        발생중인 병아리의 사이뇌에서 NPY 양성반응세포의 분포

        This study deals with the distribution and morphological study of the neurons with the neuropeptide Y (NPY) in developing chick brain. The developing brains of Korean native chicks at embryonic days 8 (E8), E10, E12, E14, E16, E18, and E20 were used. The chicks were perfused with 4% paraformaldehyde through the left ventricle and aorta. The brains were removed and transferred into 30% sucrose, and then cut in a cryostat into $60{\mu}m$ in thickness. The sections were immunostained with free-floating and avidin-biotin complex (ABC) methods. The numerous neurons with NPY were first observed in nucleus rotundus of diencephalon at E8. In particular, neurons in nucleus rotundus had the well-developed processes. At E12, the neurons with NPY were distributed widely in diencephalon; nucleus septalis lateralis, medialis, nucleus magnocelluaris preopticus dorsalis, medialis, ventralis, nucleus preopticus medialis, nucleus dorsolateralis anterior thalami pars magnocellularis, and nucleus paraventricularis magnocellularis (paraventricular nucleus) except nucleus rotundus. From E12 to E20, positive neurons were oval-shaped, changed gradually into the spherical- and multipolar-shaped. The shapes of processes were also omnidirectional and the number of those were less than in telencephalon. As the chicks developed, the morphology of neurons with NPY showed the tendency to increase in their sizes and numbers.

        한국산 무우로부터 Far Migrating Anionic isoperoxidase A3 의 정제 및 성질

        A far migrating anionic isoperoxidase, named A₃, was isolated from Korean radish (Raphanus sativas L.) root. Purification of the enzyme was accomplished by CM-cellulose chromatography, DEAF-Sephacel chromatography and Sephadex G-75 gel filtration. The enzyme was a glycoprotein having molecular weight of approximately 50,000 as determined by SDS-PAGE and Sephadex G-150 gel filtration and the pI value was 3.0. The optimum pH of the enzyme was 6.0 for guaiacol and H₂O₂and the Km values for guaiacol and H₂O₂ were 8 mM and 1 mM, respectively. Substrate studies with natural phenolic compounds were performed and their Km values were determined. In comparison with other Korean radish isoperoxidases, A₃ had higher affinity for esculetin. A particular interest is that the enzyme showed as sigmoidal substrate saturation curve against scopoletin despite of its single Subunit structure.

