Melphalan Modulates the Expression of E7-Specific Biomarkers in E7-Tg Mice.

<P>HPV oncoproteins are selectively retained and expressed in HPV infected carcinoma cells. The E7 oncoprotein interacts with the tumour suppressor Rb, and leads to the progression of oncogenesis. In a previous study, E7 biomarkers were identified in E7 Tg mice. In this study, in order to investigate whether a genotoxic carcinogen would modulate carcinogenesis in the E7-Tg mice, an anticancer drug, melphalan, was intraperitoneally injected into E7-Tg mice for eight weeks at two-day intervals and then genes and proteins were analysed using Omics approaches and RT-qPCR. RT-qPCR was performed to confirm whether E7 biomarkers would be modulated by melphalan treatment in E7-Tg mice, revealing that up-regulated E7 markers such as cyclin B1, CD166, and actin alpha1 were down-regulated, whereas expression of down-regulated E7 markers such as vimentin was restored by melphalan treatment. These results suggest that melphalan inhibits carcinogenesis via modulating E7-specific genes and proteins expressed in the lung tissues of E7 Tg mice.</P>

<P><B>Abstract</B></P> <P><B>Background</B></P> <P>Zonulin is acknowledged as the only physiological mediator established to reversibly regulate intestinal permeability through modulation of intercellular tight junctions. We aimed to determine whether there are differences in zonulin levels between 74 subjects with overweight or obesity and 76 with normal-weight and to assess correlations of circulating zonulin levels with anthropometric measures and obesity-related biomarkers.</P> <P><B>Methods</B></P> <P>We assessed anthropometric and laboratory measures, including body mass index (BMI) z-score, blood pressure, liver enzymes, lipid profiles, and insulin resistance. Serum zonulin levels were measured using an enzyme-linked immunosorbent assay.</P> <P><B>Results</B></P> <P>The mean age of the participants was 12.8 ± 1.5 years. Circulating serum zonulin levels were significantly increased in subjects with overweight/obesity compared with those of normal-weight (<I>P</I> = 0.03). Zonulin levels were significantly and positively associated with BMI z-score, alanine aminotransferase levels, triglyceride, fasting insulin, and insulin resistance as indicated by the homeostatic model assessment of insulin resistance (HOMA-IR) (all <I>P</I> < 0.05). In multivariate linear regression analysis, alanine aminotransferase was significantly and positively associated with circulating zonulin levels in adolescents with overweight or obesity (<I>P</I> < 0.01) after controlling for the effect of potential confounding factors. BMI z-score tended to be positively associated with serum zonulin levels in this subgroup analysis (<I>P</I> = 0.06).</P> <P><B>Conclusions</B></P> <P>Serum zonulin is a biomarker associated with hepatic metabolic disturbances in young adolescents with overweight or obesity. The positive relationship suggests a potentially relevant pathophysiological mechanism linking zonulin to hepatic metabolism in this age group of young adolescents with overweight or obesity.</P> <P><B>Highlights</B></P> <P> <UL> <LI> Serum zonulin is a biomarker associated with hepatic metabolic disturbances in young adolescents with overweight or obesity. </LI> <LI> There may be a potentially relevant pathophysiological mechanism linking zonulin to hepatic metabolic dysfunction even in young adolescents. </LI> </UL> </P>

Many approaches for the application of nano-sized particles to the human body as nanotechnology have been recently developed. The size of nanoparticles is related to their useful character and also plays a key role in toxicity. Since this surface area can interact with biological components of cells, nanoparticles can be more reactive in than larger particles. In the present study, a fluorescence dye-labeled 50, 100 and 200 nm-sized silica particle suspension was intravenously injected into mice to identify the toxicity, tissue distribution and excretion of silica nanoparticles in vivo. Incidence and severity of inflammatory response was transiently increased with injection of 200 and 100 nm silica nanoparticles within 12h. But there was no significant response related to injection of 50 nm particles. The silica particles of 50, 100 and 200 nm were cleared via urine and bile. The 50 nm silica anoparticles cleared to urine and bile than 100 nm and particles of 200 nm existed at lower concentration than other two smaller particles in urine and feces. Silica nanoparticles were trapped by macrophages in the spleen and liver and remained there until 4 weeks after the single injection.
Comparison of Cultural Method of Listeria monocytogenes in Salad, Salmon and Sashimi

This study was to compared efficacy between primary enrichment and secondary enrichment for detecting Listeria monocytogenes (L. monocytogenes) in salad, salmon, and sashimi that have different levels of background microflora. Strain of L. monocytogenes (ATCC 19115) was spiked in vegetable salad (n=96), fruits salad (n=48), salmon sashimi(n=48) and tuna sashimi (n=144). A total of 384 samples were tested using Fraser broth as a secondary enrichment broth in comparison with only LEB enrichment broth. Comparison of detect limits according to L. monocytogenes culture method for five sample types showed equivalent detection capabilities when incubated in LEB, LEB-Fraser, UVM-Fraser except PALCAM-Fraser broth in 10<SUP>0</SUP>-10¹ CFU/25g. However, it is thought that further consideration will be needed to unify L. monocytogenes culture methods for all samples.

The strain of rasH2 mice carrying human c-Ha-ras gene had been developed to use for rapid carcinogenicity testing. The mice develops various kinds of tumors including lymphoma when induced by carcinogen N-methyl-N-nitrosourea (MNU). In this study, 30 rasH2-Tg mice were treated with 75 ㎎/㎏ of MNU to induce lymphoma in 26 weeks. A total of 22 mice (13 male, 9 female) developed lymphoma. Immunohistochemistry for localization of cell surface antigens were performed to subclassify the lymphomas. Antibodies against BOB.1 and oct-2 were applied for B cell lymphoma and CD3 for T cell lymphoma. In T cell lymphoma, CD3 were expressed in cytoplasmic membranes of most tumor cells, whereas oct-2 and BOB.1 were localized in some cells in B cell lymphoma. Most of lymphomas cases expressed T cell marker CD3. The reason T cell lymphomas so frequently developed in rasH2 mice by MNU treatment should be further elucidated.
