        환삼덩굴 ( Humulus Japonicus ) 화분에 대한 피부 반응도 및 특이 IgE 의 측정에 관한 연구

        N/A To assess the clinical contribution of Humulus japonicus pollen which is thought to be an important causative pollen of the pollinosis in Autumn as are ragweed and sagebrush in Korea, a total of 340 patients with allergic respiratory disease visiting the allergy clinic in Severance Hospital, Yonsei University College of Medicine from March 1986 to February 1987 were studied. These patients (bronchial asthma 108, allergic rhinitis 97, bronchial asthma with allergic rhinitis 41 and other 94 cases) were tested for their skin reactivity to the extracts of Korean and Torii`s Humulus japonicus pollen, Dermatophagoides farinae, sagebrush, and rag- weed. Micro-ELISA was used to determine the Humulus japonicus-specific IgE level in 120 sera from patients who showed a positive skin test (more than 2+) to the Torii`s Humulus japonicus. The following results were obtained: 1) The global results of the skin prick test to the extract of Korean pollen of Humulus japonicus were negative in 64.0%, 1+ in 22.1%, 2+ in 11.8%, 3+ in 1.8%, and 4+ in 0.3% of 340 cases. The positive skin test rate (more than 2+) among the disease entities for the Korean and Toriis extracts of Humulus japonicus pollens respectively were 14.9% and 8.4/o in bronchial asthma; 17.5% and 21.6% in allergic rhinitis; 17.1% and 26.9% in bronchial asthma with allergic rhinitis; and 7. 7% and 6.3% in the others 2) The positive skin test rate for Korean Humulus japonicus in those patients who showed a positive skin test to Torii`s Humulus japonicus was 42.6% and the positive skin test rate of for Torii`s Humulus japonicus in those patients who showed a psotive skin test to Korean Humulus japonicus was 42.6% also. The correlation between the skin test results of the extracts of Korean and Torii`s Humulus japonicus pollen was very weak (r=0.41, p<0.05). 3) The positive skin test rates for Korean Humulus japonicus in patients who showed positive (more than 2+) and negative (less than 1+) skin tests to D. fariae, sagebrush and ragweed were 23.5%, 6.3% and 33.3%, 9.6% and 46.4%, 9.4% respectively (p<0.01). 4) The cut-off value of optical density (OD) in ELISA for Humulus japonicus-specific IgE was calculated from 30 negative control sera by computing its mean and standard deviation (Mean+2.S.D.=0.012) and forty-five (37.5%) of 120 patients' sera were ELISA positive. And the correlation between wheal size resulting from the skin prick test and the OD value of ELISA for the specific IgE of Humulus japonicus pollen was very weak (r=0.23 with Korean; r=0.12 with Torii`s). 5) The ELISA positive rates in patients who showed a positive skin test (more than 2+) to Korean and Torii s Humulus japonicus respectively were 46.2% and 50.0 %; and the ELISA positive rate in the negative skin test group was 40.3% and 32.5%. 6) The ELISA inhibition test with sera from 4 patients with high OD value (more than 0.1) showed that 2 patients were definitely inhibited upon the addition of the extract of Korean Humulus japonicus pollen extract, demonstrating Humulus japonicus specific-IgE in the patients sera. 7) The correlation between total serum IgE level and the OD value of ELISA was very weak (r=0.17), showing little effect of a high total IgE level on the results of ELISA. The ELISA positive rate was 41.7% in 12 sera with total serum IgE level of more than 500 IU/ml and 40.0% in 75 sera with a level of less than 500 IU/ml. The author could conclude that both skin test reactivity and OD value using micro-ELISA with the extract of the Korean pollen of Humulus japonicus were relatively low and consequently the allergenicity of it was minimal with the above data. The ultimate outcome in the evaluation of the skin test result with OD value in micro-HLISA to determine the specific IgE level must await further exhaustive research using a more highly purified extract of Humulus japonicus pollen.

        Characterization of tyrosinase inhibitory constituents from the aerial parts of <i>Humulus japonicus</i> using LC-MS/MS coupled online assay

        <P><B>Abstract</B></P> <P>In the screening of natural products for the development as cosmetic ingredients, the EtOAc-soluble fraction of <I>Humulus japonicus</I> showed tyrosinase inhibitory activity. HPLC-MS/MS coupled online tyrosinase assay of EtOAc-soluble fraction of <I>H. japonicus</I> characterized the twenty-eight constituents including two unknown ones and their tyrosinase inhibitory activity. Fractionation of <I>H. japonicus</I> using various chromatographic techniques yielded thirty-eight compounds. The chemical structures of isolated compounds were identified by spectroscopic analysis. As characterized by HPLC-MS/MS analysis, we isolated twenty-four predicted compounds and further identified two unknown ones, named humulusides A (<B>1</B>) and B (<B>2</B>). Additional ten compounds were also identified by purification. Tyrosinase inhibitory activity of isolated compounds were evaluated, which was closely correlated with the results from HPLC-MS/MS coupled online tyrosinase assay. Consistent with predicted data, two major compounds, <I>trans</I>-<I>N</I>-coumaroyltyramine (<B>14</B>) and <I>cis</I>-<I>N</I>-coumaroyltyramine (<B>15</B>) showed tyrosinase inhibition with IC<SUB>50</SUB> values of 40.6 and 36.4 μM. Taken together, <I>H. japonicus</I> is suggested as whitening ingredient in cosmetic products.         Growth Rate and Nutrient Content Changes of Humulus japonicus