        아가리쿠스로부터 분리한 $\beta$-glucan과 그 올리고당류의 HT-29 인체 대장암 세포에 대한 항암 활성에 관한 연구

        칡을 첨가하여 배양한 아가리쿠스버섯(Agaricus blaxei Murill)으로 부터 열수추출, gel filtration chromatography, DEAE ion exchange chromatography를 통하여 아가리쿠스 $\beta$-glucan (AG)을 추출하였다. 추출한 아가리쿠스 $\beta$-glucan에 Bacillus megaterium 유래의 endo-$\beta$-(1$\rightarrow$6)-glucanase를 처리하여 올리고당류(AO)를 얻었다. 이렇게 얻은 AG와 AO를 이용하여 HT-29 인체 대장암 세포에 대한 항암 활성을 조사한 결과, 암세포의 성장 억제 효과는 시료의 처리 농도에 의존적으로 증가하였으며, apoptosis assay에서 암세포의 apoptosis 유발이 농도에 의존적으로 증가되었다. 또한, 암세포의 세포 주기를 분석한 결과, apoptosis 발생을 뜻하는 G0 (sub-G1)기와 G1기의 비율이 증가한 반면 S기와 G2/M기는 대조군에 비해 감소되었다. 이러한 결과를 바탕으로 암세포의 apoptosis 증가에 대한 AO의 작용이 어떤 유전자와 연관이 있는지를 알아보기 위하여 cDNA microarray를 통해 유전자의 발현율을 검색한 결과, apoptosis의 내부 외부 경로에 영향을 주는 유전자(TNESE9, TNFRSF9, FADD, CASP8, BAD, CRADD, CASP9 등)의 발현이 증가되었고 세포 분열 주기의 진행과 관련된 유전자(CCND2와 CDK2)의 발현이 감소되었으며, 세포 분열 주기를 지체시키는 유전자 (CDKN2A)의 발현은 증가되었다. 또한, 사이토카인을 암호하는 유전자 (IL6, IL18, IL6R 등)와 tumor suppressor와 관련된 유전자 (CEACAM1, TP53BP2, IRF1및 PHB)의 발현이 2배 이상 증가된 것을 확인할 수 있었다. 따라서 HT-29 인체 대장암세포에 대한 AO의 성장 억제 작용은 G0/Gl기를 지체시켜 암세포 증식을 억제하고 apoptosis에 의해 암세포를 사멸시키는 항암 활성을 나타내는 것으로 확인되었으며, 특허 AO가 AG보다 현저한 활성을 보였다. 더 나아가 아가리쿠스 $\beta$-glucan (AG)과 올리고당류 (AO)는 항암 활성을 가진 대체 의약 소재로 개발될 수 있을 것으로 기대된다. [ $\beta$ ]-Glucans (AG) were prepared from Agaricus blazei cultured in the medium fortified with the roots of Pueraria spp. by repeated extraction with hot water, gel filtration chromatography and DEAE ion exchange chromatography. Oligosaccharides (AO) were derived from the hydrolysis of AG by an endo-$\beta$-(1$\rightarrow$6)-glucanase from Bacillus megaterium. The anti-HT-29 human colon cancer activity of AG or AO was investigated using MTT assay, apoptosis assay, cell cycle analysis, and cDNA microairay. AG and AO both inhibited proliferation and growth of HT-29 cells, and stimulated apoptosis of the cells in a dose-dependent manner. In cell cycle analysis, treating HT-29 cells with AG or AO resulted in the increase of cells in the G0 (sub-G1) and G1 phase. Especially, AO was more effective in inducing G0/G1 cell cycle arrest than AG. To screen the genes involved in the increase of apoptosis, the gene expression profile of the HT-29 cells treated with AO was examined by cDNA microarray. While several genes involved in cell cycle progression (CCND2 and CDK2) were down-regulated, many genes involved in apoptosis (TNFSF9, TNFRSF9, FADD, CASP8, BAD, CRADD, CASP9 etc), cell cycle inhibitor (CDKN2A), immune response (IL6, IL18, IL6R etc), and tumor suppressor (CEACAM1, TP53BP2, IRF1, and PHB) were up-regulated. These results suggest that AO could inhibit the proliferation and growth of HT-29 cells by G0/G1 cell cycle arrest and induction of apoptosis.

        Ag Pastes의 분산 특성 및 스크린 인쇄된 OTFTs용 전극 물성

        We have fabricated the source-drain electrodes for OTFTs by screen printing method and manufactured Ag pastes as conductive paste. To obtain excellent conductivity and screen-printability of Ag pastes, the dispersion characteristics of Ag pastes prepared from two types of acryl resins with different molecular structures and Ag powder treated with caprylic acid, triethanol amine and dodecane thiol as surfactant respectively were investigated. The Ag pastes containing Ag powder treated with dodecane thiol having thiol as anchor group or AA4123 with carboxyl group(COOH) of hydrophilic group as binder resin exhibited excellent dispersity. But, Ag pastes(CA-41, TA-41, DT-41) prepared from AA4123 fabricated the insulating layer since the strong interaction between surface of Ag powder and carboxyl group(COOH) of AA4123 interfered with the formation of conduction path among Ag powders. The viscosity behavior of Ag pastes exhibited shear-thinning flow in the high shear rate range and the pastes with bad dispersion characteristic demonstrated higher shear-thinning index than those with good dispersity due to the weak flocculated network structure. The output curve of OTFT device with a channel length of 107 ${\mu}m$ using screen-printed S-D electrodes from DT-30 showed good saturation behavior and no significant contact resistance. And this device exhibited a saturation mobility of $4.0{\times}10^{-3}$ $cm^2/Vs$, on/off current ratio of about $10^5$ and a threshold voltage of about 0.7 V.

