        Heme oxygenase-1 induced by desoxo-narchinol-A attenuated the severity of acute pancreatitis via blockade of neutrophil infiltration

        <P><B>Abstract</B></P> <P>Heme oxygenase-1 (HO-1) has an anti-inflammatory action in acute pancreatitis (AP). However, its mechanism of action and natural compounds/drugs to induce HO-1 in pancreas are not well understood. In this study, we investigated the regulatory mechanisms of HO-1 during AP using desoxo-narchinol-A (DN), the natural compound inducing HO-1 in the pancreas. Female C57/BL6 Mice were intraperitoneally injected with supramaximal concentrations of cerulein (50 μg/kg) hourly for 6 h to induce AP. DMSO or DN was administered intraperitoneally, then mice were sacrificed 6 h after the final cerulein injection. Administration of DN increased pancreatic HO-1 expression through activation of activating protein-1, mediated by mitogen-activated protein kinases. Furthermore, DN treatment reduced the pancreatic weight-to-body weight ratio as well as production of digestive enzymes and pro-inflammatory cytokines. Inhibition of HO-1 by tin protoporphyrin IX abolished the protective effects of DN on pancreatic damage. Additionally, DN treatment inhibited neutrophil infiltration into the pancreas via regulation of chemokine (C-X-C motif) ligand 2 (CXCL2) by HO-1.         방사선사진의 인덱스-순서 분석을 통한 치아경조직의 탈염화 검출

        Purpose : The purpose of this study was to develop the radiographic technique for detecting the demineralization which is known as indication of dental caries. Materials and Methods : This technique was based on the comparing of multiple radiographs which was irradiated by multiple X-ray spectra. For the meaningful comparing, the multiple radiographs were reconstructed to the dosimetrically consistent images using a standard material. The difference of resulting images of same target with multiple spectra represents the difference of response of material as regards the spectra. Results : We have found about 10% of demineralization of dental hard tissues particularly in the proximal region through the analyzing of differences. Conclusion : Most intriguing thing in this investigation was that the method to analyze difference shows us to an anatomic structure of dental hard tissues even if absolute values of optical density were excluded during the procedures.

        Bucillamine prevents cisplatin-induced ototoxicity through induction of glutathione and antioxidant genes

        <P>Bucillamine is used for the treatment of rheumatoid arthritis. This study investigated the protective effects of bucillamine against cisplatin-induced damage in auditory cells, the organ of Corti from postnatal rats (P2) and adult Balb/C mice. Cisplatin increases the catalytic activity of caspase-3 and caspase-8 proteases and the production of free radicals, which were significantly suppressed by pretreatment with bucillamine. Bucillamine induces the intranuclear translocation of Nrf2 and thereby increases the expression of γ-glutamylcysteine synthetase (γ-GCS) and glutathione synthetase (GSS), which further induces intracellular antioxidant glutathione (GSH), heme oxygenase 1 (HO-1) and superoxide dismutase 2 (SOD2). However, knockdown studies of HO-1 and SOD2 suggest that the protective effect of bucillamine against cisplatin is independent of the enzymatic activity of HO-1 and SOD. Furthermore, pretreatment with bucillamine protects sensory hair cells on organ of Corti explants from cisplatin-induced cytotoxicity concomitantly with inhibition of caspase-3 activation. The auditory-brainstem-evoked response of cisplatin-injected mice shows marked increases in hearing threshold shifts, which was markedly suppressed by pretreatment with bucillamine <I>in vivo</I>. Taken together, bucillamine protects sensory hair cells from cisplatin through a scavenging effect on itself, as well as the induction of intracellular GSH.</P>

        Morin exerts cytoprotective effects against oxidative stress in C2C12 myoblasts via the upregulation of Nrf2-dependent HO-1 expression and the activation of the ERK pathway

