        Dosimetric evaluation of magnetic resonance imaging-guided adaptive radiation therapy in pancreatic cancer by extent of re-contouring of organs-at-risk

        Purpose: The safety of online contouring and planning for adaptive radiotherapy is unknown. This study aimed to evaluate the dosimetric difference of the organ-at-risk (OAR) according to the extent of contouring in stereotactic magnetic resonance image-guided adaptive RT (SMART) for pancreatic cancer. Materials and Methods: We reviewed the treatment plan data used for SMART in patients with pancreatic cancer. For the online contouring and planning, OARs within 2 cm from the planning target volume (PTV) in the craniocaudal direction were re-controlled daily at the attending physician's discretion. The entire OARs were re-contoured retrospectively for data analysis. We termed the two contouring methods the Rough OAR and the Full OAR, respectively. The proportion of dose constraint violation and other dosimetric parameters was analyzed. Results: Nineteen patients with 94 fractions of SMART were included in the analysis. The dose constraint was violated in 10.6% and 43.6% of the fractions in Rough OAR and Full OAR methods, respectively (p = 0.075). Patients with a large tumor, a short distance from gross tumor volume (GTV) to OAR, and a tumor in the body or tail were associated with more occult dose constraint violations—large tumor (p = 0.027), short distance from GTV to OAR (p = 0.061), tumor in body or tail (p = 0.054). No dose constraint violation occurred outside 2 cm from the PTV. Conclusion: More occult dose constraint violations can be found by the Full OAR method in patients with pancreatic cancer with some clinical factors in the online re-planning for SMART. Re-contouring all the OARs would be helpful to detect occult dose constraint violations in SMART planning. Since the dosimetric profile of SMART cannot be represented by a single fraction, patient selection for the Full OAR method should be weighted between the clinical usefulness and the time and workforce required.

      • (-)-Loliolide isolate from Sargassum horneri alleviates the photoaging of the skin in UVB-exposured HaCaT cells

        In this study, we evaluated the protective activity of (–)- Loliolide isolated from Sargassum horneri against ultraviolet B (UVB)-induced cellular damages in HaCaT cells. The results showed that (–)-Loliolide improved the cell viability by reducing the intracellular reactive oxygen species (ROS) generation in UVB-exposured HaCaT cells. In addition, (–)-Loliolide inhibited apoptosis via the reduction of the sub-G1 cell population and the apoptotic body formation by modulating the expression levels of Bax/Bcl-2, compared to the only UVB-exposured HaCaT cells. (–)- Loliolide uppressed the activation of nuclear factor (NF)-κB signaling by regulating the phosphorylation of IκB-α and NF-κB p65 as well as the translocation of NF-κB p65 into nucleus. Furthermore, the pre-treatment of (–)-Loliolide led to the activation of nuclear factor erythroid 2-related factor 2 (Nrf2)/heme oxygenase 1 (HO-1) signaling. Interestingly, the protective capacities of (–)-Loliolide against UVB-induced cellular damages were significantly blocked by the inhibition of HO-1 signaling.       • Synthesis and characterization of nanofiber-type hydrophobic organic materials as electrodes for improved performance of PVDF-based piezoelectric nanogenerators

