      • AH-66 Hepatoma 세포의 Histone에 관한 연구

        In attempt to investigate histone fractions of tumor cells, histone fractions HI, H2a, H2b, H3 and H4 were prepared from isolated nuclei of AH-66 hepatoma cells and histone fractions were characterized by amino acid analysis and by polyacrylamide disc gel electrophoresis. The results obtained were as follows; 1, The five histone fractions found in most tissues were also present in the AH-66 hepatoma cell histories. 2. The yield of whole histone was 6.46mg per g of packed cells. 3. The yield of DNA was 14.78mg per g packed cells. Consequently the DNA to histone ratio was 1:0.44. 4. The relative amounts of five fractions, i.e., H1, H2a, H2b, H3 and H4 were 15.64% 34.83%, 21,21%, 11.76% and 16.56%, respectively. The data presented in this report show that there is a considerable variation in the relative amounts of histone fractions as compared to those of calf thymus. 5. The electrophoretic mobility of individual histone fractions gave almost similar patterns to those of corresponding fractions of calf thymus. 6. Amino acid analysis of the individual histone fractions showed that the over-all compositions were similar but not identical to those of corresponding fractions from calf thymus and other tumor cells. 7. Histone fractions HI, H2b. H3 and H4 were found to contain detectable amounts of ε-N-monomethyllysine. Usually the methylated derivatives of histidine have been reported in histone Hi fraction of Sarcoma 180 ascites tumor cells and Walker 256 carcinosarcoma. However, the methylated histidine residue was not found in all histone fractions from AH-66 hepatoma cells. 8. The evidence was presented that AH-66 hepatoma cells contained five major histone fractions which were homologous to corresponding five fractions of calf thymus.

        3차 다항식을 이용하여 구성된 곡선을 Piecewise cubic polynomial curves(PCPC)라 하며, 이 개념을 이용하여 제시된 제어점들을 통과하면서 곡면이 구성되도록 하는 방법이 Piecewise cubic polynomial splines(PCPS)이다. 본 논문에서는 PCPS법을 이용하여 스플라인 곡면을 구성하는 새로운 알고리즘을 개발하였으며, 여러가지 방법에 의하여 구성된 곡면들의 특성을 비교 분석하였다.

        Objective : The use of herbal therapy is becoming an increasingly attractive approach for the treatment of various inflammatory disorders. The Alpiniae officinari Rhizoma is popular in Aisa as a traditional herbal medicine. Alpiniae officinari Rhizoma is a species of the ginger family(Zingiberacease). Method : This study was performed to investigate the anti-inflammatory effect of Alpiniae officinari Rhizoma extract by the methods of "carrageenan induced paw edema" and "Lipopolysaccharide-induced inflammatory mediators in mouse macrophage RAW 264.7 cells". Result : We suggest that Alpiniae officinari Rhizoma extract decreased paw volume induced by plantar injection of carrageenan. Also Alpiniae officinari Rhizoma extract inhibited nitric oxide, prostaglandin E₂production and induced nitric oxide synthase, cyclooxygenase-2 protein expression in Mouse macrophage RAW 264.7 cells stimulated with lipopolysaccharide. Conclusion : This study shows that Alpiniae officinari Rhizoma extract seems to have anti-inflammatory effect by inhibition of nitric oxide, prostaglandin E_(2) production and nitric oxide synthase, cyclooxygenase-2 protein expression.

        1930년대 신식요리강습회로 본 상류층의 '식생활개선'

        This paper scrutinizes a specific social factor which was the Koreans" acceptance of a particular consumption pattern from abroad. The purpose of such acceptance was to improve Koreans" dietary lifestyle. Previous studies focused their attention upon the consumption?repressive propaganda of the Joseon Governer General office. But it should be noted that cooking classes promoting new kind of diets were arranged at the time, and that they targeted the affluent class, eventually serving as a Consumer movement encouraging purchase of modern diets. The Lifestyle Improvement Movement of the 1920s tried to implant westernized diets on the Koreans" lives. And the Korean population rejected that movement, as it ignored both traditional foods and the public"s purchasing power. In response, the Lifestyle Improvement Movement developed new foods fusing traditional food and westernized Japanese spices in the 1930s. The rich people became the targets, primarily because of their purchasing power. As a part of the Lifestyle Improvement Movement in Seoul, YWCA invited women from prosperous families to cooking classes. And in rural areas, commercial shops took advantage of this atmosphere by offering cooking classes. As a result, modern food became symbols of the high class figures, while Korea was being assimilated into the Japanese food market. Hence, Korean traditional food underwent a phase of change. This paper traced the historical origin of what led to the popularization of western diets in the Koreans" lifestyle.

