Anti-Inflammatory Activity of Phenylethanoids from Acanthus ilicifolius var. xiamenensis

Acanthus ilicifolius var. xiamenensis is a traditional herbal medicine in China. In this study, the anti-inflammatory activities of active ingredients of A. ilicifolius var. xiamenensis were investigated in RAW 264.7 cells and Freund's complete adjuvant-induced arthritic rats. Results showed that n-butanol extract exerted antiarthritic potential by reducing paw edema, arthritis score, and altered hematological and biochemical parameters in experimental rats. Phytochemical studies on n-butanol extract resulted in the isolation of five alkaloids (1–5) and five phenylethanoids (6–10). The anti-inflammatory assay of compounds 1–10 on lipopolysaccharide (LPS)-treated RAW 264.7 cells indicated that phenylethanoids 9 and 10 exhibited notable inhibitory activities. The result indicated that compounds 9 and 10 attenuated inflammation by decreasing the production of nuclear factor kappa-B (NF-κB) p65, inhibitory subunit of NF kappa B alpha, Janus kinase 2 (JAK2), signal transducer and activator of transcription 3 (STAT3), and inducible nitric oxide synthase in LPS-mediated RAW 264.7 macrophages. Phenylethanoids 9 and 10 increased the expression of interleukin-10 and endothelial nitric oxide synthase. Therefore, compounds 9 and 10 showed anti-inflammatory activity by regulation of NF-κB and JAK/STAT signaling pathways.

PurposeNumerous barriers hinder individuals with mental illness from seeking medical assistance in rural regions, yet a comprehensive understanding of these challenges remains elusive. This meta-synthesis aims to understand the barriers and facilitators in medical help-seeking among rural individuals with mental illness. MethodsWe systematically searched seven databases [PubMed, CINAHL, Medline (OVID), PsycINFO (OVID), Cochrane, Embase, and ProQuest] in May 2023 and included the studies if they reported the barriers or/and facilitators to seek healthcare in rural patients with mental illness. We conducted hand search and citation search on Google Scholar for literature supplements. Thematic analysis was employed. ResultsThe study included 27 articles reporting on the barriers and facilitators to seeking medical help in this population from 2007 to 2023. We ultimately identified themes at three levels: navigating the terrain of vulnerability and empowerment (the individual with mental illness), navigating the terrain of external environment (the external environment) and connectivity within the healthcare ecosystem for mental health (the health service system). ConclusionsWe must design more effective strategies to improve mental healthcare access for rural patients, considering cultural nuances and health service utilization patterns. This requires a multi-level approach, tailored to the unique needs of diverse populations.

This paper aims to establish an automatic and accurate pore identification method for pervious concrete. The residual module and mixed loss functions were introduced to the original UNet network to obtain the improved UNet. CT scanning was conducted on the six groups of pervious concrete samples with different aggregate sizes to obtain the initial dataset. The initial dataset was marked and enhanced, and then the pore recognition model was trained. The influence of image brightness and contrast on pore identification was analyzed. The fusion algorithm was used to improve the robustness of the model. The results show that during model training, R-UNet began to converge 20 epochs earlier than UNet and the loss value was smaller. Moreover, the maximum increase of mIoU and mDice was 10.3% and 11.7% respectively, and the maximum decrease of mHD was 14.1%. The fusion algorithm could improve the segmentation accuracy of pores in brightness anomaly images. Compared with threshold segmentation method, the method proposed in this paper could improve the accuracy of pore edge segmentation and the “fine pores” identification, and reduced the pore identification defects. The value of mHD was decreased by 48.7% − 72.4%, and the efficiency of pore identification was greatly improved.
Esterification of free fatty acids in a rotor-stator spinning disc reactor

Fatty acid methyl esters (FAMEs) were produced by the esterification of free fatty acids (FFA) with methanol, and sulfuric acid as the catalyst in a rotor-stator spinning disc reactor (RSSDR). The RSSDR, which shows excellent mixing efficiency and fast phase separation, was used as a novel continuous-flow esterification reactor. The influence of the variables (e.g., rotational speed, volume flow rate, rotor-stator distance, methanol-FFA molar ratio, catalyst dosage, and temperature) on esterification conversion () and productivity of FAMEs (PFAME) were investigated. It was found that the experimental parameters have a great impact on the and PFAME in the RSSDR system, due to the effect of micromixing intensity and residence time distribution. Furthermore, to compare with other traditional esterification reactors, the values of , PFAME, and PFAME per unit reactor volume (PFAME/VR) in the RSSDR were also employed to assess the performance for the production of FAMEs. It shows that the maximum values of PFAME, and PFAME/VR attained were 0.14mol/min and 3.06×102 mol/(mL min), respectively. Therefore, the RSSDR is proven to be an effective esterification reactor with high esterification conversion in comparison to conventional esterification reactors.
Dimensional synthesis of an Inspection Robot for SG tube-sheet

To ensure the operational safety of nuclear power plants, we present a Quadruped Inspection Robot that can be used for many types of steam generators. Since the Inspection Robot relies on the Holding Modules to grip the tube-sheet, it can be regarded as a hybrid robot with variable configurations, switching between 4-RRR-RR, 3- RRR-RR, and two types of 2-RRR-RR, and the variable configurations bring a great challenge to dimensional synthesis. In this paper, the kinematic model of the Inspection Robot in multiple configurations is established, and the analytical solution is given. The workspace mapping is analyzed by the solution-space, and the workspace of multiple configurations is decomposed into the workspace of 2-RRR to reduce the analysis complexity, and the workspace calculation is simplified by using the envelope rings. The optimization problem of the manipulator is transformed into the calculation of the shortest contraction length of the swing leg. The switching performance of the Inspection Robot is evaluated by stride-length, turning-angle, and workspace overlap-ratio. The performance indexes are classified and transformed based on the proportions and variation trends of dimensional parameters to reduce the number of optimization objective functions, and Pareto optimal solutions are obtained using an intelligent optimization algorithm.

This study focused on the antibacterial effects of the endophytic fungi producing naringenin from Dalbergia odorifera T. Chen against Staphylococcus aureus. The antibacterial activity was measured by the inhibition diameters, minimum inhibitory concentration (MIC), and minimum bactericidal concentration (MBC). The time-killing curve was also used to evaluate its antibacterial efficacy. The results of antibacterial activity determinations showed that endophytic fungi secondary metabolites can inhibit the growth of five pathogenic bacteria (S. aureus, Escherichia coli, Salmonella enteritidis, Pseudomonas aeruginosa, and Bacillus subtilis) and the most sensitive strain was S. aureus that had the MIC and MBC values of 0.13 and 0.50 mg/mL, respectively. The membrane permeability study was measured by a DNA leakage assay and electrical conductivity assay. Furthermore, the whole-cell protein lysates and DNA fragmentation assay was evaluated. The morphology of S. aureus treated with the endophytic fungi products was observed by scanning electron microscopy (SEM). The probable antibacterial mechanism of endophytic fungi secondary metabolites was the increased membrane permeability that leads to leaks of nucleic acids and proteins. SEM results further confirmed that the extracts can interfere with the integrity of S. aureus cell membrane and further inhibit the growth of bacteria, resulting in the death of bacteria. This study provides a new perspective for the antibacterial functions of endophytic fungi secondary metabolites for biomedical applications.
