        Genome sequence of carotenoid producing Sphingobacteriaceae bacterium SH-48 isolated from freshwater in Korea

        그람 음성이며 막대모양의 Sphingobacteriaceae bacterium SH-48은 삼척 소한천에서 분리하였다. SH-48에 대한 유전체 분석을 실시하였으며, G + C 비율이 38.4%인 5,650,162 bp 크기의 염기서열을 얻었다. 유전체 특징은 카로티노이드 생합성 유전자인 crt 유전자 클러스터를 보유하고 있어 균주의 잠재적 중요성을 보여준다. 이러한 유전체 정보는 카로티노이드 생합성 경로에 대한 새로운 정보를 제공한다. We sequenced the genome of the Sphingobacteriaceae bacterium SH-48 isolated from the Sohan stream in Republic of Korea by using a dilution-to-extinction culturing method. The sequences were assembled into a draft genome containing 5,650,162 bp with a G + C content of 35.4% and 4,856 protein-coding genes in 2 contigs. This strain contains the carotenoid biosynthesis genes crtY, crtZ, crtD, crtI, crtB, and crtH as gene clusters. This genomic information provides new insights into the carotenoid biosynthesis pathway.

        Complete genome sequence of Betaproteobacteria strain GR16-43 isolated form a freshwater pond in South Korea

        그람 음성이며 긴 막대 모양의 betaproteobacteria에 속하는 GR16-43을 한강 발원지 검룡소에서 분리하였다. GR16-43 균주에 대한 유전체분석을 실시하였으며, G + C 비율이 67.12%인 4,806,848 bp 크기의 염기서열을 얻었다. 유전체 특징은 황산화와 관련된 다량의 유전자를 보유하고 있어 균주의 잠재적 중요성을 보여준다. 이러한 결과는 GR16-43 균주가 빈영양 담수 환경에서의 적응 연구를 위한 유전체 정보를 제공한다. A betaproteobacterium strain GR16-43 was isolated from a surface layer of the Geomnyong Pond in Republic of Korea by a dilution-to-extinction culturing method. We report the whole genome sequence of the strain GR16-43, which contains 4,806,848 bp with a G + C content 67.12%, and to include 4,424 protein-coding genes and 47 transfer RNA genes. The genome was determined to contain the genes encoding carbon monoxide dehydrogenase, nitrate reductase, nitrite reductase, nitric oxide reductase, and the sulfur oxidation (sox) gene cluster, highlighting the potential importance of the bacterial group represented by the strain in the cycling of inorganic elements. These results indicate that strain GR16-43 genome showed several traits indicating adaptation of the bacteria to living in freshwater environments.

        미생물활성에 의한 시멘테이션 작용을 이용한 모래지반의 안정화

        To evaluate bio-cementation of microbial on sands, laboratory test was conducted using acrylic cubic molding boxes ($5cm{\times}5cm{\times}5cm$). It was incubated the microbial, called Bacillus Pasteurii, according to Park et al (2011, 2012). and applied 50ml each specimen. Two type of sand samples used were Jumoonjin sand and common sand (well graded). These sands were molded in acrylic boxes with the relative density of 30 % and 60 % respectively. Microbial were poured onto the samples molded in acrylic boxes and cured at the room temperature and humidity. After 7, 14 and 21days, it was measured the compressive strength, pH, EC, and density and it were observed SEM and XRD to verify the effect of bio-cementation. It was found that bio-cementation was increased a strength of sands and it was appeared that strengths were related to the type of sand and relative density. Therefore it was confirmed the solidification of sands using the bio-cementation by microbial activation and the usefullness of acrylic molding boxes when tests were conducted on the soil of sands.