      • 계지의 항염 효과에 관한 연구

        Objectives : Cinnamomi Ramulus (CR), the young twing of Cinnamomi loureirii nees, has been used for treating symptoms related to pain. rheumatic arthritis and inflammation in Korean herb medicine. This study was carried out to investigate the anti-inflammatory effect of CR in vivo and in vitro. Methods : Extracts of CR were prepared and the chemical components of the extracts were examined by gas chromatography-mass spectrometry (GC-MS). The extracts were administrated to the rat paw edema model induced by carrageenan to evaluate the anti-inflammatory effect of CR. The expressions of nitric oxide (NO). prostaglandin E2(PGE2), inducible nitric oxide synthase (iNOS) and cyclooxygenase (COX)-2 were also quantified in lipopolysaccharide (LPS)-induced RAW 264.7 macrophages to survey the effect of CR in vitro. The main components were cinnamaldechyde and coumarin. Results : We examined the anti-inflammatory activity of the 80% ethanol extract of Cinnamomi Ramulus in vivo by using carrageenan-induced rat paw edema model. Maximum inhibition of 54.9% was noted at the dose of 1000㎎/㎏ after 2 hours of drug administration in carrageenan-induced rat paw edema and this showed a potent anti-inflammatory effect. Conclusions : The results showed that Cinnamomi Ramulus suppressed dose-dependently LPS-induced NO production in RAW 264.7 macrophages and also decreased iNOS protein expression. Cinnamomi Ramulus also showed a significant inhibitory effect in LPS-induced PGE2 production and COX-2 expression.

        체성 연부 조직에 생긴 평활근육종: 1예 보고

        체성 연부 조직에 생기는 평활근육종은 매우 드물다. 66세 남자 환자가 방사선 소견상 경계가 좋은 양성 종양이 의심되어 절제술을 시행 받았다. 육안조직소견상 경계가 좋은 둥근 종괴이나, 현미경 소견에서 세포충실도가 증가되고 부분적으로 중증도의 핵이형성을 보였다. 또한 면역조직화학염색상 Smooth muscle actin과 Desmin에 동시에 양성이어 평활근세포 기원으로 생각되었다. 저자들은 괴사가 없고 유사분열은 단 한 개만이 관찰되었으나 중증도의 핵이형성을 보였기에 평활근육종으로 진단한 증례를 경험하였기에 문헌고찰과 함께 보고하는 바이다. Leiomyosarcoma of somatic soft tissue is a rare tumor compared with retroperitoneal lesion. We report a case of a leiomyosarcoma of the somatic soft tissue in a 66-year-old man. He presented as an enlarging mass in the left thigh for eight months. Radiologic examinations revealed a well defined round mass, suspicious of benign tumor, such as hemangioma or leiomyoma. He underwent surgical resection. The mass was 3.0 cm in diameter, and it was confined within adductor longus muscle without any connection to adjacent neurovascular bundles. Histologic examination showed moderately cellularity and focal marked atypia with a fascicular growth pattern of spindle cells showing blunt ended nuclei. In addition, the proliferation index was 2~3% by immunohistochemistry using monoclonal antibody MIB-1. But only one definite microscopic mitotic feature was found. Although it showed low mitotic activity without necrosis, this case was diagnosed leiomyosarcoma according to marked cellular atypism.