        The growth pattern and the seasonal changes in nutrient contents of Humulus japonicus were investigated. Stem length of H. japonicus reached the maximum from 240 to 260 Julian date and the median value was found at 255 Julian date. The maximum leaf area was observed from 235 to 248 Julian date and the median value was at 240 Julian date. The maximum growth rate of the stem length ranged from 205 to 227 Julian date. The leaf area showed the maximum growth rate from 196 to 214 Julian date. The median date in the growth rate of the stem length and leaf area was 212 and 205 Julian date, respectively. The growth rate of H. japonicus was related to rainy season and precipitation. Phosphorus and sodium contents of H. japonicus were correlated with maximum potential rate of relative growth. Although stem biomass of H. japonicus was $ 1.5{\sim}3.5$ times larger than that of leaf, N content of the leaf ($4.48{\sim}5.27%$) was about 2 times higher than that of the stem ($2.00{\sim}3.62%$). High content of N might be responsible for the high growth rate of H. japonicus in summer. This result provides valuable information for appropriate timing for the removal of H. japonicus.

        환삼덩굴(Humulus japonicus) 추출물의 항결핵 효과

        환삼덩굴의 항결핵효능을 알아보기 위하여 두 가지의 실험을 실시하였다. 먼저 환삼덩굴의 메탄올 추출물을 만들어 결핵균에 직접 작용시켰을 때 아주 우수한 결핵균살상효과를 보였다. 두번째로 탐식구에 감염된 결핵균에 대한 증식 억제효능이 있는지 알아 보기 위해 사람의 대식구에 메탄올 추출물을 작용시킨 후 결핵균을 감였시켰을 때 대조군에 비해 현저한 결핵균 증식억제 효과를 보였다. 환삼덩굴의 메탄올 추출물로부터 여러가지 용매를 통해 다섯 가지의 분획물을 얻어 같은 실험을 실시하였다. 이들 분획물 중 결핵균 직접 살상효과를 보인 것은 헥산과 에틸아세테이트층이었고, 탐식구에 감염된 결핵균에 대한 증식억제 효능을 보인 것은 부탄올과 물층이었다. 이처럼 환삼덩굴은 결핵균에 대해 직접적인 살상효과 뿐 아니라 탐식구에 감염되어 있는 결핵균의 증식을 억제하는 효과도 보여 주었는데 이와 같은 결과로 볼 때 환삼덩굴로부터 새로운 결핵치료물질 개발을 기대해 볼 수 있을 것으로 기대된다. The present study aimed to evaluate the in vitro antimycobacterial effects of hop plant, Humulus japonicus. Methanol extract of H. japonicus (MeOH extract) showed strong direct bactericidal effects against Mycobacterium tuberculosis in vitro. Furthermore, the MeOH extract significantly inhibited M. tuberculosis growth in human macrophages. When five fractions obtained from MeOH extract were examined using the same methods, the hexane and ethyl acetate fractions showed bactericidal effects against M. tuberculosis in vitro, whereas the butanol and water fractions inhibited M. tuberculosis growth in macrophages. Because H. japonicus extract exhibited antimycobacterial activity against both free M. tuberculosis in culture medium and intracellular M. tuberculosis in human macrophages, this plant might be a good candidate for development of a new anti-tuberculosis drug.

        Online LC-DPPH 측정 시스템을 활용한 환삼덩굴 추출물의 Radical 소거 활성 성분 분석

        Humulus japonicus is a species that disrupts the ecosystem by interfering with the survival of other plants due to its ability to grow attached to other objects or plants and its prolific reproductive capacity. In order to increase the utilization of H. japonicus, an analysis was conducted on the major antioxidant active compounds present in H. japonicus extract. 1,1-Diphenyl-2-picrylhydrazyl (DPPH) radical scavenging activity was measured simultaneously with liquid chromatograph (LC) separation, and then UV spectrum measurement and mass spectrometer (MS) analysis were performed for the identified radical scavenging active compound peak. Through literature review and reference standard analysis, it was confirmed that the active compound was luteolin-7-O-glucoside. In quantitative analysis, the content of luteolin-7-O-glucoside in the extract was found to be 0.702 mg/ml, which is about 1.4% of the dried H. japonicus sample. Luteolin-7-O-glucoside is more water-soluble than luteolin, the aglycone of this compound, and has been reported to have various physiological activities, so it should be treated importantly in research using H. japonicus.

