        태충 · 삼음교의 침 자극이 Streptozotocin으로 유발된 당뇨쥐의 신장 손상에 미치는 영향

        Objectives : This study was performed to investigate the effects of LR3 and SP6 acupuncture on renal damage in streptozotocin(STZ)-induced diabetic mice. Methods : ICR male mice were stabilized for a week and divided into four groups: a normal mice group(N), no-acupuncture diabetic mice group(Control), LR3 acupuncture diabetic mice group(LR3), and SP6 acupuncture diabetic mice group(SP6). Diabetes was experimentally induced by intraperitoneal injection of STZ(150 mg/kg) in citrate buffer(pH 4.5). For two weeks, LR3 and SP6 acupunctures were administered bilaterally at each point once a day. After two weeks, the animals’ weight was measured and they underwent a laparotomy. Serum glucose and blood urea nitrogen(BUN) were measured from the blood taken from the heart. We measured glucose, reactive oxygen species(ROS), peroxynitrite(ONOO-) and thiobarbituric acid reactive substances(TBARS) in the kidney and compared expression levels of superoxide dismutases(SOD), glutathione peroxidase(GPx), nuclear factor-kappa B(NF-kB), cyclooxygenase-2 (COX-2), inducible nitric oxide synthase(iNOS) and Interleukin-1 beta(IL-1β). Results : BUN significantly decreased in LR3, SP6 compared to the control group. LR3 showed significantly decreased glucose compared to the control group. LR3, SP6 significantly decreased in ROS and ONOO- compared to the control group. LR3 significantly decreased in TBARS compared to the control group. SP6 significantly increased in expressions of SOD-1, catalase, and GPx compared to the control group. LR3, SP6 significantly decreased in COX-2 compared to the control group. SP6 significantly decreased in IL-1β compared to the control group. Conclusions : This study suggests that LR3 acupuncture may be effective in controlling glucose and lipid peroxidation and that SP6 acupuncture may have anti-oxidative and anti-inflammatory effects on renal damage in STZ-induced diabetic mice.

        태충(太衝)(LR3)에 대한 영수(迎隨) 및 념전보사(捻轉補瀉)가 intraluminal filament 삽입술(揷入術)에 의하여 유발(誘發)된 백서(白鼠)의 focal ischemia에 미치는 영향(影響)

        Objectives: Acupuncture using a tonification or sedation techniques method is used as a controlling the medication for an early stroke in the Korean medicine. LR3 has indicatons of headache, vertigo, facial paralysis, apoplexy, epiepsy as the source acupoint of a Liver meridian. So this study is aims to investigate the anti-apoptotic and neuroprotective effects of acupuncture on the focal ischemia induced by intraluminal filament insertion in rats. Methods: The focal ischemia was induced by intraluminal filament insertion into middle cerebral artery. The animals were divided into seven groups (n=8 in each group): Normal, intactness group; Conrol group, no therapy group after being ischemia induced; MA-1, acupuncture perpendicularly without Tonification or Sedation techniques at LR3 after being ischemia induced; MA-2, acupuncture obliquely towards the knee at LR3 after being ischemia induced; MA-3, acupuncture obliquely towards the toe at LR3 after being ischemia induced; MA-4, acupuncture obliquely towards the knee and rotate 9 times in a clockwise direction at LR3 after being ischemia induced; MA-5, acupuncture obliquely towards the toe and rotate 6 times in a counterclockwise direction at LR3 after being ischemia induced. The anti-apoptotic and neuroprotective effects of Acupuncture techniques of tonification or sedation at LR3 are observed by mGluR5, Bax, Cresyl violet, ChAT-stain and NGF. Results: The intensity of mGluR5 and the density of ChAT was increased in MA-1 group. The intensity of Bax was decreased in MA-3, MA-4 group. The density of neurons stained by Cresyl violet and ChAT was increased in MA-2, MA-3, MA-4, MA-5 group. The density of neurons stained by NGF was only increased in MA-4 group. Conclusions: Our study suggests that acupuncture perpendicularly without Tonification or Sedation techniques and obliquely towards the knee and rotate 9 times in a clockwise direction(Tonifying technique) at LR3 after being ischemia induced at LR3 shows anti-apoptotic and neuroprotective effects on cholinergic neuron in focal cerebral ischemia of the stroke in rats.

