        Myricetin Inhibits the Release of Glutamate in Rat Cerebrocortical Nerve Terminals

        The excessive release of glutamate is a critical element in the neuropathology of acute and chronic brain disorders. The purpose of the present study was to investigate the effect and possible mechanism of myricetin, a naturally occurring flavonoid with a neuroprotective profile, on endogenous glutamate release in the nerve terminals (synaptosomes) of the rat cerebral cortex. The release of glutamate was evoked by the K+ channel blocker 4-aminopyridine (4-AP) and measured by one-line enzyme-coupled fluorometric assay. We also used a membrane potential-sensitive dye to assay the synaptosomal plasma membrane potential, and a Ca2 + indicator Fura-2 to monitor cytosolic Ca2 + concentrations ([Ca2 + ]C). Results show that myricetin inhibited 4-AP-evoked glutamate release, and this effect was prevented by chelating extracellular Ca2 + ions and the vesicular transporter inhibitor bafilomycin A1. However, the glutamate transporter inhibitor dl-threo-beta-benzyl-oxyaspartate had no effect on myricetin action. Myricetin did not alter the synaptosomal membrane potential, but decreased 4-APinduced increases in the cytosolic free Ca2 + concentration. Furthermore, the myricetin effect on 4-AP-evoked glutamate release was prevented by blocking the Cav2.2 (N-type) and Cav2.1 (P/Q-type) channels, but not by blocking intracellular Ca2 + release. These results suggest that myricetin inhibits glutamate release from cerebrocortical synaptosomes by attenuating voltage-dependent Ca2 + entry. This implies that the inhibition of glutamate release is an important pharmacological activity of myricetin that may play a critical role in the apparent clinical efficacy of this compound.

        Monitoring changes in the genetic structure of Brown Tsaiya duck selected for feeding efficiency by microsatellite markers

        Objective: Few studies have genetically monitored chickens over time, and no research has been conducted on ducks. To ensure the sustainable management of key duck breeds, we used microsatellite markers to monitor Brown Tsaiya ducks over time genetically. Methods: The second, fourth, sixth to eighth generations of the Brown Tsaiya duck selected for feeding efficiency and control lines were included in this study to investigate the genetic variations, effective population size, population structure and the differentiation between populations over time with 11 microsatellite markers derived from Brown Tsaiya duck. Results: The results showed there were a slight decrease in the genetic variations and an increase in within-population inbreeding coefficient (F<sub>IS</sub>) in both lines, but no consistent increase in F<sub>IS</sub> was observed in each line. The effective population size in the second and eighth generations was 27.2 for the selected line and 23.9 for the control line. The change in allele richness showed a downward trend over time, and the selected line was slightly lower than the control line in each generation. The number of private alleles (N<sub>p</sub>) in the selected line were higher than in the control line. Moderate differentiation was observed between the second and eighth generations in the selected line (F<sub>ST</sub> = 0.0510) and the control line (F<sub>ST</sub> = 0.0606). Overall, differentiation tended to increase with each generation, but genetic variation and structure did not change considerably after six generations in the two lines. Conclusion: This study provides a reference for poultry conservation and helps to implement cross-generation genetic monitoring and breeding plans in other duck breeds or lines to promote sustainable management.

      • Multifunctional Natural Polysaccharides as Promising Composites for Emerging Intelligent Packaging

        Traditionally, food packaging is used to enhance food quality and provide consumers with product descriptions. As a new type of food packaging technology, the new generation of intelligent packaging can sensitively sense changes in the environmental atmosphere of stored food, so as to make a visual response to monitor the quality and freshness of food. This work summarizes the latest research on the development, properties and applications of natural polysaccharide-based smart food packaging materials. Firstly, the intelligent packaging system and polysaccharide-based intelligent packaging are summarized, and several representative polysaccharides are introduced. Secondly, different sensing methods that can be used to detect changes in food properties, such as those based on changes in time, temperature, humidity, oxygen levels, pH, chemical composition, or microbial contamination, are described. Then, the application of polysaccharide-based intelligent packaging materials in the food industry is discussed. Finally, the challenges and future directions of polysaccharide-based intelligent food packaging are discussed.