        이하선 심층엽에 발생한 다형선종의 보존적 심층엽 이하선 절제술: 증례보고

        Parotid deep lobe tumors usually has been treated by total parotidectomy. But there is functional and aesthetic side effects such as post parotidectomy depressions, variable aesthetic deformities, facial nerve injury and Frey's syndrome. Conservative limited deep parotidectomy may result in fewer side effect. Preservation of the superficial lobe for deep lobe tumors could decrease the incidence of complications without any problems in the treatment effect. Additionally, the parotid function preservation and cosmetic appearance after operation also satisfy both the patients and surgeons. We report a case of pleomorphic adenoma of the deep lobe which has been successfully treated by conservative deep parotidectomy.

        항균활성 보유 Penicillium rubefaciens NNIBRFG5039의 최적배양 조건

        본 연구에서는 methicillin resistant Staphylococcus aureus subsp. aureus (MRSA) KCCM 40510 및 Bacillus cereus KCTC 3624 균주에 대한 항균활성을 보유한 균류Penicillium rubefaciens NNIBRFG5039를 경북 상주시 도남동 공기 중으로부터 분리‧동정하였고, 배양조건에 따른 균사체 생육 및 항균활성을 비교하였다. 그 결과, P. rubefaciens NNIBRFG5039는 PDB 배지, 배양 온도 30℃, 초기 pH 6.5로 배양하였을 때 항균활성이 가장 높게 나타나는 것을 확인하였다. 최적조건하에서 5L fermenter를 이용하여 배양시간에 따른 균사체 건조중량 및 항균활성을 비교한 결과, 배양 5일째에 생장 및 항균활성이 가장 높았다. P. rubefaciens NNIBRFG5039의 배양여액의 항균활성 스펙트럼을 조사한 결과, methicillin-resistant Staphylococcus aureus subsp. aureus CCARM 3089·3090·3091·3095와 KCCM 40510, Bacillus cereus KCTC 3624, B.subtilis KACC 10111, Filobasidium neoformans KCTC 7902, Enterococcus faecalis KCCM 11814에 대해서도 항균활성을 가지는 것을 확인하였다. 또한, P. rubefaciens NNIBRFG5039의 배양여액으로부터 항균활성 후보물질을 각종 크로마토그라피법으로 순수분리하고, NMR과 ESI-MS 등의 기기분석을 실시하여 구조를 (S)-6-hydroxymellein (1)으로 동정하였다. In screening for antimicrobial activity against methicillin-resistant Staphylococcus aureus (MRSA) KCCM 40510 and Bacillus cereus KCTC 3624, NNIBRFG5039 was isolated from the air in Sangju-si, Gyeongsangbuk-do. Based on a high sequence similarity of the internal transcribed spacer (ITS) region, NNIBRFG5039 was determined to be closely related to Penicillium rubefaciens CBS 139145. The optimal media, initial pH, and temperature for mycelial growth and antimicrobial activity of P. rubefaciens NNIBRFG5039 were determined as follows: potato dextrose broth (PDB), pH 6.5, and 30°C, respectively. Under the optimal culture conditions, maximum mycelial growth (12.4 g L-1) and antibacterial activity (7.5 mm zone of inhibition against MRSA KCCM 40510, and 5.0 mm zone of inhibition against B. cereus KCTC 3624) were observed in a 5 L stirred-tank fermenter. We also isolated the antimicrobial compound from an ethyl acetate fraction, and its chemical structure was identified as (S)-6-hydroxymellein (1) by ESI-MS, 1H-NMR, and 13C-NMR. Consequently, the extract from P. rubefaciens NNIBRFG5039 may be used in functional materials for antimicrobial-related applications.

