        적합수혈후 초래된 지연성 용혈작용이라 추정한 1 례 보고

        Hemolytic transfusion reactions may occur after the administration of donor bloods even when they have been shown compatible apparently by crossmatch tests. Such episodes present a diagnostic challenge and raise serious doubts about our understanding of blood group incompatibility. Fever, hemoglobinuria, oliguria and marked fall in hematocrit values developed in a patient 30 hours after she had received one unit of apparently compatible blood during operation under general anesthesia. Results of routine serologic studies at the time of the transfusion reaction were normal. She died on 40 hours after operation.

        Current Research Trends on Improvement of Balloon Flower Production and Processing

        Balloon flower (BF, Platycodon grandiflorum (Jacq.) A.DC.) is a monotypic species of the bellflower family (Campanulaceae), which is native to Asia. In terms of popular culture of east asian country, BF has been in the limelight as flowers, medicine and food. Platycosides (saponins) from the roots of BF are reported to have a wide range of efficacy and characterized by a structure containing a triterpenoid aglycone and two sugar chains. Saponins from BF are of pharmaceutical significance, and their applications are increasing with evidence of their efficacy. As the cultivation area of BF is continually increased in Korea, research for preventing root rot incidence and nitogen fertilizer application on yield of saponin are reported and also storaging, steam heating and bio-transformational method are developed. There are several biological effects of compounds from BF. saponin from BF has cholesterol lowering effects, neuroprotective activity, cytotoxic effects against cancer cells, and polysaccharide has immuno-boosting effects and pheolic compounds from aerial part of BF has anti-oxidative effects. However, The purchase decision factor of consumers in market is cultivate year of BF and producing country are also important as contents of platycodins in BF. This paper reviews the origin of plants, harvest and post harvest processing methods that maximize the value in terms of efficacy of extracts and platicoside from BF.

        Fracture Healing Effects of Locally-Administered Adipose Tissue-Derived Cells

        Purpose: Although the applications of adipose tissue-derived cells (ADCs) in regenerativemedicine have been investigated, the role of ADCs in fracture healing remains unclear. In this study, we examined the fracture-healing effects and survival of transplanted ADCs using micro-computed tomography (CT) and bioluminescenceimaging (BLI). Materials and Methods: Luciferase-expressing ADCs were suspended in solubilized basement membrane preparation (SBMP) and xenografted on defects in the right femur of nude mice (n=5). SBMP alone was grafted on a defectin the contralateral femur. Serial in vivo micro-CT and BLI were performed for 20 days. Ex vivo BLI images of both femurs were obtained. Differences in the Hounsfieldunit (HU), HUratio, and luciferase activities were compared using Wilcoxon signed-rank tests and non-parametric longitudinal analyses (p<0.05). Results: In vivo BLI revealed a signal drop on day 2, reconstitution on day 5, and continuous decrement thereafter. Ex vivo BLI revealed residual activity in the ADC-implanted and adjacent areas. No activity was detected in the contralateral femur. The overall increment rate of normalized HUs was higher for ADC-treated femurs than for SBMP-treated femurs. Cell migration to distant injury sites was not detected. Conclusion:Enhanced bone density in the implant area suggests that ADCs have fracture-healing effects.

        Treatment Outcomes of Transurethral Macroplastique Injection for Postprostatectomy Incontinence

        Purpose: We investigated the efficacy of transurethral injection of Macroplastique bulking agent (Uroplasty) for male stress urinary incontinence (SUI) after prostate surgery. Materials and Methods: This retrospective review included men with SUI treated by transurethral injection for symptoms resulting from prostate surgery. Patients were evaluated at 1 month and 6 months after injection by determining the number of pads used per day and changes in incontinence symptoms. Treatment success was defined as use of 1 pad or fewer per day combined with subjective symptom improvement. Results: The study population comprised 30 men with a mean age of 66.1±5.3 years. Of the 30 patients, 24 (80.0%) underwent prostate cancer surgery and the remaining 6 (20.0%) underwent surgery for benign prostatic hyperplasia. The preinjection pad number was 2.9±1.9 pads per day. After injection treatment, the mean follow-up period was 9.3±12.7 months and the success rate was 43% (13/30) at 1 month and 32% (6/19) at 6 months. Injection was more likely to result in a successful outcome in patients with no preinjection radiation treatment history and higher abdominal leak point pressure (ALPP) than in those with a previous history of radiation treatment and lower ALPP, although this result was not statistically significant. Acute urinary retention occurred in 5 patients (17%). Conclusions: Transurethral Macroplastique injection treatment is a relatively noninvasive treatment method for male SUI with a success rate of 43% at 1 month and 32% at 6 months. Patients with a higher ALPP and no previous history of radiation therapy may experience better treatment outcomes.

