        Wingate test 및 Supramaximal exercise를 이용한 무산소성 운동능력의 측정시 나타난 변인의 관련성을 살펴본 본 연구를 요약하면 다음과 같다. Wingate test시 혈중젖산농도 최고치는 9.04±0.62mM로서 Supramaximal exercise시 보다 유의하게 높았으며, 혈장 LDH 및 CPK활성도는 두가지 운동부하후 모두 유의한 변화는 나타나지 않았다. 근전도상의 적분치, 중간파형빈도 및 평균파형빈도도 유의한 변화는 없었다. 등속성 수축시의 최대토오크는 양쪽 다리 모두에서 초당 60도의 각속도에서가장 높았다. Wingate test시의 최대파워는 등속성 수축시 최대토오크 및 Wingate test시 중간파형빈도의 직접적인 영향을 받는 것으로 나타났으며, 평균파워는 Wingate test시의 적분치 변화, 듣는 쪽 슬관절의 등속성 수축시최대토오크 및 Supramaximal exercise시 혈장 LDH 활성도의 직접적인 영향을 받는 것으로 나타났다. 이상의 결과에서 무산소성 운동능력과 관련지어 그 기전을 살피고자 할 때 체내 화학적 변화, 전기생리적 변화 및 발현능력의 복합성을 고려해야 하며 최대누적 산소결핍량의 활용가능성은 측정강도 결정방법을 중심으로한 계속적인 연구가 요구된다. The present study examined the relationship of variables related to anaerobic capacity with Wingate test and supramaximal exercise. Peak value of blood lactate concentration with Wingate test showeda significantly higher than suprammaximal exercise, and plasma LDH & median power frequency and mean power frequency were not any difference between two tests. Peak torque of isokinetic contractionh on knee joint showed a highest value in 60 degrees of angular velocity among a various angular velocity. Peak power of Wingate test was related to peak torque of isokinetic contraction and median power frequency of EMG with Wingate test, and mean power of Wingate test was related to integrated EMG with Wingate test, peak torque with isokinetic contraction of dominant knee joint and plasma LDH activity with supramaximal exercise. In this result, when it analyzed the mechanism of anaerobic capacity, ti was required the consideration of complexity according to biochemical alteration, electrophysiological alteration and revelation of performance, Appicapability of maximal accumulated O₂deficit was demaned a continuous study with the focus of exercise intensity.

        전복(Haliotis discus hannai) 추출물의 혈압강하, 항산화능 및 항혈전능에 대한 in vitro 효과

        본 연구에서는 전복육질과 내장의 추출방법에 따른 추출물의 혈압강하, 항산화, 항혈전 효과에 대한 in vitro 효과를 구명하고자 하였다. 전복육질(abalone body)과 내장(visceral portion)의 80% ethanol 추출물은 ACE(angiotensin converting enzyme)활성에 대해 높은 억제효과를 나타내었으며, 전복육질 추출물의 경우 농도의 증가에 따라 증가되는 경향을 나타내었다. 그러나 내장추출물은 농도증가에 따라 큰 차이가 없는 것으로 나타났다. 수용성추출물의 ACE활성 억제효과는 농도별에 따라 증가되는 경향을 나타내었으며, 육질과 내장 간에 큰 차이가 있는 것으로 나타났다. 아질산염 소거활성으로 평가한 항산화 효과는 80% ethanol 추출물의 경우, 전복육질에서 농도증가에 따라 증가된 수준을 나타내었으나 그 수치는 낮은 값을 나타내었다. 내장의 경우, 낮은 농도에서는 육질과 비슷한 수준을 유지하였으나 농도가 증가함에 따라 높은 항산화활성을 나타내었다. 수용성추출물의 항산화효과는 농도증가에 따라 증가된 수준을 나타내었으나 육질과 내장에 큰 차이는 보이지 않았다. 항혈전 효과는 80% ethanol 추출물에서 육질의 prothrombin time이상대적으로 내장의 prothrombin time보다 높게 나타났다. 수용성추출물의 경우, 항혈전 효과가 거의 없었으며, 전복의육질이나 내장에 따른 특별한 차이점 또한 없는 것으로 나타났다. 수용성추출물을 48시간 냉장온도에서 저장한 후의 전복육질과 내장의 ACE 활성에 대한 억제효과는 육질과 내장간에 큰 차이가 있었으나 0 time의 값과 큰 차이를 나타내지 않았다. 항산화 효과는 육질과 내장의 경우 농도증가에 따라 직선적인 증가경향을 나타내었으나, 육질에 비해 내장에서 높은 항산화능이 있는 것으로 나타났다. 전복육질과 내장의 prothrombin time은 큰 차이가 없었으며, activated partial thromboplastin time 또한 큰 차이가 없는 것으로 나타났다. The purpose of this study was to examine the in vitro effects of body and visceral portion of Haliotis discus hannai on angiotensin converting enzyme (ACE) activity and antioxidant and anticoagulant capacity. Extracts from both abalone body and visceral portion using 80% ethanol showed a high ACE?inhibitory effect. While ACE-inhibitory effect of extracts of the body part was dose?dependent, visceral extracts did not show any difference by the level of concentration. ACE?inhibitory effect of the visceral portion was much higher than that of the body. Antioxidant capacity was increased with increasing concentration of 80% ethanol body extracts although the capacity was low. The 80% ethanol visceral extracts showed a similar level of antioxidant capacity to the body extract in low concentration. Water extracts showed a dose-dependent increase in the activity. There was no significant difference in the antioxidant activity between the body and the visceral part. Anticoagulant capacity of 80% ethanol extracts, which was measured using prothrombin time (PT), was higher in the body part than the visceral part. Water extracts of Haliotis discus showed no any significant effect on anticoagulant capacity. The in vitro effects were also examined after Haliotis discus was refrigerated for 48 hours. Higher ACE-inhibitory effect was observed for the visceral portion than the body, in particular, before the sample was refrigerated. Antioxidant effect of Haliotis discus increased with increasing level of the sample before it was refrigerated. However, there was a significant difference between the body and the visceral part, which showed significantly higher capacity. There was no significant difference between the body and visceral part in PT regardless of refrigeration. While activated partial thromboplastin time (APTT) showed no significant difference between body and visceral part, there was a significant difference in the capacity between before and after the refrigeration, which showed much lower coagulant capacity.

