두메오리나무잎에서 flavonid 화합물의 분리 및 물오리나무잎에서 분리한 diarylheptanoid 화합물의 Nitric Oxide 생성억제활동
자작나무과(科) Betulaceae의 오리나무속(屬, Alnus species)에 속하는 식물은 17종이상이 우리나라에 자생하는 것으로 알려졌다. 민간 및 한방에서는 오리나무 Alnus japonica의 수피를 赤楊이라 부르며, 청열(淸熱), 강화(降火)하는 작용이 있어서 비출혈(鼻出血) 등의 목적에 쓰였고, 설사 그외의 외상출혈 및 숙취 등에도 응용한 것으로 나타나 있다. Ainus속 식물에는 diarylheptanoid계열을 비롯한 Flavonoid, tannin 등 페놀성화합물이 함유된 것으로 알려져 있으며, 그 外 dammarane type의 triterpenoid도 주목을 받아왔다. 본 연구자는 국내자생종중의 하나인 두메오리나무(Alnus maximowiczii Call)의 잎으로부터 페놀성 화합물을 분리하고 그 화학구조의 규명을 시도하여 3종의 flavonoid를 분리하였다. 이들은 각종 이화학적 성상과 IR, UV, MS, NMR등의 기기분석 자료를 해석하여 quercetin-3-O-β-D-glucopyranoside(1), quercetin-3-O-α-L-rhamnopyranoside(2) 및 myricetin 3-O-α-L-rhamnopyranoside(3)로 동정하였다. 이들 flavonoid 화합물은 두메오리나무잎에서는 처음으로 분리되었다. 또한 물오리나무(Alnus hirsuta Turcz)잎에서 분리된 diarylheptanoid의 Nitric Oxide 생성억제 효과를 실험하였으며 그 결과 hirsutanonol과 oregonin이 활성화된 RAW 264.7cell에서 iNOS protein의 발현억제에 기인하는 Nitric Oxide(NO) 생성억제 효과를 나타내었다. 따라서 hirsutanonol과 oregonin은 강력한 NO 생성억제약물임을 알 수 있었다. Alnus species are deciduous board-leaved trees or shrubs found in the damp place and mountain area and more than 17 species are grow wild in Korea. The barks of Alnus japonica have been used for oriental traditional medicine as the remedies for fever, hemorrhage and diarrhea. The chemical constituents of these plants are well known as diarylheptanoid, tannin and triterpenoid. Phytochemical examination of the leaves of Alnus maximowiczii Call, one of the indigenous Alnus species grows in Korea, has led to the isolation of three flavonoid. Structure of these flavonoids were elucidated as quercetin-3-O-β-D-glucopyranoside (1), quercetin-3-O-α-L-rhamnopyranoside (2) and myyricetin-3-O-α-L-rhamnopyranoside (3) respectively by means of physico-chemical and spectral evidences. Four diarylheptanoids which were isolated from the leaves of Alnus hirsuta Turcz and have been assessed for nitric oxide (NO) production inhibitory effects in vitro. Hirsutanonol and oregonin showed inhibition of NO synthesis in dose-dependent manners by murine macrophage-like RAW 264.7 cells stimulated with interferon-γ (IFN-γ) plus lipopolysaccharide (LPS). Their 50% inhibitory concentrations (IC50) were 14.3 and 3.8 μM, respectively. The inhibitory effects of these compounds on NO synthesis were due to suppression of iNOS mRNA expression as determined by Northern and Western blotting. Hirsutanonol and oregonin were found to be potent inducible nitric oxide synthase (iNOS) inhibitors.
