      • A Study on Storage of a Major Herbal Medicine Material, Peucedani Radix

        To develop optimal storage method of root of Peucedani Radix, which has been grown as major cultural herbal medicine material, root of Peucedani Radix was stored for 10 months with different packing materials and sealing methods. The loss in dry weight as influenced by packing materials and sealing methods was the lowest at vacuum packing and followed by complete sealing methods with transparent polyethylene film. The ratio of root rot during the storage period was not significantly different between packing materials but was significantly different between sealing conditions. Conclusively vacuum packing and complete sealing with polyethylene film appears to be optimal for storage of Peucedani Radix.

        전호(前胡)가 Bleomycin에 의한 폐 섬유화에 미치는 영향

        Objectives : Idiopathic pulmonary fibrosis (IPF) is chronic fibrotic interstitial pneumonia and the pathogenesis is unknown. Peucedani Radix is well-known for the treatment of respiratory diseases and pulmonary hypertension. This study was to evaluate the effectiveness of Peucedani Radix on the bleomycin-induced lung fibrosis model (BLFM) in mouse. Methods : We induced lung fibrosis by intratracheal instillation of bleomycin in C57BL/6J. We compared two groups BLFM without Peucedani Radix (group I) and BLFM with Peucedani Radix (group II). We performed bronchoalveolar lavages (BAL) and obtained lung specimens from both group I and II on the 7th (A) and 21st (B) day, and also for the normal group. We compared with group I and II to find BAL by using ANOVA test and to find pathologic symptoms by using semiquantitative histological index (SHI). Results : In BAL, total cell counts, lymphocytes, and neutrophils was increased in both group I and II comparing with normal group. However, lymphocyte level was decreased more in group IIB than group IB. It was statistically significant. In microscopic findings, scores of SHI in normal group, group IB and IIB were 0.33, 4.47, and 1.96 each. Conclusions : Peucedani Radix might have inhibitory effect on lung fibrosis by reducing inflammatory cells in bleomycin-induced lung fibrosis model in mouse.

        방풍, 식방풍의 대사체 프로파일링을 통한 지표성분 선정 및 분석법검증

        방풍(Saposhnikoviae Radix, SR) 과 식방풍(Peucedani Japonici Radix, PR)은 각각 다른 기원식물을 가지며 속과 종이 다르지만 한국, 중국, 및 일본에서 대표적인 전통 한약재로 오랫동안 혼용되어 사용되었다. 본 연구에서는 방풍과 식방풍의 판별 마커를 확인하기 위하여 UPLC-QTOF/MS를 이용한 대사체 프로파일링 및 다변량 통계분석을 진행하였다. 그 결과, 5-Omethylvisammioside와 peucedanol을 각각 방풍과 식방풍의 지표성분으로 선정하였으며, UPLC를 이용하여 분석법을 검증하였다. 제안된 방풍 및 식방풍의 지표성분에 대한 분석법 검증은 방풍과 식방풍의 분류와 품질 평가 및 성분 검증을 위한 효과적인 방법이 될 것으로 기대된다. Saposhnikoviae Radix (SR) and Peucedani Japonici Radix (PR) have been used as the main traditional herbal medicines in Korea, China and Japan. In this study, ultraperformance liquid chromatography coupled to quadrupole time of flight mass spectrometry (UPLC-QTOF/MS)-based metabolomics was applied to evaluate the quality of SR and PR using the marker compounds. In the S-plot of SR and PR, 5-O-methylvisammioside and peucedanol were selected as a marker compound for SR and PR, respectively. Also, an UPLC method was established and well validated for marker compounds of SR and PR. These results suggested that the established analysis method could be used one of the good methods for the classification and quality assessment of SR and PR.

        식방풍의 미백 및 주름개선 효과 탐색

        Peucedani japonici Radix is the root of Peucedanum japonicum Thunberg and has been used for folk medicine for curing palsy andalleviation of fever, and recently as its leaves are also used for food, its production and consumption are increasing. In the results ofconducting experiments to confirm the efficacy of Peucedani japonici Radix for whitening and anti-wrinkle, by using MeOH extract, itwas divided into CHCl3 and 30% MeOH Fr., and 60% MeOH Fr., MeOH Fr., H2O Fr., and from 30% MeOH Fr., two substances wereisolated and from CHCl3 Fr., one substance was isolated, and as a result of confirming the structure through the results of 1H-NMR, 13C-NMR, FAB-MS, it was confirmed that these substances were guanosine, adenosine, and scopoletin. In order to evaluate the efficacy ofthese isolated compounds for whitening and anti-wrinkle, experiments on tyrosinase inhibition activity and elastase inhibition activitywere conducted. As a result of the experiments, it was confirmed that tyrosinase inhibition activity had relatively superior tyrosinaseinhibition activity efficacy in scopoletin (IC50=67.21.g/mL), compared with kojic acid (IC50= 26.96.g/mL) which was used as thepositive control group. And in the anti-wrinkle efficacy, it was confirmed that scopoletin (IC50=375.12.g/mL), although elastaseinhibition activity efficacy was low, had relatively superior inhibition efficacy, compared with ursolic acid (IC50=66.98.g/mL) whichwas used as the positive control group. Through these results, it was confirmed that Peucedani japonici Radix could be used as mult-functional material for whitening and anti-wrinkle.