        가토의 하악관절에 Collagenase 주입을 통한 관절염 유발 모델에 관한 연구

        Purpose: The purpose of this study is to induce artificial arthritis on rabbit TMJ by injecting collagenase. Materials and Methods: An experimental animal model of arthritis induced by surgical method or intraarticular injection of chemical agent like LDH, papain, ketorolac. Surgical method is complex and needs a long time in inducing arthritis. Intra-articular injection of chemical agent like LDH, papain, ketorolac is simple. But chemical agent like LDH, papain, ketololac needs multiple injections to induce arthritis and mechanism inducing arthritis was known. Collagenase destroys helical domain of type II collagen in extracellular matrix produced by chondrocyte and then induces arthritis. We injected collagenase (0.5, 1.0, 2.0 mg) into the temporomandibular joint of rabbit. In the control group saline was intra-articularly injected. The condylar cartilage, disk and synovia were histologically examined at 1, 2, 4, 6 weeks after the initiation of collagenase injections. Results: Four weeks after injection of 2.0 mg collagenase, we could see histologic change like arthritis. In other groups, we couldn't see arthritis-like change. Conclusion: In our study, we produce arthritis on temporomandibular joint of rabbit by using injection of collagenase in temporomandibular joint of rabbit. And this experimental osteoarthritis is a useful animal model.

        Burkholderia sp. OS17의 항균활성 증진을 위한 배양최적화

        본 연구는 담수환경에서 항균활성을 보유한 미생물을 발굴하고, 활성 증진을 위해 배양조건을 최적화하는 것이다. 상주시 중동면 오상저수지에서 시료를 채취하여 38종의 미생물을 순수분리하였다. 16S rRNA 염기서열 분석에 근거하여 Proteobacteria강(22종), Actinobacteria강(7종), Bacteroidetes강(6종), Firmicutes강(3종)으로 구성되어있는 것을 확인하였다. 메티실린내성 황색포도상구군 등 10종의 유해미생물에 대한 항균활성을 보유한 Burkholderia sp. OS17 균주를 선발하였다. 항균활성 증진을 위한 상용배지, 온도, 초기 pH별 생육 및 항균활성 비교실험을 수행하였다. OS17 균주는 YPD 배지, $35^{\circ}C$, pH 6.5로 배양했을 때 가장 활성이 높았다. LB, NB, TSB, R2A 배지와 $20^{\circ}C$, $25^{\circ}C$ 배양했을 때는 생장은 가능하나 항균활성이 전혀 없었다. 이전결과를 바탕으로 YPD 배지, $35^{\circ}C$에서 배양하면서 5 L fermenter를 이용하여 생육, 항균활성, pH 확인을 통해 배양 48시간을 최적 배양시간으로 선정하였다. 항균활성을 보유한 미생물의 배양 최적화는 항균물질 생산에 영향을 미치고, 이는 상업적 응용에 이점으로 작용할 수 있다. In this study, we isolated and identified bacteria from freshwater and soil collected from Osang reservoir, to screen antimicrobial bacteria against various pathogenic bacteria. 38 strains were isolated and assigned to the class Proteobacteria (22 strains), Actinobacteria (7 strains), Bacteroidets (6 strains), and Firmicutes (3 strains) based on 16S rRNA gene sequence analysis. Among them, strain OS17 showed a good growth inhibition against 5 methicillin-resistant Staphylococcus aureus subsp. aureus strains and Bacillus cereus, Bacillus subtilis, Filobasidium neoformans. As a result of the 16S rRNA gene sequence analysis, strain OS17 show the high similarity with Burkholderia ambifaria $AMMD^T$, B. diffusa $AM747629^T$, B. tettitorii $LK023503^T$ 99.8%, 99.7%, 99.6%, respectively. We investigated cell growth and antimicrobial activity according to commercial culture medium, temperature, pH for culture optimization of strain OS17. Optimal conditions for growth and antimicrobial activity in strain OS17 were found to be: YPD medium, $35^{\circ}C$ and pH 6.5. When the strain was cultured in LB, NB, TSB, R2A media at $20^{\circ}C$ and $25^{\circ}C$, the antimicrobial activity did not show. Culture filtrate of strain OS17 showed antimicrobial activity against 5 MRSA strains, Bacillus cereus, Bacillus subtilis, and Filobasidium neoformans with inhibition zones from 2 to 8 mm. Optimal reaction time was 48 h in YPD medium, 100 rpm and 0.3 vvm in 2 L-scale fed-batch fermentation process for antimicrobial activity. Culture optimization of strain OS17 can be improved on antimicrobial activity. Therefore, the antimicrobial activity of Burkholderia sp. OS17 had potential as antibiotics for pathogens including MRSA.

