Use of transrenal DNA for the diagnosis of Tuberculosis

There have been attempts to use biological samples other than sputum for the diagnosis of TB. Urine is promising clinical specimen because of its availability, ease of access, processing and storage, and the low infection risk to healthcare workers during sample collection. In this study we evaluated the outcomes of urine diagnostics in hospitalized MDR, XDR pulmonary TB patients. In order to detect TB-specific transrenal DNA, 10 urine samples were collected during the study period, and quickly added to 1 ml of a stabilization buffer to a final concentration of 10 mM, in order to minimize degradation of soluble DNA by nucleases. DNA was extracted from 8ml of whole urine and eluted in 25ul with an automated system based on magnetic silica particle technology. We used droplet PCR, it might be an ideal alternative to conventional real-time PCR for microorganism detection. We aimed to assess the capacity of ddPCR for detecting low levels of circulating Mycobacterium tuberculosis DNA target (known as IS6110). ddPCR was found to provide results in good agreement with the other methods tested and to be highly reproducible.In this study, the results show that ddPCR could be used to measure low levels of MTB DNA, and it has the potential to be used to diagnose pulmonary TB based on clinical samples. The application of the Tr-DNA test could provide a significant advance in the diagnosis of TB. The availability of new method using urine that is more convenient and accessible may greatly usefulness for individual hospitalized patients for monitoring their treatment process.

Every artist tries to attempt to search for 'self' through his own medium. It is the same with a writer, especially a poet. W.B.Yeats, a poet and poetic dramatist, had also tried to achieve 'Unity of Being' through his poems and poetic dramas. But in the case of Yeats, the development process of his ideas and theories is said to have played an important part in his search for self, that is, Unity of Being. The purpose of this paper is to examine and define the search for self through the development process of his ideas and thoeries. Yeats's dissatisfaction with science, materialism and empiricism led him to join the Dublin Hermetic Society in 1886 in which he acquired "the doctrine of reincarnation", "the theory of correspondences," and the idea that "all existences are arranged cyclically" from Madame Blavatsky, the leader of the Society. Turning against the Society's asceticism and Blavatsky's dogmas, Yeats took part in the so-called "the Order of Golden Dawn" led by MacGregor mathers. Yeats was initiated into Mathers' Kabbalism, European Rosicrucianism. So he came to believe that there exists the symbolic system in the process of human soul. He also studied William Blake and was interested in Blake's ideas of "the four Zoas" and "the four fold vision." He developed Blake's ideas into his own theory of "Quarternity" as well as into his idea of four Faculties (Will, Mask, Craetive Mind, and Body of Fate). In 1925, he compiled the ideas and theories which he learned from various sources into a systematic organiation in A Vision, in which he described his vision chiefly in terms of the idea of the "Great Wheel". Yeats's vision of cyclical progress of human soul revealed in the idea of the "Great Wheel" is thus not only the ground of his understanding of human life and national history but a part of his search for self, his Unity of Being.
Complete genome sequence of probiotic Lactobacillus johnsonii IDCC9203 isolated from infant feces

Lactobacillus johnsonii IDCC9203 (= KCTC 10923BP) is a probiotic with anti-inflammatory properties, both in vitro and in vivo. Here, we report the complete genome sequence of the IDCC9203 strain, which was isolated from infant feces. The genome was determined using PacBio and Illumina sequencing. This strain comprises a circular chromosome of 1,898,461 bp, and its guanine-cytosine (GC) content is 34.70%. These results will provide an understanding of this strain’s functional properties and safety assessments.
A Different Perspective on the P-value Problem with Big Data

Recently, information system researchers have become used to handling big data comprising more than 10,000 observations. Researchers using statistical inference in such very large samples have often found that p-values quickly go to zero as the sample size increases. This study investigates the p-value problem with large sample via central limit theorem. The results revealed that strong positive dependency of big data may be the main cause behind the problem.
