A protein-polysaccaride fraction Kp(53.9% polysaccharide, 14.2% protein) was separated from the shake-cultured mycelia of a basidiomycetous fungus, Phellinus linteus, and its antitumor activity against sarcoma 180 in ICR mice was investigated. When administered after the tumor implantation. Kp exerted antitumor activity by inhibiting the growth of the sarcoma 180 solid tumor by 71.5% and increasing the life span of the sarcoma 180 ascitic mice by 51.5% at 100 ㎎/㎏. In pretreatment tests, in which Kp was administered once daily for 9 days before the tumor implantation. Kp inhibited the growth of the solid and ascites form of sarcoma 180, respectively, by 35.4% and by 80.3% at 100 ㎎/㎏. However, Kp showed no in vitro cytotoxic activity against a murine leukemia L1210 and a human gastric tumor SNU.1 upto the concentration of 200 ㎍/㎖. From these results, it is clear that the antitumor activity of Kp is exerted through its immunomodulating activity on the host's immune system.

The ether, methanol, and physiological saline extracts of the dried carpophores of ten basidiomycetous fungi, which were collected in the conifer woods of Chungnam National University at Taechon, were subjected to an antibiotic activity test by disc-plate method. Among them, the methanol extracts of Marasmius maximus, Hirschioporus abietinus, and Russula sanguinea and the the ether extracts of M. maximus and H. abietinus showed antibiotic activities against Sarcina lutea; and the methanol and ethanol extract of M. maximus showed antibiotic activity against Staphylococcus aureus. None of the extracts, however, showed any activity against gram negative bacteria, Serratia marcescens and Proteus vulgaris.

The ether, methanol, and physiological saline extracts of the dried carpophores of ten basidiomycetous fungi, which were collected in the conifer woods of Chungnam National University at Taechon and Keyryoung Mountain Area in Chungcheungnamdo, were subjected to an antibiotic activity test by disc-plate method. Among them, the methanol extracts of Inocybe trechispora, Lactarius velumis, and Laccaria laccata and the ether extracts of I.trechispora showed antibiotic activities against Sarcina lutea; and the methanol extracts of I. trechispora and L. velumis and physiological saline extract of Coriolus versicolor showed antibiotic activities against Staphylococcus aureus; and the methanol extract of L. velumis showed antibiotic activity against Escherichia coli. None of the extracts, however, showed any activity against Serratia marcescens and Proteusvulgaris.

The ether, methanol, and physiological saline extracts of the dried carpophores of ten basidiomycetes, which were collected from several locations of Korea, were subjected to a screening test for antibiotic activity by disc-plate method using Sarcina lutea, Bacillus subtilis, Escherichia coli, and Serratia marcescens as target organisms. As the result, the methanol extract of five basidiomycetes-Amanita rebescens, Amanita solitaria, Lactarius vellereus, Latarius subzonarius and Scleroderma verrucosum-were found te have antibiotic activity. Among them, the methanol extracts of A. rubescens and L. vellereus exerted strong activity against both of the gram positive and negative bacteria. The ether extracts of A. rubescens and S. verrucosum also showed activity against S. lutea. The physiological saline extract, however, showed no activity at all.
Regulation of Taurine Transporter Activity by Glucocorticoide Hormone

