      • 경연 대회를 통한 초ㆍ중고생의 창의력 신장 효과에 관한 연구

        미래 사회를 대비하기 위한 개인의 개성을 존중과 창의적인 능력을 지닌 인간을 양성을 목적으로 충청남도 지역의 초 ㆍ 중고등학생들을 대상으로 한 창의력 경연대회가 공주대학교 과학교육연구소에서 개최되었다. 충청남도 초 ㆍ중 ㆍ 고등학생들을 대상으로 실시한 창의력 경연대회는 창조적 아이디어를 내어 스스로 문제를 해결해 나갈 수 있는 교육 프로그램을 제공하는 좋은 기회였음이 확인되었으나, 일부의 팀에서만 참신하고 재미있는 아이디어들이 제안되었다. 여학생의 참가자 수가 상당히 증가하였으며, 대체적으로 입상권의 학생들의 상당 부분이 여학생이라는 것이 특이할만하다. 학년이 올라감에 따라서 창의적인 아이디어를 내는 양이나 질에 있어서 뒤떨어지는 것으로 분석되었으며, 토론 학습의 적응력이 미흡하고 발표력 및 청취력에 있어서도 떨어지는 것으로 나타났다. 따라서 중 ㆍ고등학교의 학교교육에서도 학생들에게 프로젝트형 탐구학습 프로그램의 적용은 물론 토론식 수습을 통한 학생들의 사고의 전환의 기회를 제공해야 할 것으로 사료된다. The creativity competition was held to foster the creativity of elementary and secondary school students in the Institute of Science Education, Kongju national University. It was found that the creativity competition effected on the improvement of inventing creative ideas and problem solving activities. But, only some competitors participated in semifinal and final contests, presented original and interesting ideas. More female students participated in competition than last year. More female participants were awarded a prize than males. Totally, participants were defective in discussion and communication, and presentation and listening ideas. Increasing grades correlated inversely with creativity and originality. There is need of inquiry project teaming programs and presenting opportunities of conversion of thinking by discussing instructions in school

      • 충북 남부지역 중소 제조기업의 경쟁력 제고를 위한 경영 및 애로기술 실태조사 연구

        본 연구에서는 충북 남부 3개군 지역(옥천 · 보은 · 영동)의 중소 제조기업의 경영실태 및 애로기술을 대상으로 실증조사 되었다.·지역의 특정상황의 전제하에 지방 중소기업들의 실질적인 문제점들을 발견하고, 그 개선 방안을 탐색함은 물론 산 · 학 · 연 컨소시엄 사업의 성공적인 수행을 위해 시도되었다.세부적으로는 중소 제조기업의 경영 및 애로기술 특성을 일반적 사항, 경영애로요인, 경영전략, 현장개선기법, 정보화, 제품 및 공정 기술상의 애로기술 현황파악의 6개 영역으로 조사되었다. 조사결과들은 추후 산 · 학 · 연 공동기술개발 사업에 있어 창의적, 틈새적 과제발굴 및 신제품 기술개발의 토대마련이 될 뿐 아니라, 지역경제 발전을 위한 산 · 학 관의 지원 전략 수립에 적극 활용 될 것으로 판단된다.

