Solar-driven biocatalytic C-hydroxylation through direct transfer of photoinduced electrons

<P>Despite the immense potential of P450s, the dependence on the nicotinamide cofactor (NADPH) and NADPH-P450 reductase (CPR) limits their employment in the chemical industry. Here, we present a visible light-driven platform for biocatalytic C-hydroxylation reactions using natural flavin molecules, especially flavin mononucleotide, as a photosensitizer. By employing visible light as a source of energy instead of the nicotinamide cofactor, the bacterial CYP102A1 heme domain was successfully applied for photobiocatalytic C-hydroxylation of 4-nitrophenol and lauric acid - in the absence of NADPH and CPR. We present a proof of concept that the photoactivation of flavins is productively coupled with the direct transfer of photoinduced electrons to the P450 heme iron, achieving photobiocatalytic C-hydroxylation reactions.</P>

Streptomyces avermitilis produces clinically useful drugs such as avermectins and oligomycins. Its genome contains approximately 33 cytochrome P450 genes and they seem to play important roles in the biosynthesis of many secondary metabolites. The SAV_7130 gene from S. avermitilis encodes CYP158A3. The amino acid sequence of this enzyme has high similarity with that of CYP158A2, a biflaviolin synthase from S. coelicolor A3(2). Recombinant S. avermitilis CYP158A3 was heterologously expressed and purified. It exhibited the typical P450 Soret peak at 447 nm in the reduced CO-bound form. Type I binding spectral changes were observed when CYP158A3 was titrated with myristic acid; however, no oxidative product was formed. An analog of flaviolin, 2-hydroxynaphthoquinone (2-OH NQ) displayed similar type I binding upon titration with purified CYP158A3. It underwent an enzymatic reaction forming dimerized product. A homology model of CYP158A3 was superimposed with the structure of CYP158A2, and the majority of structural elements aligned. These results suggest that CYP158A3 might be an orthologue of biflaviolin synthase, catalyzing C-C coupling reactions during pigment biosynthesis in S. avermitilis.

S-668 IgG autoantibody to FcεRIα in chronic urticaria patients with doubtful clinical implication

Background: Chronic Spontaneous Urticaria (CSU) is common skin disorder characterized by persistent or recurrent itchy wheals and pruritus of at least 6 weeks duration where the autoimmune mechanism is involved. In this study, we aimed to evaluate the level of IgG autoantibody against FcεRIα (IgG to FcεRIα) in CSU patients by dot-blot immunoassay and investigate the association with other clinical parameters including autologous serum skin test (ASST), urticaria activity score (UAS) and other antibodies. Method: We enrolled 125 CSU patients and 63 healthy controls (NC). We detected serum IgG autoantibody to FcεRIα by using rapid Dot-Blot immunoassay. The cut-off value was determined as mean intensity plus 2 standard deviations of values from NC Result: We detected IgG autoantibody to FcεRIα in 27.2% of CSU patients, significantly higher than NC group (1.6%, p<0.001). There was no difference in frequency of IgG anti-FcεRIα between CSU patients with positive and negative ASST response (61.8% vs 47.3% p=0.149). A significant female preponderance among CSU patients with positive IgG to FcεRIα was observed (94.1% vs 65.9%, p<0.001). However, there was no significant association of IgG anti- FcεRIα with other clinical parameters including UAS, the presence of anti-thyroid antibody and ANA, atopic status, serum total IgE levels and clinical response to antihistamines. In contrast, the C3 level was significantly different according to the presence of anti- FcεRIα antibody (109.8±19.9 vs 123.5±31.3, p= 0.027). Conclusions: The prevalence of FcεRIα autoantibody was significantly higher in CSU patients, particularly in female compared to NC. However, IgG anti-FcεRIα positivity was low (27.2%) and clinical implication has not been established in CSU. It seems that the anti-FcεRIα autoantibody might be involved in complement activation rather than in a direct degranulation of mast cell or basophil. <P>The aim of this new work was to improve the dissolution rate of poorly water-soluble cilostazol (CLT) by adsorbing dissolved drug molecules onto the surface of undissolved carriers via reprecipitation and deposition process as the solvent (methylene chloride) was evaporated. The adsorption mixtures of CLT with Aerosil 300 and lactose monohydrate provided better drug dissolution rate as compared to mannitol. However, Aerosil-based adsorption powders were unable to be compressed into tablet due to low and unsatisfactory compressibility. The optimized CLT-loaded tablets containing lactose-based adsorption powders displayed almost complete dissolution within 90 min and showed significantly higher dissolution in intestinal fluid (pH 6.8) containing 0.2% sodium lauryl sulfate than the control tablet and the commercial reference product Pletaal (R) (Otsuka). Differential scanning calorimetry (DSC) and powder X-ray diffraction (PXRD) indicated that the crystalline property of CLT remained in adsorption powders with diminutive peak intensity. Fourier transform infrared spectroscopy (FT-IR) spectra indicated that the intermolecular hydrogen bond was formed between drug molecule and lactose monohydrate. Field-emission scanning electron microscope (Fe-SEM) clearly confirmed that CLT crystals with reduced size around 10 mu m were adsorbed and deposited onto the surface of adsorbent material.</P>
아스피린 과민성 천식 환자에서 TLR3 유전자 다형성에 따른 IgG 아형 결핍 및 천식 중증도의 연관성 연구