Human taurine transporter has 12 transmembrane domains and its molecular weight is 69.6 kDa. The long cytoplasmic carboxy and amino termini might function as regulatory attachment sites for other proteins. Six potential protein kinase C phosphorylation sites have been reported in human taurine transporter. In this report, we studied the effects of phorbol 12-myristate 13-acetate (PMA) and glucocorticoid hormone on taurine transportation in the RAW 264.7, mouse macrophage cell line. When the cells were incubated with [³H]taurine in the presence or absence of Na^+ ion for 40 min at 37℃, the [³H]taurine uptake rate was 780-times higher in the Na^+-containing buffer than in the Na^+-deficient buffer, indicating that this cell line expresses taurine transporter protein on the cell surface. THPl, a human promonocyte cell line, also showed a similar property. The [³H]taurine uptake rate was not influenced by the inflammatory inducing cytokines such as interleukin-1, gamma-interferon or interleukin-1 +gamma-interferon, but was decreased by the PMA in the RAW 264.7 cell line. This suggests that activation of protein kinase C inhibits taurine transporter activity directly or indirectly. The inhibition of [³H]taurine uptake by PMA was time-dependent. Maximal inhibition occurred in one hr stimulation with PMA. Increasing the treatment time beyond one h reduced the [³H]taurine uptake inhibition due to the depletion or inactivation of protein kinase C. The cell line also showed concentration-dependent [³H]taurine uptake under PMA stimulation. The phorbol-ester caused 23% inhibition at the concentration of 1 pM PMA. The inhibition was significant even at a concentration as low as 10 nM PMA The reduced [³H]taurine uptake could be recovered by treatment with glucocorticosteroid hormone. Deacamethasone led to recover of the reduced taurine uptake induced by phorbol-ester, recovering maximally after one hr. This may suggest that macrophage cells require higher taurine concentration in a stressed state, for the secretion of glucocorticoid hormone is increased by hypothalamo-pituitary-adrenocortical (HPA) axis activation in the blood stream.

Analysis of the culture filtrates of lysergic acid alkaloid-producing fennentations at intervals during growth of the cultures showed that phosphate is most rapidly depleted from the medium in fermentations that produce the highest levels of alkaloid. In view of the known interference with phosphate metabolism by arsenate, sodium arsenate was added to the culture medium with the intent of creating, in effect, a phosphate deficiency. The addition of arsenate at levels between one-fiftieth and one-twentieth the molar concentration of phosphate resulted in increases in alkaloid production up to 100%. Experiments with tentimes the usual concentration of phosphate showed that the ratio of arsenate to phosphate rather than the absolute concentration of arsenate was most significant in obtaining this effect. Increasing the concentration of phosphate alone (up to twenty times the usual concentration) resulted in substantial increases in alkaloid production. The effect of arsenate on both the rate and the extent of phosphate uptake was determined. Among several inhibitors of electron transport and/or oxidative phosphorylation, only the uncouples of oxidative phosphorylation (dinitrophenol) was effective in increasing alkaloid production.

To find anititumor metabolites in Korean basidiomycetes, the shake-cultured mycelia of eight of the higher fungi were extracted with hot water and the extracts, after being partially purified, were subjected to in vivo antitumor test. When administered i. p. at the dose of 30mg/kg/day for ten consecutive days into the female ICR mice, which had been implanted with $1{\times}10^{6}$ / cells of sarcoma 180 twenty four hours before the first injection, the extracts of Agaricus campestris, Lyophyllum decastes, Lyophyllum ulmarium, Armillaria Tabescence and Calvatia exipuliformis respectively showed inhibition ratios of 64.1%, 65.45, 60.-%, 53.0 and 49.3%. These five species were selected for further study, whereas the extracts of Phallus impudicus, Coprinus comatus and Pholiota squarrosa whih showed the inhibition ratios of 31.2%, 33.5% and 19.0% were discontinued.

Antitumor components were obtained from the cultured mycelia of Pleurotus pulmonarius by ethanol precipitation. The protein-bound polysaccharide was subjected to DEAE-Sephadex column chromatography and Sephadex G-200 gel filtration. The antitumor fraction C₁ was isolated. The inhibition ratio of fraction C₁ was 81.8% in the doses of 10 ㎎/㎏/day for 10 days. The antitumor fraction C₁ consisted of a polysaccharide and a protein. The protein-moiety was composed of 14 amino acids. From the peritoneal cell populations in the mice given antitumor fraction C₁, the injection of the fraction caused the influx of peritoneal macrophages at two days when compared with those of soluble starch. This was named pulmonaran after its species name.