        Simultaneous analysis of seven marker compounds from Saposhnikoviae Radix, Glehniae Radix and Peucedani Japonici Radix by HPLC/PDA

        A new combination of high performance liquidchromatography (HPLC) method coupled with photodiodearray (PDA) analysis has been developed for the simultaneousquantitative determination of seven major components inSaposhnikoviae Radix (SR), Glehniae Radix (GR) and PeucedaniJaponici Radix (PR), namely peucedanol 7-O-b-Dglucopyranoside(1), prim-O-glucosylcimifugin (2), cimifugin(3), 40-O-b-D-glucosyl-5-O-methylvisamminol (4), bergapten(5), sec-O-glucosylhamaudol (6), and imperatorin (7). Clearseparation of these seven components were achieved on aPhenomenex Kinetex C18 (250 9 4.6 mm, 5 lm) column bygradient elution of water (A) and methanol (B) as mobilephase. The flow rate was 1.0 mL/min and the UV detectorwavelength was set at 254 nm. The method was successfullyused in the analysis of SR, GR, and PR with relatively simpleconditions and procedures, and the results were satisfactory forlinearity, recovery, precision, accuracy, stability and robustness. The results indicate that the established HPLC/PDAmethod is suitable for the classification of SR, GR, and PR.

        UPLC를 이용한 국내 유통품 전호의 성분패턴 비교 연구

        This study aims to compare and analyze the difference in components of Peucedani Radix distributed in Korea using ultra-performance liquid chromatography (UPLC). Eighteen samples distributed in Korea as Jeonho or Asam were collected and used for this study. Three identified standard medicines, Peucedanum praeruptorum, Angelica decursiva, and Anthriscus sylvestris, were used for comparative analysis of major components with collected samples. In the case of 7 samples from Korea, K1―K7, they were identified as A. sylvestris because they all showed a similar component pattern and included deoxypodophyllotoxin (3). However, it has been confirmed that A. sylvestris is sold under the same Korean name as Jeonho, P. praeruptorum, and A. decursiva, which are the original plants of Peucedani Radix in the Korean Pharmacopoeia. On the other hand, among 11 samples from China, C1―C11, eight samples showed similar component patterns and contained the same marker component (4―6), so they were identified as P. praeruptorum, but the remaining three samples, C2, C4, and C8, showed different component patterns from the three standard medicines.

        前胡藥鍼이 Ovalbumin-induced Asthma Mouse Model에 미치는 영향

        Objectives : The aim of this study was to investigate the effect of Peucedani Radix herbal acupuncture(PR-HA) at St36(joksamni) on ovalbumin-induced asthma in mice. Methods : C57BL/6 mice were sensitized and challenged with OVA(ovalbumin) for 12 weeks(once a week) C57BL/6 mice were injected, inhaled and sprayed with OVA for 12 weeks (3times a week). One of the two experimental groups was just treated with needle-prick on St36 and the other group was treated with 1% concentrations of PR-HAS at St36, for the later 8 weeks (3times /week). Results : 1. The weight and total cells of lung of the mice group treated with PR-HA decreased significantly compared with those of control group. 2. Total Leukocytes and Eosinophils in BALF of the mice group treated wtih PR-HA decreased significantly compared with those of control group. 3. Eosinophils in BALF of the mice group treated wtih PR-HA in Photomicrographs decreased significantly compared with those of control group. 4. According to Histological analysis of lung sections, it decreased significantly adhension of collagen in PR-HA than those of control group 5. The concentration of IgE, IL-4, IL-5, in BALF and IL-4, IL-5, Il-13 in serum of the mice group treated with PR-HA decreased significantly compared with that of control group. 6. The number of Gr-1+/CD11b+, CD11b+, CD3-/CCR3+, CD4+, CD3e+/CD69+ , CD23+/B220+ cells in the lungs of the mice group treated with PR-HA decreased significantly compared with those of control group. 7. The cytokine's manifestation of mRNA of the mice group treated with PR-HA with RT-PCR decreased significantly compared with that of control group. Conclusion : We conclude that PR-HA is effective on OVA-induced asthma of C57BL/6 mouse.

