        Genetic Recombination by Protoplast Fusion in Streptomyces scabies

        The double mutants for protoplast fusion and genetic mapping on the chromosome were obtained as Streptomyces scabies L1(leu met) and N1(nic his) using UV irradiation. The protoplast formation and regeneration of these two strains were also investigated to progress the strain improvement by means of protoplast fusion. Protoplasts were obtained by treatment with 1 ㎎/㎖ of lysozyme in the P reduced medium. Each mutant was treated with 44% of PEG 1000 for protoplast fusion, and recombination frequency showed to be 1.4×10^(-3). Relative recombinant frequencies in each markers by means of selective media could be used for genetic analysis and gene mapping.

        Some Undescribed Cladosporium, Alternaria, Curvularia and Eurotium repens in Korea

        한국의 토양과 서울 시내의 공기로부터 진균류를 분리하였스며 그 중에서 다음 10군을 미기록 종으로 보고한다 : Cladosporium sphaerospermum, C. herbarum, C. colocasiae, Alternaria chlamydospora, A. cheiranthi, A. citri, Curvularia ovoidea, C. inaequalis, C. affinis, and Eurotium repens. The following ten species collected from the air of Seoul City and the soils in Korea are reported as undescribed to Korean fungal flora: Cladosporium sphaerospermum, C. herbarum, C. colocasiae, Alternaria chlamydospora, A. cheiranthi, A. citri, Curvularia ovoidea, C. inaequalis, C. affinis, and Eurotium repens.

        항생물질 생산 방선균의 역가 개량에 관하여

        An improved procedure for the rapid purification of glucose-6-phosphate dehydrogenase from extracts of Saccharomyces cerevisiae was developed by using affinity chromatography. Among six affinty media tested, NADP$^{+}$-agarose and Affi-gel Blue were more effective than others (i.e., Affi-gel Red, AMP-agarose, ATP-agarose, and NAD$^{+}$-agarose). Conditions to desorb the enzyme bound to the affinity media were examined to increase the purity as well as yield. The best result was obtained when the column was developed with a linear gradient of KCl (0-1.0M). In case of Affi-gel Blue, introduction of NAD$^{+}$ (15mM) washing step prior to the salt gradient was most effective to remove NAD$^{+}$-binding proteins. For a large scale preparation of G-6-P dehydrogenase higher recovery was obtained by Affi-gel Blue than NADP$^{+}$-agarose, however, the purity of the enzyme was decreased by 10 times if the former was used as the affinity medium. The capacity of Affi-gel Blue for G-6-P dehydrogenase was found to be 5 times higher than that of NADP$^{+}$-agarose. Furthermore Affi-gel Blue could be reused repeatedly and its preparation is relatively easier and less expensive than NADP$^{+}$-agarose.X> +/-agarose.

        The stage phobia at piano performance is very familiar condition to every piano player, and it can be significant interference in the life of a piano player. Anxiety or tension is a normal psychophysiological symptom, but it becomes a stage phobia when it occurs in a specific situation such as piano performance. There can be personal differences, but it can be amplified by irrational belief, personality, competitive desire and individual background. So it is necessary to understand one`s personal characteristics influencing and amplifying anxiety, and then the stage phobia can be overcome by modifying the personal characteristics. To practice sincerely and continuously has significant meaning to every piano player to perform his or her music satisfactorily. To accept the meanings and symptoms of the stage phobia, to find out the causes of amplifying the stage phobia, to modify it, and to practice continuously and sincerely can be very effective strategies of stage phobia.

        Chondroid Syringoma on Face

        Chondroid syringoma is a rare mixed tumor of the skin which is composed of both mesenchymal and epithelial cells. Its incidence at less than 0.1% and is frequently located on the head and neck. Chondroid syringoma is easily confused with epidermal cysts. Since malignant forms of chondroid syringoma have been reported, accurate and timely diagnosis is important for proper management. We report clinical and histological features of chondroid syringoma in 5 patients treated at our institution. In most of the cases, chondroid syringoma presented as a round, firm, nodular or cystic lesion that had well marginated heterogeneity in sonography. Clinically, all of the lesions were removed by simple excision. Microscopically, all five tumors were well circumscribed and consisted of epithelial, myoepithelial, and stromal components. The epithelial component formed tubules lined by one or more rows of eosinophilic epithelial cells. The outer layer of tubules appeared to be flattened myoepithelial cells. The stroma is myxoid and contained spindle shaped myoepithelial cells. We expect that the clinical, sonographic, and histological data from our report may help clinicians who are confronted with various kinds of analogous facial lesions to decide the most proper management for their patients.

      • 高靈附近 洛東層群 砂岩의 堆積岩石學的 硏究

        This paper deals with Sedimentological Petrology of Nagdong Group (Upper Jurassic(?) to Lower Cretaceous) at Goreong area in Gyeongsang Province, Korea. The Nagdong Group has four Chronological subdivisions, that is Yeonhwadong, Hasandong, Dongmyeong, and Chilgog formations. Such subdivisions are based on mainly rock color and lithological features. 78 sandstone samples were taken and studied upper ordinary polarizing microscope with great emphasis of mineral constituents and grain size parameter. The obtained results may be summerized as the following. The type of sandstone is arkose, subarkose, and great amount of graywacke (according to Dapples sandstone classification, 1953). The sandstone are also moderately well sorted to moderately sorted (according to Folk and Ward, 1957) and near symmetrical to fine skewed (according to Folk and Ward, 1957). Kurtosis of the sandstone belongs to meso kurtic to lepto kurtic (according to Folk and Ward, 1957). Grain size parameter analysis shows that the sandstones are deposited sediments under fluviatile environment. Mineral constituents identified also shows that the provenance of the Nagdong Group in the thesis area is polygenetic terrain of Metamorphic rocks with little amouts of granitic and Sedimentary rocks.

        효모세포의 당과 아미노산의 운반에 관한 연구 II

        Saccharomyces cerevisiae J170, a mutant, was used for $DL-^{14}C-leucine$ uptake during the sporulation and vegetative stage. $^{14}C-Leucine$ uptake into yeast cells appeared the highest at pH 6.0, indicating the same result of glucose transport, $^{14}C-Leucine$ uptake in sporulation period was higher than in growth phase, showing the evidence that leucine is more required for protein synthesis. This tendency has the evidence tht leucine is more required for protein synthesis. This tendency has the evidence that leucine is more required for protein synthesis. This tendency has been also supported from the result of Km values of leucine uptake in two stages of yeast. Leucine uptake was inhibited by 2,4-dinitrophenol in two stages of yeast. This means that leucine transport system is associated with energy dependent in both stages. The contents of all amino acid in growth phase cells were higher than those of sporulation stge cells, and those of methionine and tyrosine were showed in trace during the sporulation stage. In contrast, the content of glutamic acid in sporulation stage was compared with those of other amino acids.