Purpose: Toll-like receptor 3 (TLR3) recognizes to viral double-stranded RNA and is involved in antiviral defenses. A probable role of TLR3 gene variants in the pathogenesis of aspirin-intolerant asthma (AIA) has been suggested. AIA patients present more frequent asthma exacerbations in which respiratory viral infections could be an exacerbating factor. IgG subclass deficiency was commonly present with bronchial asthma. Based on previous findings, we investigated whether TLR3 variants could affect IgG3 subclass deficiency in AIA.Methods: We enrolled 279 AIA patients, 403 aspirin-tolerant asthma (ATA) patients, and 315 normal healthy controls (NC) in this study. TLR3 polymorphism at the promoter region -299698G>T was genotyped. The serum levels of IgG subclasses were determined by the single radial immunodiffusion method. Expressions of IgG3 and TLR3 on Epstein-Barr virus transformed-B cells isolated from asthmatic patients were evaluated by flow cytometry to investigate B-cell functions.Results: The TLR3 -299698 T allele was significantly associated with severity and IgG3 deficiency in the AIA group (P=0.044 and P=0.010, respectively), but not in the ATA group. IgG3 expression on B cells from asthmatics with IgG3 deficiency was significantly lower compared to those without (P=0.025). There was a positive correlation between IgG3 expression levels on B cells and serum IgG3 levels (r2=0.434, P=0.002).Conclusion: These results suggest that the TLR3 -299698G>T polymorphism may be associated with IgG3 subclass deficiency and severity in AIA. FCT 1 : Beneficial effects of early treatment of nevus of Ota with a 1064-nm Q-switched Nd:YAG Laser

Nevus of Ota, caused by dermal melanocytosis, is cosmetically troublesome in Asian patients. Q-switched laser systems including Nd:YAG lasers were used to treat this condition. The lesions of young age are proportionally smaller and the skin is thinner allowing for better penetration of the laser. This is a retrospective study of 31 Korean patients (Fitzpatrick skin type IV) with nevus of Ota treated with Q-switched Nd:YAG laser. Patients received a series of 6-32 treatment sessions with 1064-nm Q-switched Nd:YAG laser at settings of 5-8 mm spot, 1.9-5.0 J/cm2 mean fluences and given at 2- to 3-week intervals. The fluence of laser treatments was set at 2.5 J/cm2 and adjusted based on patient response to the previous treatment session and sensitivity to pain. Treatment was applied until the lesions showed mild erythema. Age at the first treatment was significantly correlated with the degree of improvement. The earlier patients started the first treatment, the less mean numbers of treatment session were required to reach moderate, marked, and near total improvement (r=0.417, 0.498 and 0.467, p<0.05). Besides, by starting treatment early, low mean fluences were required to reach same end points in each session (r=0.586, p<0.001). Adverse effects were minimal but post-inflammatory hyperpigmentation was observed in one patient. It was clinically effective and safe for early treatment of nevus of Ota with a Q-switched Nd:YAG laser.