        식방풍의 성분분리 및 생리활성

        In this study, isolation of antioxidative compounds was performed for development of anti-oxidizing agent. $CHCl_3$, $H_2O$, 30%, 60% MeOH, MeOH fractions were examined antioxidative activity by DPPH, test of inhibition on NO production. It was revealed that 30%, 60% MeOH and $CHCl_3$ fractions had significant antioxidative activity. In 30% MeOH and 60% MeOH, $CHCl_3$ fraction, six compounds were isolated and elucidated as adenosine(I), guanosine(II), peucedanol 7-O-$\beta$-D-apiofuranosyl(1$\rightarrow$6)-$\beta$-glucopyranoside(III), peucedanol 7-O-$\beta$-D-glucopyranoside(IV), peucedanol(V) and scopoletin(VI) by physicochemical data and spectroscopic methods. (Negative FAB-MS, $^{1}H-NMR$, $^{13}C-NMR$). The results from antioxidative activity screening for the each compound showed that compound IV was relatively superior antioxidant ability. In anti-inflammatory activation assay, compound III, IV, VI had concentration-dependent-activity and compound IV had superior anti-inflammatory ability. These results suggest that Peucedani Radix might be developed as a potent anti-oxidative, anti-inflammatory agents and ingredients for related functional foods.

        Simultaneous Determination and Recognition Analysis of Coumarins in Angelica decursiva and Peucedanum praeruptorum by HPLC-DAD

        Peucedani Radix is the root of Angelica decursiva Franchet et Savatier (=Peucedanum decursivum Maximowicz) or Peucedanum praeruptorum Dunn in several Asian countries. The coumarins contained in Peucedani Radix were quantitatively analyzed using HPLC-DAD to develop a simultaneous determination for the quality control of A. decursiva and P. praeruptorum. For quantitative analysis, four major coumarins contained in these medicinal plants were assessed. Nodakenin (1), nodakenetin (2), praeruptorin A (3), and praeruptorin B (4) were separated with a Phenomenex Luna C18 column (5 μm, 4.6 × 250 mm) under the gradient conditions using distilled water with 0.1% phosphoric acid and acetonitrile with 0.1% phosphoric acid as the mobile phase, at a flow rate of 1.0 ml/min and a detection wavelength of 330 nm. This method was fully validated for linearity, accuracy, precision, recovery, and limit of detection and quantification. As a result, A. decursiva and P. praeruptorum were clearly classified by the quantification of four major coumarins in extracts. Also, the pattern recognition analysis based on HPLC indicates that all of the samples were largely clustered into two groups. Therefore, it is possible to distinguish between A. decursiva and P. praeruptorum and contribute to quality control.

        난알부민으로 유도된 천식 마우스에서 식방풍(植防風)의 알레르기 면역반응 조절효과 연구

        Objectives: The root of Peucedanum japonicum Thunberg (Peucedani Japonici Radix; PJR) has been traditionally used as an herbal medicine for the treatment of anti-headache, anti-paralysis, anti-cancer, vascular protection, and blood pressure regulation. In this study, we investigated the anti-allergic effect of PJR water extract on ovalbumin (OVA)-induced allergic asthma in mice. Methods : Mice were sensitized at days 1, 8 and 15 with OVA and airway challenged at days 22, 24, 26, 28, and 30 to induced allergic asthma. PJR-W extract at doses of 100 and 300 ㎎/㎏/body weight (bw) was orally administered during OVA challenge once per a day. The levels of allergic mediators such as immunoglobulin (Ig) E, and Th1/Th2 cytokines (IFN-γ and IL-4) were measured in the sera of mice by ELISA. The histological change of lung tissue was observed with hematoxylin and eosin (H&E) staining. Results : The administration of PJR-W extract significantly decreased the serum levels of IgE, IL-4, and IFN-γ compared with those of OVA control group. In H&E staining, PJR-E extract inhibited OVA-induced airway inflammation and the inflammatory cells infiltration in the peribronchial regions of the lung. Conclusions : These results indicate that PJR-W extract has an anti-inflammatory and anti-allergic effect on allergic response through the down-regulation of allergic mediators, suggesting that this herb may be used as a useful source for the treatment of allergic inflammatory diseases such as asthma.
