기호음료 성분의 Angiotensin-I 전환효소 저해작용

The present study was conducted to investigate Angiotensin I-converting enzyme(ACE) inhibition activity of the components of traditional tea materials in Korea. Angiotensin I-converting enzyme(ACE) inhibition activity of water soluble fractions obtained from the samples were strong in Zingiberis rhizoma, Acantopanacis cortex, Schizandrae fructus, Perilla semen, Cassiae torae semen, Zizyphy fructus in order. ACE inhibition activity of fractions obtained from methanol extract of Cassiae torae semen were strong in ethyl acetate fraction, ethyl ether fraction, water fraction, chloroform fraction in order. Compound C showed the strongest ACE inhibition activity among compound A, B, C, D separated from Cassiae torae semen, but Compound C separated from Cassiae torae semen was lower than bradykinin in the ACE inhibition activity. 전통 기호음료 성분에 의하여 나타나는 기능특성을 조사하기 위한 연구의 일환으로 결명자,들깨, 대추, 모과, 오미자, 오갈피 및 생강 추출물을 ion-exchange chromatography, 유기용매에 의한 분획, silica gel column chromatography, thin layer chromatography에 의하여 여러 가지 획분으로 분획하여 Angiotensin I 전환효소(ACE) 저해작용을 조사한 결과는 다음과 같다. 기호음료 원료에서 추출한 수용성 획분의 ACE 저해작용은 생강>오갈피>오미자>들깨>결명자>모과>대추의 순으로 나타났다. 일반 가정에서 널리 이용되고 있는 결명자에서 분리한 compound C 는 ACE 저해제로 알려져 있는 bradykinin에 비하여 ACE 저해작용이 비교적 낮았다.
다양한 정제방법에 의한 전기영동용 한천유래 아가로즈의 제조 및 DNA분리 특성

제주도산 우뭇가사리로부터 agar를 제조하여 분석용 등급의 agarose 제조를 위하여 cetylpyridium chloride (CPC), polyethylene glycol (PEG), chitosan을 처리하여 agaropectin을 제거하여 agarose를 분리, 정제하였다. 3가지 방법중 chitosan을 처리하여 정제한 agarose는 황산기 함량이 가장 낮았으며, electroendosomosis도 가장 낮은 수치를 나타내어 분석용 agarose 제조를 위한 가장 효율적인 방법으로 나타났으며 실제 DNA 전기영동 실험에서도 가장 우수한 resolution을 나타내었으며 교차면역 전기영동에서도 시판되고 있는 제품에 비하여 품질이 양호함을 알 수 있었다. The present study was conducted to investigate the preparative methods of agarose for gel electrophoresis from agar. Naturally occuring agar consists of two main polysaccharides, the neutral polysaccharide agarose and the acid sulphated polysaccharide agaropectin. The sulphate and carboxyl functions of the agar are accumulated in the agaropectin. The hydrophilic, non-ionogenic, rigid and transparent gel matrix of the agarose was found to be suitable for gel electrophoresis gel filtration and affinity chromatography. Agar was purified by chitosan treatment, cetylpyridinium chloride (CPC) treatment, and polyethylene glycol (PEG) treatment. Yields of agarose purified from agar with chitosan, CPC and PEG were 56.7%, 55.6% and 62.3%. It was proper to treat with chitosan in preparative methods of agarose for gel electrophoresis from agar.
