Rhodopseudomonas sphaeroides의 고정화균체에 의한 수소생산의 효율적 기질공급

Rhodopseudomonas sphaeroides B6 세포를 agar gel 중에 고정했을 때, 수소생산을 위한 최적 agar 농도는 2%(w/v)였다. B6의 2% agar gel 고정화균체(300㎖ gel: 2.85 ㎎ dry cells/㎖)와 비고정세포(1ℓ culture: 0.87㎎ dry cells/㎖)에 있어서 초기의 최고 수소 생산활성은 각각 47.5 및 48.O ㎖/hr/culture로 거의 같았다. 그러나, 수소생산이 거의 정지된 배양후기에 lactate의 제한공급(10mmole)에 의한 비고정세포의 활성회복은 50% 이하로 감소되었지만, 고정화세포의 활성은 거의 초기상태로 회복되었다. B6의 고정화균체를 이용하여 12시간 주기의 광조사 조건으로, 매 1ℓ의 수소생산시 마다 그에 소비된 만큼의 기질에 상당하는 9.3mmole의 DL-lactate와 1.86 mmole의 L-glutamate 함유 기본배지를 주기적으로 공급한 결과, 228시간의 배양기간 동안 명암의 반복 조건에도 불구하고 평균 510 ㎖/day/300㎖ ge1(2.9 ㎎ dry cells/㎖)의 속도로 수소생산이 지속되었다. 이 결과는 광합성세균에 의한 수소생산에 있어서, 보다 효율적 기질공급과 가변적인 태양광 이용 조건에 대응되는 자동배양 시스템의 구성 가능성을 시사한다. The Photosynthetic bacterium, Rhodopseudomonas sphaeroides strain B6 was immobilized on agar gel. The optimum concentration of agar for hydrogen production was 2% (w/v). Maximum rates of hydrogen production by immobilized (300㎖ of gel; 2.85㎎ dry cells/㎖) and free cells (1ℓ culture; 0.87㎎ dry cells/㎖) were 47.5 and 48.0 ㎖/hr/culture, respectively. However, when both cultures were fed by 10mmoles of lactate as limited electron donor at the later period of incubation, the activity of hydrogen production by free cells was significantly decreased but, immobilized cells continued hydrogen production with almost the same initial rate. We examined hydrogen production by immobilized cells of strain B6 under periodic illumination for 12 hr-intervals. When the culture was periodically fed by basal medium containing 9.3 mmoles of DL-lactate and 1.86 mmoles of L-glutamate as consumed electron donor and nitrogen source, respectively, for every one liter of hydrogen produced, hydrogen was evolved continuously with the average rate of 510 ㎖/day/300 ㎖ gel (2.9 ㎎ dry cells/㎖) during the incubation time for 228 hr.
Rhodopseudomonas sphaeroides 고정화균체의 자동조절 옥외 반연속배양에 의한 수소생산

광합성세균에 의한 광의존 수소생산에 있어서 상시 가변적인 태양광 이용조건에 대응되는 자동조절 반연속배양장치를 구성하였다. 이는 pulse 발생장치를 설비한 가스메터에 의해 일정의 수소생산량이 측정되면, 수소생산에 소비된 만큼의 기질이 timer와 연결된 정량펌프에 의해 공급될 수 있는 시스템이다. 이 배양장치를 이용하여, 옥외조건에서 Rhodopseudomanas sphaeroides B6의 agar gel 고정화균체에 의한 수소생산 실험을 시도하였다. 일조와 기후의 격심한 변동에도 불구하고 배양이 잘 조절되어 수소생산성이 유지되었다. 이는 광합성세균의 수소생산을 위한 옥외반연속배양의 자동조절 가능성을 나타내는 것이다. For the photoproduction of molecular hydrogen by photosynthetic bacteria in outdoor conditions, we constructed automatically controlled semi-continuous culture system. When the amout of hydrogen gas produced can be measured by a gas meter with a pulse generator, the same amout of substrate consumed for hydrogen production could be supplied by micropump related with timers. Using the apparatus, we examined hydrogen production with immobilized cells of Rhodopseudomanas sphaeroides B6 in outdoor conditions. In spite of severe fluctuation of weather and illumnination, the culture was maintained under good control with regard to hydrogen productivity. It was possible to automate the semi-continuous outdoor culture of photosynthetic bacteria for hydrogen production.
신경로추적자로 바이러스를 이용한 흰 쥐 귀밑샘지배 중추신경핵의 면역조직화학적 동정