        Changes in prostate-specific antigen kinetics during androgen-deprivation therapy as a predictor of response to abiraterone in chemonaïve patients with metastatic castration-resistant prostate cancer

        Purpose: Metastatic castration-resistant prostate cancer (mCRPC) has a poor prognosis. Abiraterone acetate (AA), enzalutamide, and chemotherapy are first-line treatments for patients with mCRPC. This study examined prognostic factors for AA response in the form of prostate-specific antigen (PSA) kinetics throughout androgen-deprivation therapy (ADT) in chemonaïve patients with mCRPC. Materials and Methods: We retrospectively included data from 34 chemonaïve patients with mCRPC who had received AA at some point between January 2017 and December 2018. We separated patients into two study arms according to the decrease in PSA percentages after use of AA for 3 months. We correlated PSA kinetics parameters with response and compared the two study groups with respect to PSA kinetics. Results: The patients’ median age was 77 years. In the total group of patients, 64% had a response to AA, whereas 35% did not. The ratio of the PSA level at nadir to the level during ADT was significantly higher in the AA-sensitive group (19.78 vs. 1.03, p=0.019). Conclusions: Patients who experienced a dramatic change in PSA level during ADT were more likely to be resistant to AA after progression to mCRPC. Chemotherapy rather than AA might be more suitable as a first-line treatment for these patients.

        Antiviral activity of Rheum palmatum methanol extract and chrysophanol against Japanese encephalitis virus

        Rheum palmatum, Chinese traditional herb, exhibits a great variety of anti-cancer and anti-viruses properties. This study rates antiviral activity of R. palmatum extracts and its components against Japanese encephalitis virus (JEV) in vitro. Methanol extract of R. palmatum contained higher levels of aloe emodin, chrysophanol, rhein, emodin and physcion than water extract. Methanol extract ($IC_{50}=15.04{\mu}g/ml$) exhibited more potent inhibitory effects on JEV plaque reduction than water extract ($IC_{50}=51.41{\mu}g/ml$). Meanwhile, $IC_{50}$ values determined by plaque reduction assay were $15.82{\mu}g/ml$ for chrysophanol and $17.39{\mu}g/ml$ for aloe-emodin, respectively. Virucidal activity of agents correlated with anti-JEV activity, while virucidal $IC_{50}$ values were $7.58{\mu}g/ml$ for methanol extract, $17.36{\mu}g/ml$ for water extract, $0.75{\mu}g/ml$ for chrysophanol and $0.46{\mu}g/ml$ for aloe-emodin, respectively. In addition, $10{\mu}g/ml$ of extract, chrysophanol or aloe emodin caused 90 % inhibition of JEV yields in cells and significantly activated gamma activated sequence-driven promoters. Hence, methanol extract of R. palmatum and chrysophanol with high therapeutic index might be useful for development of antiviral agents against JEV.

        Comparative Analysis of the Genomes of Bombyx mandarina and Bombyx mori Nucleopolyhedroviruses

        The Bombyx mandarina nucleopolyhedrovirus (BomaNPV) S1 strain can infect the silkworm, Bombyx mori,but is significantly less virulent than B. mori nucleopolyhedrovirus (BmNPV) T3 strain. The complete nucleotide sequence of the S1 strain of BomaNPV was determined and compared with the BmNPV T3 strain. The circular, double stranded DNA genome of the S1 strain was 126,770 nucleotides long (GenBank accession no. FJ882854), with a G+C content of 40.23%. The genome contained 133 potential ORFs. Most of the putative proteins were more than 96% identical to homologs in the BmNPV T3 strain, except for bro-a, lef-12,bro-c, and bro-d. Compared with the BmNPV T3 strain, however, this genome did not encode the bro-b and bro-e genes. In addition, hr1 lacked two repeat units, while hr2L, hr2R, hr3, hr4L, hr4R, and hr5 were similar to the corresponding hrs in the T3 strain. The sequence strongly suggested that BomaNPV and BmNPV are variants with each other, and supported the idea that baculovirus strain heterogeneity may often be caused by variation in the hrs and bro genes.

        Development of human IgE biosensor using Sezawa-mode SAW devices

        This paper reports Sezawa-mode surface acoustic wave (SAW) devices with via-isolated cavity to construct the allergy biosensor. To fabricate Sezawa-mode SAW devices, the RF magnetron sputtering method for the growth of piezoelectric ZnO thin films are adopted and influences of the sputtering parameters are investigated. The optimal substrate temperature of 300 C, RF power of 120 W and sputtering pressure of 2 Pa were used to deposit piezoelectric ZnO films with a smooth surface, uniform grain size and strongly c-axis-orientated crystallization. A back-etched SAW resonator is used in this study. The wet etching of (100)-oriented silicon wafers is used to form a back-side cavity which is critical to the formation of a hopper cavity for holding bio-analytes. The remaining membrane structure silicon thickness was 25 mm. In this report, the chrome (Cr, 12 nm)/gold (Au, 66 nm) layer was initially deposited onto the sensing area of SAW devices as the binding layer for biochemical sensor. The resonance frequency of the Sezawa-mode SAW device is 1.497 GHz. The maximum sensitivity of the Sezawa-mode is calculated to be 4.44 106 cm2/g for human immunoglobulin-E (IgE) detection. The stability for human IgE detection is calculated to be 80% and the variation of the stability 3% was obtained after several tests.

