        Physicochemical properties analysis of bamboo salt on chicken emulsion sausage

        Quality characteristics of chicken emulsion sausage manufactured with various levels of NaCl and 9 times heated bamboo salt (0.3%, 0.6%, 0.9%, and 1.2% respectively) were examined. The pH value of chicken emulsion sausage was increase tendency with increasing amount of bamboo salt, on the contrary in case of NaCl sample were decrease tendency with increasing amount of NaCl. Both before and after heating, redness of chicken emulsion sausage with bamboo salt treatments were upward trend with increasing amount of bamboo salt. water holding capacity (WHC) of 1.2% NaCl sample was significantly higher than 0.3%-0.9% (p < 0.05), but 0.9%, 1.2% bamboo salt samples were significantly higher than 0.3, 0.6 % (p < 0.05). Water loss of 1.2% NaCl and 0.9% bamboo salt samples were significantly lower than other treatment (p < 0.05). Protein solubility values significantly increased amount of bamboo salt and NaCl (p < 0.05), and samples of 0.9% NaCl and 0.6% bamboo salt values show similar values. Cooking yield of samples were increased tendency with increasing amount of NaCl and bamboo salt. Also viscosity values of sample containing 1.2% bamboo salt sample showed higher viscosity than other treatments. These results show that containing 1.2% NaCl chicken emulsion sausage and 0.9% bamboo salt chicken emulsion sausage were similar physicochemical properties. Therefore, bamboo salt is suitable for manufacturing chicken emulsion sausage.

        Antioxidation and Cancer Cell (HT-29) Antiproliferation Effects of Rubus coreanus Miquel Bamboo Salt

        Bamboo is used by Korean as an important medicinal material. It has many different medicinal efficacy from its stub, roots and leaves. Rubus coreanus Miquel (raspberry) is thought to contain compounds which are important towards having health benefits. Rubus coreanus Miquel bamboo salt sample was prepared and showed in vitro antioxidant, antimutagenic and anticancer effects in this study. The radical scavenging effect of Rubus coreanus Miquel bamboo salt on DPPH radical were tested. The Rubus coreanus Miquel bamboo salt sample revealed better scavenging activities on DPPH radical than the other salt samples at concentrations of 10 and 20%. Antimutagenic effects of Rubus coreanus Miquel Bamboo salt were investigated and compared with other salt samples. Though solar salt exhibited comutagenicty with MNNG, bamboo salt showed antimutagenicity against MNNG. The Rubus coreanus Miquel bamboo salt showed more increased antimutagenicity than the bamboo salt. MTT assay was carried out with the salts treated HT-29 cancer cells. The Rubus coreanus Miquel bamboo salt showed better anticancer activities (69%) than purple bamboo salt (44%) and solar salt (11%) at a concentration of 1%, and Rubus coreanus Miquel bamboo salt also exhibited better anticancer activities than other salt samples at the low concentration of 0.5% (p<0.05). The analysis of apoptosis related genes was carried out using RT-PCR to check the anticancer effect in the colon cancer cells and showed the same anticancer cancer effects as shown in MTT assay. From the experimental results, Rubus coreanus Miquel bamboo salt exhibited better health functional effects than common bamboo salts and solar salt. (Cancer Prev Res 15, 306-312,2010)

        Bamboo salt attenuates CCl₄-induced hepatic damage in Sprague-Dawley rats

        Bamboo salt, a Korean folk medicine, is prepared with solar salt (sea salt) and baked several times at high temperatures in a bamboo case. In this study, we compared the preventive effects of bamboo salt and purified and solar salts on hepatic damage induced by carbon tetrachloride in Sprague-Dawley rats. Compared with purified and solar salts, bamboo salts prevented hepatic damage in rats, as evidenced by significantly reduced serum levels of aspartate aminotransferase, alanine aminotransferase, and lactate dehydrogenase (P < 0.05). Bamboo salt (baked 9×) triggered the greatest reduction in these enzyme levels. In addition, it also reduced the levels of the proinflammatory cytokines interleukin (IL)-6, interferon (IFN)-γ, and tumor necrosis factor (TNF)-α. Histopathological sections of liver tissue demonstrated the protective effect of bamboo salt, whereas sections from animals treated with the other salt groups showed a greater degree of necrosis. We also performed reverse transcription-polymerase chain reaction and western blot analyses of the inflammation-related genes iNOS, COX-2, TNF-α, and IL-1β in rat liver tissues. Bamboo salt induced a significant decrease (~80%) in mRNA and protein expression levels of COX-2, iNOS, TNF-α, and IL-1β, compared with the other salts. Thus, we found that baked bamboo salt preparations could prevent CCl₄-induced hepatic damage in vivo.

