Effect of Ionizing Radiation on Rat Tissue: Proteomic and Biochemical Analysis

<P>Reactive oxygen species (ROS), generated by ionizing radiation, has been implicated in its effect on living tissues. We confirmed the changes in the oxidative stress markers upon irradiation. We characterized the changes in the proteome profile in rat liver after administering irradiation, and the affected proteins were identified by MALDI-TOF-MS and ESI-MS/MS. The identified proteins represent diverse sets of proteins participating in the cellular metabolism. Our results demonstrated that proteomics analysis is a useful method for characterization of a global proteome change caused by ionizing radiation to unravel the molecular mechanisms involved in the cellular responses to ionizing radiation.</P>

악성코드에 대한 보다 바람직한 예방은 클라이언트측이 아닌 서버측에서 이루어지는 것이다. 본 논문에서는 새로운 개념 의 서버용 안티바이러스 엔진인 SAVE 1.0을 설계 구현하여 제시한다. SAVE는 기본적으로 1개의 AV모니터와 다수의 AV에이전트로 구성되어 있는데 다중 CPU를 장착한 병렬처리서버의 특징을 직접 활용하기 위하여 관리자가 다중쓰레드 기법을 직접 지정할 수 있도록 하고 있다 악성코드를 5가지 부류로 구분하여 바이러스 시그너쳐 DB를 구축 제공함으로써 부류별 진단을 지정할 수 있도록 하고 있으며 실시간 네트워크 패킷 검사를 통한 악성코드 유입 점검 기능도 제공한다. 웹 기반의 관리자 인터페이스는 뛰어난 사용자 편의성을 제공하고 있다. 성능실험결과 악성코드 검색율은 국내외 상용제품과 비교해서 매우 우수한 것으로 나타났으며 검색속도에 있어서도 서버의 CPU 수 증가에 거의 선형 비례하여 향상되는 특징을 보이고 있다. It is more desirable to prevent and detect the malicious codes in server system rather than in client PCs. In this paper, we suggest a new anti-virus engine, SAVE 1.0, which is executed on a parallel processing server. SAVE consists of 1 AV monitor and several AV agents. A system administrator can control the degree of multi-thread directly in order to enhance the utilization of parallel processing servers. Virus signature DBs in SAVE are classified into 5 categories, which support system administrator's options to use the classifier or not. By real-time checking of network packets, the influx of malicious codes can be prevented. Web-based GUI is one of the convenient functions provided in SAVE. In performance tests, we have confirmed that SAVE has higher virus detection ratio than any other AV engine, and SAVE has a desirable feature as an software of parallel processing servers since the performance of SAVE is improved almost linearly as the CPUs in a parallel processing server increases.

Various cytokines including IL-1β, IL-2, IL-4, IL-10, IL-12p40, IFN-γ, and TNF-α have been recently reported to play important roles in both host and donor cell activations during xenotransplantation. Interleukin (IL)-18, a proinflammatory cytokine, can induce IL-4 and IL-13 production in T cells, NK cells, B cells, mast cells, and basophils. IL-18 has the capacity to stimulate innate immunity and both Th1-and Th2- mediated responses, but little is known to involve in the xenograft rejection. In order to investigate the role of IL-18 in xenograft rejection, we transplanted the pig kidney (PK15) cells to C57BL/6 mouse with or without intraperitoneal injection of mouse recombinatant IL-18. It was analyzed the population of T cell, B cell and NK cell in the mice transplanted with PK 15 cells and recombinant IL-18 by flow cytometry. We found that splenic CD3+ T cells were increased in mice injected PK15 cells with recombinant IL-18. These results suggest that recombinant IL-18 is critical in the xenograft rejection by increasing T cell population of the recipients.

생쥐에서 치주조직 형성 과정에 따른 치주인대 세포-특이 PDLs17 단백질 발현 변화

PDLs17, PDL-specific gone, had previously identified the cDNA for a novel protein from cultured PDL fibroblasts using subtraction hybridization between gingival fibroblasts and PDL fibroblasts. The purpose of this study was to observe the immunohistochemical localization of PDLs17 on the developing periodontium of mice. Developing mice (post-natal mice; P4, P10, P15) were recovered and decapitated, and the heads were fixed overnight at in a freshly prepared solution of 4% paraformaldehyde. Some specimens were decalcified for 2-4 weeks in a solution containing 10% of the disodium salt of ethylenediamine-tetraacetic acid (EDTA). Next, tissues were dehydrated, embedded in paraffin and sectioned serially at 6㎛ in thickness. Polyclonal antiserum raised against PDLs17 peptides. SEAVHETDLHDGC, were made, and immunostained the tooth, periodontium, developing bone and bone marrow of developing mice. The results were as follows. 1. Immunohistochemical analysis showed that PDLs17 is Preferentially expressed in the developing tooth follicle, but not in tooth pulp and overlying gingival mucosa of 4-day-old developing mouse. PDLs17 also expressed in preosteoblast-likec ells in the developing alveolar bone of 4-day-old mouse. 2. PDLs17 expressions were observed in developing tooth follicle, tooth pulp and alvolar bone of 10-day-old developing mouse. 3. PDLs17 protein were observed in PDL cells, tooth pulp and surrounding alveolar bone of 15-day-old tooth-eloping mouse. However, there were no PDLs17 expression in ameloblasts and odontoblasts. These results suggested that PDLs17 might expressed at the initial stage of differentiation when the undifferentiated mesenchymal cells in the bone marrow and PDL, differentiatiation from tooth follicle. However, more research needs to be performed to gain a better understanding of the exact function of PDLs17 during the differentiation of tooth pulp cells and PDL cells.

The ameloblastic fibrosarcoma is the malignant counterpart of the ameloblastic fibrosarcoma in which the mesenchymal element has become malignant. Clinically it frequently occurs in the 3rd and 4th decades, and more frequent in the mandible than in the maxilla. Radiographic features are apparent multiocular radiolucency with ill-defined border. The authors experienced two cases of ameloblastic fibrosarcoma of the mandible in a 26-year-old male and a 48-year-old female patients who suffered from pain and swelling on the affected area. And we discussed the clinical, radiological and histopathological features of this disease with a brief review of the literatures.
