        지황의 건조 방법에 따른 무기성분, 환원당, Catalpol 및 Benzo[α]pyrene의 함량 변화

        This experiment was carried out in order to collect the basic data on the standardization of the manufacturing process of Rehmannia glutinosa Libosch. var. purpurea Makino drying. By the drying methods of R. glutinosa, the content of water, inorganic components, reducing sugar, catalpol and benzo[α]pyrene were investigated. The water content was 15.6~17.2% when R. glutinosa was dried by cold-warm air moisture absorption drying method (CAMAD) at 60℃ during 6 days. Among of the inorganic components of R. glutinosa the K content was the most followed by P, Na, Ca and Mg. The reducing sugar content of R. glutinosa by the hot air drying method (HAD) was much more than that by the CAMAD. The catalpol content of R. glutinosa was not different by the drying temperature when it was dried by the CAMAD. The catalpol content of the large size tuber (about 50.0 g/unit) showed a tendency to increase from 60℃ until 70℃ drying temperature, but that of the small size tuber(about 4.0 g/unit) was decreased as being a trend as the drying temperature high when R. glutinosa was dried by the HAD, But the catalpol content R. glutinosa had a tendency to drop significantly at drying temperature above 80℃. The benzo[α]pyrene content was little detected when R. glutinosa was dried by both the SLD and the CAMAD, and the sampling by the HAD indicated within the scope of 5 μg/kg which was the scope to regulate by Korean food and drug administration. In conclusion, it seemed that an appropriate drying temperature of R. glutinosa by the CAMAD and the HAD was about 60℃ and about 70℃, respectively, when we consider the catalpol content and benzo[α]pyrene detection in the manufacturing process of drying R. glutinosa.

      • Genome-wide identification of 31 cytochrome P450 (<i>CYP</i>) genes in the freshwater rotifer <i>Brachionus calyciflorus</i> and analysis of their benzo[α]pyrene-induced expression patterns

        <P><B>Abstract</B></P> <P>While marine invertebrate cytochrome P450 (CYP) genes and their roles in detoxification mechanisms have been studied, little information is available regarding freshwater rotifer CYPs and their functions. Here, we used genomic sequences and RNA-seq databases to identify 31 <I>CYP</I> genes in the freshwater rotifer <I>Brachionus calyciflorus</I>. The 31 <I>Bc-CYP</I> genes with a few tandem duplications were clustered into CYP 2, 3, 4, mitochondrial, and 46 clans with two marine rotifers <I>Brachionus plicatilis</I> and <I>Brachionus koreanus</I>. To understand the molecular responses of these 31 <I>Bc-CYP</I> genes, we also examined their expression patterns in response to benzo[α]pyrene (B[α]P). Three <I>Bc-CYP</I> genes (<I>Bc-CYP3044B3</I>, <I>Bc-CYP3049B4</I>, <I>Bc-CYP3049B6</I>) were significantly upregulated (<I>P</I> <0.05) in response to B[α]P, suggesting that these <I>CYP</I> genes can be involved in detoxification in response to B[α]P exposure. These genes might be useful as biomarkers of B[α]P exposure in <I>B. calyciflorus</I>. Overall, our findings expand the repertoire of known CYPs and shed light on their potential roles in xenobiotic detoxification in rotifers.</P>

        당귀, 작약, 천궁, 삼백초, 인진쑥 및 산조인을 첨가한 한방차의 팽화 온도별 고형분 용출율과 Benzo[α]pyrene의 변화

        팽화 처리온도를 각각 140~220℃로 한 한방차 제품의 성분변화를 분석한 결과는 다음과 같다. 처리온도의 상승에 따라 일부 탄화가 발생하며 조회분 함량이 상대적으로 상승하는 소폭의 변화가 있었고, 조단백질 및 조지방 함량은 거의 변화가 없는 것으로 나타났으며 수분함량은 감소하였다. 한방차의 고형분 용출률은 0.18~0.27%(w/w)로 나타내었는데, 팽화온도가 상승할수록 증가하였다. 고형분의 용출은 온도가 화학적 변화보다 물리적 변화에 의해 식품의 원재료 성분인 탄수화물, 단백질, 지방 등이 천연 상태에서 상호가교 결합이 물리적인 힘으로 어느 정도 파괴되어 성분의 용출이 용이해지기 때문인 것으로 생각된다. 벤조피렌 함량은 0.18~0.24ppb로 처리온도, 원재료에 따라 B(α)P 함량에 차이가 발생한 것으로 나타났다. The following study is the result of herbal teas puffed at different temperatures between 140~220℃. There was change of single breadth that some carbonization occurs according to rise of processing temperature and crude ash content rises relatively, and crude protein and crude fat content had hardly changed and moisture content decreased. The solid elution rate of the herbal teas appeared by 0.18~0.27% (w/w), it increased as puffing temperature rises. The reason for the increase in solid elution rates is due to the breakage of cross bridges between the raw materials in the herbal tea which are carbohydrates, proteins, lipids and etc. after treatments of physical changes rather than chemical ones. Benzopyrene content happened difference in B(α)P content according to processing temperature, raw material by 0.18~0.24 ppbs.

