        Effects of controlled atmosphere on browning, redox metabolism and kernel quality of fresh in-hull walnut (Juglans regia L.)

        The green hull of fresh walnuts ( Juglans regia L.) is highly susceptible to browning and accompanied with deterioration ofthe walnut quality, and the currently available preservation technologies still do not meet the market threshold supply of freshwalnut. This study investigated the eff ect of controlled atmosphere (CA) storage alone and combination with 1-Methylcyclopropene(1-MCP) on hull browning and kernel quality of fresh in-hull walnut. Fresh in-hull walnuts were treated with air(Control), CA (5% O 2 , 7.5% CO 2 ), 1-MCP (0.5 μL L −1 ) + CA and stored at 0 ± 0.5 °C for 100 d. Browning incidence, hullredox metabolism, kernel color and walnut oil quality were analyzed. The results showed that CA promoted the activities ofsuperoxide dismutase (SOD), catalase (CAT), and ascorbate peroxidase (APX), maintained total phenol (TP), γ-aminobutyricacid (GABA), ascorbic acid (ASA), and glutathione (GSH) contents, inhibited ethylene production, reduced reactive oxygenspecies (ROS) accumulation, and delayed the hull browning. Besides, CA attained the highest kernel quality throughinhibited the decrease of kernel color value and moisture content, and reduced acid value (AV) and peroxide value (POV)of walnut oil. While 1-MCP + CA treatment increased the activities of antioxidant enzymes, the combination weakened theother positive eff ects of CA. However, hull browning and kernel deterioration in 1-MCP + CA were less than that observedin air control. Overall, CA was an eff ective method for the preservation of fresh in-hull walnut at 0 ± 0.5 °C.

      • Identification and Functional Analysis of Differentially Expressed Genes Related to Metastatic Osteosarcoma

        Background: To explore the molecular mechanisms of metastatic osteosarcoma (OS) by using the microarray expression profiles of metastatic and non-metastatic OS samples. Materials and Methods: The gene expression profile GSE37552 was downloaded from Gene Expression Omnibus database, including 2 human metastatic OS cell line models and 2 two non-metastatic OS cell line models. The differentially expressed genes (DEGs) were identified by Multtest package in R language. In addition, functional enrichment analysis of the DEGs was performed by WebGestalt, and the protein-protein interaction (PPI) networks were constructed by Hitpredict, then the signal pathways of the genes involved in the networks were performed by Kyoto Encyclopaedia of Genes and Genomes (KEGG) automatic annotation server (KAAS). Results: A total of 237 genes were classified as DEGs in metastatic OS. The most significant up- and down-regulated genes were A2M (alpha-2-macroglobulin) and BCAN (brevican). The DEGs were significantly related to the response to hormone stimulus, and the PPI network of A2M contained IL1B (interleukin), LRP1 (low-density lipoprotein receptor-related protein 1) and PDGF (platelet-derived growth factor). Furthermore, the MAPK signaling pathway and focal adhesion were significantly enriched. Conclusions: A2M and its interactive proteins, such as IL1B, LRP1 and PDGF may be candidate target molecules to monitor, diagnose and treat metastatic OS. The response to hormone stimulus, MAPK signaling pathway and focal adhesion may play important roles in metastatic OS.

        Determinants of Coopetition Relationships in International Joint Ventures for High-Speed Rail Projects

        The mileage of high-speed rail (HSR) is expected to grow in the foreseeable future. Fierce international competition and HSR business uniqueness have facilitated the broad application of international joint ventures (IJVs) globally. However, the interaction between competition and cooperation complicates IJVs, and IJVs partners’ implementation has rarely been informed concerning the fluent operation of IJVs for HSR projects. This study aims to examine the determinants of IJVs coopetition for HSR projects. We identified 35 variables along with their clusters through literature review and pilot study, and collected data from the international HSR industry using a questionnaire survey. Eight clusters verified by the confirmatory factor analysis come from three levels: macro-level, project-level, and firm-level. Based on the partial least squares structural equation modeling (PLS-SEM), a path model for the coopetition relationship has been established, demonstrating that macro-level and project-level determinants affect the coopetition relationship via the firm-level determinants. Furthermore, the path model indicates recommendations corresponding to various situations (isolating situation, partnering situation, contending situation, and adapting situation) for IJVs members to find a way to improve the coopetition relationship. Thus, this study not only contributes to the existing knowledge body on coopetition theory, but also enhances HSR contractors’ comprehension of the coopetition relationship determinants as well as the interactions.