        심정격(心正格)혈위(穴位), (소충(少衝),대돈(大敦), 소해(少海),음곡(陰谷)에 대한 영수(迎隨)및 념전(捻轉) 보사(補瀉) 침자(鍼刺)가 국소 뇌허혈 유발 흰쥐의 항뇌세포고사와 신경보호 효과에 미치는 영향

        Objectives: Acupoints of LR1(Dadun)·HT9(Shaochong), KI10(Yingu)·HT3(Shaohai) are used as controlling the diseases from heart problems. Especially, when they are used, tonification or sedation techniques method is taken as a controlling the medication for an early stroke in the Korean medicine. So the aim of this study is to investigate the anti-apoptotic and neuroprotective effects of Acupuncture techniques of tonification or sedation at LR1·HT9, KI10·HT3 on the focal ischemia induced by intraluminal filament insertion in rats. Materials and Methods: The focal ischemia was induced by intraluminal filament insertion into middle cerebral artery. AT3; acupuncture therapy at right LR1, HT9(twirling forward with the thumb of right hand 9 times), KI10·HT3(twirling forward with the forefinger of right hand 6 times) AT-4; acupuncture therapy at right LR1, HT9(forward direction, twirling forward with the thumb of right hand 9 times), KI10·HT3(opposite direction, twirling forward with the forefinger of right hand 6 times) inserting to the direction of route of the each meridian. The anti-apoptotic and neuroprotective effects of Acupuncture techniques of tonification or sedation at LR1, HT9, KI10·HT3 were observed by Bax, Bcl-2, mGluR5, Cytochrome c, Cresyl violet and ChAT-stain. Results: The intensity of Bax and Bax/Bcl-2 ratio were increased in ACU-2 and ACU-4 group but decreased in ACU-3 group. The intensity of mGluR5 was increased in ACU-1, ACU-2, ACU-3 and ACU-4 group. The intensity of cytochrome c was decreased in ACU-3 and ACU-4 group. The density of neurons stained by Cresyl violet and ChAT were increased in ACU-1, ACU-3 and ACU-4 group. Conclusions: Our study suggests that acupuncture therapy of tonification at LR1, HT9 by twirling forward with the thumb of right hand 9times, sedation at KI10·HT3 by twirling forward with the forefinger of right hand 6 times after perpendicularly inserting needle shows anti-apoptotic and neuroprotective effects on cholinergic neuron in focal cerebral ischemia of the stroke in rats.

        태충혈과 삼음교혈의 침 자극이 Streptozotocin으로 유발된 당뇨쥐의 간 손상에 미치는 영향

        The Effects of LR3 and SP6 Acupuncture on Liver Damage of Streptozotocin-induced Diabetic Mice Sung Jin Kim, Yun Kyu Lee, Hyun Jong Lee and Jae Soo Kim* Dept. of Acupuncture & Moxibustion medicine, College of Korean Medicine, Daegu Haany University Objectives : The purpose of this study was to investigate the effect of LR3 and SP6 acupuncture on liver damage of streptozotocin-induced diabetic mice. Methods : Male ICR mice were divided into four groups, consisting of the normal mice group(N), acupuncture-free diabetic mice group(Con), LR3-acupuncture diabetic mice group(LR3) and SP6-acupuncture diabetic mice group(SP6). The following measurements were taken: Body weight, food intake and water intake for 2 weeks; liver weight, and glucose levels in the serum and liver; ALT and AST in the serum; reactive oxygen species(ROS), reduced glutathione(GSH) and oxidized glutathione(GSSG) in the liver; and lastly, receptor for advanced glycation endproducts( RAGE), Nε-carboxymethyl lysine(CML), Nε-carboxyethyl lysine(CEL), phosphorylation of inhibitory kappa B alpha(p-IκBα), nuclear factor-kappa B(NF-κB), activator protein-1(AP-1), cyclooxygenase-2(COX-2), inducible nitric oxide synthase(iNOS), tumor necrosis factor-alpha(TNF-α), β-actin, cytochrome c and caspase in the liver. Results : The liver weight and GSH/GSSG ratio were significantly increased in SP6 compared to Con. The glucose levels in the liver were significantly decreased in LR3 compared to Con. The generation of ROS and GSSG were significantly decreased in SP6 compared to Con. The expressions of RAGE, CML, AP-1, TNF-α, cytochrome c and caspase 3 were significantly decreased in LR3 compared to Con. The expressions of p-IκBα, NF-κB, AP-1, COX-2, iNOS and caspase 3 were significantly decreased in SP6 compared to Con. Conclusion : It is predicted that LR3 acupuncture is related to reduced glucose levels in the liver and expressions of AGE, and that, SP6 acupuncture is related to reduced oxidative stressrelated transcription factors and inflammation-related proteins. Therefore, we suggest that LR3 and SP6 acupuncture have protective effects on the liver of streptozotocin-induced diabetic mice by preventing apoptosis.

