B(a)P, DMBA 및 DMN의 세포매개성 돌연변이 형성기전에 있어 마우스 Strain차 및 Cytochrome P-450 Isozymes의 역할

화학적 발암 및 돌연변이 유발물질인 B(a)P, DMBA 및 DMN의 세포매개성 돌연변이 유발작용이 마우스 strain차 및 Cytochrome P-450 isozymes에 의하여 어떤 영향을 받는 지를 실험 관찰하여 보고하는 바이다. The change in mutagenic activities of chemical carcinogens, 7,12-dimethyl benz(a)anthracene(DMBA) benzo(a) pyrene (B(a)P) and N-nitrosodimethylamine (DMN) have been investigated using V79 chinese hamster cells as target cells in non-induced, 3-methylcholanthrene(MC) -induced and phenobarbital (PB) -induced C57 Black, C3H and B6 mice, respectively. The number of 6-thioguanine-resistant mutants showed remarkable dose-dependent increase by treatment of DMBA and B(a) P in every 3-MC-induced mice irrespective of mouse strain. In PB-induced mice, number of the resistant mutants to 6-TG were slightly higher than non-induced group by treatment of DMBA in every strain. On the contrary, compared to the control group, number of the resistant mutants to 6-TG were reduced in the B(a)P-treated groups. DMN had produced 6-thioguanine-resistant mutants proportionately as a function of concentration, but was not affected by 3-MC and PB pretreatment, These results suggest that 3-MC and PB induced the different kinds of cytochrome P-450 isozymes and these isozymes may not be responsible for converting DMN into active mutagenic products.
마우스 회장 평활근에 미치는 lithium의 이완작용에 관한 연구

We studied the relaxant effects of isoproterenol and lithium on the mouse ileal smooth muscle which were set at 1gm initial tension or contrated by carbachol (3×10-5M). At the same time, we also investigated whether the above effects are changed or not by the pretreatment of propranolol, β-receptor blocker. The results obtained were summarized as follows; 1. The relaxant effects of isoproterenol and lithium on the mouse ileal segment which were set at 1gm initial tension were prominent and these effects had increasing tendencies dose-dependently. 2. Same effects were also observed at the carbachol-induced contracted mouse ileal segment. 3. The relaxant effects of isoproterenol may be inhibited competitively, but those of lithium inhibited noncompetitively by the pretreatment of propranolol(3×10-6M) In conclusion, isoproterenol and lithium are seemed to have prominent relaxant effects on mouse ileal smooth muscle which was tensed by 1gm initial tension or contracted by carbachol but mechanism of actions were different between both drugs. The former may act through 7·receptor but the latter through other mechanisms.

We investigated the reproductive capacity of starry flounder Platichthys stellatus from the Uljin (UJ) marine ranching area and compared it to that of P. stellatus from the coastal waters of Pohang (PH). In UJ, female gonadosomatic index (GSI) peaked in October (3.14±0.87) and male GSI was high in October and December. In PH, female GSI peaked in January (18.64±2.15) while male GSI began to increase in October and remained high until March. Most ovaries of UJ females were immature with perinucleus oocytes, although the testes of UJ males were ripe in January. Both the ovaries and testes of PH starry flounders were ripe from January to March. The plasma estradiol-17b (E2) levels of UJ females were highest in October (4.09±1.90ng/mL) although the testosterone (T) levels of UJ males were highest in December (3.81±0.78ng/mL) and decreased gradually until April. The 17a, 20b-dihydroxy-4-pregnen-3-one (17α20βP) levels of UJ females were not detected. The E2 levels of PH females were highest in December (36.25±33.07ng/mL) and 17a20bP levels were highest in March (5.51±0.95ng/mL). The T levels of PH males were highest in December (4.03±1.34ng/mL) and decreased gradually until October. Taken together, these results suggest that most females from UJ did not reach maturation with a spawning period that was considered to be between December and January.