        Bamboo Salt Has In Vitro Anticancer Activity in HCT-116 Cells and Exerts Anti-Metastatic Effects In Vivo

        Bamboo salt is a traditional food widely used in Korea. The in vitro anticancer effects of this salt were evaluated in HCT-116 human colon cancer cells using a 3-(4,5-dimethyl-2-thiazolyl)-2,5-diphenyltetrazolium bromide (MTT) assay. A 1% salt concentration of bamboo salt baked nine times (9×) inhibited the growth of HCT-116 cells by 53%, which was higher than salt baked three times (3×) or once (1×; 44% and 41%, respectively) and much higher than solar sea salt (Korean sea salt) and purified salt (22% and 18%, respectively). To elucidate the inhibitory mechanisms underlying the anticancer effect of the salt samples in cancer cells, expression of genes associated with apoptosis, inflammation, and metastasis was measured with reverse transcription–polymerase chain reaction and Western blotting. Bamboo salt (9×) significantly induced apoptosis in cancer cells (P<.05) by upregulating Bax, caspase-9, and caspase-3, and downregulating Bcl-2. The expression of genes associated with inflammation (NF-κB, iNOS, and COX-2) was significantly downregulated (P<.05) by 9× bamboo salt, demonstrating its anti-inflammatory properties. The 9× bamboo salt also exerted a greater anti-metastatic effect on cancer cells than the other salts as demonstrated by decreased mRNA expression of MMP genes and increased expression of tissue inhibitors of metalloproteinases, which was confirmed by the inhibition of tumor metastasis induced in colon 26-M3.1 cells in BALB/c mice. In contrast, purified and solar salts increased metastasis in the mice. Our results demonstrated that 9× bamboo salt had the most potent in vitro anticancer effect, induced apoptosis, had anti-inflammatory activities, and exerted in vivo anti-metastatic effects. Additionally, the anticancer, anti-inflammatory, and anti-metastatic effects of the 1× and 3× bamboo salts were stronger than those of the purified and solar salts.

        Bamboo salt has been used for the purpose of prevention and treatment of various diseases in Korea. Present study was carried out to ascertain the effects of purple bamboo salt upon anti-allergic effect, anti-inflammatory activity and immune-enhance effect as well. Purple bamboo salt significantly inhibited the ear swelling response and histamine release induced by compound 48/80 in mice and rat peritoneal mast cells. Purple bamboo salt (0.01 ∼ lg/kg) also dose-dependently inhibited the passive cutaneous anaphylaxis by oral administration. Purple bamboo salt (1 mg/mL) in hibited phorbol 12-myristate 13-acetate plus calcium ionophore A23187-stimulated tumor necrosis factor (TNF)-${\alpha}$, interleukin (IL)-1${\beta}$ and IL-6 secretion, by 67.04${\pm}$0.08%, 68.01${\pm}$1.85%, 69.48${\pm}$0.54%, respectively. In addition, purple bamboo salt inhibited the expression of TNF-${\alpha}$ mRNA in HMC-1 cells. Finally, we investigated the effect of purple bamboo salt in the forced swimming test (FST) and the change of purple bamboo salt-mediated cytokine production from MOLT-4 cells. At the 7th, immobility time was significantly decreased in the purple bamboo salt-administration group (35.4 ${\pm}$5.9 s for 1 g/kg) in comparison with the control group (93.2 ${\pm}$ 15.45). After FST, the content of glucose in the blood serum was increased and the levels of blood urea nitrogen, lactic dehydrogenase was decreased in purple bamboo salt-administration group. However, it had no effect on the elevation of CK and TP level. Purple bamboo salt (1 mg/mL) significantly increased the interferon (IFN)-${\gamma}$ and IL-2 level compared with media control (about 3.7-fold for IFN-${\gamma}$, about 3.5-fold for IL-2, p〈0.05) but did not affect the IL-4.