        Objectives and Methods: This study was performed to investigate the effect of electro -acupuncture at four gates(bilateral LR3 and LI4) on brain activity in normal subjects using fMRI. Results and Conclusions: 1. fMRI signal increase by electro-acupuncture at Lt. LR3 was observed in Rt. Middle frontal gyrus in group average as well as more than half of the subjects. 2. fMRI signal decreases by electro-acupuncture at Lt. LR3 were observed in Rt. Superior frontal gyrus, Rt. Middle temporal gyrus, Rt. Cingulate gyrus in group average as well as more than half of the subjects. 3. fMRI signal increases by electro-acupuncture at Lt. LI4 were observed in Lt. Superior frontal gyrus, Lt. Middle frontal gyrus, Lt. Inf. Semi-Lunar Lobule(cerebellum), Rt. Middle frontal gyrus, Rt. Cingulate gyrus in group average as well as more than half of the subjects. 4. fMRI signal decreases by electro-acupuncture at Lt. LI4 were observed in Lt. Middle frontal gyrus, Lt.         태충의 해부학적 구조와 효율적인 자침법에 관한 연구

        Background: The aim of this study is to provide the anatomical information and physi- ological effect of LR3 with its needling depth and retention time. Methods: We investigated the information of LR3 from literature and related research. Then we searched the clinical effect and needling depth, angle, retention time of LR3 from on-line databases like ‘DBPIA’, ‘KISS’, ‘OASIS’, ‘RISS’, PUBMED’ and ‘Google Scholar’ (2005-2023). Results: The needling depth of LR3 is 7-13 mm in the classic test, that of contemporary re- search is 10-20 mm. When stimulating LR3, the relative anatomical structures are dorsalis pedis artery and medial terminal branch of deep peroneal nerve. Conclusion: The effect of LR3 is originated from stimulation of deep peroneal nerve near- by dorsalis pedis artery. It activates the parasympathetic nerve through the induction of somatic autonomous reflex which leads to the effect of blood vessel expansion, intestinal movement activation, and blood sugar drop.