The structures of the new compounds were determined by spectroscopic methods to be divarin-3-O-${\beta}$-glucopyranoside (4), and lariciresinol-9-O-${\beta}$-xylopyranoside (6). Compounds 1 and 3 exhibited moderate cytotoxicity against two human cancer cell lines (SK-OV-3 and HCT15) with $ED_{50}$ values ranging from 8.84 to $8.79{\mu}M$.

        환삼덩굴로부터 분리한 Luteolin-7-O-$\beta$-D-Glucoside의 활성산소 소거능

        We investigated the active oxygen scavenging activity and active compound from Humulus japonicus. The ethyl acetate and butanol fraction exhibited strong scavenging effects on hydroxyl radical. Furthermore active compound was isolated and purified using Amberlite XAD-2 column chromatography and preparative HPLC from ethyl acetate fracton, and major compound was identified as a luteolin-7-O-$\beta$-D-glucoside by the MS, UV, 1H-NMR and 13C-NMR analyses. Luteolin-7-O-$\beta$-D-glucoside exhibited strong scavenging effect on active oxygens such as superoxide radical, hydroxyl radical and hydrogen peroxide.

        <P><I>Humulus japonicus</I> Siebold & Zucc. (HJ) has traditionally been administered to patients with pulmonary disease, skin disease and hypertension in Korea, and it is considered to exert anti-inflammatory, antioxidant, antimicrobial and anti-mycobacterial effects. However, its effects against Alzheimer's disease (AD) have yet to be explored. Thus, this study was carried out to investigate whether HJ has a beneficial effect on the progression of AD in an animal model. A methanolic extract of HJ (500 mg/kg/day) was intragastrically administered to 5-month-old APP/PS1 transgenic (Tg-APP/PS1) mice for 2.5 months. Novel object recognition and Y-maze alteration tests were used to assess cognitive function, and an immunohistochemical assay was performed to assess amyloid β (Aβ) deposition, tau phosphorylation and gliosis. An <I>in vitro</I> assay using a microglial cell line was also performed to investigate the anti-inflammatory effects of HJ. Our results revealed that HJ significantly decreased the mRNA and protein expression levels of tumor necrosis factor-α (TNF-α), interleukin (IL)-1β, IL-6 and inducible nitric oxide synthase (iNOS) induced by lipopolysaccharide in the microglial cell line. The administration of HJ for 2 months improved the cognitive function of Tg-APP/PS1 mice. HJ notably reduced the area occupied by Aβ and neurofibrillary tangles, and the number of activated astrocytes and microglia in the cortex of Tg-APP/PS1 mice. The findings of our study suggest that HJ has the therapeutic potential to inhibit the progression of AD and to improve cognitive deterioration in Tg-APP/PS1 mice.</P>

        자생식물 쑥, 돌외 및 환삼덩굴의 지역별, 재배방법별 생육 특성과 주요 약리 성분

        우리 나라에서 자생하고 있는 식물중 기능성식품원료로 이용 가능성이 높은 쑥, 한삼덩굴, 돌외의 주년생산과 우량계통 선발을 위한 기초자료를 얻고자 이들에 대한 수집종간 및 재배방법간의 재배학적 특성과 성분을 분석(分析)하였던 바 얻어진 주요 결과는 다음과 같다. 1. 쑥의 생산량(건초량)은 수원에서 수집한 뺑쑥에서 제일 많았으나 정유함양(精油含量)은 음성에서 수집한 약쑥과 음성에서 수집한 쑥은 각각 0.15%, 옹진에서 수집한 쑥은 0.12%, 뺑쑥은 0.09%이었다. 2. 돌외의 수량은 관행재배에 비해 지주(支柱)를 세워 덩굴손이 지주를 감아 신장시킨 재배에서 48% 많았고, 돌외의 지상부에서는 rutin이 함유되어 있는 것으로 나타났다. 3. 환삼덩굴의 수량은 계룡상에서 수집한 종이 음성에서 수집한 종에 비해 12% 많았고, 환삼덩굴의 지상부에서 phenol성(性) 성분인 isoquercitrin이 함유되어 있는 것이 정성적으로 확인되었다. This study was conducted to obtain basic information on growth characteristics and major components of three Korean endemic resource plant species, Artemisia sp., Gynostemma pentaphyllum Makino and Humulus japonicus S. et Z. , growing in different areas in Korea. Three geographical types of artemisia, namely Pangssuk, Ongjinssuk and Yakssuk which were collect in Suwon, Ongjin and Eumsong, respectively were compared in yield, and Pangssuk artemisia was higher in yield than the others. However, essential oil content was the highest in Yakssuk artemisia. Dry weight of areal parts of G. pentaphyllum was higher in the cultural method with props for tendril growth than in the conventional cultural method without props. Rutin was detected in areal parts of G. pentaphyllum. Dry weight of areal parts of H. japonicus collected at the Kyeryong mountain in Chungnam province was higher than that collected in Eumsong. A phenolic compound isoquercitrin was detected in areal parts of H. japonicus.
