Genotyping-by-sequencing (GBS) is an outstanding technology for genotyping and single nucleotide polymorphism (SNP) discovery compared to next generation sequencing (NGS) because it can save time when analyzing large-scale samples and carries a low cost per sample. Recently, studies using GBS have been conducted on major crops and, to a greater extent, on fruit crops. However, many researchers have some problems due to low GBS efficiency resulting from low quality GBS libraries. To overcome this limitation, we developed an efficient GBS library construction method that regulates important conditions such as restriction enzymes (RE) digestion and a PCR procedure for grapevine. For RE digestion, DNA samples are digested with ApeKI (3.6U) at 75℃ for 5 hours and adapters are ligated to the ends of gDNA products. To produce suitable PCR fragments for sequencing, we modified the PCR amplification conditions; temperature cycling consisted of 72℃ (5 min), 98℃ (30 s), followed by 16 cycles of 98℃ (30 s), 65℃ (30 s), 72℃ (20 s) with a final extension step. As a result, we had obtained optimal library construct sizes (200 to 400 bp) for GBS analysis. Furthermore, it not only increased the mapping efficiency by approximately 10.17% compared to the previous method, but also produced mapped reads which were distributed equally on the19 chromosomes in the grape genome. Therefore, we suggest that this system can be used for various fruit crops and is expected to increase the efficiency of various genomic analysis performed.
한국형 유산균 Bifidobacterium 속 균주의 항생물질에 대한 감수성

Minimal inhibitory concentrations (MICs) of Bifidobacterium spp. strains (Bifidobacterium breve K-110, B. breve K-111 and B. infantis K-525) isolated from healthy Korean against antituberculosis agents and fluoroquinolones were determined. From the MICs it was found that Bifidobacterium breve K-110, B. breve K-111 and B. infantis K-525 were susceptible to rifampicin and fluoroquinolenes and resistant to other antituberculosis agents.
3D Magic Mirror 작품을 이용한 인터랙티브 아트 설계 및 구현

Digital art is recent genre from high technology and cultural development and one of the most unlimited growing fields. An audience becomes more interested in digital art as time goes by. Computer technology results in interaction development in interactive art. In this paper, we use 「Magic Mirror」, which is worked by Maurits Cornelis Escher, for creating interactive art 3D 「Magic Mirror」. Maurits Cornelis Escher was a Dutch graphic artist. 3D 「Magic Mirror」 is out of existing 2D 「Magic Mirror」 and has an infinite sense of space. We make to broaden vision of appreciating 「Magic Mirror」 and sublimate hided artistic merit in novel artistic merit. An optical illusion is one of special features in works of Escher. Before, A cause of optical illusion in 「Magic Mirror」 isn't found out by an audience easily. However, in 3D 「Magic Mirror」, an audience has a find it in no time at all then we make it obvious. And we added another fresh artistic merit which is animating objects in 3D 「Magic Mirror」. 새로운 예술인 디지털 아트는 시간이 흐를수록 관객이 많은 흥미를 느끼고 기술적으로나 예술적으로나 무한한 발전을 거듭하고 있는 예술분야 중 하나이다. 기술의 발전은 인터랙티브 아트에서 작품과관객과의 상호작용에 날개를 달아주는 결과를 낳았는데 본 논문에서는 기하학적 원리와 수학적 개념을 토대로 자신의 상상에서 비롯된 내적 이미지를 표현한 네덜란드 판화가․화가인 Maurits Cornelis Escher의 「Magic Mirror」를 사용하여 인터랙티브 아트 작품을 구현한다. 2D 작품인 Magic Mirror에서벗어나 3D로 구현하여 작품을 보는 시각을 넓히고 평면적 작품에 숨겨졌던 예술적 가치를 승화시켜내어 새로운 예술적 가치를 이끌어내었다. Escher 작품의 큰 특징 중 하나인 착시의 원인을 「Magic Mirror」에서는 관객이 쉽게 찾을 수 없었으나 3D아트로 재창조시켜 원인이 분명해질 수 있었고 이 오브젝트들을 애니메이팅 시켜 또 다른 예술적 가치를 더하였다. 관객의 직접 참여를 유도하는 인터랙션에 초점을 두어 작품을 구현하여 단지 전시만을 목적으로 하던 「Magic Mirror」에서 벗어나 3D 「 Magic Mirror」는 관객과 소통할 수 있었다.
Agrobacterium에 의한 오이 형질전환에서 자엽절 절편의 이용

Agrobacterium tumefaciens-mediated cotyledonary-node explants transformation was used to produce transgenic cucumber. Cotyledonary-node explants of cucumber (Cucumis sativus L. cv., Eunsung) were co-cultivated with Agrobacterium strains (EHA101) containing the binary vector (pPZP211) carrying with CaMV 35S promoter-nptII gene as selectable marker gene and 35S promoter-DQ gene (unpublished data) as target gene. The average of transformation efficiency (4.01%) was obtained from three times experiments and the maximum efficiency was shown at 5.97%. A total of 9 putative transgenic plants resistant to paromomycin were produced from the cultures of cotyledonary-node explants on selection medium. Among them, 6 transgenic plants showed that the nptII gene integrated into each genome of cucumber by Southern blot analysis. Agrobacterium과 자엽절 절편 공동배양으로 오이 형질전환체를 생산하였다. 오이 자엽절 절편 (c.v. Eunsung)은 선발 마커로서 nptII유전자와 유용유전자로서 DQ유전자가포함된 pPZP211를 EHA101에 형질전환하여 공동 배양하였다. 3회 반복실험으로부터 평균 형질전환 빈도는 4.01%를, 최대 빈도는 5.97%를 보여 주었다. Paromomycin항생제 저항성을 갖는 9개의 식물체를 선발과정을 통해 얻었으며, Southern blot 분석에 의해 6개 식물체의 genome에nptII유전자자 안정적으로 도입되어 있음을 확인할 수 있었다.
Lactobacillus plantarum MKHA15와 Leuconostoc mesenteroides MKSR을 첨가한 발효 채소 주스의 특성

The objective of this study was to develop fermented vegetable juices that possess antidiabetic and antioxidant activities. Lactobacillus plantarum MKHA15 (MKHA15) and Leuconostoc mesenteroides MKSR (MKSR) were applied to ferment onion, cabbage, and tomato juices at 37oC and 30oC for 72 h, respectively, and their functionality was tested using the 12 h hour-fermented juice by MKHA15, and 48h hour-fermented juice by MKSR. Inhibition of α-glucosidase activity was observed in all fermented juices. The onion juice fermented by MKHA15 showed significantly higher α-glucosidase inhibition activity compared to other juices. All juices showed more than 70% inhibition of α-amylase activity. The DPPH radical scavenging activity of onion juice fermented by MKSR showed significantly lower activity than cabbage and tomato juices; however, no difference was observed between the types of starter cultures. The SOD-like activity of cabbage juice fermented by MKSR was the highest among the fermented juices. The juices fermented by MKHA15 showed higher reducing power than those by MKSR. Therefore, we believe that cabbage, onion and tomato juice fermented by MKHA15 and MKSR would be useful in probiotic juices, as they possess antidiabetic and antioxidant activities.