      • 곁사슬형 Polyethylene Glycol을 가진 Adenosine Deaminase 거대분자 결합체의 생화학적 특성

        혈색소, 홀몬 및 효소들과 같은 단백질은 질병 치료에 흔히 이용되고 있으나 이들은 생체 내로 투여하였을 때 치료제로서의 안정성과 항원성이 문제가 되어 그 효능을 기대하기 힘든 경우가 적지 않다. 저자들은 이러한 결점을 개선해 보고자 그 첫 단계로 비항원성 합성 고분자 화합물인 곁사슬형 polyethylene glycol(PEG)을 단백질의 실험모델로 사용한 효소 adenosine deaminase(ADA)에 결합시켜 이 복합체(PEG-ADA)의 물리적 및 생화학적 성질에 대하여 조사하였다. 연구 결과 곁 사슬형 PEG-ADA는 PEG로 인해 ADA가 변형되었지만, 효소활성이 유지되고 있었으며 단백질 분해효소에 대해서도 안정되어 있음을 보여 주었다. The enzyme is usually limited to use as a therpeutics because of its own instability and antigenicity. Adenosine deaminase(ADA), which was selected as a model of enzymes in this study, was modified by a pendant-type polyethylene glycol(PEG) in order to reduce the nzyme's own disadvantages, and the authors studied about the biochemical characteristics of the pendant type PEG-ADA conjugate to elucidate whether this conjugate could be a more improved and effective therapeutic enzyme haying stability and non-antigenicity. Calf adenosine deaminase was modified with pendant-type polyethylene glycol using cyanuric chloride as a coupler. The covalent attachmant of PEG to ADA altered the kinetic profile of the enzyme in which the value of Km for adenosine was decreased. The Km value of the ADA and PEG-ADA10 were 30 μM, 25 μM against adenosine, respectively. But the PEG-ADA20 exhibited reasonable retention of catvlytic activity with an increased Km, which was 50 μM against adenosine. The ADA and PEG-ADA showed optimum in acidic direction. The ADA and PEG-ADA had same temperture optima of 50℃. Thermal inactivation at the temperature above optima was essentially the same between the native and modified enzymes. PEG appeared to have no effect of stabilizing enzymes on high temperature. This suggested that PEG attachment caused some destabilization of the adenosine deaminase structure. PEG-ADA also demonstrated a resistance to the proteolytic digestion by trypsin and chymotrypsin. The electrophoretic mobility in polyacrylamide gels of PEG -ADA was reduced as compared to the native enzyme. The unmodified enzymes showed single band, whereas the modified ones formed a mumber of bands throughout the gels. The decreased electrophoretic mobility of PEG-ADA might be due to a charge-shielding effect of the hydrophilic PEG shell surrounding the enzyme. In addition, the increased size of the adducts might serve to retard migration in the polyacrylmide gels. The alteration of the pH dependance and the dramatically decreased mobility in polyacrylamide gel electrophoresis of the PEG-enzyme pointed up the effect of PEG modification on the apparent charge of the enzyme molecules. It was believed to be due to a change in conformation dictated by the charge of the PEG-enzyme or imposed by the covalent attachment of PEG to the enzyme. These results showed that the biochemical properties of adenosine deaminase were altered after chemical modification by pendant type PEG. The preparation of PEG-ADA retained the enzyme activity and had resistance against proteolytic enzymes. The authors concluded that the pendant-type PEG-ADA might be available in the medical field in the future.

      • 고혈압치료제의 in vitro및 in vivo 이용률간의 상관성 연구(Ⅱ)

        의약풍이 생체내로 흡수되는_파정에서 생리학적 인자 및 제제학적 인자들에 의해 생체이용률이 달라져 약물의 흡수과정을 연구함으로써 약물의 생체이용률을 예측하는 것이 가능하다. 그러나 제제학적 측면에서 용출시험과 흡수과정인. 장관막 투과도 나아가서 생체이용률간의 띤구가 미흡한 상태이다. 따라서 약물의 흡수과정을 연구하벼 생체이용률을 예측하기 위하여 장관막 투과도 및 혈중농도사이의 상관관계를 예견할 수 있는 모델을 개발하고자 1999년도 연구사업으로 고혈압치료제중 베타-차단약 (아테놀올, 메토프롤올, 프라프라놀꼴)을 선택하여 웅출시험과 장관막 투과도계수를 측정하여 사람의 흡수을을 예측할 수 있는 장관막 투파도 모델을 확림하였고, 2000년도에는 계속사업으로서 각 약물에 대한 횐쥐에서의 생체이용를 파라메타를 측정하고 1999년도 연구사업결과에서 얻은 장관막 투과도 시험결과를 가지고 생체이용률과의 상관섞을 구하고자 하였다. 각 베타-차단약을 경구 및 정맥투여하고 시간별로 채혈한 후 분리한 혈장으로부터 각 약물에 대한 EfLC를 이용한 분석준건을 확지하여 혈중농도를 측정하고 생체이용률 파라메타(AUC, Cma, Tme, 등)를 구하였다. 횐쥐의 소장을 이용하여 diffusion chamber 법으로 측정한 장관막 투과도계수와 각 생체이용률 파라미터 사이의 상관성을 구찬 결과 양호한 상관관계를 보였다. 이와 같은 연구결과를 통하여 약물의 특성에 따라 동물의 장관막 투도. 모델을 이용하여 사람의 생체이용률도 예측할 수 있을 것으로 사료된다.         Evaluation of Antioxidant Activities of Peptides Isolated from Korean Fermented Soybean Paste, Chungkukjang

