        반복적 스트레스가 흰쥐의 Opioid성 신경전달에 미치는 영향

        연구목적: 반복적인 스트레스에 노출된 실험동물에서 opioid성 신경전달의 변화양상을 검색하고 내인성 opioid peptide의 변화양상을 알아보기 위하여 본 연구를 시행하였다. 방 법: 본 연구에서는 웅성 흰쥐에게 하루 2시간, 2주간의 반복적인 부동화 스트레스를 가한 후 뇌의 각 부위에서 opioid 수용체의 mu-, delta- 및 kappa- 아형의 최대결합력 및 친화력의 변화 양상을 관찰하였다. 결 과: 반복적 부동화 스트레스 노출군에서는 대조군에 비하여 delta-아형 수용체의 수가 시상하부(hypo-thalamus) 및 선조체(striatum)에서 증가하였다. 본 연구결과 관찰된 선조체에서의 delta-아형 수용체의 상향조절과 관련지어 delta-아형 수용체의 내인성 효현제인 enkephalin의 뇌조직 내에서의 활성도를 검색하였을 때, 반복적 스트레스 노출군에서는 대조군에 비하여 그 함량이 감소하였다. 이러한 뇌조직내 enkephalin 활성도 감소 현상은 수용체 변화와 상응하게 시상하부 및 선조체에서 관찰되었으며, 이러한 수용체의 조절 현상은 내인성 수용체 효현제인 enkephalin의 활성도의 감소에 따른 생체의 적응 현상으로 해석되었다. 결 론: 이상의 결과로부터 반복적인 스트레스는 opioid 수용체 delta-아형을 상향조절하였으며, opioid 수용체 delta-아형의 내인성 효현제의 활성도를 감소시킴을 관찰하였다. Objectives : Research into emotional or behavioral stress typically focuses upon the hypothalamic-pituitary-adrenal axis. It is well established that the hypothalamic-pituitary-adrenocortical axis is subject to inhibitory control by opioids in a variety of animal species including pigs. Exposure to acute stress induces the upregulation of opioid receptors and the release of endogenous peptides which mediate the stress-induced analgesia. There is some literature substantiating that repeated stress can lead to changes in opioidergic neurotransmission. However, the changes are highly variable. This study was designed to observe the modulatory effect of repeated immobilization stress on opioidergic neurotransmission. Methods : Male Sprague-Dawley rats weighing 150-200g were forced to suffer immobilization stress for 2 hours on each of 14 successive days. Then we examined the maximum binding capacity and affinity of each opioid subtypes(mu, delta, kappa). Results : Repeated immobilization stress increased the binding of [³H]DPDPE on the delta-subtype opioid receptor in the striatum and hypothalamus. Saturation experiments followed by scatchard analyses of the results showed an increase in the density of delta-subtype opioid receptors, but the affinity of the delta-subtype opioid receptor remained unchanged. Repeated immobilization stress reduced enkephalin activity of striatum and hypothalamus. Conclusions : From these results, it could be concluded that repeated immobilization stress up-regulated the delta-subtype opioid receptors and reduced the activity of enkephalin, an endogenous ligand for the delta-subtype opioid receptor.

      • 부유대역 용융응고법으로 제조된 YBa_2Cu_3O_x 산화물초전도체의 미세구조와 전기적 특성

        The YBa_2Cu_3O_x-20 wt% Y_2BaCuO_5 composition designed in YBCO system was synthesized by solid state reaction method. The optimum condition of hot zone temperature and growth rate in floating-zone melt were 1080 ℃ and 2.5 mm/hour, respectively. In samples growth as these condition, the low angel boundaries in grain direction and small amount of amorphous phase known as decreasing characteristics of superconducting materials between grain boundaries in microstructure are investigated by polarized light microscopy on the whole. The critical current density(Jc) at 77k of sample using powder fabricated with no CeO_2 and doped 5 wt%CeO_2 were 8,000 A/㎠(Jc : 561A) and 12,000 A/㎠(Jc: 826A) at 0 T by direct current transport method, respectively.

