The purpose of this study is to get preeminent data from comparative analysis of multi-family housing unit plans between Korea and China nowadays. Even though Korea and China belong to common cultured band in Asia, there are many different Housing aspects and unit plans. Through this study we want to grasp the characteristics of both nation's apartment unit plans and to contribute for advance into the China apartment market. Analysis target is constructing and lotting out apartment at South Korea and China in 2004, and the space syntax model is used as an analysis tool. First, analyze kind, number and special quality of each room by use area, and second, analyze area and scale of the rooms. Through the integration analysis we know the grade of rank and centrality of each rooms, and search difference of South Korean and Chinese apartment.
Weissella cibaria가 생산하는${\alpha}$-Galactosidase 및 ${\beta}$-Glucosidase의 특성

대두의 발효를 통하여 생리활성을 가지고 있는 이소플라본 aglycone 함량을 높이기 위한 ${\beta}$-glucosidase와 대두에 다량 함유되어 있는 stachyose, rafinose와 같은 난소화성 oligosaccharides를 분해하기 위해 ${\alpha}$-galactosidase 효소 분비 미생물을 김치로부터 ${\alpha}$-galactosidase와 ${\beta}$-glucosidase를 생산하는 미생물을 탐색하였다. 탐색과정을 위해서 선별한 미생물을 16S rDNA sequencing 동정한 결과, Weissella cibaria 동정되어 Weissella cibaria K-M1-4로 명명하였다. Weissella cibaria K-Ml-4를 대두 액체배지에서 18시간동안 배양한 후, 생산한 효소는 배양액을 에탄을 침전, DEAE sepharose, sephacryl S-100HR column chromatography 통하여 ${\alpha}$-galactosidase의 경우, 정제도 5.3배, 수율 3.5% 그리고 ${\beta}$-glucosidase의 경우, 정제도 4.4배, 수율 2.9%로 정제되었다. ${\alpha}$-Galactosidase 효소특성은 $60^{\circ}C$에서 최대 활성을 나타내었으며, $80^{\circ}C$에서 30분 처리시 43% 잔존활성을 보였다. pH 8.0에서 최대 활성을 나타내었으며, pH 5.0-9.0에서 안정하였다. 금속이온에 대한 영향에서 $Fe^{2+}$과 $Cu^{2+}$을 첨가하였을 때 효소 활성이 증가하였다. p-Nitrophenyl-${\alpha}$-D-galacto-pyranoside (PNPG) 기질에 대한 Km은 0.98 mM이었고, Vmax는 $1.81{\mu}$mole/min 이었다. ${\beta}$-Glucosidase 효소 특성은 $50^{\circ}C$에서 최대 활성을 나타내었으며, $80^{\circ}C$에서 30분 처리시 46% 잔존활성을 보였다. pH 7.0에서 최대 활성을 나타내었으며, pH 5.0-9.0에서 안정하였다. 금속이온에 대한 영향에서 $Fe^{2+},\;Co^{2+},\;Cu^{2+}$을 첨가하였을 때 효소 활성이 증가하였다. p-Nitrophenyl-${\beta}$-D-gluco-pyranoside (PNPG)에 대한 Km값은 1.24mM이었고, Vmax는 $6.81{\mu}$mole/min 이었다. A strain producing ${\alpha}$-galactosidase and ${\beta}$-glucosidase was isolated from Kimchi. The isolated strain was identified as Weissella cibaria by 16S rDNA analysis and designated as Weissella cibaria K-M1-4. The enzyme activity of ${\alpha}$-galactosidase and ${\beta}$-glucosidase reached the maximum in the soy medium at $37^{\circ}C$ for 24 hr. The enzymes were purified by ethanol fractionation, DEAE sepharose fast flow, and sephacryl S-100HR column chromatography. ${\alpha}$-Galactosidase specific activity was shown by 576 Units/mg protein and the yield was 3.5% of the total activity of crude extracts. ${\beta}$-glucosidase specific activity was shown by 480 Units/mg protein and the yield was 2.9% of the total activity of crude extracts. The optimum temperature for ${\alpha}$-galactosidase was $60^{\circ}C$ and 43% of its original activity remained when it was treated at $80^{\circ}C$ for 30 min. For ${\alpha}$-galactosidase shows the optimum pH of 8.0 and is fairly stable between pH5.0 and pH9.0. The enzyme activity was increased in the presence of $Fe^{2+}$ and $Cu^{2+}$. The value of Km and Vmax for the enzyme were 0.98 mM and $1.81{\mu}$mole/min, respectively. The ${\beta}$-glucosidase has the optimum temperature of $50^{\circ}C$ and 46% of its original activity remained when it was treated at $80^{\circ}C$ for 30min. 다양한 침지처리가 대두의 미생물 오염 및 발아율에 미치는 영향