        Effect of Thinning on Defoliated ‘Fuyu’ Persimmon on Fruit and Tree Development in the Current and the Following Years

        Questions arise as to what to do with the fruits of persimmon trees when they are severely defoliated by a typhoon early in the season. In this study, the leaves of 5-year-old, field-grown ‘Fuyu’ trees were defoliated 75% on Sept. 3, 1999 and Sept. 6, 2001. Zero (control), 25, 50, and 75% of their fruits were then thinned on these trees, changing the leaf-to-fruit ratio to about 5, 7, 10, and 20, respectively. The ratio of non-defoliated intact trees was maintained at 20 throughout the season. Fruit weight and color increased to the level of intact trees when fruit thinning exceeded 25%, although the leaf-to-fruit ratio was as low as 7. On the other hand, fruit soluble solids responded poorly to the increase in fruit thinning on defoliated trees; even a 75% thinning could not restore them to the level of intact trees. Results suggested that fruit thinning could improve fruit size and color, but have a limited impact in increasing fruit soluble solids when the trees had lost 75% of their leaves in early September. In the following year, measurements of shoot growth, leaf area, fruit set, and fruit characteristics did not show consistent differences attributable to the thinnings on 75%-defoliated trees the previous year.

        <P><B>Highlights</B></P><P>► Yeast Rps3p cleaves not only AP sites and pyrimidine dimers but also 8-oxo-G. ► Yeast Rps3p could cleave pyrimidine dimers without glycosylase activity. ► Asp154 and Lys200 are identified as important residues for the catalytic activity.</P> <P><B>Abstract</B></P><P>Yeast ribosomal protein S3 has multifunctional activities that are involved in both protein translation and DNA repair. Here, we report that yeast Rps3p cleaves variously damaged DNA that contains not only AP sites and pyrimidine dimers but also 7,8-hydro-8-oxoguanine. This study also revealed that Rps3p has a β-lyase activity with a broad range of substrate specificity which cleaves phosphodiester bonds of UV or oxidatively damaged DNA substrates. Mutation analysis of the yeast Rps3 protein including introduction of domain deletions and residue replacements identified the residues Asp154 and Lys200 are important for the catalytic activity. In addition, the repair enzyme activity of yeast Rps3p was confirmed by complementation in <I>xth</I>, <I>nfo Escherichia coli</I> cells in which the DNA repair process is defective.</P>