        <P><B>Abstract</B></P> <P>Poly(3,4-ethylenedioxythiophene) (PEDOT) derivatives are synthesized by oxidative polymerization using sodium dodecyl sulfate (SDS) as an anionic surfactant dopant. The resulting polymeric materials featuring nanofiber-type one-dimensional (1D) structures are identified as poly(2‐butyl‐2,3‐dihydrothieno[3,4‐b][1,4]dioxine:dodecyl sulfate (PEDOT-C<SUB>4</SUB>:DS) and poly(2‐hexyl‐2,3‐dihydrothieno[3,4‐b][1,4]dioxine:dodecyl sulfate (PEDOT-C<SUB>6</SUB>:DS). The ratio of the DS anion doped into PEDOT-C<SUB>4</SUB>:DS and PEDOT-C<SUB>6</SUB>:DS is 0.16 and 0.23, respectively. The contact angle of water on the PEDOT-C<SUB>4</SUB>:DS and PEDOT-C<SUB>6</SUB>:DS films is 76.6° and 87.7°, respectively, showing hydrophobic properties similar to that with water on PVDF. It facilitated the fully uniform film formation due to excellent surface matching. Peeling force of PEDOT-C<SUB>4</SUB>:DS and PEDOT-C<SUB>6</SUB>:DS is stronger than the one of PEDOT:PSS‐CNT composite. GIWAX analysis showed that PEDOT-C<SUB>4</SUB>:DS formed the highly ordered edge-on structure and PEDOT-C<SUB>6</SUB>:DS formed the bimodal orientation consisting of edge-on structure mainly and face-on structure slightly. The electrical conductivity (σ<SUB>PEDOT‐C4:DS</SUB>=50.0 S cm<SUP>−1</SUP>) of PEDOT-C<SUB>4</SUB>:DS is 41.7 times higher than that of PEDOT:PSS (σ<SUB>PEDOT:PSS</SUB>=1.2 S cm<SUP>−1</SUP>).</P> <P>The output signals (maximum voltages/currents) of piezoelectric nanogenerators (PNGs, electrode/PVDF/electrode) using these materials as electrodes are PNG-1 (PEDOT:PSS‐CNT composite) 1.25 V/128.5 nA, PNG-2 (PEDOT-C<SUB>4</SUB>:DS) 1.54 V/166.0 nA, and PNG-3 (PEDOT-C<SUB>6</SUB>:DS) 1.49 V/159.0 nA. Of these, PNG-2 & PNG-3 show maximum piezoelectric output power of 63.0 nW and 59.9 nW at 9 MΩ compared to PNG-1 (41.0 nW at 10 MΩ). They are enhanced up to 53.7%.       • 강화학습 기반 직산 분리 모델 개발

        직달일사와 산란일사를 분류하는 과정은 보다 정밀한 방위별 일사량 및 발전량을 예측하거나 이를 기반으로 건물에서 예측 제어를 위해 필수적으로 선행되어야 한다. 하지만 대부분의 직산분리 모델은 해외의 기상 발생패턴을 통계적으로 분석해 개발되었기 때문에 우리나라의 지역적 특성을 반영하지 않는 경우 계산된 일사 값은 큰 오차를 보인다. 이에 본 연구에서는 강화학습 알고리즘을 통해 발생되는 오차를 학습하여 기존 직산분리 모델의 정확도를 개선하는 직산분리 모델을 제안하였다. 제안된 모델은 오랜기간 동안 누적된 기상 데이터가 없는 현장을 가정하여 약 2주 동안의 기상데이터만을 학습하였다. 시뮬레이션 결과 제안된 모델은 학습기간과 유사한 겨울철(2월) 16W/㎡ 수준의 오차를 보였으며, 여름철(7월)에는 27W/㎡ 수준의 오차를 보이며 기존 직산분리 모델의 오차를 개선하는 성능을 보였다.

      • Phlorofucofuroeckol A isolated from Ecklonia cava improves the inflammation in TNF-α/IFN-γ stimulated keratinocytes and relieves edema in a TPA-stimulated mice model

        This study investigated the effect of phlorofucofuroeckol A (PeA) isolated from Ecklonia cava against inflammatory response in the tumor necrosis factor-α (TNF-α)/interferon-γ (IFN-γ) stimulated keratinocytes and a TPA-stimulated ear edema mice model. The result showed PeA reduced the generation of intracellular reactive oxygen species (ROS) without the cytotoxicity at the used all concentrations in TNF-α/IFN-γ -stimulated keratinocytes. Also, PeA significantly down-regulated the mRNA expression and the secretion of inflammatory cytokines and chemokines. In addition, PeA effectively inhibited the formation of apoptotic body and the population of sub-G1 DNA content by suppressing the activation of mitogen-activated protein kinase (MAPK) and nuclear factor (NF)-κB signaling. Moreover, PeA activated the nuclear factor erythroid 2-related factor 2 (Nrf2)/heme oxygenase 1 (HO-1) signaling in TNF-α/IFN-γ-stimulated keratinocytes. In further study, PeA inhibited the ear edema by reducing the expressions of inflammatory mediators in TPA-stimulated mice model. This work (Grants No. M01201820150306) was supported by KIMST.