This Study was carried out to investigate the secondary cental nuclei innervating rat parotid gland. PRV-Bablu as a neuronal tracer was injected into the left parotid gland and brains obtained through cardiac perfusion ere treated by immunohistochemical staining The results were as follows : 1. The secondary central nuclei innervating rat parotid gland were paraventricular nucleus and cental part of amygdaloid complex largrly in diencephalon. 2. The paraventricular nucleus and cental part of amygdaloid complex in diencephalon showed morphological asymetry between PRV-Bablu injected site and uninjected one. 3. The Ratio between total neurons and PRV-Bablu infected nerons in paraventricular nucleus was 27.62+16.23% in left and 12.78+8.69% in right. 4. The Ratio between total neurons and PRV-Bablu infected nerons in central part of amygdaloid nucleus was 14.25+9.26% in left and 8.35+6.26% in right.
가토(家兎)에서 nitrite에 의한 methemoglobinemia에 미치는 selenium의 영향(影響)

The protective of influences of sodium selenitc ($Na_2SeO_3$) against the methemoglobinemia with sodium nitrite were investigated on hemoglobin, methemoglobin, glutathione peroxidase and NADH-methemoglobin reductase activity in rabbits which were given 0,1,3 and 9ppm sodium selenite of drinking water for a week. Dietary selenium did not alter total hemoglobin in the blood of rabbits. Selenium was found to decrease nitrite-induced methemoglobin in a dose-dependent manner. The glutathione peroxidase activity was also increased by selenium in all the experimental groups. However, the NADH-methemoglobin reductase activity by selenite did not show significant differences as concerns the methemoglobinemia. These results showed that selenium could inhibit nitrite-induced methemoglobinemia. Its influence of inhibition is suggested that the effect of the reduction of methemoglobin was greatly stimulated by glutathione peroxidase activity.
Guinea pig에서 alcohol과 paraquat에 의한 갑상선 독성에 미치는 selenium의 방어 효과

This study examined the effect of alcohol(AL) and/or paraquat(PQ) on serum TSH, thyroid hormones and enzyme activities, and the protective effect of selenium(SE) againse alcohol and/or paraquat-induced thyroid toxicity in guinea pigs. The experomental group consisted of control, 15% alcohol(AL), 4ppm sodium selentite(SE), 200ppm paraquat(PQ), AL+PQ, AL+SE, PQ+SE and AL+PQ+SE mixed in drinking water-fed guinea pigs for 4 weeks. The morphological changes of thyroid gland were studies on paraffin-embedded sections stained with H-E stain. Body weight losses, high serum concentration in TSH and cholesterol, and low values on triiodothyronine($T_3$), thyrozine($T_4$), free $T_4$ and alkaline phosophatase(ALP) were produced in the groups fed AL and/or PQ. We also noted that AL+PQ-fed group was marked increase in serum TSH. In AL or AL+PQ-fed groups when cpmpared to control group had increased the ratio of thyroid weight to body weight(ratio Twt/Bwt), whereas the ratio Twt/Bwt was decresed in SE or PQ-fed groups. However, the serum TSH, $T_3$,$T_4$ free $T_4$ and cholesterol values, and the ratio Twt/Bwt were reversed in groups given the combination of SE, compared with AL and/or Pq-fed groups, also ALP values were reversed in groups given the combination of SE, compared with AL or AL+PQ-fed groups. In microscope, morphological changes showed a remarkable between the AL or PQ-fed group and controls. In AL+PQ+SE-fed guinca pig, follicular colloid is high density in thyroid follicle and increased in connective tissue around the thyroid cells, and thyroidal epithelia were composed of cuboidal or columnar epithelium. The indicated that the morphological changes of thyroid were direct action in the thyroid cell. The results of this study confirmed that the toxic effect of AL or PQ on thyroid occur independently of changes in liver function, and that SE confers marked protection against AL or PQ-induced thyroid toxicity.

The Internet is an important tool in contemporary society. This study was carried out to investigate current use of internet and to suggest effective development of it in the field of physical therapy. A sample of physical therapy members in the internet responded to the questionare by means of e-mail from 26, April to 14, May, 1999. Of the 98 subjects, 26 completed the questionnaire, a response rate of 26.53%. The results were as follows. The most important problem to use internet in the field of physical therapy was poor information and abscence of database. The exchange of information among members, search database, virtual lecture system and distance treatment counseling in the future were expected. Ail members, professors of university and college, physical therapists in clinic and students must cooperate to construct database and useful information.