      • 안양천 등 관내하천의 수질오염분석 및 생태학적 분석에 관한 연구(II)

        안양천을 비롯한 10개 지천에 대한 환경오염 분석을 하였다. 전편(Ⅰ)에 이어 안양천의 관내지류인 산본천, 당정천, 오전천, 왕곡천, 통미천을 중심으로 하여 수질오염분석과 중금속오염도 조사 및 생태학적 분석에 대하여 연구하였다. 아울러 안양천 및 그 지류의 환경진단과 함께 하천을 살릴수 있는 몇가지 제안을 하였다. 안양천 및 지류의 수질오염분석결과 측정기간에 BOD 값이 평균 25.5 mg/l으로, 최저 17.0mg/l에서 최고 36.4mg/l으로 환경기준치를 초과하고 있다. COD는 평균 18.9mg/l으로 최저 12.7mg/l에서 최고 24.2mg/l로서 환경기준치인 5급수 수질을 초과하고 있으나 다른 수질의 지표에 비해 비교적 양호한 값을 나타내고 있다. SS는 평균 23.8mg/l으로 3급수 이내의 수질을 유지하고 있다. 총질소(T-N)는 평균치가 16.7mg/l으로 상류는 기준치 이하를 나타내고 있으나 중·하류는 5급수 이상의 높은 수치를 보이고 있어 제거대책이 강구되어야 할 것이다. 하천퇴적물에 대한 중금속 오염도 분석결과 5개항목 모두 기준치를 초과하여 높게 나타났으며 특히 아연(Zn)의 농도는 매우 심각한 것으오 분석되었다. 분석결과는 Zn>Cu>Pb>As>Cd순으로 나타났다. 생태학적 분석에 있어서는 안양천의 상류에서 하류에 이르기까지 어류는 서식하지 못하고 있으며 하류일부지역에서 오염에 강한 극소수 어류가 발견되었다. 식물도 하류로 갈수록 오염에 비교적 강한 식물이 군락을 이루고 있었다. Water pollution analysis about Anyangchun river and rivers within the province, investigation of a contamination level of heavy metal and ecological analysis were studies. The result of water pollution analysis Anyangchun river and its tributaries during the period of measure says BOD was average 25.5mg/l, ranging from minimum 17.0mg/l to maximum 36.4mg/l which exceeds and environmental standard level COD was average 18.9mg/l, ranging from minimum 12.7mg/l to maximum 24.2mg/lwhich exceeds fifth class of water quality, and environmental datum level, but represents relatively better value than the other base point of water quality. SS was average 23.8mg/l, which maintains water quality of below the third class. Total Nitrogen was average 16.7mg/l ; the upper water was seen below a base point, but the middle and lower showed exceeds fifth class of water quality. Therefore, a proper measure to remove nitrogen will have to be deviced. According to the analysis result of heavy metal contamination level about the river deposit, all of 5 items exceeded the environment base point. Specially, concentration of Zinc was considered very serious. The result of analysis was proven in the order to Zn>Cu>Pb>As>Cd. In ecological analysis, fish couldn't living from the upper water to the lower one in Anyang river with in province and a small minority of fish were discovered in part of the lower water. Hereby we suggested the alternative to decline water pollution in Anyang watercourse.

        알코올 사용 장애 환자에서 알코올 사용의 심각도에 따른 불안 및 우울 증상과의 연관성

        Objectives:The purpose of this study was to examine the relationships of anxiety& depressive symptoms according to the severity of alcohol use in patients with alcohol use disorder. Methods:A total of 85 patients with alcohol use disorders were enrolled in this study. The severity of alcohol use was measured by the Korean version of alcohol use disorders identification test(AUDIT-K)& National Alcohol Screening test(NAST). Anxiety and depressive symptoms were measured by using the Beck Anxiety Inventory(BAI) and the Beck Depression Inventory(BDI), respectively. Pearson’s correlation test was applied to correlate among the severity of alcohol use, anxiety and depressive symptoms. Results:The mean scores of AUDIT-K, NAST, BAI and BDI were 22.22(±8.28), 6.24(±3.25), 18.64(±13.08), and 22.05(±10.13), respectively. Pearson’s correlation test showed that between the score of AUDIT-K and NAST, BAI or BDI, there were positively significant correlations(r=.534, p<.01 r=.231, p<.05 r=.334, p<.01), respectively. And in cases of≥26 score of AUDIT-K, there were much more positive correlations with BAI(r=.425, p<.05). Conclusion:The results of this study demonstrated that patients with alcohol use disorders have high co-morbid anxiety and depressive disorders. Overall, the greater severity of alcohol use may be associated with more anxiety and depressive symptoms. However, it is suggested that in the patients with severe alcohol use, correlation appeared to be positive only for anxiety symptoms.