      • Improvement of skin inflammation and moisturizing effect of UVB-exposed HaCaT cells of low molecular weight fucoidan obtained from Sargassum siliquastrum

        The present study evaluated the effect of a low molecular weight fucoidan obtained from a celluclast extract of Sargassum siliquastrum (S. siliquastrum) against improvement of skin inflammation and moisturizing effect caused by ultraviolet B (UVB) exposure in HaCaT cells. First of all, we prepared the four kinds of polysaccharides (SSS1, SSS2, SSS3 and SSS4) from the celluclast extract of S. siliquastrum by a step gradient ethanol precipitation. Among them, SSS4 consisted of the higher sulfate content showed the higher antioxidant activities. Also, UVB protective effect confirmed that SSS4 reduced intracellular ROS production while improving cell viability. In addition, SSS4 inhibited the activation of MAPK and NFκB signaling in UVB-exposed HaCaT cells. Moreover, SSS4 significantly increased the amount of hyaluronic acid by modulating the expression levels of skin barrier-related proteins and moisturizing factors. This study suggested that SSS4 is a fucoidan and has potential therapeutic effects on skin damage caused by UVB exposure in HaCaT cells.         북한이탈주민의 초점집단면접을 활용한 북의 직업관 연구

        근대의 산물인 직업 및 노동의 의미와 가치는 자신의 생계유지를 위한 재화 확보 화동, 사회적 역할의 분화를 통한 사회적 기여 활동, 자아의 성취 및 자아실현이라는 세 가지 가치로 분화했다. 근대사회에서 개인들의 직업관은 이들 중 어떤 측면을 보다 중요하게 여기는가에 따라 달라지며 이는 남과 북도 마찬가지이다. 남과 북은 각기 다른 정체-경제체제를 지향했기 때문에 직업관도 다를 수밖에 없다. 이에 본 연구에서는 북한이탈주민을 대상으로 한 초점집단면접을 통해 북의 직업관을 파악해 보고자 하였다. 초점집단면접은 인터뷰 참가자들이 자유롭게 토론하면서 서로의 발화를 상호확인하고 교차 검증하는 과정을 거치게 된다는 점에서 개별 인터뷰보다 더 객관적인 데이터를 확보할 수 있다는 장점을 갖기에 채택한 연구방법이다. 해방 이후, 농업사회에서 근대 산업사회로 이행하기 위해 북에서는 사회-국가에 대한 기여라는 의미에서 노동에 가치를 부여했다. 하지만 근대적인 노동에 익숙하지 않은 노동자를 근로대중 인민으로 변모시키기 위해서는 근대적 노동규율을 확립하고 법 제도를 개혁하는 등의 국가통제 방식이 요구되었다. 이후 공산주의적 새 인간을 내세우면서 자발적으로 국부(國富)를 증진시키는 인간형을 창출해 나갔다. 여기에 애국주의적 서사를 통해 애국심을 고취하고 국가 차원의 혁명에 집단적으로 참여하는 혁명주체로 노동자들을 계속적으로 호명하였다. 하지만 자아실현의 의미가 축소되고 개인의 삶과 멀어진 ‘사회 기여 노동’은 배급제의 붕괴라는 사회 시스템의 와해 앞에서 토대를 잃어버리고 오히려 ‘생계 유지 노동’라는 관점에서의 노동 및 직업의 의미가 현실성을 지니게 되었다. 북의 직업관은 고난의 행군 전후로 완전히 달라졌음을 보였으며, 고난의 행군 이후 직업 선택의 가장 중요한 기준은 자신의 생계를 해결해 줄 수 있는 일이냐의 여부였다. ‘돈’의 가치가 부각되면서 북 사회를 움직이는 ‘정치자본’ 하에서 ‘경제자본’이 출현하고 있었고, 이 균열을 메우는 장치가 뇌물인 것으로 확인되었다. 여전히 강고하게 작동하는 정치자본의 영향 아래 정치적 배신만은 해서는 안 된다는 보편적 사고체계를 가짐에도 불구하고, 묵인할 수 있을 정도의 일탈과 침묵은 균열을 메우는 장치로 작동하고 있었다. 최근 들어 북이 시장화되면서 남쪽처럼 생계나 사적 욕망에 따른 경쟁에 의한 직업관의 변화가 일어나고 있지만 아직까지도 북쪽의 직업관은 남쪽 사람들과 만났을 때, 충돌을 야기할 가능성이 높다. 하지만 남쪽이든 북쪽이든 직업관에서 상대적으로 결여하고 있거나 부차화했던 것은 자아 성취 또는 자아실현이란 의미에서의 직업이다. 이것은 남 또는 북의 문제가 아니라 바로 근대적인 노동의 가치, 노동중심, 생산중심의 노동관 또는 직업관이 가지고 있는 문제이기도 하다. 