        A Comparison Study of Kinematic and Dynamic Models for Trajectory Tracking of Autonomous Vehicles Using Model Predictive Control

        Efficient trajectory tracking approaches can enable autonomous vehicles not only to get a smooth trajectory but to achieve a lower energy dissipation. Since vehicle model plays an important role in trajectory tracking, this paper investigates and compares the performance of two classical vehicle models for trajectory tracking of autonomous vehicles using model predictive control (MPC). Firstly, a two-degree-of-freedom kinematic model and a three-degree-of-freedom yaw dynamic model are established for autonomous vehicles. Meanwhile, in order to carry out tracking control more effectively and smoothly, the tire slip angle has been taken into account by the dynamic model. Then, we design two MPC controllers for trajectory tracking, which are based on the kinematic model and the dynamic model, respectively. The performances of two MPC controllers are evaluated and compared on the Carsim/Matlab joint simulation platform. Experimental results demonstrated that, under low-speed working conditions, both two MPC controllers can follow the reference trajectory with high accuracy and stability. However, under high-speed working conditions, the tracking error of the kinematic model is too large to be used in the real trajectory tracking problem. On the contrary, the controller based on the dynamic model still performs a good tracking effect. In addition, this study offers guidance on how to select a suitable vehicle model for autonomous vehicles under different speed working conditions.

        Pre-harvest ethylene control affects vase life of cut rose ‘Carola’ by regulating energy metabolism and antioxidant enzyme activity

        We studied the role of ethylene control in regulating energy metabolism, antioxidant enzyme activity, and vase life of cutrose Rosa hybrida ‘Carola’. Rose fl owers at stage II were sprayed with one of the following solutions: water (control), 10μL L −1 1-methylcyclopropene (1-MCP), or 0.5 g L −1 2-chloroethanephosphonic acid (ethephon). After harvest, ethyleneproduction rate, respiration intensity, energy charge (EC), activities of energy metabolism-related and antioxidant enzymes,and malondialdehyde (MDA) content were measured. Results showed that 1-MCP enhanced the activities of superoxidedismutase, H + -adenosine triphosphatase, Ca 2+ -adenosine triphosphatase, succinic dehydrogenase, and cytochrome c oxidase,increased adenosine triphosphate (ATP) content, maintained high EC levels, inhibited respiration intensity, reduced peroxidase(POD) and polyphenol oxidase (PPO) activity and MDA accumulation, and prolonged vase life. Ethephon promotedethylene production and respiration intensity, increased POD and PPO activity, reduced ATP content and EC levels, andaccelerated senescence. Our results support a novel role for ethylene control in regulating senescence of ‘Carola’.

        Isoflavanones from the Stem of Cassia siamea and Their Anti-tobacco Mosaic Virus Activities

        Two new isoflavanones, (3R) 7,2',4'-trihydroxy-3'-methoxy-5-methoxycarbonyl-isoflavanone (1) and (3R) 7,2'-dihydroxy-3',4'-dimethoxy-5-methoxycarbonyl-isoflavanone (2), together with six known isoflavanones (3-8), were isolated from the stems of Cassia siamea. The structure of 1-8 was elucidated by spectroscopic methods including extensive 1D- and 2D-NMR techniques. Compounds 1, 2, 5-8 were evaluated for their anti-tobacco mosaic virus (Anti-TMV) activity. The results showed that compounds 1 and 6 showed potential anti-TMV activity with inhibition rates of 24.6% and 26.9%, respectively. Compounds 2, 5, 7, 8 also showed anti-TMV activity with inhibition rates in the range of 11.8-18.6%.