        <P>In the present study, we investigated the cytoprotective efficacy of morin, a natural flavonoid, against oxidative stress and elucidated the underlying mechanisms in C2C12 myoblasts. Our results indicated that morin treatment prior to hydrogen peroxide (H2O2) exposure significantly increased cell viability and prevented the generation of reactive oxygen species. H2O2-induced comet-like DNA formation and gamma H2AX phosphorylation were also markedly suppressed by morin with a parallel inhibition of apoptosis in C2C12 myoblasts, suggesting that morin prevented H2O2-induced cellular DNA damage. Furthermore, morin markedly enhanced the expression of heme oxygenase-1 (HO-1) associated with the induction and phosphorylation of nuclear factor-erythroid 2-related factor 2 (Nrf2) and the inhibition of Kelch-like ECH-associated protein 1 (Keapl) expression. Notably, these events were eliminated by transient transfection with Nrf2-specific small interfering RNA. Additional experiments demonstrated that the activation of the Nrf2/HO-1 pathway by morin was mediated by the extracellular signal-regulated kinase (ERK) signaling cascade. This phenomenon was confirmed with suppressed Nrf2 phosphorylation and consequently diminished HO-1 expression in cells treated with a pharmacological inhibitor of ERK. Collectively, these results demonstrated that morin augments the cellular antioxidant defense capacity through the activation of Nrf2/HO-1 signaling, which involves the activation of the ERK pathway, thereby protecting C2C12 myoblasts from H2O2,-induced oxidative cytotoxicity.</P>

        <P>Natural phytochemicals of plant origin, including flavonoids, have been found to be potent antioxidants providing beneficial effects against oxidative stress-related diseases. The present study was carried out to investigate the antioxidant properties of morin, a flavonoid originally isolated from the flowering plants of the Moraceae family. Superoxide dismutase (SOD)-like activity and 2,2'-azino-bis-(3-ethylbenzothiazoline-6-sulfonic acid) (ABTS(center dot+)) radical scavenging activity were determined. We also investigated the cytoprotective effects of morin against hydrogen peroxide (H2O2)-induced DNA damage and apoptosis in V79-4 Chinese hamster lung fibroblasts. Our results demonstrated that morin had strong scavenging effects against ABTS' radicals with enhanced SOD activity, which varied in a dose-dependent manner. Morin was found to reduce H2O2-induced intracellular reactive oxygen species generation and nuclear DNA damage, and it recovered cell viability damaged by H2O2 via inhibition of mitochondrial dysfunction-mediated apoptosis. Notably, the treatment of V79-4 cells with morin markedly enhanced the expression of heme oxygenase-1 (HO-1) but not quinone oxidoreductase-1, which was associated with the increased expression and phosphorylation of nuclear factor-erythroid 2-related factor 2 (Nrf2) and the downregulation of Kelch-like ECH-associated protein 1 expression. Based on our findings, we conclude that morin effectively ameliorated oxidative stress-induced DNA damage through intrinsic free radical scavenging activity and activation of the Nrf2/HO-1 pathway.</P>

      • 탄소섬유시트에 의한 슬래브 보강에 관한 실험적 연구

        Recently, Carbon Fiber Sheets(CFS) have been used for strengthening the deteriorated RC beams and slabs because of its resistant capacity of corrosion and easy repairing works. In this study, the static tests are performed on RC slabs strengthened with CFS. Test results show that ultimate strength of specimens strengthened with CFS is Increased as 6∼37% comparing to the control specimen. Test parameters are the shape of strengthening, the number of sheet layer and the dry condition of concrete. Deflection, failure load, strains of reinforcing bar, concrete and carbon fiber sheet are measured from tests(one points loading). The failure mode and the most suitable spacing and width of CFS are discussed from these measured data. Each of one-on-one strengthening of a pure interval and width of Carbon Fiber Sheet(CFS) 25 centimeters specimen has the advantage of the ultimate load. Because strengthening method of regular intervals than method of the whole check un aspect of crack progres. It has the advantage of an aspect of maintenance management and it has load-resisting similar to method of the whole surface strengthening as a layer is large in number. Because moisture contents of concrete have an effect on strengthening capacity, when CFS was bonded, the primer affirmed for moisture will be developed.