        Robust Nonlinear Control of a 6 DOF Parallel Manipulator:Task Space Approach

        This paper presents a robust nonlinear controller for a 6 degree of freedom (DOF) parallel manipulator in the task space coordinates. The proposed control strategy requires information on orientations and translations in the task space unlike the joint space or link space control scheme. Although a 6 DOF sensor may provide such information in a straightforward manner, its cost calls for a more economical alternative. A novel indirect method based on the readily available length information engages as a potential candidate to replace a 6 DOF sensor. The indirect approach generates the necessary information by solving the forward kinematics and subsequently applying alpha-beta-gamma tracker. With the 6 DOF signals available, a robust nonlinear task space control (RNTC) scheme is proposed based on the Lyapunov redesign method, whose stability is rigorously proved. The performance of the proposed RNTC with the new estimation scheme is evaluated via experiments. First, the results of the estimator are compared with the rate-gyro signals, which indicates excellent agreement. Then, the RNTC with on-line estimated 6 DOF data is shown to achieve excellent control performance to sinusoidal inputs, which is superior to those of a commonly used proportional-plus-integral-plus-derivative controller with a feedforward friction compensation under joint space coordinates and the nonlinear controller under task space coordinates.<br/>

      • Human rpS3 is involved in DNA repair and cell cycle control

        In the cellular response to DNA damaging agents, cells undergo cell cycle arrest or apoptosis against irrepairable DNA damage. RpS3 is known to function as UV DNA repair endonuclease III and ribosomal protein S3. In this study, we used normal and rpS3-overexpressed 293T cells to examine the role of rpS3 in response to DNA damaging agents. When 293T cells transfected with rpS3 were irradiated with UV, the pattern of cell cycle was dramatically changed in comparison with un-transfected 293T cells. We also found that the expression of rpS3 in normal cells was increased by treatment with DNA damaging agents. By means of Western and Northern blot analyses in rat tissues, we showed the expression pattern of rpS3 protein and its mRNA. These data suggest that DNA repair and cell cycle arrest are interrelated to each other through rpS3, and the increased expression of rpS3 seems to regulate the cell cycle arrest by DNA damaging agents.

      • Dependence of Activated Carbon Consumption to Water Quality Parameters and Environmental Factors

        정수처리과정에서 냄새 및 맛 유발원을 제거하기 위한 활성탄 소비량과 여러가지 수질인자 및 기상조건과 같은 환경인자와의 관계에 관한 정보를 수집하기 위해 본 연구를 수행하였다. 1992년부터 2001년까지 상수처리장에서 수집된 자료와 동일지역의 기상자료를 수집하여 통계적으로 분석하였다. 연구결과에 의하면 정수처리시 활성탄 소비량과 몇몇 수질항목 및 환경인자 사이의 매우 의미있는 상관관계가 있는 것으로 나타났다. 특히 활성탄의 월간사용량은 총인, 수온, 기온, 그리고 강우량과 정비례관계에 있는 것으로 나타났으며, 반면에 용존산소와는 음의 상관관계를 나타내었다. 특별히 주목할 것은 이들 인자들과의 상관관계가 동월보다는 전월의 인자들과 더욱 크게 나타났다는 점이다. 이는 호수의 수질을 관리하기 위하여 적절한 수단을 제공하는 면에서 중요하다고 할 수 있다. 이것은 또한 활성탄의 수급조절의 판단자료로 활용할 수 있을 것이다. This study was done to collect information on relations between activated carbon consumption for elimination of taste and odor in a water treatment plant and various water quality parameters or environmental factors such as meteorological conditions. Statistical analyses were done on the water quality data collected at a water treatment plant and on the regional meteorological data for the period from 1992 to 2001. The study reveals that the correlation between the amounts of activated carbon consumed in the water treatment plant and some of the water quality parameters or the environmental factors is very significant. In particular the monthly average consumption of activated carbon shows linear dependencies on factors such as total phosphorous, water and air temperature, and precipitation rate. On the contrary, it also shows a negative dependency on dissolved oxygen. What is most noticeable is the fact that these dependencies are more profoundly related to the important from the point of view of providing adequate measures for the water quality management of lakes. Also it will be applicable in the adjustment of the budget for the supply of activated carbon.

        Ischemic necrosis of the cirrhotic liver is rare because the liver has dual blood supply and an extensive collateral system. However, it may occur in the condition of sudden reduction of hepatic arterial flow such as acute blood loss or shock Recently, we experienced two cases of ischemic necrosis of the liver assoicated with cirrhosis. Those were presented as space-occupying lesions on the radiological study. In contrast with hepatocellular carcinoma, these lesions showed neither hypervascularity nor deposition of injected lipiodol. Repeated percutaneous fine needle cytology revealed no evidence of malignant cells, Furthermore, short-term follow-up examination showed decrease in size and/or extent of the lesions, It is important to differentiate these lesions from tumorous condition because interventional trial such as transarterial chemoembolization may further aggravate the ischemic lesion and unnecessary surgery under false impression of hepatocellular carcinoma in cirrhotic patients should be avoided.