        A soybean plastidic ATP/ADP transporter gene, GmAATP, is involved in carbohydrate metabolism in transgenic Arabidopsis

        The plastidic ATP/ADP transporter (AATP) imports adenosine triphosphate (ATP) from the cytosol into plastids, resulting in the increase of the ATP supply to facilitate anabolic synthesis in heterotrophic plastids of dicotyledonous plants. The regulatory role of GmAATP from soybean in increasing starch accumulation has not been investigated. In this study, a gene encoding the AATP protein, named GmAATP, was successfully isolated from soybean. Transient expression of GmAATP in Arabidopsis protoplasts and Nicotiana benthamiana leaf epidermal cells revealed the plastidic localization of GmAATP. Its expression was induced by exogenous sucrose treatment in soybean. The coding region of GmAATP was cloned into a binary vector under the control of 35S promoter and then transformed into Arabidopsis to obtain transgenic plants. Constitutive expression of GmAATP significantly increased the sucrose and starch accumulation in the transgenic plants. Real-time quantitative PCR (qRT-PCR) analysis showed that constitutive expression of GmAATP up-regulated the expression of phosphoglucomutase (AtPGM), ADP-glucose pyrophosphorylase (AGPase) small subunit (AtAGPase-S1 and AtAGPase-S2), AGPase large subunit (AtAGPase-L1 and AtAGPase-L2), granule-bound starch synthase (AtGBSS I and AtGBSS II), soluble starch synthases (AtSSS I, AtSSS II, AtSSS III, and AtSSS IV), and starch branching enzyme (AtSBE I and AtSBE II) genes involved in starch biosynthesis in the transgenic Arabidopsis plants. Meanwhile, enzymatic analyses indicated that the major enzymes (AGPase, GBSS, SSS, and SBE) involved in the starch biosynthesis exhibited higher activities in the transgenic plants compared to the wild type (WT). These findings suggest that GmAATP may improve starch content of Arabidopsis by up-regulating the expression of the related genes and increasing the activities of the major enzymes involved in starch biosynthesis. All these results suggest that GmAATP could be used as a candidate gene for developing high starch-accumulating plants as alternative energy crops.

        Two New Diphenylethylenes from Arundina graminifolia and Their Cytotoxicity

        Two new diphenylethylenes, gramniphenols H and I (1 and 2), together with six known diphenylethylenes (3- 8), were isolated from Arundina graminifolia. The structures of 1-8 were elucidated by spectroscopic methods including extensive 1D- and 2D-NMR techniques. Compounds 1 and 2 were evaluated for their cytotoxicity against five human tumor cell lines. Compound 1 showed cytotoxicity against PC3 cells with IC50 value of 3.5 μM. Compound 2 showed cytotoxicity against NB4 and PC3 cells with IC50 values of 3.6 and 3.8 μM, respectively.

      • Comparison of Gadobenate Dimeglumine and Gadopentetate Dimeglumine for Breast MRI Screening: a Meta-analysis

        Background: As a common and essential contrast medium at present, gadobenate dimeglumine has shown better performance than some other agents when applied to Breast Magnetic Resonance Imaging Screening (Breast MRI Screening). Nevertheless, reports on the diagnostic performance of these two mediums (gadobenate dimeglumine and gadopentetate dimeglumine) are not completely consistent. Objective: To assess the diagnostic value of gadobenate dimeglumine and gadopentetate dimeglumine for Breast MRI Screening in patients suffering from breast cancer and to provide more convinced evidence to guide clinical practice in terms of appropriate contrast agents. Data Sources and Review Methods: Original articles in English and Chinese published before January 2013 were selected from available databases (The Cochrane Library, PUBMED, EMBASE, Chinese Biomedical Literature Database, Chinese Scientific Journals Full-text Database, Chinese Journal Full-text). The criteria for inclusion and exclusion were based on the standard for diagnosis tests. Meta-Disc software (Version 1.4) was used for data analysis. Then, the area under curve (AUC) of SROC and the spearman rank correlation of sensitivity against (1-specificity) were calculated. Results: Total of 17 researches involving 1934 patients were included. The pooled sensitivity of gadobenate dimeglumine and gadopentetate dimeglumine were 0.99 (0.97, 1.00) and 0.93 (0.88, 1.00) respectively. The pooled specificity for these two contrast agents were 0.924 (0.902, 0.943) and 0.838 (0.817, 0.858) respectively, and the AUC of SROC curve were 0.9781 and 0.9215 respectively. Conclusions: Gadobenate dimeglumine can be regarded as a more effective and feasible contrast medium for Breast MRI Screening. At least 5% differences in diagnostic performance are usually considered as clinically relevant.