        칼륨죽염의 in vitro 항암 기능성 증진 효과

        칼륨을 첨가하여 제조한 죽염의 항암효과를 일반죽염 및 천일염, 정제염과 비교하여 칼륨 첨가의 영향을 알아보고자 하였다. DPPH radical 소거능을 측정한 결과, 칼륨죽염과 죽염의 radical 소거능이 높았고, 칼륨죽염의 소거능이 가장 높게 나타났다. 정제염과 천일염의 소거능은 상대적으로 낮았고 칼륨죽염과 20배 이상의 차이가 있었다. MNNG를 사용한 Salmonella Typhimurium TA100 돌연변이 실험결과 소금은 모두 보돌연변이 효과가 있었지만, 칼륨죽염의 보돌연변이 효과가 다른 소금보다 3~46% 낮게 나타났다. HCT-116 암세포에 성장 억제 효과에 있어서도 위와 비슷한 경향을 나타내었는데, 칼륨죽염과 일반죽염의 암세포 성장 억제효과와 암세포의 apoptosis 유도 효과가 천일염 및 정제염에 비해 높았고 그중 칼륨죽염의 in vitro 항암 기능성이 가장 우수하였다. 이상의 결과로부터 죽염의 항암효과는 천일염이 정제염보다 좋으며, 이러한 죽염의 항암효과를 칼륨을 첨가함으로써 더욱 증진될 수 있는 것으로 나타났다. 본 연구에서 죽염의 항암 기능성이 증가한 주요 원인은 칼륨이라고 생각되어지며, 이것이 죽염으로 제조될 때 그 효과가 더 증진되는 것으로 보인다. Potassium added with bamboo salt showed better antioxidative effects than bamboo salt, solar salt, or purified salt. It also showed inhibitory effects on the mutagenicity of MNNG (N-methyl-N-nitro-N-nitrosoguanidine) in a Salmonella Typhimurium TA100 tester strain. At concentrations of 1.25 and 2.5 mg/plate, potassium bamboo salt and bamboo salt showed weaker co-mutagenicity effects than either purified salt or solar salt, respectively. Anticancer effects of salts were evaluated using MTT assay in HCT-116 human colon carcinoma cells. At a 1% salt concentration, the growth inhibitory rate of potassium bamboo salt was 54%, higher than that of 1 time baked bamboo salt (36%). However, purified salt and solar salt showed relatively lower inhibitory effects of 19% and 23%, respectively. To determine the inhibitory mechanisms of potassium bamboo salt, the expression levels of Bax and Bcl-2 genes in HCT-116 cells were determined by RT-PCR. Potassium bamboo salt significantly increased Bax and decreased Bcl-2 expression levels unlike bamboo salt, purified salt, and solar salt (p<0.05). Therefore, addition of potassium to salt decreased co-mutagenicity and increased in vitro antioxidative and anticancer effects.

        죽염 된장의 발효 특성 및 기능성 증진 효과

        The aim of this study is carried out to evaluate the fermentation characteristics and increased functionality when doenjang was prepared with bamboo salts. Grain type mejus were fermented with mixed starter cultures of Aspergillus oryzae, Bacillus subtilis, and Lactococcus lactis. These mejus were mixed with different kinds of saltspurified salt (PD), sea salt (SD), one-time baked bamboo salt (1×BD), three-time baked bamboo salt (3×BD), and nine-time baked bamboo salt (9×BD)- when doenjangs were prepared. For doenjang fermentation period of 8 weeks at 37°C, the fermentation characteristics of all the groups were compared. The amino type nitrogen content and enzyme activities (protease and α-amylase) in the samples were significantly increased. In DPPH radical scavenging activities and hydroxyl radical scavenging activities, 9×BD (47% and 69%) showed the highest scavenging activities compared to PD (40% and 49%), SD (42% and 57%), 1×BD (42% and 64%) and 3×BD (45% and 65%) (P<0.05). The anticancer effects of doenjang in HT-29 cancer cells indicated all the groups, especially doenjang prepared with bamboo salts were higher than the others (P<0.05). Apoptosis related genes of Bax and Bcl-2, as well as inflammation related genes of iNOS and COX-2 were regulated by the treatment of doenjangs in HT-29 cancer cells. SD, 1×BD, 3×BD, and 9×BD increased the expression level of Bax and decreased the expression level of Bcl-2, iNOS, and COX-2. These results suggest that sea salt and bamboo salt especially bamboo salt could improve fermentation characteristics and functionality of doenjang and play an important role in regulating apoptosis and inflammation related genes in cancer cells.

      • Effect of Bamboo salt-pro on carries-inducing properties of Streptococcus mutans

        We studied the effect of Bamboo salt-pro on the growth and acid production of S. mutans. The growth of S.mutans was reduced by the presence of the Bamboo salt-pro (1 mg/ml) and NaCl (1 mg/ml) significantly, and the positive control group (1 % of NaF) also exhibited antibacterial activity significantly. Bamboo salt-pro (1 mg/ml) reduced the rate of acid production by S. mutans. Bamboo salt alone did not demonstrate such a reduction in acid production at the concentration of 1 mg/ml. The inhibitory action of Bamboo salt-pro on acid production was found at a concentration of 1 mg/ml, but bamboo salt alone was not at a concentration of 1 mg/ml. In addition, we investigated the anti-inflammatory effect of Bamboo salt-pro on human mast cell line HMC-1. Bamboo salt-pro (0.1 and 0.01 mg/ml) inhibited significantly the secretion of inflammatory cytokine, tumor necrosis factor-a with $59.47{\pm}0.15%$, $51.98{\pm}0.16%$ respectively. Our results suggest that Bamboo salt-pro importantly contributes to the prevention or treatment of periodontitis and other oral diseases and inflammatory diseases.