다양한 조건으로 대두를 침지하여 대두 발아시 미생물에 의한 오염을 최대한 방지하고 발아율을 높이는 조건을 검토하기 위해 대두를 멸균수, 0.1%의 자몽씨 추출물과 0.1%의 비타민 B₁ 유도체(AS5, vitagen)용액 등에 25℃, 35℃, 45℃, 55℃에서 3~12시간 동안 침지시킨 후 25℃, 상대습도 95%의 조건에서 3일 동안 발아시키며 발아율을 관찰하였다. 침지전 대두의 일반 미생물수는 6.0×10? CFU/g, 대장균군수는 3.0×10? CFU/g이었고, 대부분의 침지조건에서 미생물오염 정도는 감소하였다. 침지 대두의 발아율은 침지시간이 길어질수록 미침지 대두보다 발아율이 떨어졌고 45℃이상에서는 급격히 떨어져 55℃에서는 발아가 거의 일어나지 않음이 관찰되었다. 대두의 발아율은 침지용액의 종류와 침지시간 및 온도에 의해 영향을 받았으며, 대두의 미생물 오염감소와 초기발아를 유도를 위한 최적 조건은 자몽씨 추출물을 사용하여 35℃에서 6시간 침지시켰을 때였다. The presoaking conditions of soybeans in various solution for decreasing microbial contamination and improving germination were investigated. Soybeans were treated with presoaking solutions (sterilized water, 0.1% grape fruit extract, 0.1% vitamin B₁ derivate solution AS5 and vitagen) at 25℃, 35℃, 45℃ and 55℃ for 3~12 hr and germinate at 25℃, 95%RH for 3 days. Total bacterial count and coliform count of soybeans were 6.0×10? CFU/g and 3.0×10? CFU/g, respectively, and the most of presoaking treatments decreased the microbial contamination. The increase of presoaking time induces low germination ratio compared with unsoaked soybeans and the germination of soybeans was not detected at 55℃. The germination of soybeans was effected by presoaking solutions, temperature, and time. The optimal presoaking condition for decreasing microbial contamination and promoting initial germination was using grape fruit extract at 35℃ for 6 hr.
紫河車가 卵巢摘出 白鼠의 性호르몬 및 脂質含量에 미치는 影響

Placenta Hominis(PH) have been used in oriental medicine for many centuries as a Yang-tonifying drug. Therefore, the purpose of this Study was to investigate the effects of PH on the sex hormone changes and lipid metabolism in the experimental model. The experimental model of menopause was induced by bilateral ovariectomy in rats. We were used PH extract with freeze-dried and 200±20(g) female SD Rats for this Study. The level of serum total estrogen, serum progesterone, serum luteinizing hormone(LH), serum total cholesterol, serum triglyceride, serum HDL-cholesterol and body weight were measured. The results of this Study were obtained as follows ; 1. Body weight was decreased significantly in all treated group of PH as compared with control group. 2. Serum total estrogen level was increased significantly in only treated group of PH(40℃) extract as compared with control group. 3. Serum progesterone level was increased significantly in only treated group of PH(40℃) extract as compared with control group. 4. Serum LH level was decreased in all treated group of PH as compared with control group. 5. Serum total cholesterol level was decreased in all treated group of PH as compared with control group. 6. Serum HDL-cholesterol level was increased significantly in only treated group of PH(40℃) extract as compared with control group. 7. Serum triglyceride level was decreased significantly in only treated group of PH(40℃) extract as compared with control group. These results suggest that PH could be applied effectively menopausal syndrome, and would be used by powder or pill.

The various extracellular enzymes produced by lactic acid bacteria isolated from kimchi were assayed to improve the shelf-life of kimchi. Peroxidase was not detected in all tested lactic acid bacteria and small amount of ascorbic acid oxidase was detected in Pediococcus pentosaceus and Lactobacillus brevis. In case of α-amylase, 27-8 and 20.9 unit/mg were shown in Pediococcus acidilactici and Pediococcus pentosaceus, respectively but β-amylase and protease activities were very low. The enzyme related to textural property of kimchi, pectinesterase showed low activity but polygalacturonase activity was 0.28 unit/mg in Lactobacillus homohiochii and 0.27 nit/mg in Lactobacillus plantarum.
