      • 151종 생약제 추출물이 B형 간염 바이러스 표면 항원(HBsAg)과 항체(HBsAb)의 결합반응 그리고 HBV DNA polymerase 활성에 미치는 효과 : 한국산 각종 생약제를 이용한 B형 간염치료제의 개발

        세계인구 43억명 중 B형 간염 바이러스 보균자는 2억 1천 5백만명이며 이중 78%인 1억 6천만명 이상이 아시아 대륙에 집중되어 있다. 한국도 B형 바이러스 간염의 침윤지대이다. 본 연구는 B형 바이러스 보균자 혹은 만성간염을 치료할 수 있는 생약제를 탐색할 목적의 일환으로 한국에서 시판하는 생약제 151종을 구입하여 증류수로 추출하고 여과한 후 냉동건조하여 HBs 항원에 대한 결합능, HBV DNA polymerase억제능, tumor necrosis factor(TNF) 유리에 미치는 영향, 그리고 galactosamine 유발 간염에 대한간장보호 작용을 조사하였다. 151종의 생약제중 0.16㎍/㎖ 농도에서 간염 vaccine 내에 함유된 HBs 항원과 결합하는 생약제는 지유, 가자, 복분자, 대황, 정향, 빈량, 산수유, 소목, 마황, 쇄양, 후박, 목과, 양강, 오약, 산사, 목단피 등 16종이었다. 이들 생약제중 대황(Rheum palmatum L. 大黃), 가자(Terminalia chebula R. 訶子), 지유(Sanguisorba officinalis L. 地楡), 복분자(Rubus coreanus M. 覆盆子)는 HBV DNA polymerase 활성도를 비교적 강하게 억제하였으며 빈량(Areca catechu L. 빈랑), 정향(Eugenia caryophyllata T. 丁香), 목과(Chaenomeles japonica L. 木瓜), 산수유(Cornus officinalis S. 山茱萸)는 약하게나마 HBV DNA polymerase 활성을 억제하였다. 또한 HBV DNA polymerase를 억제한 대황, 가자, 지유, 복분자, 빈량, 정향, 마황, 소목, 산수유를 대상으로 TNF 유리에 미치는 영향을 조사한 결과 소목을 제외한 8종의 생약제 모두가 대식세포를 자극하여 TNF를 유리시켰다. 그리고 장차 임상에 사용함을 전제로 HBV DNA polymerase를 억제한 대황, 가자, 지유, 복분자, 빈량, 정향, 마황, 소목, 산수유를 galactosamine 유발 간염(백서)에 투여하여 간기능 검사에 미치는 효과를 조사하였으나 간기능 검사상으로는 특별한 보호작용을 나타내지 못하였으며 생약제 단독 투여로서는 간기능에 대하여 나쁜 영향을 주지 않았다. Hepatitis B virus infection is major cause of acute and chrome hepatitis, cirrhosis of liver, and hepatocellular carcinoma. There are over 200 million carriers of hepatitis B virus in the world, nearly 80 % of whom live in the South East Asia, Korea is also highly endemic area of hepatitis B virus infection. This study was performed to evaluate the effect of extracts of traditional Korean herbal medicines on hepatitis B virus. 151 traditional herbs were extracted, filtered and lyophilized. Using 151 herbal extracts we tested binding capability to the HBs Ag, inhibition of HBV DNA polymerase, release of tumor necrosis factor (TNF), and protection against galactosamine induced liver damage. Among 151 herbs, 16 herbs (Sanguisorba officinalis L., Terminalia chebula R., rubus coreanus M., Rheum palmatum L., Eugenia caryophyllata T., Areca catechu L., Cornus officinalis S., Caesalpinia sappan L, Ephedra sinica S., Cynomorium songaricum R., Machilus thunbergii S., Chaenomeles japonica L., Alpinia officinarum H., Lindera strychnifolia V., Crataegus pinnatifida B., Paeonia suffruticasa A. ) had binding capability to HBs Ag which contained in the commercial hepatitis vaccine, at 16 ug/㎖ of concentratioa Among the above 16 herbs, Rheum palmatum L., Terminalia chebula R., Sanguisorba officinalis L., Rubus coreanus M. significantly inhibited the HBV DNA polymerase activity, and Areca catechu L., Eugenia caryophyllata T., Ephedra sinica S., Caesalpinia sappan L., Chaenomeles Japonica L., Cornus officinalis S. also inibited the HBV DNA polymerase activity but less effective. All of the herbs, except Caesalpinia sappan L., which inhibited the HBV DNA polymerase activity, actively stimulated the mononuclear cells to release tumor necrosis factor(TNF) in whole blood culture system. The herbs which inhibited the HBV DNA polymerase activity, did not show any protective effect in galactosamine-induced hepatitis in rats, but there was no direct toxic effect when herbs were administered alone.