        The objectives of present study were to characterize the peptides which were isolated from Korean fermented soybean paste, chungkukjang, and to determine their antioxidant activities. Four fractions were collected from the methanol extract of chungkukjang by using a recycling preparative HPLC. Among fractions, Fr-2 was identified to be highly potent free radical scavenging activity in the assay of 1,1-diphenyl-2-picryl-hydrazyl (DPPH) and nitroblue tetrazolium(NBT)-reduction inhibition. Base on antioxidant effects, fraction Fr-2 was employed for the refraction with a prep-column and separated into five fractions of which two fractions were identified to have higher antioxidant activity. To confirm the amino acid constituents of antioxidant fractions Fr-2-2 and Fr-2-3 were analyzed, and eight kinds of amino acids such as aspartic acid, threonine, serine, glutamic acid, glycine, lysine, histidine, and arginine were identified as the constituent amino acids. Antioxidant activities of the separated peptides were further assessed cell viability with 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyl terazolium bromide (MTT), and fluorescence-activated cell sorting (FACS) analysis of H4IIE cells treated with hydrogen peroxide (H2O2). Chungkukjang peptides have shown their ability to protect H4IIE rat hepatoma cells against H2O2- induced oxidative stress by concentration and time-dependent manner. Therefore, These results indicated that fermented soybean paste chungkukjang will be promoted the antioxidant and radical scavenging activities, and beneficial for health. The antioxidant peptide fractions Fr-2-2 and Fr-2-3 were denominated as P-NICS-1 and P-NICS-2, respectively. However, further studies were required to clarify their amino acid sequences and molecular properties, and physiological significances.

      • 시험관내 용출 및 장관막 투과도와 생체이용률과의 상관성

        A biopharmaceutics drug classification system for correlation between in vitro dissolution and in vivo bioavailability is proposed based on recognizing that drug dissolution and gastrointestinal permeability are the fundamental parameters controlling the rate and extent of drug absorption. The objective of this study was to assess whether in vitro dissolution projects of immediate-release beta-Mocker tablets can be correlated with intestinal membrane permeability and/or in vivo bioavailability. In vitro dissolution of the beta-Mocker tablets was examined using KP VII Apparatus II methods at various pH. Intestinal membrane permeability was determined in vitro using the diffusion chamber method. Bioavailability parameters were cited from literatures. The dissolution profiles did not accurately represent the in vivo bioavailablity. However there were good correlations between intestinal membrane permeability and log P (noctanol/buffer). The correlations obtained in this study indicted that in vitro diffusion chamber method could be used to predict intestinal absorption in vivo.

        Optimization of Radiator Position in an Internally Radiating Photobioreactor : A Model Simulation Study

        This study focused on the optimization of the illumination method for efficient use of light energies in a photobioreactor. In order to investigate the effect of radiator position, a model simulation study was carried out using Synechococcus sp. PCC 6301 and an internally radiating photobioreactor as a model system. The efficiency of light transfer in a photobioreactor was analyzed by estimating the average light intensity in a photobioreactor. The simulation results indicate that there exists an optimal position of internal radiators, and that the optimal position varies with radiator number and cell concentration. When light radiators are placed at the optimal position, the average light intensity is about 30% higher than that obtained by placing radiators at the circumstance or center of a photobioreactor. The method presented in this work may be useful for improving light transfer efficiency in a photobioreactor.

        We report a case of cutaneous sarcoidosis which occurred in a 32-year-old female who had, in addition to some cutaneous nodules, recently developed an acute spontaneous erythematous area of induration in old scars on her forehead and lower lip. Histopathologic findings revealed well-demarcated islands of epitheloid cells with a few giant, cells. In addition, slight admixture of lyrnphoid cells with present. at the margins of the epitheloid cell granulomas. Complete regression of the skin lesions was obtained with systemic steroid therapy without recurrence for some 8 months. (Kor J Dermatol 1992; 30(1): 126-130)