        중온 침출수 재순환 혐기성 소화 시스템을 이용한 음식물류 폐기물 처리

        본 연구에서는 침출수 재순환 시스템을 적용한 중온 혐기성소화를 이용하여 음식물류 폐기물을 분해하여 메탄가스를 생산하였다. 실험은 36℃로 유지되는 항온수조 내에 생물반응조와 침출수 저장조로 구성된 2개의 동일한 시스템(System A, System B)을 사용하였고, 생물반응조 하단 30 mm위에는 스크린이 있어 고액분리를 하여 침출수 저장조로 침출수를 이송하였다. 침출수 재순환은 매일 수행하였으며, 침출수 재순환 시에는 생물반응조 하단에서 침출수 저장조로 2.5 L를 30분간 이송한 뒤 다시 침출수 저장조에서 생물반응조 상부로 2.5 L를 30분간 주입하였다. 주입된 음식물류 폐기물은 수집되기 전 한 번 세척하였으며 반응조에 주입되기 전에 36℃로 온도를 올렸다. System A에 49.1 g VS, System B에 54.0 g VS을 2주 간격으로 투입하였다. 저해인자로 측정된 항목은 NH₄⁺-N과 염도였으며, 두 가지 항목의 농도 모두 시스템에 끼친 영향은 없는 것으로 나타났다. System A는 112일간, System B는 140일 동안 운전하였는데, 각 시스템에서 인발된 슬러지는 없었다. 음식물류 폐기물의 혐기성 소화를 통한 평균 메탄 발생량은 System A의 경우 0.439 L CH4/g VS, System B의 경우 0.368 L CH4/g VS로 나타났다. In this study, mesophilic anaerobic digestion of source separated food waste was carried out by leachate recirculation system and methane gas was produced. Two systems - system A and B were fabricated and placed within water bath to maintain 36℃. Each system was comprised of an anaerobic bioreactor and a leachate tank. Leachate in bioreactor was separated through the screen located at 30 mm above the bottom and a pump was installed to transfer collected leachate to the leachate tank. Everyday, 2.5 L of the leachate was pumped from the bioreactor to the leachate tank for 30 min and transferred leachate was pumped back to the top of the bioreactor for 30min, sequentially. Source separated food waste used for this experiment was washed by water before transferring to the laboratory. Transferred food waste was warmed to 36℃ before being fed to bioreactors. System A was fed to 49.1 g VS (Volatile Solids) and System B was fed to 54.0 g VS at every two weeks, respectively. NH₄⁺-N and salinity were monitored to see the inhibition toward anaerobic bioreaction and it was found that concentrations of these materials were not high enough to affect the bioreaction. Although the food waste was fed biweekly for 112 days and 140 days at system A and B, respectively, there was no sludge withdrawal from each system. Average methane productions rates were 0.439 L CH4/g VS and 0.368 L CH4/g VS for system A and B, respectively.

        Public Health Policy and Citizen Participation Citizen participation is citizen's public behavior which directly involves in and influences policy process. The types of citizen participation can be categorized into institutional participation and noninstitutional participation. Institutional participation means that the government institutionally legitimizes citizen participation in administrative system. Institutional participation includes cooperative participation and autonomy. The examples of cooperative participation are public hearing, commissioner for administration, citizen committee, professional committee, administrative council, etc. Noninstitutional participation includes citizen's campaign and bargaining. Citizen participation in public health policy process has been activated in Korea with democratization. At present, the types of citizen participation in public health policy includes various public health committees, public hearing, lawsuit for public good, civil petition, report system, citizen's proposal, claim for inspection of the public health administration, monitoring, citizen's campaign, etc. Among them, the various public health committees are operated on the basis of health related laws. According to the laws, it appears that there are many high ranking officials in the committees, and committee members are mostly appointed by the ministers or governors. Therefore, it can be said that the committees are operated by the will of the government. The committees are not characterized as decision making organizations. The function of them is only counseling. It is difficult to expect real participation through institutional participation. Recently, Korean people have formed various citizen organizations related to public health for active citizen campaign. The organizations have actively participated in the public health policy proess and greatly affected change or adoption of public health policy including health insurance, separation of dispensary from medical pratice, etc. To enhance the citizen participation in public health policy, it is important to introduce ombudsman system, lawsuit system by a representative, and direct citizen participation system. The people's awareness level of citizen participation must also increase and the bureaucrats' behavior needs to be changed. It is necessary to activate freedom of public administration information and enlarge local autonomous right. Citizen participation can be also enhanced greatly through development of citizen's campaign.
