The present study was aimed to investigate the effects of latency period on alveolar ridge distraction using the intraoral distraction device, Nine adult mongrel dogs of either sex, weighing about 15kg, were used. They were divided into 3-day, 5-day, and 7-day groups according to the latency period. The left upper and lower premolars and first molars were extracted, Twelve weeks after the extraction, an osteotomy was performed and the distraction device was applied. After the latency period, the distraction was applied at a rate of 1.2mm for 8 days. The distraction device was left in place for 2 weeks to allow consolidation and was then removed. The animals were sacrified at 8 weeks after completion of distraction and were examined macroscopically, radiographically, and histologically. After completion of the distraction, the alveolar crest protruded prominently, showing the vertical augmentation of the alveolar bone. Soft tissues were broken down after the 6th day of distraction in 3-day latency group, and premature union occurred on the 7th day of distraction in the 7-day latency group. The average distance was 9.40±0.3mm in 3-day latency group, 9.35±0.1mm in 5-day latency group, and 8.85±0.1mm in 7-day latency group. In the radiograph taken at 8 weeks after distraction, there was slight bone resorption around the medial and distal edges of the alveolar bone segment, and a new bone deposition was observed in the neighboring alveolar crest area in all groups. Fibrous tissues were present in a part of the buccal cortical bone area of the distraction gap, and the woven and lamellar bones were observed in the distracted gap. There were bony bridges in the distraction gap in all animals examined. These results suggest that optimal time of latency period on alveolar ridge distraction using the intraoral distraction device is about 5 days in dogs, and about 7days in human beings.
Ribosomal DNA의 ITS영역 염기서열 분석에 의한 기주특이적 독소 생성 Alternaria spp.의 계통분류학적 유연관계

기주특이적 독소 생성 Alternaria spp.와 비병원성 A. alternata의 계통분류학적 유연관계를 규명하기 위하여 기주특이적 독소 생성 Alternaria 6균, 비병원성인 A. alternata 그리고 비교균으로서 소형포자를 형성하는 A. brassicicola, 대형포자를 형성하는 A. solani의 ribosmal DNA의 5.8S를 포함한 ITS영역의 염기 서열을 분석하였던 바 다음과 같은 결과를 얻었다. 1. Alternaria spp.의 rDNA ITS영역의 염기서열 크기는 548∼562bp였으며 5.8S rDNA에서는 비교종간에 서로 일치하였고 ITS I에서는 종간 뚜렷한 차이를 보였다. 2. 기주특이적 독소 생성 Alternaria spp.의 rDNA의 ITS영역의 염기서열은 비병원성인 A. alternata의 염기서열과는 서로 일치하였으나 A. brassicicola와는 3%, A. solani와는 7%의 염기서열 차를 보였다. 3. 기주특이적 독소 생성 Alternaria spp.는 비병원성 A. alternata와 진화적으로 매우 가까운 유연관계를 나타내었다. The regions of internal transcribed spacer(ITS) including 5.8S ribosomal DNA(rDXA) from six host-specific toxin(HST) producing Alternaria spp. nonpathogenic A. alternata. A. solani and A. brassicicola were sequenced to assess their relationship in taxonomy. There was no difference in the sequences of ITS including 5.8S rDNA among the HST-producing Alternaria spp. and nonpathogenic A. alternata. However, the sequences of HST-producing Alternaria spp. were different from those of A. solani and A. brassicicola by about 7% and 3%, respectively. These results suggest that HST-producing Alternaria spp. are evolutionarily very close to non pathogenic A. alternata although they have host-specific pathogenecity.

