글루탐산나트륨 첨가배지에서 재배된 새송이버섯의 특성 및 생리활성 연구

새송이 버섯 (Pleurotus eryngii) 재배배지에 글루탐산나트륨 (monosodium glutamate, MSG)을 혼합 첨가하여 재배한 자실체의 아미노산 및 γ-aminobutyric acid (GABA) 함량 변화, 그리고 항산화 작용 등의 생리활성을 분석하여 새송이버섯의 기능성 향상 가능 여부를 조사하였다. 실험 결과 MSG 첨가량이 증가함에 따라 arginine, glutamic acid, alanine, glycine 등 아미노산은 함량이 증가되었으며 histidine, serine, methionine, isoleucine, leucine, phenylalanine 등은 함량 변화는 거의 없었다. 한편 MSG 첨가량을 증가시킴에 따라 자실체의 GABA함량은 전체적으로 증가되었으며 MSG 6 g/bottle 첨가군에서 자실체내에 가장 높은 GABA함량 (1.18 mg/g)을 얻을 수 있었다. 또한 배지에 MSG첨가량을 증가시킴에 따라 자실체 추출물의 DPPH (1,1-diphenyl-2-picrylhydrazyl) radical 소거능 및 아질산염 소거능 (nitrite scavenging activity)도 증가됨을 알 수 있었다. 이러한 결과로부터 새송이버섯을 재배할 때 배지에 소량의 저렴한 MSG (~US $1/kg)를 첨가하면 아미노산 조성 및 항산화작용 등 생리활성이 개선되는 고 기능성 새송이버섯 식품 소재가 개발될 수 있을 것으로 사료된다. This study was performed to investigate the composition of amino acids and biological properties with the ethanol extract of fruiting bodies of Pleurotus eryngii grown on the sawdust compost mix (400 g sawdust plus 200 g rice bran) supplemented with various dosages of monosodium glutamate (MSG). Amino acid composition analyses showed that arginine, glutamic acid, alanine and glycine increased as the dosage of MSG was increased, whereas histidine, serine, methionine, isoleucine, leucine and phenylalanine did not increase. γ-Aminobutyric acid (GABA) content increased significantly up to 1.18 mg/g extract when 6 g MSG was supplemented to the sawdust mix. Antioxidant activity of the extract was estimated and compared to the standard antioxidant (ascorbic acid). The antioxidant property such as 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging activity increased with the extract samples of increasing dosage of MSG. Although the extract showed low levels of nitrite scavenging activity, this activity increased up to 1.5-2.0 fold when MSG was supplemented to the sawdust mix above a dosage of 0.5 g. The results obtained from the present investigation would appear that Pleurotus eryngii grown on the MSG-enriched sawdust mix can be used more effectively as one of potential sources of functional foods.

No Synergistic Effect of Silibinin and Morin in a Kainic Acid-Induced Epileptic Mouse Model

Temporal lobe epilepsy (TLE) is the most common form of localization-related epilepsy, with the highest prevalence rate in adulthood. Recently, we reported the beneficial effects of the individual treatment with flavonoids such as silibinin and morin in kainic acid (KA)-treated mouse model for TLE. In this study, we investigated whether there is a synergistic effect of co-treatment with silibinin and morin on the susceptibility to seizure, the frequency of spontaneous recurrent seizures (SRSs), and granule cell dispersion in the dentate gyrus, which could be partially controlled by treatment with each flavonoid in the animal model for TLE. Unfortunately, we did not observe any synergistic effect against the susceptibility of seizure and SRS induced by KA treatment. However, the combination of these flavonoids showed similar antiepileptic effects compared with treatment with each one individually. Therefore, although silibinin and morin are not suitable for combination therapy, our results still suggest that these flavonoids can be used as potent therapeutic compounds for preventing epileptic seizures.