      • 야생 노랑제비꽃(Viola orientalis W.Becker)의 형태적 특성 및 Callus유도에 관하여

        부산근교의 고산 지대에 자생하는 야생 노랑제비꽃(Viola orientalis W. Becker)의 외부 형태적 특성과 줄기 조직으로부터 callus형성에 미치는 식물 생장조절제의 영양에 관한 실험을 실시하였던 바 그 결과를 요약하면 다음과 같다. 잎은 근생엽과 줄기상의 잎으로 구별할 수 있었으며, 엽장, 엽폭, 그리고 엽면적 각각 3.9㎝, 4.7㎝ and 15㎠로 모두 근생엽이 skjawl 줄기상의 잎보다 컸다. 그러나 엽형지수에 있어서는 근생엽이 0.83으로 가장 작았고 줄기상의 가장 작은 잎이 1.30으로 제일 크게 나타났다. 식물 생장조절물질의 단용에서 callus 유도는 Auxin의 NAA5×10??, 5×10??, 5×10??M에서 양호하였다. Auxin과 Cytokinin의 혼용에서 callus유도는 NAA 5×10??M 및 5×10??M에 BA 5×10??, 5×10??, 5×10??M의 전구에서 양호하였다. Callus 성장이 가장 양호한 조합은 NAA 5×10??M + BA 5×10??M 및 NAA 5×10??M + BA 5×10??M이었다. This experiment was conducted to study morphological feature and effect of plant growth regulators on callus induction of wild viola(Viola orientalis W. Becker). The results obtained were summarized as follows: In morphological feature, there was two distinctive leaf-forming types : one was rosette type derived from subterranean stem and the other was leaves formed on stem. Generally rosette type leaves were larger than others and the leaf length, leaf width, and leaf area were 3.9㎝, 4.7㎝ and 15㎠ on average, respectively. In the leaf shape index(the ratio of leaf length/width), the smallest value(0.83) was shown on rosette leaf while the greatest value(1.30) was determined from leaves on stems. Callus induction from the stem segments of wild viola was relatively higher at NAA concentrations of 5x10??M, 5x10??M or 5x10??M than at the same ones of other regulators used. The most effective combined concentrations of Auxin and Cytokinin for the callus formation were 5×10??M of NAA plus 5×10??M of BA or 5×10??M of NAA plus 5×10??M of BA.

        진동시험에 의한 참외의 진동특성

        During a long journey of agricultural products from the production area to markets, the quality of agricultural products was always affected by some degree of vibration. The vibration input during the transportation may cause serious agricultural product injury, and this damage is particularly severe whenever the vegetable inside package is free to bounce, and is vibrated at its resonant frequency. The objectives of this study were to determine the resonant frequency of the oriental melon and to investigate the relationships between resonant frequency and physical properties of the oriental melon such as mass, volume and major and minor axes. In this study vibration testing device was constructed to determine the vibration response of the oriental melon in frequency ranges of 5 to 150 Hz. The computer program for controlling the vibration shaker and the function generator and measuring the vibration characteristics of the oriental melon was developed. The ranges of resonant frequency and peak acceleration at resonance of the oriental melon were 51 to 73 Hz and 1.24 to 1.92 G-rms, respectively. The resonant frequency and the peak acceleration decreased with the increase of the sample mass, volume, major and minor axes of the oriental melon. Multiple regression models for resonant frequency and peak acceleration of the oriental melon as a function of mass, major axis and minor axis of the sample were developed and analyzed.

        플라즈마 산화 처리된 65/35황동의 산소 분압별 부식특성에 관한 연구

        Brasses are the most widely used copper-based alloys because of the good corrosion resistance, formability, high strength and excellent hot-working properties. However, dezincification occurs when one or more components of an alloy are more susceptible to corrosion than the rest and, as a result, are preferentially dissolved. In order to enhance the corrosion resistance, the oxide film was deposited on the 65/35 brass by pulsed dc plasma oxidation with various pressure. The corrosion behavior of plasma oxidized brasses with various oxygen pressure was investigated by electrochemical methods (potentiodynamic polarization test, potentiostatic test, electrochemical impedance spectroscopy) and surface analyses (x-ray diffraction, scanning electron microscopy, auger electron spectroscopy, electron probe micro analyzer). Particular attention was paid to the effects of oxygen pressure and oxide thickness on the coating properties related to the corrosion behavior. From the results of electrochemical test, it was found that the oxidized brass under condition of 1.5 Torr oxygen pressure was effective in improving corrosion resistance. It was attributed to the protective oxidized layer (ZnO). Finally, an optimized oxidation condition for corrosion protection was found to be 1.5 Torr oxygen pressure and 1 pun thickness.         Lymphangioma is an abnormal collection of congenitally isolated lymphatic system, which is found mostly in early childhood. Although this may occur in any part of the body mainly including the head, neck, axilla and abdomen, there are rare reports about mediastinal lymphangioma especially in old age. We hereby report a case of solitary mediastinal lymphangioma con. rmed pathologically in a 66-year-old woman. A 66-year-old woman who underwent a kidney transplant surgery 20 years ago was referred to pulmonary division because of a newly developed right upper mediastinal nodule on simple chest X-ray, which was not shown one year ago. She had no symptoms such as cough, dyspnea and chest pain. She was a never smoker. Chest computed tomography (CT) scan demonstrated about a 25mm well-de. ned low density nodule at right upper anterior mediastinum. Initial impression of CT scan was a benign cystic mass, such as bronchogenic cyst, or cystic change of neurogenic tumor. We observe her for 1 year. Because size of the nodule increased from 25 to 34mm in follow-up chest CT scan compared with previous CT scan. Therefore anterior mediastinal tumor was completely resected by video-assisted thoracosopy. Its immunohistochemical stain revealed positive for D2-40 and CD31, which supported the histological diagnosis of cystic lymphangioma. We report the 3rd case of solitary mediastinal cystic lymphangioma in Korea as it is an extremely rare lesion in old age. This can contribute to literature due to its rarity and unusual location. And we recommend that clinicians should consider cystic lymphangioma as a possible diagnosis for the patients with increasing mediastinal mass asymptomatically.