        팽화홍삼분말이 벤조피렌을 투여한 마우스의 항산화 효소 활성에 미치는 영향

        팽화홍삼분말이 B(α)P의 투여에 의한 간 독성이 유발된 마우스에서의 글루타치온 및 과산화지질 함량, 항산화 효소 활성에 미치는 영향을 살펴보았다. B(α)P 투여에 의해 증가된 혈청 내 ALT와 AST의 활성이 팽화홍삼분말을 투여하면 감소하였다. 간 조직중의 SOD, catalase 그리고 GSH-Px의 활성도 B(α)P 투여로 유의적으로 증가되었다가, 팽화홍삼분말의 투여로 이들 활성이 유의적으로 감소하였고, 반면, 글루타치온 함량과 GST 활성은 B(α)P 단독군에서는 감소되었다가 팽화홍삼분말 투여 시 유의적인 증가를 보였다. 그러나 cytochrome P-450 활성과 지질과산화물 함량은 B(α)P 투여 시 증가되었다가 팽화홍삼분말의 투여 시 유의적으로 감소되었다. 특히 홍삼분말보다 팽화시킨 홍삼분말에서 더 큰 항산화능을 보였다. 이상의 결과로 팽화홍삼분말은 항산화 효소의 활성에 의한 B(α)P에 의한 간 손상에 대한 보호효과를 가지는 것으로 사료된다. In order to determine the effects of puffing red ginseng (PRG) powder on the antioxidant enzyme activities of hepatotoxicity in benzo(α)pyrene[B(α)P]-treated mice, the mice were divided into 5 groups. The dried red ginseng were prepared by puffing conditions of moisture content 10% and puffing pressure 5 kgf/㎠, and then powdered. PRG powder was injected i.p. once a day for 5 successive days, followed by the administration of B(α)P treatment on the fifth day. We also evaluated the relationship between lipid peroxidation and PRG powder on oxidative stress. The increased activities of superoxide dismutase, catalase, and glutathione peroxidase observed following B(α)P-treatment were reduced by the treatment of PRG powder. Whereas the glutathione content and glutathione S-transferase activity depleted by B(α)P were significantly increased, the B(α)P-associated elevation of cytochrome P-450 activities and lipid peroxide content were reduced as the result of PRG powder treatment. Especially, PRG powder had higher antioxidant activities than RG powder. These results suggest that puffing red ginseng powder can protect against B(α)P intoxicification through its antioxidant properties.

        헛개나무 잎 추출물의 벤조피렌 유발 간 독성에 대한 보호효과

        헛개나무 잎 추출물의 간 손상 억제 작용을 확인하고자, B(α)P 투여로 간 독성이 유발된 마우스에서 과산화지질의 생성 및 관련 효소의 활성 변화를 살펴본 결과 B(α)P 투여로 인해 혈청 ALT와 AST의 활성, 간 조직 중의 과산화지질 함량, cytochrome P450 함량, SOD, catalase 그리고 GSH-Px의 활성이 유의적으로 증가하였고(p<0.05), GSH 함량과 GST 활성은 감소하였다. 반면 헛개나무 잎 메탄올 추출물의 전 처리로 인해 ALT와 AST의 활성, 과산화지질 함량, cytochrome P450 함량 그리고 항산화 효소인 SOD, catalase 및 GSH-Px의 활성이 유의적으로 감소하였으며(p<0.05), GSH 함량과 GST 활성은 증가하였다. 이상의 결과로 헛개나무 잎 메탄올 추출물은 생체 내에서 자유기로 인해 야기되는 간장의 산화적 손상을 효과적으로 억제할 수 있을 것으로 사료된다. This study was carried out to investigate the protective effect of Hovenia dulcis Thumb leaves extract on liver damage in benzo(α)pyrene (B(α)P)-treated mice. Hovenia dulcis Thumb leaves methanol extract was intraperitoneally injected once daily for 5 successive days, followed by treatment with B(α)p. The elevated activities of serum aminotransferase and hepatic cytochrome P450 by B(α)p were decreased by pretreatment with Hovenia dulcis Thumb leaves extract. Hovenia dulcis Thumb leaves extract also significantly prevented the elevation of hepatic malondialdehyde content and depletion of glutathione content induced by B(α)P. In addition, the increased activities of superoxide dismutase, catalase and glutathione peroxidase after B(α)P-treatment were decreased. On the other hand, glutathione S-transferase activity was increased by pretreatment with Hovenia dulcis Thumb leaves extract. These results suggest that Hovenia dulcis Thumb leaves extract have a protective effect on liver damage by B(α)P through the mechanisms of decreasing lipid peroxide and activities of free radical generating enzymes.

