◈The purpose of report is to study the vision of leisure culture through changes of modern Korean family structure. Changes on modern family structure is embodied with changes on ‘form, action and value’ by possessive relation between husbands and wives and the changes are defined in a view of organizational theory called self-organization. Due to changes of possessive relation of the actual Korean family structure, 1)a new custom family reorganization in terms of ‘form’ has been taking place, 2)a group and an individual culture have been coexisting in terms of ‘action’ 3)and in terms of ‘value’, individualizaton has accomplished between family members. The Korean family structure that has changed into ‘functional organizations for survival has come up as new consumption unitsThe visions of new industry and culture of sports leisure, that have risen through changes of Korean family structure, must be analyzed in social structural way. 단삼(Salvia miltiorrhiza) 추출물의 치주질환유발 세균의 생육억제 및 Collagenase 저해 활성

단삼(Salvia miltiorrhiza) 추출물의 치주질환유발 세균에 대한 생육저지 활성 및 collagenase저해 활성을 측정하였다. 단삼 에탄올 추출물은 치주질환유발 세균에 대해 우수한 생육저지 활성을 나타내었다. 유기용매 분획물중, hexane에서 생육억제능이 가장 우수하였다. 치주질환유발 세균 C. curvus, C. rectus, E. corrodens, F. nucleatum, P. gingivalis, P. intermedia 및 W. succinogenes에 대한 단삼 추출물의 최소생육저해농도(MIC)는 200,50,50,250,150,250및 200 ${\mu}g$/ml로 각각 우수한 생육억제능을 나타내었다. 단삼 유기용매 분획물의 collagenase저해 활성은 동일 농도의 minocycline에 비해 우수한 저해 활성을 나타내었으며, chloroform분획물의 경우 88.2%의 저해 활성을 나타내었다. This study was carried out to evaluate the inhibition of growth and collagenase activity of Salvia miltiorrhiza Bunge against microorganisms causing periodontal diseases. The ethanol extract of Salvia miltiorrhiza showed sig-nificant growth inhibition against microorganisms causing periodontal diseases. Ethanol extract was further fractionated with organic solvents in the order of hexane, chloroform and ethyl acetate. Among the fractions tested, the hexane fraction showed the highest cell growth inhibition. The minimal inhibitory concentration (MIC) of the extract against C. curvus, C. rectus, E. corrodens, F nucleatum, P. gingivalis, P. intermedia and W. succinogenes were 200, 50, 50, 250, 150, 250 and 200 ${\mu}g$/ml, respectively. The inhibition of collagenase activity by organic solvent fractions were higher than that of minocycline, and the inhibition ratio of collagenase activity was $88.2{\pm}2.1$ % in the chloroform fraction.
백강균(Beauveria bassiana)의 균사체 최적 배양조건 및 효소활성

백강균(Beauveria bassiana DGUM 34001)은 $24^{\circ}C$의 온도와pH 7.0의 초기 pH에서 최적의 균사체 생육을나타내었다. 사용한 복합배지 중 mushroom complex배지(MCM)에서 가장 우수한 균사생육을 나타내었다. Czapek-Dox 한천배지를 최소배지로 각각 탄소원, 질소원 및 인산원의 영향을 조사한 결과, glucose, soytone 및 sodium phosphate ($NaH_2$$PO_4$)에서 가장 우수한 균사 생육을 보여주었다. MCM액체배지에서 균사 배양 후 세포외 효소 활성을 측정한 결과,$\alpha$-amylase, lipase, chitinase, CMCase및 pretense의 비효소활성은 각각 297.0, 0.058, 0.33, 0.21 및 22.8 units/mg protein이었다. Casein과 soluble chitosan을 첨가할 경우 세포외 분비 pretense와 chitinase의 효소 활성 이 증가되었다. The optimum temperature and pH for mycelial growth of B. bassiana DGUM 34001 were $24^{\circ}C$ and pH 7.0, respectively. Among the complex media used, mushroom complex medium (MCM) was the most favorable for mycelial growth. When Czapek-Dox medium was used as a minimal medium, glucose was an excellent source for carbon and energy. Soytone and sodium phosphate were favorable constituent for culture medium as a source of organic nitrogen and phosphorus, respectively. When the fungus was grown in MCM broth, the specific activity of extracellular enzyme of ${\alpha}$-amylase, lipase, chitinase, CMCase and pretease were 297.0, 0.058, 0.33, 0.21 and 22.8 units/mg protein, respectively. When various sources of organic nitrogen and chitin were supplemented to determine the production of enzymes, casein and soluble chitosan enhanced the production of extracellular protease and chitinase.
송이의 세포외 분비 $\beta$-Glucosidase 효소의 특성

Cellulose 분해에 관련된 $\beta$-Glucosidase 효소 활성의 특성 파악을 위하여 송이균사(Tricholoma matsutake DGUM 26001)의 액체 배양시 세포외로 분비되는 $\beta$-Glucosidase 효소를 부분정제하여 그 특성을 조사하였다. 효소 활성에 미치는 적정 온도는 55-$70^{\circ}C$이었고 최적 온도는 $65^{\circ}C$이었다. 적정 효소활성에 영향을 주는 적정 pH는 3.0-5.0 범위였으며 최적 pH는 4.0이었다. Salicin을 기질로 최적 조건하에서 $\beta$-Glucosidase 효소의 비활성도는 18.7 unit/mg protein이었다. 열안정성은 $60^{\circ}C$이하의 온도에 60분간 열처리시 약 90%이상의 효소활성을 유지하였다. $Fe^{++}$이온은 효소활성을 촉진하였으나, $Hg^{++}$ 및 $Cu^{++}$이온은 효소활성을 매우 억제하였다. Salicin에 대한 효소활성을 100으로 하였을 때, cellobiose는 48.6%의 상대적 효소활성을 나타내었으며, cellobiose에 대한 Km값 및 Vmax값은 각각 0.12mM과 0.02umol/min었다. In order to determine the characteristics of $\beta$-glucosidase associated with cellulose degradation, the enzyme produced extracellularly by the mycelia of Tricholoma matsutake DGUM 26001 in culture broth was partially purified. The enzyme activity was maintained in the range of temperatures trom 55 to $70^{\circ}C$ and its optimum temperature was $65^{\circ}C$. The $\beta$-glucosidase enzyme showed relatively high activity in the range of pH 3.0-5.0 and its optimum pH was 4.0. Under the optimal conditions, the specific activity of $\beta$-glucosidase for salicin as a substrate was 18.7 unit/mg protein. After thermal treatment of the enzyme at $55^{\circ}C$ for 60 min, more than 90% of the enzyme activity was still sustained. Iron($Fe^{++}$) stimulated enzyme activity, whereas mercury($Hg^{++}$) and copper($Cu^{++}$) inhibited. Compared to salicin as a substrate, the relative activity for cellobiose was observed to be 48.6%. The apparent $K_m$ and $V_{max}$ of the enzyme with cellobiose were 0.12 mM and 0.02 umol/min, respectively.