<P><B>Background:</B> T-cell immunosenescence, a hallmark of an aging immune system, is potentially linked to the risk of developing cancer and other aging-related diseases. Chronic infection by cytomegalovirus (CMV) has been widely studied as a risk factor for T-cell immunosenescence, but the role of persistent chemicals has never been examined. As a typical example of persistent chemicals, we evaluated whether organochlorine pesticides (OCPs) are related to T-cell immunosenescence in the general population.</P><P><B>Methods:</B> Serum concentrations of β-hexachlorocyclohexane, <I>p,p</I>′-DDT, <I>p,p</I>′-DDE, and trans-nonachlor were measured in 95 Korean adults ages 30 to 64 years. T-cell immunosenescence was assessed by the frequencies of CD8<SUP>+</SUP>CD57<SUP>+</SUP>, CD8<SUP>+</SUP>CD28<SUP>−</SUP>, CD4<SUP>+</SUP>CD57<SUP>+</SUP>, and CD4<SUP>+</SUP>CD28<SUP>−</SUP> T lymphocytes in 20 mL of fresh peripheral blood.</P><P><B>Results:</B> The senescence of CD8<SUP>+</SUP> T lymphocytes was the most consistently associated with OCPs. For quartiles of measurements of OCPs, adjusted mean percentages of CD8<SUP>+</SUP>CD57<SUP>+</SUP> and CD8<SUP>+</SUP>CD28<SUP>−</SUP> T lymphocytes in the CD8<SUP>+</SUP> T lymphocyte population were 23.9, 27.6, 31.0, and 38.7 (<I>P</I><SUB>trend</SUB> < 0.01) and 25.6, 27.3, 28.0, and 35.5 (<I>P</I><SUB>trend</SUB> = 0.02), respectively. When we compared the strength of the associations among OCPs, CMV IgG titer, and age, OCPs showed the strongest association with markers of immunosenescence. Importantly, the association between OCPs and immunosenescence markers was more prominent among participants without known risk factors, such as a young age or low CMV immunoglobulin G titer.</P><P><B>Conclusions:</B> Chronic exposure to low-dose OCPs may be a new risk factor for T-cell immunosenescence.</P><P><B>Impact:</B> T-cell immunosenescence may be one possible mechanism linking low-dose OCPs and many chronic diseases. <I>Cancer Epidemiol Biomarkers Prev; 27(10); 1159–67. ©2018 AACR</I>.</P>

Biochemical characterization of variants of canine CYP1A1 using heterologous expression

<P>Cytochrome P450 1A1 (CYP1A1) is a heme-containing mono-oxygenase involved in metabolism of environmental contaminants. Two variants of dog CYP1A1 with a single residue difference were identified and designated Sap1 and Sap2. Compared with Sap1, Sap2 had a Trp50Leu substitution. The biochemical characteristics of the variants were comparatively analyzed using heterologous expression in <I>Escherichia coli</I>. The membrane fraction of <I>E. coli</I> expressing Sap2 exhibited higher CYP holoprotein and heme contents than the Sap1-containing membranes, although the level of total CYP1A1 protein (i.e., apoprotein + holoprotein) was comparable between the groups. As normalized to holo-CYP content, the Sap2-expressing membranes showed lower CYP1A1-specific enzyme activities, such as 7-ethoxyresorufin <I>O</I>-dealkylation (EROD), than the Sap1 group. In single substitution variants of residue 50, proteins with hydrophobic residues having mass similar to Leu exhibited lower EROD activities than those with hydrophobic residues having larger mass than Leu. In addition, variants with polar or charged residues having mass similar to Leu showed activities that were comparable to those of Sap2. Taken together, these findings suggest that the Trp50Leu substitution leads to an enhancement of holo-CYP1A1 formation, but diminishes the enzyme activity because of the small size of Leu compared with Trp.</P>

Hodgson's bats are critically endangered in South Korea. This study analyzed the concentrations of elements in liver, kidney, and intestine tissues from a Hodgson's bat found dead in the wild. The concentrations of essential elements followed the order Fe > Zn > Cu > Mn > Se in the three tissues. Hg was detected at the highest concentrations among the non-essential elements analyzed in the liver and kidney tissues, while As was the most highly concentrated non-essential element in the intestine. To the best of our knowledge, this is the first study of tissue element concentrations in Hodgson's bats.
