The oral micro-organism Streptococcus mutans synthesizes extracellular glucosyltransferase(GTFs) which polymerize the glucose moiety of sucrose into glucan polymers. These glucan was known to cause dental caries. The study was set up to elucidate the chemical properties of polysaccharides obtained from the cell wall(CWP) and extracellular polysaccharides(ECP) of S. mutans KCTC 3065 polysaccharides. The recovery rate(%) of extracellular and cell wall polysaccharides were purified of 0.032% and 0.13% of total polysaccharide(dry weight), respectively. CWP fraction was determined to be approximately average 70 kilodalton by gel filtration, contains 62.2% of D-glucose and 38.9% of galactose. Whereas, ECP appears approximately average 60kD by gel filtration, and contains exclusively α-D-glucose, fructose and mannose.
영지의 ${\beta}$-glucan성 다당류에 의해 활성화된 흰쥐 간내 Kupffer 세포의 NO, TNF-${\alpha}$ 및 TGF-${\beta}$ 형성

Ganoderan (GAN), an immunomodulating ${\beta}$-glucan from mushroom Ganoderma lucidum, was evaluated for its ability to induce formation of nitric oxide (NO), tumor necrosis factor-${\alpha}$(TNF-${\alpha}$) and transforming growth factor (TGF-${\beta}$) from rat Kupffer cell in vitro. Hepatic macrophages activated by GAN significantly elevated concentration of NO and TNF-${\alpha}$ in cultured medium, but not significantly elevated that of TGF-${\beta}$. GAN-activated Kupffer cells secrete 14.9${\mu}$M (p<0.01) of NO and 2619.5${\rho}$g/ml (p<0.01) of TNF-${\alpha}$after 36hr of incubation at 37$^{\circ}C$. The results revealed that GAN enhanced 4-fold production of NO and 19 fold formation of TNF-${\alpha}$ compared to the control. The proliferation of GAN-activated Kupffer cells was inhibited as compared with its negative control. Comparing the activity among glucans derived from microorganisms, highly branched zymosan, glucomannan from Saccharomyces cerevisiae, significantly increased TNF-${\alpha}$ and NO production. These results indicate that the ${\beta}$-glucan from G. lucidum activates rat Kupffer cell and secretes NO and TNF-${\alpha}$. It also suggest that rat Kupffer cell posses certain receptor for ${\beta}$-anomeric glucan.

The dietary fiber(GLD-06), β-glucanic polysaccharide, from the mycelium of Ganoderma lucidum was fractionated by the sequential use of salts and hot extraction and obtained 0.35% of yield from the mycelial culture medium.

Antitumor polysaccharides were known to activate complement system and incease pecific serum proteins in mouse, and some researcher reported that antitumor activity of polysaccharides might be correlated and macrophage. The b-glucan of G. ucidum induces potent antitumor evaluate the bioactivities of extracellular polysaccharide obtaioned from the mycelia of G. lucidum. The water-insoluble rome (EXO-WI) showed the highest antitumor activity(92.1% inhibition rate) in carcoma- bearing mice, whereas, have relatively little activites at anticomplementary activity. In examination of macrophage activation, Raw 264.7 cells activated with EXO-WI incrased 3.5-fold(24 hrs) formation of nitric oxide levels and about 4-folds production of TNF – a compared with a control with medium only. EXO-WI was composed with exclusively b-D-glucose and 14.7% protein moiety. These results indicate that the EXO-WI purified from myelia of G. lucidum may be of a useful immunopotentiator in the treatment of cancer and immune disease.

The extract of Basidiomycetes induces potent antitumor immunity in tumor-bearing mice. The study was set up to elucidate the anti-cancer effects of various mushroom extract against human cancer cell lines. The mushroom extract was prepared from the fruit body of many types of Basidiomycetes: Poria cocos, Ganoderma lucidum, Ganoderma oregonence. Ganoderma applanatum, Ganoderma tusgae. Fomitella fraxinea. Laetiporus sulphureus, Phellinus lintenus, Agaricus brazei. Most extracts showed cytotoxicity on human cancer cell line: hepatocytoma (SNU 387), human colon carcinoma (SNU C4)and human gastric adenocarcinoma (SNU-1). The highest anticancer effect(83.1% cytotoxicity) of tested samples occurs with Ganoderma applanatum extract on human hepatocytoma SNU 387 cell line. In conclusion, the low molecular extract(<10,000 M. W) of Basidiomycetes hot extract was higher anticancer effect than that the high molecular()10.000 M. W) extract on A549, SNU-1, SNU C4. and SNU 387 human cancer cell line.