        반복배열된 토마토 phenylalanine ammonia-Iyase(p AL X1, PAL X2) 유전자의 구조해석

        토마토의 genome내에는 적어도 5개 이상의 PAL유전자 좌가 존재한다는 사실을 이 등(1992)이 이미 genomic Southern blot hybridization으로 확인하여 보고하였다. 그러나 본 연구에서 제작한 genomic DNA libraries를 대상으로 검색한 결과 기존에 보고된 PAL유전자 이외에 약 15 kb 와 10 kb에 해당하는 큰 EcoRI 단편을 확보 할 수 있었다. 이들 단편을 BamHI, HindIII등 9종의 제한효소를 사용하여 Southern blot hybridization을 행한 결과 PAL X1의 경우는 모든 효소에 대하여 2개의 단편이 hybridization 되었으며, 특히 BamHI으로 절단하여 얻은 3개의 단편중 두 개는 PAL5 유전자의 exon 2 부위에서 취한 oligomer(18 mer)와 primer extension 반응이 진행되어서 약 200 bp의 PAL유전자와 아주 높은 상동성을 갖는 염기서열이 확인되었다. 따라서 PAL X1유전자는 2 copy의 유전자가 나란히 존재하거나 아니면 염색체 재배열이 진행된 것으로 추정된다. 이러한 결과는 적어도 7개 이상의 PAL유전자 좌가 토마토 염색체내에 존재하는 것으로 사료된다. We observed the structure of phenylalanine ammonia-lyase gene (PAL) which is one of the best studied plant defense-related genes responding to pathogen infection by producing suberin, lignin, and phytoalexins. In tomato, at least 5 different genetic loci have been identified by genomic southern blot hybridization and nucleotide sequence analyses of partially cloned gene fragments (Lee et al. 1992). However, our results suggest that two other isoforms designated as PAL X1 and PAL X2 are located on the chromosome in tomato plant. Furthermore, the preliminary results obtained from southern blot hybridization analyses of subcloned fragment digested with several restriction endonuclease indicated that PAL X1 and PAL X2 clones contain at least two copies of PAL gene and partial nucleotide sequence analyses of each subcloned fragment with the same primer taken from known nucleotide sequence of PAL5 gene indicated that they are located side by side on the same chromosome.

        당귀(Angelica spp.)의 기원분석에 관한 분자생물학적 연구 현황 및 향후과제

        당귀는 우리나라를 포함하여 중국과 일본 등 아시아국가에서 유용하게 이용되는 한약재이다. 그러나 국가마다 그 기원을 달리하기 때문에 혼 오용이 심하고 국제시장에서 혼란을 불러일으킬 소지가 다분하다. 따라서 오래전부터 형태학적, 세포유전학적 분석과 지표성분을 이용한 화학적 판별 마커의 개발에 관한 연구가 많이 진행되어 왔다. 또한 최근에는 다양한 재배환경과 수확 후 가공 및 처리방법에도 비교적 안전한 유전자 단편의 염기서열 비교분석을 통한 분자생물학적 기술을 적용한 판별기술의 개발에 관한 연구결과 들이 발표되고 있다. 그러나 아직까지 이들 기술의 실용화를 통한 현장 적용에는 한계가 있으며 보다 많은 후속연구가 수행되어야 한다. 이에 본 논문에서는 현재까지의 연구결과를 바탕으로 얻어진 문제점을 논하고 향후 필요한 추가 연구 과제들에 관하여 기술하였다. The dried root of Angelica species is used in traditional Chinese medicine in East Asia, particularly in Korea, China and Japan. Since the plant origin differs in these countries, they are often misused or adulterated in the commercial markets, resulting in distrust among the consumers. Enormous efforts have therefore been focused to distinguish the origin for the Angelica genus, by using morphological or cytogenetical analyses, and chemical markers based on biochemical analyses of secondary metabolites. DNA is considerably stable against different cultivation conditions, and to treatment and processing after harvesting of plants. Hence, several researches have been filed for the development of molecular markers, based on the single nucleotide polymorphisms in specific regions of DNA. However, there are several obstacles for application in the commercial markets, concerning the reproducibility, accuracy, sensitivity, and rapidity of these tests. In this review, we summarize the research achievements that help classify the origin of Angelica species, in particular, Angelica gigas Nakai. A. sinensis(oliv.) Diels, A. acutiloba Kitag., and A. acutiloba var. sugiyamae Hikino. Further researches are required for practical applications.
