%, crude protein 27.85% and 12.77%, moisture 3.61% and 4.69%, crude fat 1.21% and 4.80%, crude ash 3.41% and 2.63%, respectively. Total amino acid contents in the protein of feral peach seed were 3,444.35 mg%, and the major amino acids were aspartic acid(681.10 mg%), glutamic acid(495.48 mg%), alanine(283.66 mg%), serine(251.36 mg%), proline(229.80 mg%), lysine(192.31 mg%) and leucine(191.34 mg%), respectively. Total amino acid contents in the protein of feral peach pulpy substance were 1,064.02 mg%, and the major amino acids followed aspartic acid(250.15 mg%), glutamic acid(129. 63 mg%), lysine, proline, leucine, alanine and serine, in a decreasing order. The richest total amino acid content contained in feral peach seed and pulpy substance was aspartic acid, followed by glutamic acid. The amount of free amino acids of feral peach seed were 6,215.34 ms%, and the major free amino acids were glutamic acid(827.25 mg%), threonine, valine and $\beta-aminobutyric$ acid, respectively. Free amino acid contents of pulpy substance were 683.82 mg%, and the major free amino acids were glutamic acid(339.49 mg%), serine proline, alanine and $\gamma-amino-n-butyric$ acid. Especially, in the case of glutamic acid, it was highest. The compositions of major total fatty acid in the lipid feral peach (Prunus persica Batsch var. davidiana Max.) seed and pulpy sabstance were linoleic acid($C_{18:2}$, n-6) and linolenic acid($C_{18:3}$, n-3), particularly.

        쏘팔메토(Saw Palmetto) 열매 추출물의 이화학적인 특성 연구

        FT-IR, GC/MS, and ATR-FT-IR analyses were performed to confirm the physicochemical characteristics of saw palmetto fruit (SPF) extract. FT-IR analysis of the standard product showed that the band corresponding to the carbonyl bond of free fatty acid was stronger than the band of acyl-glyceride. Sample E was identified as having the same trend as the standard sample. Fatty acid composition analysis revealed that the main fatty acids in the standard sample were lauric acid and oleic acid. The content of lauric acid ranged from approximately 30% to 38% in samples B, C, D, and E, while the content of oleic acid ranged from approximately 29% to 34%. The GC/MS analysis confirmed that the standard SPF extract consisted of fatty acids and fatty acid ethyl esters. Sample E demonstrated a similar pattern to the standard samples in terms of oleic acid, lauric acid, and fatty acid esters. ATR-FT-IR analysis indicated that only sample E was predicted to contain 100% saw palmetto extract. Therefore, these study findings can be considered fundamental data for analyzing the physicochemical characteristics of the composition of SPF extract.

        감마선 조사가 건조-발효소시지의 Trans 지방산, 유리아미노산 및 관능검사 특성에 미치는 영향

        본 연구는 감마선 조사가 건조-발효햄의 조직감, 지방산, 유리아미노산 및 관능적 특성에 미치는 영향을 조사하기 위해 실시하였다. 경도(hardness) 및 부착성(adhesiveness) 의 경우 조사 처리구가 유의적으로 낮게 나타났다(p<0.01). 검성(gumminess)의 경우에는 2 및 4 kGy처리구가 대조구 및 1 kGy처리구보다 유의적으로 높은 값을 나타내었다(p<0.01). 지방산의 경우 처리구의 포화지방산(SFA)(C14:0, C16:0, C18:0)은 유의적으로 높았고(p<0.05), 불포화지방산 (UFA)(C16:1, C18:1 및 C18:3), trans 지방산, n-3 및 n-6 지방산 의 함량은 4 kGy처리에서 감소하였다(p<0.05). 감마선 조사처리는 phosposerine, aspartic acid, glutamic acid를 증가 시켰으나, lysine은 감소시켰다(p<0.05). 관능검사의 경우, 황취(sulfur odor) 및 지방산화 맛(fat pungent flavor)은 4 kGy에서 가장 높게 나타났으며(p<0.05), 곰팡이 냄새(moldy odor) 및 곰팡이 맛(moldy flavor)은 낮게 나타났다(p<0.05). 이상의 결과 감마선 조사가 조직감, 지방산조성 및 관능적 특성에 영향을 미쳤다. In this study, the effects of gamma-irradiation (1, 2 and 4 kGy) on texture profiles, fatty acid composition, free amino acids and sensory evaluation of dry-fermented sausage were evaluated. The hardness and adhesiveness of irradiated samples were significantly lower (p<0.01) than that of the control, while the gumminess of samples irradiated with 2 and 4 kGy was higher than the other tested samples (p<0.01). The sample irradiated at 4 kGy had a higher saturated fatty acid (SFA) content (C14:0, C16:0, C18:0) and significantly lower (p<0.05) unsaturated fatty acid content (UFA) (C16:1, C18:1, C18:3), trans, n-3 and n-6 fatty acid than the non-irradiated sample. Gamma-irradiation resulted in an increase (p<0.05) in phosposerine, aspartic acid and glutamic acid and a decrease (p<0.05) in glutamic acid in free amino acids. For the samples irradiated at 4 kGy, the sulfur odor and fat pungent flavor were significantly higher (p<0.05) and the moldy odor and moldy flavor were significantly lower than the control sample. In conclusion, gamma-irradiation had a significant effect on the texture profiles, fatty acid composition and sensory evaluation of dry-fermented sausage.

      • Borage oil restores acidic skin pH by up-regulating the activity or expression of filaggrin and enzymes involved in epidermal lactate, free fatty acid, and acidic free amino acid metabolism in essential fatty acid-deficient Guinea pigs

        <P><B>Abstract</B></P> <P>Borage oil (BO) reverses a disrupted epidermal lipid barrier and hyperproliferation in essential fatty acid deficiency (EFAD). However, little is known about its effect on skin pH, which is maintained by epidermal lactate, free fatty acids (FFAs), and free amino acids (FAAs) which is generated by lactate dehydrogenase (LDH), secreted phospholipase A2 (sPLA2), or filaggrin degradation with peptidylarginine deiminase-3 (PADI3). We hypothesized that BO restores skin pH by regulating epidermal lactate, FFA metabolism, or FAA metabolism in EFAD. To test this hypothesis, EFAD was induced in guinea pigs by a hydrogenated coconut oil (HCO) diet for 8 weeks, followed by 2 weeks of a BO diet (group HCO + BO). As controls, groups HCO and BO were fed HCO or BO diets for 10 weeks. In group HCO + BO, skin pH, which was less acidic in group HCO, was restored; and epidermal lactate and total FFAs, including palmitate, stearate, linoleate, arachidate, behenate, and lignocerate, were higher than in group HCO. LDH and sPLA2 (mainly the PLA2G2F isoform) activities and protein expressions were similar between groups HCO + BO and BO. Epidermal acidic FAAs, as well as filaggrin and PADI3 protein and mRNA expressions were higher in group HCO + BO than in group HCO. Oleate, total FAAs including other FAAs, and LDH and sPLA2 mRNA expressions were not altered between groups HCO and HCO + BO. Basic FAAs were not altered among groups. Dietary BO restored acidic skin pH and increased epidermal levels of lactate, most FFAs, and acidic FAAs by up-regulating LDH, sPLA2, filaggrin, and PADI3 activities as well as protein or mRNA expressions in EFAD.</P>