        Pantoea sp. S3W-11의 항균활성과 배양 최적화

        In this study, 46 bacteria strains were isolated from water and soil collected from Geumjin-ri, Ganggu-ri and Namseock-ri in Yeongdeok-Gun to screen antibacterial bacteria against various pathogenic bacteria. Among them, strain S3W-11 showed an excellent growth inhibition against methicillin-resistant Staphylococcus aureus, Bacillus cereus and Escherichia coli. As a result of the 16S rRNA gene sequence analysis, strain S3W-11 showed the high similarity with Pantoea vagans LMG 24199<SUP>T</SUP>, Pantoea agglomerans DSM 3493<SUP>T</SUP>, Pantoea conspicua LMG 24534<SUP>T</SUP> 99.64%, 99.58%, 99.18%, respectively. The effects of culture medium composition, temperature, and initial pH on the cell growth and antibacterial activity were confirmed for culture optimization of strain S3W-11. Optimal conditions for growth and antibacterial activity in strain S3W-11 were found to be: YPD medium, 25℃ and pH 6.5. When the strain was cultured in LB, NB, TSB, and R2A media, the antibacterial activity has not been confirmed. Optimal reaction time was 48 h in YPD medium, 100 rpm and 0.3 vvm in 2 L-scale fed-batch fermentation process for antimicrobial activity. Strain S3W-11 can be improved on antibacterial activity in optimal culture conditions (YPD medium, 25℃ and pH 6.5). Therefore, the antibacterial activity of Pantoea sp. S3W-11 had potential as antibiotics for pathogens including MRSA.

      • 미생물활성에 의한 모래의 안정화공법 연구

        연약지반 및 사면 안정에 대한 강도증가를 위해 사용하는 시멘트로 인하여 경화도의 높은 증가를 기대할 수 있다. 하지만 시멘트를 사용함으로 인하여 시멘트에 함유된 유독물질인 Cr6+으로 인한 토양의 2차 오염 확산과, 경화도 효과에 따른 시멘트의 소비량 및 시멘트 생산 시 발생하는 이산화탄소 배출량이 가장 많은 것으로 지적받고 있어 시멘트사용의 저감과 이산화탄소의 발생을 줄이는 기술개발이 필요하다. 따라서 본 연구에서는 지중에 존재하는 미생물의 생체광물형성작용으로 생성된 탄산칼슘으로 지반의 고결화(solidification)작용을 일으켜 시멘트의 사용을 대체하면서, 성토나 절토사면의 친환경적 보강은 물론이고 전국적으로 15,000여개소가 존재하는 것으로 알려진 농업용 흙댐의 유지보수 등에 적용하고자 하는 것을 목적으로 하고있다. 이에 본 연구에서는 생체광물형성작용으로 탄산칼슘을 생성시키는 미생물인 Bacillus pasteurii균을 이용하여 미생물의 대사활동에 의해 생성된 광물질인 탄산칼슘으로 지반을 고결시키는 효과를 구명하고자, 실내실험이 용이한 모래를 대상으로 기본적인 실험을 수행하였다. 실험에서는 표준사와 일반모래 2종류를 상대밀도 30%, 60%로 5cm x 5츠 x 5cm의 정육면체 아크릴 상자에 채워넣고, Bacillus pasteurii균 50ml 을 주입하여 공시체를 성형하였다. 10일 그리고 20일이 지난 시료에 대해서 압축강도 실험과 CaO함량 분석, 새롭게 형성된 광물질(탄산칼슘)을 확인하기 위한 전자현미경(SEM)분석, X선 회절분석(XRD)을 실시하였다.