        한국산 무우 묘목의 성장에 따른 Peroxidase isozyme 들의 조직특이성 발현

        Changes in the pattern of isoperoxidases and the specific activity of peroxidases were examined from cotyledone, hypocotyl and root portion of the Korean radish seedlings grown for 3, 5 and 7 days. The specific activity of peroxidases was very high in the root compared with that of hypocotyl and cotyledone portion of the seedlings and peroxidase isozyme patterns in these three organs were changed during entire growth periods. Total cellular RNA was isolated from hypocotyl and root portion of the seedlings with guanidium thiocyanate and guanidine HCl method. The total RNA was further fractionated by oligo (dT)-cellulose chromatography. For translation of peroxidase mRNAs in cell-free protein synthesizing system, rabbit reticulocyte lysate was tested and the optimum translational conditions of peroxidase mRANs were determined. The poly(A) +RNA and poly(A)-RNA isolated from hypocytyl and root portion of the seedlings grown for 5 and 7 days were translated in vitro and the correlation between the changes in the expression pattern of isoperoxidases and qualitative and quantitative changes in peroxidase mRNA populations was investigated. The peroxidase activity of poly(A)-RNAs was much higher than that of poly(A)+RNA in seedlings grown for 7 days when compared with that of poly(A)-RNAs from seedlings grown for 5 days.

        담배조직배양으로부터 isoperoxidase 의 분리 및 성질

        Two anodic isoperoxidases, named isoperoxidase A₃ and A₄, were isolated from tobacco callus line, Nicotiana tabaccum L., var. Virginia 115. Isolation of the enzymes was accomplished using DEAE-cellulose ion exchange chromatography and Sephadex G-150 gel filtration. The molecular weight of isoperoxidase A₃ was approximately 38,000 as determined by SDS polyacrylamide gel electrophoresis and 38,500 estimated with Sephadex G-150 gel filtration. The molecular weight of isoperoxidase A₄ was 43,000 as determined by SDS polyacrylamide gel electrophoresis and Sephadex G-150 gel filtration. The optimum pHs for guaiacol with isoperoxidase A₃ and A₄ were pH 6.0 and pH 7.0, respectively, and these two isoperoxidases had same K_m value of 5 mM for guaiacol. Substrate studies with natural phenolic compounds such as scopoletin, esculetin, ferulic acid and chlorogenic acid were performed and the K_m values for these substrates were 0.18 mM, 0.24 mM, 0.26 mM and 0.35 mM, respectively, in case of isoperoxidase A₃. Such K_m values did not differ greatly from those of isoperoxidase A₄ from Virginia strain and known scopoletin peroxidase A_f from WR-132 strain. In comparison with isoperoxidase A₃ from W-38 strain and A_f from WR-132 strain which were known as scopoleticn peroxidases, these two enzymes from Virginia strain showed notable differences in scopoletin oxidizing ability in terms of relative substrate oxidation ratios.

        정상 성인에서 복부 드로우-인 기법을 동반한 복부근 활성화가 파악력에 미치는 영향

        Purpose: The grip strength is influenced by various factors, such as position of the upper extremity, characteristics of the hand, and general physical condition. In this study, we investigated whether abdominal muscle activity in combination with the abdominal drawing-in maneuver has any effect on the grip strength in healthy young adults. Methods: This study included 31 healthy subjects (16 males, 15 females). We used surface electromyography and pressure bio-feedback unit for this experiment. All Subjects were placed in a cock-lying position with comfort and the grip strength was measured. On the following day, the pressure bio-feedback unit was placed beneath their fifth lumbar vertebra, and the, grip strength was measured again. This time, the measurement was taken while drawing-in their abdomen below the navel gently and gradually, while maintaining a neutral pelvic position. Results: The grip strength was significantly increased when subjects performed the drawing-in maneuver than when they were comfortable (p<0.05). In addition, activations of the rectus abdominal muscles significantly increased (p<0.05). Conclusion: In the current study, we found that abdominal muscle activity, in combination with the abdominal drawing-in maneuver, affected the grip strength, positively. Therefore, we suggest that this result should be considered when evaluating the grip strength.
