S6K1 Phosphorylation of H2B Mediates EZH2 Trimethylation of H3: A Determinant of Early Adipogenesis

S6K1 has been implicated in a number of key metabolic responses, which contribute to obesity. Critical among these is the control of a transcriptional program required for the commitment of mesenchymal stem cells to the adipocytic lineage. However, in contrast to its role in the cytosol, the functions and targets of nuclear S6K1 are unknown. Here, we show that adipogenic stimuli trigger nuclear translocation of S6K1, leading to H2BS36 phosphorylation and recruitment of EZH2 to H3, which mediates H3K27 trimethylation. This blocks Wnt gene expression, inducing the upregulation of PPARγ and Cebpa and driving increased adipogenesis. Consistent with this finding, white adipose tissue from S6K1-deficient mice exhibits no detectable H2BS36 phosphorylation or H3K27 trimethylation, whereas both responses are highly elevated in obese humans or in mice fed a high-fat diet. These findings define an S6K1-dependent mechanism in early adipogenesis, contributing to the promotion of obesity.

진동수주형 파력발전장치는 수주, 터빈, 발전기 그리고 전력변환 장치 등이 연계된 복잡한 물리적인 특성을 나타낸다. 본 연구는 1/4 스케일의 모형시험을 통해 진동수주와 터빈의 물리적인 관계의 도출에 중점을 두고 있다. 진동수주실과 연성된 터빈의 공력특성은 오리피스를 활용하여 모사하였다. 진동수주실 성능평가에 핵심요소인 터빈효과는 오리피스를 통과하는 유속과 압력강하로 대표할 수 있다. 진동수주형 파력발전장치의 터빈효과는 모형시험에서 계측된 유속과 압력강하로부터 비선형적인 관계를 갖는 것을 확인하였다. The oscillating-water-column wave energy converter represents the complex physical characteristics associated with the water column, turbines, generator, and power converter. This study focuses on the derivation of the physical relationship between the water column and turbine based on the 1/4 scale model test. The aerodynamic characteristics of the OWC ducted turbine were simulated using an orifice. The turbine effect, a key element in the OWC-chamber performance evaluation, can be represented by the flow rate and pressure drop through the orifice. The turbine effect of OWC-WEC was confirmed to have a non-linear relationship from the measured flow rate and pressure drop in the model test.

The authors had treated 66 cases of cleft lip and cleft palate patients, involving 48 male and 18 female patients. There were 58 cases of unilateral cleft lip patients, involving l case of median cleft lip patient, and 8 cases of bilateral cleft lip patients. Patients with both cleft lip and cleft palate were 15 cases among 66 cases. We had performed cheiloplasty by means of Millard s, Hagedorn s, Le Mesurier s and Veau s Methods with Z-plasty and palatorrhaphy by Dorrance s and Von Langenbeck Ernst s methods.
Teager's 에너지 알고리즘을 이용한 한국어 음소인식에 관한 연구

In this paper, as a basic research for constructing a small size Korean speech recognition system we carried out endpoint detection of korean fricatives and affricates using a new energy measuring method based on moodfied Teager'senergy algorithm. Also the data obtained by Teager's energy algorithm were carried out recognition experiments with a feature parameter using neural networks. In case of endpoint detection of Korean fricatives and affricates using Teager's energy algorithm, the algorithm is not only simpler than traditional endpoint detecting algorithm but also more accurate using small computation when endpoint detecting. In our experiments, we used TDNN(Time Delay Neural Network) with input vector represented by 16 melscale FFT coefficients at a 16-msec frame rate, which was a transformed information of endpoint detecting obtained by Teager's energy algorithm. The result of endpoint detection using Teager's energy algorithm was more accurate in affricates compared to fricatives. In case of using neural networks, we obtain the same result. The length of the input vector in voice data has a good recognition rate at 140msec, as the node number of hiddej layer of neural networks increases, the recognition rate is increased.