        Degradation of Benzo[α]pyrene in Roasted Oil Seeds by Corona Discharge Plasma Jet

        Benzo[α]pyrene (BaP), a carcinogenic polycyclic aromatic hydrocarbon, is ubiquitous in nature. It is generally found in heat-treated foods like roasted sesame seeds. BaP degradation has attracted attention due to the recalcitrant nature of BaP. In this study, corona discharge plasma jet (CDPJ) was used to degrade BaP on glass slides and in food materials. The plasma discharges were generated using air as working gas under atmospheric pressure conditions and at different currents (1.00, 1.25, and 1.50 A). Optimal BaP degradation was observed upon using CDPJ generated at 1.50 A current and at 15 mm sample-to-electrode distance (STED). Under these conditions, initial BaP concentration on slides was reduced maximally by 87.09% in 30 min. The degradation kinetics were well-fitted by Weibull tail model compared with others. In food commodities (roasted sesame and perilla seeds), the average levels of BaP degradation ranged between 32.96-45.90% following CDPJ treatment for 30 min.

        로스팅 조건이 삼백초, 인진쑥, 산조인, 당귀, 작약 및 천궁을 첨가한 한방차의 벤조피렌 변화에 미치는 영향

        한방차의 roasting 온도를 80∼140℃ 범위로 하여 성분 변화를 분석한 결과, 온도의 상승에 따라 수분함량이 감소하고 일부 탄화가 발생하며 조 회분 함량이 소폭 상승하였고, 조 지방 및 조단백질 함량은 소폭 감소하였다. 한방차의 고형분 용출율은 0.15~0.32%(w/w)를 나타내었는데, roasting 온도가 상승할수록 감소하는 경향을 나타내었다. 처리온도가 80∼110℃구간에서는 큰 변화를 나타내지 않은 반면 110∼140℃구간에서는 고형분 용출율이 급격히 감소하였다. 온도가 상승할수록 용출율이 감소하는 것은 내부 조직이 치밀하여 상대적으로 용출이 어렵기 때문이다. 벤조피렌 함량은 0.29~0.51ppb으로 온도가 높을수록 B(α)P함량이 증가하였다. 처리온도와 원재료에 따라 B(α)P 함량에 차이가 발생하며, 내부온도는 약 200℃정도지만 roaster 표면의 실제 온도는 약 2000℃에 이르는데 표면과 접촉한 부분에서 B(α)P가 생성된다. B(α)P는 주로 음식을 조리, 가공할 때 식품의 주성분인 탄수화물, 단백질, 지방 등이 열분해 되어 생성되는 것으로 생각된다. The following is the study result of herbal tea roasted at different temperatures between 80∼140℃. Depending on treatment temperature the water content decreased, some carbonization occurred and crude ash content relatively increased. Also crude protein and crude fat decreased little. Solid elution rate of herbal tea showed 0.15~0.32%(w/w) and the rate of solid elution decreased with higher roasting temperature. There was no big change in 80∼110℃ treatment section but the solid elution decreased rapidly in 110∼140℃ section. The reason for decreasing solid elution rate at higher treatment temperature is because the compact inner tissue makes elution difficult. Benzopyrene content (0.29~0.51ppb) showed a tendency to increase with higher treating temperature. From this result, the B(α)P content differed depending on the treatment temperature and raw materials. In case of roasting, the actual inside temperature is around 200℃ but since the surface temperature of the roaster reaches around 2000℃ some portion of B(α)P content was presumed to be produced from the area that came in contact with this surface. When the processing which is a main component of food carbohydrate, protein, fat reason despite serve heat treatment as a whole is to be detected even though the B(α)P in this way is considered to be.