        죽염 제조공정에 따른 이산화황, 미네랄 함량 및 이화학적 특성

        자죽염(purple bamboo salt; PBS)을 제조하는 공정에 영향을 미치는 원염(갯벌천일염, MSS; 죽염, BS)의 종류, 대나무 첨가에 따른 BS의 이화학적 특성, 황 화합물, 미네랄 함량 및 환원력을 비교?분석하고, 이를 통해 PBS에 함유된 SO₂와 산화환원전위(ORP)의 발생 원인을 분석하였다. BS를 고온에서 용융시켜 제조한 PBS에서 630 ppm의 SO₂가 검출되었다. PBS의 원료 염과 RB에서는 SO₂가 검출되지 않았으나, 염화물, 황산염류, 탄산염류와 대나무(RB)를 함께 탄화시켜 SO₂ 검출 원인을 확인한 결과 황산칼륨에 의해 782 ppm이 발생하였다. BS의 SO₂는 BSRB1(13.88)~BSRB4(109.13 ppm)에서만 발생하였고 SO₃<SUP>2-</SUP>는 MSSRB4와 BSRB2~BSRB4에서만 발생하였으며, 시간이 경과함에 따라 증가되는 경향을 나타내었다. SO₄<SUP>2-</SUP>는 시간이 지남에 따라 감소하는 경향을 나타내었으며 SO₂와 SO₃<SUP>2-</SUP>가 많을수록 낮은 함량을 나타내었다. ORP는 모두 시간이 지남에 따라 증가하는 경향을 나타내었고 BS에서 더 높은 수준을 나타내었으며, BSRB4(-211.40 mV)에서 가장 높은 환원력을 나타내었다. 일반적 특성 중 불용분은 BS에서 더 높았으며 RB가 탄화되어 남은 물질들이 영향을 미친 것으로 판단된다. Ca, K, Mg의 경우 굽는 시간 경과에 따라 MSS는 감소, BS는 증가하는 경향을 나타내었다. BSRB4의 Ca는 1.4배, Mg는 1.5배 증가하였고 K 함량은 1.8배 더 높은 것으로 나타났다. 미량 미네랄은 굽는 시간 경과에 따라 MSS에서 Li, Al, Mn, Fe, Sr이 증가되었고, BS에서 Al, Fe, Ni가 증가하였다. 음이온(Cl, NO₃, Br)과 중금속(Pb, Cd, Hg, As)은 각 요인에 대한 영향을 받지 않았다. 결론적으로 BS의 기능성을 나타내는 환원력은 SO₂ 및 SO₃<SUP>2-</SUP>와 같은 저분자 황화합물에 의한 것이며, 이는 한 번 굽는 과정을 거친 BS를 사용하고 이에 RB를 첨가하여 고온에서 용융하는 시간이 길수록 이들 황화물질이 많아져 환원력이 증가된다는 것을 의미하는 것이다. The purpose of this study was to investigate the mechanisms of behind SO₂ formation and elevated cause of reducing power in purple bamboo salt (PBS) along with an analysis of physicochemical properties, content of sulfur compounds, oxidation reduction potential (ORP), mineral contents of salt type (MSS, mudflat solar salt; BS, bamboo salt), and addition of raw bamboo (RB). SO₂ content of 630 ppm was detected in PBS. SO₂ was not detected in MSS, BS, or RB, whereas SO₂ (782 ppm) from K₂SO₄ was detected after heating a NaCl, KCl, MgCl₂, MgSO₄, MgO, CaCl₂, K₂SO₄, and FeSO₄ with RB. SO₂ content of BS increased with baking time, and it originated from BSRB1 (13.88 ppm) to BSRB4 (109.13 ppm). SO₃<SUP>2-</SUP> originated only from MSSRB4 and BSRB2~BSRB4. Sulfate ion content decreased along with increasing SO₂ and sulfite ion contents. ORP increased with baking time of MSS and BS, and it was present at higher levels in BSRB4 (-211.40 mV) of BS than MSS. Insoluble content was higher in BS than MSS. Further, Ca, K, and Mg ion contents decreased in MSS and increased in BS with baking time. BSRB4 had 1.4 fold higher levels of Ca, 1.5 fold higher levels of Mg, and 1.8 fold higher levels of K than BS. Li, Al, Mn, Fe, and Sr in MSS as well as Al, Fe, and Ni in BS increased with baking time. Anions (Cl, NO₃, and Br) and heavy metals (Pb, Cd, Hg, and As) between MSS and BS were not significantly different. These results suggest that the reducing power of BS was due to SO₂ and sulfite ion. To increase the amounts of these compounds and reducing power, higher melting temperature and longer baking time are necessary along with BS, which is created by the addition of RB to roasted salt.