        Polyphosphate가 함유된 근관충전재가 구강세균의 성장에 미치는 영향

        Eliminating the infecting bacteria of the root canal system and preventing reinfection must be the main objectives of all endodontic works. None of commercially available root canal sealers have the properties of desirable tissue compatibility and strong antibacterial activity. The purpose of this study is to develope an ideal root canal sealer using commercially available polyphosphate (polyP), Calgon, which is known to be antibacterial and safe. For the study, resin-type AH26, zinc oxide eugenol-type Tubli-Seal, Ca(OH)2-type Apexit as base sealers for polyp (0∼3%) and para formaldehyde-containing N2 as a control base were selected. Specimens (3×4mm) of the sealers were prepared in a 37℃ incubator far 3 and 10 days and their antibacterial activity against streptococci and black-pigmented anaerobic rods was observed using an agar diffusion method. The result were as follows : 1.Among 3-day-old root canal sealers, N2 as a positive control showed the strongest antibacterial effect, followed by AH26, Tubli-Seal and, Apexit which barely showed antibacterial activity against the test bacteria. In contrast, 10-day-old AH26 showed a greater antibacterial activital than 10-day-old N2. 2.All sealer specimens showed a greater antibacterial activity against black-pigmented anaerobic rods than streptococci. Three-day-old ones appeared to be more antibacterial than 10-day-old ones except for Apexit. 3.As compared to N2, 3-day-old AH26 demonstrated a similar antibacterial activity against black-pig-mented anaerobic rods but to a lesser extent to streptococci. Ten-day-old AH26 showed a greater antibacterial activity against black-pigmented anaerobic rods than 10-day-old N2. 4.As compared to AH26, Tubli-Seal generally revealed a lower antibacterial activity but it showed a greater antibacterial activity aginst S. gordonii Challis. 5.Enhancement of antibacterial activity by polyp was more clearly observed when it was added to Ca(OH)2-based root canal sealers, Tubli-Seal and N2. 6.The addition of polyP enhanced the antibacterial activity of 3-day-old AH26 against S. gordonii G9B (16%) and Challis (29%), and P. gingivalis 2561 (24%) only. Moreover, polyp failed to increase antibacterial activity of 10-day-old AH26 against the test strains but P. gingivalis A7A1-28 (13%). 7.The addition of polyp increased the antibacterial effect of 3-day-old Tubli-Seal on several test bacteria including S. mutans GS-5 (50%), S. gordonii G9B (47%) and Challis (122%), and all the test strains of P. gingivalis (13∼35%) except for 9-14K-1. The addition of polyp to 10-day-old Tubli-Seal increased antibacterial activity of the root canal sealer against most test strains. 8.3-day-old Apexit failed to show antibacterial activity, if any very little against S. mutans GS-5 and Pr. intermedia ATCC 49046. However, polyP increased its antibacterial activity by 50 and 69%, respectively. Increase of antibacterial activity of 10-day-old Apexit by polyP was more clearly observed than that of 3-day-old one. The overall results suggest that polyP may be used in clinic as an antibacterial agent in root canal sealers. However, in order to maximize its antibacterial effect, the composition of sealers should be considered.