<P><B>Background:</B></P><P>We evaluated the association between polymorphisms of cytochrome P450 2A6 (<I>CYP2A6</I>)/excision repair cross-complementation group 1 (<I>ERCC1</I>)/X-ray repair cross-complementing group 1(<I>XRCC1</I>) and treatment outcomes of metastatic gastric cancer (MGC) patients treated with S-1/cisplatin.</P><P><B>Methods:</B></P><P>Among MGC patients (<I>n</I>=108), who received S-1 (40 mg m<SUP>−2</SUP> b.i.d., days 1–14) and cisplatin (60 mg m<SUP>−2</SUP>, day 1) every 3 weeks, we analysed the wild-type allele (<I>W</I>) and variants (<I>V</I>) of <I>CYP2A6</I> (<I>*4</I>, <I>*7, *9, *10</I>), and the polymorphisms of <I>ERCC1</I> (rs11615, rs3212986) and <I>XRCC1</I> (rs25487).</P><P><B>Results:</B></P><P>Patients having fewer <I>CYP2A6</I> variants had better response rates (<I>W</I>/<I>W vs W</I>/<I>V</I> other than <I>*1/*4 vs V</I>/<I>V</I> or <I>*1/*4</I>=66.7 <I>vs</I> 58.3 <I>vs</I> 32.3% <I>P</I>=0.008), time to progression (TTP) (7.2 <I>vs</I> 6.1 <I>vs</I> 3.5 months, <I>P</I>=0.021), and overall survival (23.2 <I>vs</I> 15.4 <I>vs</I> 12.0 months, <I>P</I>=0.004). <I>ERCC1 19442C</I>><I>A</I> (rs3212986) was also associated with response rate (<I>C/C</I>, 46.7% <I>vs C/A</I>, 55.3% <I>vs A/A</I>, 87.5%) (<I>P</I>=0.048) and TTP (4.4 <I>vs</I> 7.6 <I>vs</I> 7.9 months) (<I>P</I>=0.012). Patients carrying both risk genotypes of <I>CYP2A6</I> (<I>V</I>/<I>V</I> or <I>1/*4</I>) and <I>ERCC1 19442C</I>><I>A</I> (<I>C/C</I>) <I>vs</I> those carrying none showed an adjusted odds ratio of 0.113 (<I>P</I>=0.004) for response, and adjusted hazard ratios of 3.748 (<I>P</I>=0.0001) for TTP and 2.961 (<I>P</I>=0.006) for death.</P><P><B>Conclusion:</B></P><P>Polymorphisms of <I>CYP2A6</I> and <I>ERCC1 19442C</I>><I>A</I> correlated with the efficacy of S-1/cisplatin.</P>
Sphingomonas aestuarii sp. nov., isolated from tidal flat sediment

<P>A novel bacterium (strain K4(T)) belonging to the genus Sphingomonas was isolated from tidal flat sediment in Korea. Its morphology, physiology, biochemical features and 16S rRNA gene sequence were characterized. Colonies of this strain are yellow in colour and the cells are rod-shaped, exhibiting negative Gram staining. The strain grows at 0-5 % (w/v) NaCl and 20-35 degrees C, with optimal growth occurring at 0 % (w/v) NaCl and 30 degrees C. Phylogenetic analysis based on 16S rRNA gene sequences indicated that strain K4(T) is associated with the genus Sphingomonas. Within the phylogenetic tree, this novel strain shares a branching point with Sphingomonas asaccharolytica Y-345(T), with which it shares 97.3 % 16S rRNA gene sequence similarity. The polyamine pattern predominantly contains the Sphingomonas-specific triamine sym-homospermidine. Combined analysis of 16S rRNA gene sequences, DNA-DNA relatedness, physiological and biochemical test results identified genotypic and phenotypic differences between strain K4(T) and other Sphingomonas species. On the basis of these differentiating features, it is concluded that strain K4(T) (=KCTC 22050(T)=DSM 19475(T)) represents a novel species of the genus Sphingomonas, for which the name Sphingomonas aestuarii sp. nov. is proposed.</P>