        太白·神門 및 大敦·湧泉에 시술한 竹茹 藥鍼이 2K1C 고혈압 白鼠의 혈압에 미치는 영향

        Objectives : The purpose of this study was to compare Taebaek(SP3)․Sinmun(HT7), Daedon(LR1)․Yongcheon(KI1) on the Blood Pressure in Hypertensive RAT induced by 2K1C. Methods : This experiments was to investigate the effects of Herbal-Acupuncture at Taebaek(SP3)․Sinmun(HT7), Daedon(LR1)․Yongcheon(KI1) on the Blood Pressure, Cardiomegalic index, plasma levels of renin, plasma levels of atrial natriuretic peptide(ANP), serum levels of potassium, and serum levels of aldosterone in Hypertensive RAT induced by 2K1C. Results : The results were as follows. Ⅰ. Blood Pressure was decreased significantly after Herbal-Acupuncture of Taebaek(SP3)․Sinmun(HT7), Daedon(LR1)․Yongcheon(KI1). Ⅱ. Cardiomegalic index was decreased significantly after Herbal-Acupuncture of Daedon(LR1)․Yongcheon(KI1). Ⅲ. Plasma levels of atrial natriuretic peptide(ANP) was increased significantly after Herbal-Acupuncture of Taebaek(SP3)․Sinmun(HT7), Daedon(LR1)․Yongcheon(KI1). Ⅳ. Plasma levels of renin was decreased significantly after Herbal-Acupuncture of Taebaek(SP3)․Sinmun(HT7), Daedon(LR1)․Yongcheon(KI1). Conclusion : These results suggest that Blood pressure was decreased significantly after Herbal-Acupuncture of Daedon(LR1)․Yongcheon(KI1) in Hypertensive RAT induced by 2K1C.

        多種의 太衝(LR3) 鍼刺療法이 Intraluminal Filament 삽입술로 유발된 白鼠의 허혈성 국소 뇌손상에 미치는 영향

        Objective : This study was performed to investigate the effects of acupuncture therapy(AT, AT-9), electro-acupuncture therapy(EAT) and low level laser acupuncture therapy(LAT) at LR3 on the focal ischemia-induced by intraluminal filament insertion in rats. Methods : In the present syudy, the focal ischemia was induced by Intraluminal filament insertion into left middle cerebral artery. The subjects were divided into five groups after focal brain ischemia. (n=15, in each group) : Control with no treatment, AT with acupuncture at LR3, AT-9 with acupuncture at LR3 and rotating 9 times in a clockwise direction, EAT with electro-acupuncture at LR3 and LAT with invasive laser acupuncture at LR3. Anti-apoptotic and neuroprotective effects of acupuncture were observed by mGluR5 mRNA, Bax mRNA, Bcl-2 mRNA, Cytochrome C protein, Cresyl violet-stain and Choline acetyltransferase (ChAT)-stain in the hippocampus. Results : 1. In LAT, mGluR5, Cresyl violet-stain and ChAT-stain were increased. 2. In LAT, Cytochrome C protein was decreased. 3. In AT-9, Bax, Cytochrome C protein and the Bax/Bcl-2 ratio were decreased. 4. In AT-9, Bcl-2, Cresyl violet-stain and ChAT-stain were increased. 5. In EAT, Bcl-2 and Cresyl violet-stain were increased. Conclusions : These results suggests that LAT and AT-9 show anti-apoptotic and neuro-protective effects and that LAT and AT-9 may be useful for managing stroke by focal brain ischemia.

        태충 침자가 간 미토콘드리아 내 Complex IV에 미치는 영향

        Objectives : The liver is rich in mitochondria and it plays a key role in whole-body energy homeostasis. Mitochondria is double membrane-bound organelle that supplies energy for intracellular metabolism including Krebs cycle and beta-oxidation. Acupuncture is known to stimulate and regulate the flow of energy. To explore the effect of acupuncture on the mitochondrial respiratory chain activity in the rats' livers, the activity of mitochondrial respiratory chain complexes I to IV was observed. Methods : The rats were divided into 4 groups; Normal 1 (no acupuncture treatment and anesthesia for 5 min), Normal 2 (no acupuncture treatment and anesthesia for 10 min), MA1 (acupuncture treatment at bilateral LR3 under anesthesia for 5 min), and MA2 (acupuncture treatment at bilateral LR3 under anesthesia for 10 min). All rats were sacrificed and the livers were examined for respiratory chain change. Results : There was no difference in ubiquinon oxidoreductase, succinate dehydrogenase, and ubiquinol cytochrome C oxidoreductase after acupuncture at LR3. Acupuncture at LR3 for 10 min increased the activity of cytochrome C oxidase compared with no acupuncture groups. Conclusions : Acupuncture at LR3 mediated mitochondrial respiratory chain activity via the cytochrome C oxidase signaling pathway in the livers of rats.