        SnO_2-ZnO 계 후막센서 구조에 따른 CO 감지 특성

        CO 기체 감지 특성을 향상시키기 위해서 3 mol% ZnO를 첨가한 SnO_2와 3mol% SnO_2를 첨가한 ZnO의 적층 형태를 변화시켜 연구하였다. 적층 구조는 단일층, 복층, 그리고 이종층 구조로 후막 인쇄법을 사용하여 제작하였다. SnO_2-ZnO계에서 제 2상은 발견되지 않았다. 전도성은 SnO_2에 ZnO를 첨가하면 감소하고, ZnO에 SnO_2를 첨가하면 증가하였다. 측정 온도증가와 CO 기체 유입으로 전도성은 증가하였다. 단층 및 복층의 후막센서 구조의 감도 향상은 없었으나, SnO_2 -3ZnO-ZnO · 3SnO_2·substrate 구조의 이종층 센서의 감도는 향상되었다. 센서 구조에 관계없이 I-V 변화는 모두 직선성을 나타내서 Ohmic 접합 특성을 이루고 있었다. The sensing properties of carbon monooxide were investigated as a function of mixing ratio and the lamination structure of 3mol% ZnO-doped SnO_2 and 3mol% SnO_2-doped ZnO. The lamination structures were fabricared monolayer, double layer, and hetero layer of SnO_2, ZnO, and theirs mixture composition using thick film process. There was no second phase by the reaction of SnO_2 and ZnO. The conductance was decreased by the addition of ZnO in SnO_2, but it was increased with the addition of SnO_2 in ZnO. The conductance was increased with temperature and the inlet of CO. There was no improvement of sensitivity in the structure of mono- and double-layer. The hetero-layer structure, however, of SnO_2 · 3ZnO-ZnO · 3SnO_2 showed the higher resistivity and the highest sensitivity. Ohmic characteristics was confirmed by the linear properties for I-V measurements.

      • 광센서를 위한 ZnO:Al 투명도전막의 제조 및 특성연구

        RF 마그네트론 스퍼터링법으로 AZO(ZnO:Al)막을 증착시켰다. 타겟내의 Al_2O_3의 첨가량, 기판온도, 분위기압, RF출력 등의 증착조건의 변화에 따라 제조된 AZO막의 조성, Al 도핑양, 구조적 특성 등을 XRD, SEM, EDS 및 RBS로 조사하였다. 이와 함께 전기적 및 광학적 특성을 Hall 효과 및 광투과도 특성을 통하여 조사하였다. 증착된 AZO막은 육방정의 wurtzite 구조였으며 모든 증착조건에서 기판에 수직으로 성장하는 (002)면 방향을 가지는 c-축배향성을 나타내었다. 증착된 AZO막내의 AB 도핑량은 타겟내의 무게비에 비례하였으며 Zn : O의 조성비토 1 : 1로 거의 일정하였다. 투명도전막으로서의 최적조건은 3wt%의 Al_2O_3이 첨가된 타겟으로 기판온도가 150℃, 분위기압이 2mTorr, RF출력이 150W로 증착한 경우였으며 이 때 비저항은 4.7×10 exp (4)Ω㎝, 캐리어 농도는 7.5×10 exp (20)㎝^-3이었다. 또 두께가 1500Å인 막의 경우 550㎚에서 광투고도가 90%이상이었으며 광학적 밴드갭은 3.53eV였다. Aluminium doped zinc oxide(AZO)films were prepared by RF magnetron sputtering method. The chemical compositions, doping concentrations, and the structural char-acteristics of the AzO films were studied in accordance with various deposition parameters, such as Al_2O_3 amount, substrate temperature and working pressure, by XRD, EDS, SEM and RBS. And The electrical and optical properties of AZO films were characterized by Hall effect and optical transmittance measurements. AZO films had were hexagonal wurtzite structure and dominent c-axis orientation. The Al doping concentration in the films were monotonically increased. The atomic ration of Zn : O were 1 : 1. The substrate, working pressure and rf power for optimum condition to fabricate the transparet conductive films using a target containing 3wt% Al_2O_3 were 150 ℃, 2mTorr and 150W, respectively. The resistivity and the carrier density of the AZO film prepared under this condition were 4.7 × 10 exp (-4) Ω㎝ and 7.5 × 10 exp (20) ㎝^-3, respectively. The optical transmittance of 1500 Å thick films at 550㎚ is ∼90% and optical energy band gap was 3.53eV.