      • Fabrication and Applications of Hemicellulose-based Hydrogels

        Hemicellulose-based hydrogel is a three-dimensional networked hydrophilic polymer with excellent water retention, good biocompatibility and mechanical properties, which has attracted much attention in the field of soft materials, especially in the field of hemicellulose-based materials. Herein, recent advances and developments in hemicellulose-based hydrogels were reviewed. The preparation method, formation mechanism and properties of hemicellulose-based hydrogels are introduced from the aspects of chemical cross-linking and physical cross-linking. The differences of different initiation systems such as light, enzymes, microwave radiation, and glow discharge electrolytic plasma are summarized. The latest applications and developments of hemicellulose-based hydrogels in the fields of controlled drug release, wound dressings, high-efficiency adsorption, and sensors are summarized. Finally, the challenges faced in the field of hemicellulose-based hydrogels are summarized and prospected, in order to provide references for the research of new hemicellulose-based hydrogels.

      • A meta-analysis of genome-wide association studies for adiponectin levels in East Asians identifies a novel locus near <i>WDR11-FGFR2</i>

        <P>Blood levels of adiponectin, an adipocyte-secreted protein correlated with metabolic and cardiovascular risks, are highly heritable. Genome-wide association (GWA) studies for adiponectin levels have identified 14 loci harboring variants associated with blood levels of adiponectin. To identify novel adiponectin-associated loci, particularly those of importance in East Asians, we conducted a meta-analysis of GWA studies for adiponectin in 7827 individuals, followed by two stages of replications in 4298 and 5954 additional individuals. We identified a novel adiponectin-associated locus on chromosome 10 near <I>WDR11-FGFR2</I> (<I>P</I> = 3.0 × 10<SUP>−14</SUP>) and provided suggestive evidence for a locus on chromosome 12 near <I>OR8S1-LALBA</I> (<I>P</I> = 1.2 × 10<SUP>−7</SUP>). Of the adiponectin-associated loci previously described, we confirmed the association at <I>CDH13</I> (<I>P</I> = 6.8 × 10<SUP>−165</SUP>), <I>ADIPOQ</I> (<I>P</I> = 1.8 × 10<SUP>−22</SUP>), <I>PEPD</I> (<I>P</I> = 3.6 × 10<SUP>−12</SUP>), <I>CMIP</I> (<I>P</I> = 2.1 × 10<SUP>−10</SUP>), <I>ZNF664</I> (<I>P</I> = 2.3 × 10<SUP>−7</SUP>) and <I>GPR109A</I> (<I>P</I> = 7.4 × 10<SUP>−6</SUP>). Conditional analysis at <I>ADIPOQ</I> revealed a second signal with suggestive evidence of association only after conditioning on the lead SNP (<I>P</I><SUB>initial</SUB> = 0.020; <I>P</I><SUB>conditional</SUB> = 7.0 × 10<SUP>−7</SUP>). We further confirmed the independence of two pairs of closely located loci (<2 Mb) on chromosome 16 at <I>CMIP</I> and <I>CDH13</I>, and on chromosome 12 at <I>GPR109A</I> and <I>ZNF664</I>. In addition, the newly identified signal near <I>WDR11-FGFR2</I> exhibited evidence of association with triglycerides (<I>P</I> = 3.3 × 10<SUP>−4</SUP>), high density lipoprotein cholesterol (HDL-C, <I>P</I> = 4.9 × 10<SUP>−4</SUP>) and body mass index (BMI)-adjusted waist–hip ratio (<I>P</I> = 9.8 × 10<SUP>−3</SUP>). These findings improve our knowledge of the genetic basis of adiponectin variation, demonstrate the shared allelic architecture for adiponectin with lipids and central obesity and motivate further studies of underlying mechanisms.</P>

      • Can Local Long-term Institutional Ownership Alleviate Information Asymmetry in Bank Loan Pricing?

        This paper investigates whether informed ownership alleviates information asymmetry in bank loan pricing. We use local long-term institutional ownership (LLTIO) as a proxy for exogenous ownership that possesses geography-related soft information, and show that it is negatively associated with the spread charged by syndicate lenders. The negative relation between LLTIO and loan spread is salient only when geography-related soft information helps the lender to evaluate the borrower’s creditworthiness and when conflicts of interest between equity and debt holders are unlikely. We show that the mechanisms for the LLTIO effect include the alleviation of both moral hazard and adverse selection problems.