        Castleman' s disease is a rare, reactive lymphoid proliferation of unknown cause and pathogenesis. Depending on histololgic findings, it has been divided into two distinct histologic subtypes that include hyaline-vascular type(90%) and plasmacell type(10%). This disease occurs most commonly in mediastinal lymph nodes and cervical lymph nodes, inguinal lymph nodes respectively. But occurs rarely in retroperitoneum. We report a case of hyaline-vascular type asymptomatic Castleman' s disease of the retroperitoneum in a 47-year-old man.

      • 갑상선 세포에서 전사보조활성인자인 CBP와 CIITA의 인터페론-감마 활성 부위에 대한 조절 : CBP and CIITA on GAS(Interferon- γ Activated Site)-Mediated Transcription in Thyroid Cells

        Background : In the previous stuides, we identified that the interferon-r activated sequence (GAS) in the 5-flanking region of rat ICAM-1 gene is major element for interferon-r-inducible expression of the gene in rat thyroid cells, FRTL-5. We here, investigated the role of transcriptional coactivators, CBP(CREB binding protein) and CⅡTA (class Ⅱ transactivator) in the modulation of the activity of GAS which could interacts with signal transducers and activators of transcription-1 and 3 (STAT1 and STAT3). Methods : The expression of CBP RNA and protein were quantitated in FRTL-5 after stimulation with interferon-r (IFN-r), thyroid stimulating hormone (TSH), forskolin and methimazole. Direct association of CBP with STAT were analyzed by immunoprecipitation. The transcriptional roles of CBP and CⅡTA in the regulation of GAS were assessed by the cotransfection with their expression vectors with reporters; 5-deletion constructs of rat ICAM-1 promoter or 8xGAS-luc constructs, into FRTL-5 thyroid cells. Results : The level of CBP RNA and protein were not changed by the treatment with TSH, IFN-r, forskolin and methimazole in FRTL-5, FRT and BRL liver cells. The CBP could be directly associated with STAT1. Furthermore, the overexpression of CBP significantly increases the both promoter activities; rat ICAM-1 gene promoter which has GAS element and 8xGAS-luc cassette constructs. However, the cotransfection of CⅡTA decreased the constitutive and CBP-mediated transactivation of rat ICAM-1 promoter and 8xGAS-luc cassette constructs. Conclusion : We identified that the two tanscriptional coactivators; CBP and CⅡTA has differential roles in the regulation of transcriptional activity of GAS drived promoter. CBP increases the GAS activity through the direct binding with STAT1, but CⅡTA inhibited the CBP-mediated transactivation of GAS activity.

      • IRAS 15099-5856: REMARKABLE MID-INFRARED SOURCE WITH PROMINENT CRYSTALLINE SILICATE EMISSION EMBEDDED IN THE SUPERNOVA REMNANT MSH15-5<i>2</i>

        <P>We report new mid-infrared (MIR) observations of the remarkable object IRAS 15099-5856 using the space telescopes AKARI and Spitzer, which demonstrate the presence of prominent crystalline silicate emission in this bright source. IRAS 15099-5856 has a complex morphology with a bright central compact source (IRS1) surrounded by knots, spurs, and several extended (similar to 4') arc-like filaments. The source is seen only at >= 10 mu m. The Spitzer mid-infrared spectrum of IRS1 shows prominent emission features from Mg-rich crystalline silicates, strong [Ne II] 12.81 mu m, and several other faint ionic lines. We model the MIR spectrum as thermal emission from dust and compare with the Herbig Be star HD 100546 and the luminous blue variable R71, which show very similar MIR spectra. Molecular line observations reveal two molecular clouds around the source, but no associated dense molecular cores. We suggest that IRS1 is heated by UV radiation from the adjacent O star Muzzio 10 and that its crystalline silicates most likely originated in a mass outflow from the progenitor of the supernova remnant (SNR) MSH 15-52. IRS1, which is embedded in the SNR, could have been shielded from the SN blast wave if the progenitor was in a close binary system with Muzzio 10. If MSH 15-52 is a remnant of Type Ib/c supernova (SN Ib/c), as has been previously proposed, this would confirm the binary model for SN Ib/c. IRS1 and the associated structures may be the relics of massive star death, as shaped by the supernova explosion, the pulsar wind, and the intense ionizing radiation of the embedded O star.</P>