그러므로 문제는 남북의 직업관을 단순 비교해서 어느 한쪽의 직업관이 낫다는 식이 아니라 궁극적으로 ‘자아실현’을 중심으로, 근대적인 직업관 그 자체를 넘어서는 방안을 찾아가는 차원에서 남북의 직업관을 비교 연구하는 것이다. The meaning and value of jobs and labor, which are products of modern times, have been differentiated into three values: securing goods for sustaining one"s livelihood, social contribution activities through the differentiation of social roles, and self-fulfillment and self-realization. In modern society, individuals" views of work depend on which of these aspects are more important, which also applies to South Korea and North Korea. South Korea and North Korea have different views on their jobs as they aim for a different political and economic system. This study aimed to understand North Korea"s labor values through focus group interviews (FGI) with North Korean defectors. The FGI method is advantageous in that it undergoes a process of cross-validation of each other"s utterances while freely discussing. Therefore, the FGI method was used to produce more objective information than individual interviews. After liberation, to transition from an agricultural society to a modern industrial society, North Korea gave value to labor in the sense of contribution to society. However, to transform workers who were unfamiliar with modern labor into working people, state-controlled methods were required, such as establishing modern labor regulations and reforming the legal system. Since then, the state has voluntarily created a human form that promotes a new communist human being. Additionally, through patriotic narratives, workers were continuously called revolutionaries that inspired patriotism and collectively participated in the revolution on the national level. However, the meaning of self-realization has been reduced and the meaning of "social contribution labor" has become more realistic in the face of the collapse of the social system of the distribution system. Labor values in North Korea have completely changed before and after the march of suffering, and the most important criterion for choosing a job after the march was whether it could ensure one"s livelihood. As the value of "money" was emphasized, "economic capital" was emerging under the "political capital" that moves North Korean society, and it was confirmed that bribery was the device bridging the gap. Despite having a universal system of thinking according to which political betrayal should not be done under the influence of strong political capital, tolerable deviations and silence were working as devices to bridge the cracks. Recently, the marketization of the North has led to changes in job views resulting from competition due to livelihoods and private desires as in the South, but the North"s job views are still likely to cause conflicts when they meet those of the South. However, what is relatively lacking or secondary to the view of occupation, whether in South Korea or North Korea, is a job in the sense of self-fulfillment or self-realization. This matter surpasses the relationship between South Korea and North Korea. It concerns modern labor values, labor centers, production-oriented labor, or occupational views. Therefore, the problem does not lie in simply comparing and studying the job views of both Koreas, but in ultimately finding ways to surpass the modern view, focusing on "self-realization".
