        Study on the In-vitro Culture Method for Normal Embryonic Cell Development of Porcine Parthenogenetic Embryos

        In the early development of parthenogenetic embryo, cytoplasm and nucleic acid fragmentation may be a cause of lower embryo development. The purpose of this study was to evaluate whether embryonic development and apoptosis factors can be reduced by controlling the in-vitro culture environment by the addition of hormones, pregnancy serum and uterine milk. Our study showed that the activity of Casp-3 increased within the cytoplasm when artificially used hormones to induce the incubation environment, and PCNA's manifestation was low. However, the addition of pregnant serum appeared to lower the Casp-3 activity compared to the other groups. In addition, MMP-9 activity was increased and early embryo development and cytoplasmic fidelity were also increased. Therefore, the results of the present study showed that the use of gestational serum in the development of parthenogenetic embryo inhibit apoptosis and increases cytoplasmic reorganization by natural environmental control in in vitro culture.

      • Effect of MMPs Expression on Maturation and Cellular Growth of Blastocysts in Bovine

        The expression of MMPs in the development of the fertilized egg has a very important role in cell configuration. Objective To evaluate the clinical, the effect of differentially expressed MMPs on serum and serum - free medium on the maturation of blastocysts. The expression patterns of MMPs in serum and serum-free medium were compared at 6 h, 18 h and at the blastocyst stage using real-time PCR, ELISA and immunofluorescence. The results showed that the expression of MMPs was increased in the embryos of the serum medium, as a result of analysis of MMPs and TIMPs, MMP-2 was expressed in the cytoplasm of embryos in the serum-free medium, And it was found to be higher in expression than MMP-9. The serum medium was different from the bloodless badge: overall, TIMPs showed a higher expression in the ovarian cells than cyanosis, and TIMP-3 was more pronounced. Development rate of blastocyst according to in vitro culture method was higher than that of serum - free medium (61.22% 60/98) and serum - free medium (48.28% 28/58). Analysis of the protein release locations of MMPs and TIMPs showed that MMPs and TIMPs are highly expressed in serum mediums, focusing on the inner cell mass. However, very low expression appeared in the tropoblast. On the other hand, serum - free medium showed different expression from serum medium and TIMPs expression was generally low. Therefore, in the case of serum media, the expression of MMPs is highly expressed in the cytoplasm of the fertilized egg, increasing the reconstruction of cells.

      • Expression Analysis of Programmed Cell Death Patterns on the In-vitro Culture of Supplemented with Cell Death Regulation Factor during Development of Nuclear Transfer Embryos in Porcine

        The nature of molecular mechanisms governing embryo development is largely unknown, but recent reports have demonstrated that proper execution of programmed cell death is crucial for this process. The main objective of this study is to examine the mode of programmed cell death during nuclear transfer embryos development in porcine. In particular, the relative employment of two major pathways in programmed cell death; e.g. apoptosis (type I) and autophagy (type II) was compared. Oocytes use in the study was matured in vitro in the presence of 10% FBS maturation medium. After nuclear transfer embryos were cultured for each programmed cell death control factor [Cysteamine(Cyst : 0.4mM), 3-methyladenine(3MA : 2.5mM) and Rapamycin(RP : 100nM)] in TCM-199 medium supplemented with 0.1% BSA. In this study results of among the blastocysts development in 3MA; PCNA, MAP1LC3A and ATG5 RNA gene expression level increased in the order of IVF<Cyst < 3MA < RP. However Casp-3 and TNF-r RNA gene expression level decreased in the order of IVF < 3MA and RP< Cyst. The expression of mTOR showed a pattern opposite to that of MAP1LC3A. That is, its expression was the lowest in Cyst group. And next experiments analysis of MMP expression patterns. Analysed this MMPs enzyme activation to evaluate the effectiveness of high quality brastocyst culture in porcine. In this results of the enzymatic activity of MMP-2 and MMP-9 was assessed in culture, the level of active MMP-9 was higher expression in the medium of each 3MA and RP treatment group, with the level of another treatment group being relatively higher. These results suggest that the autophagy activation culture medium is more effective for stable and innovative nuclear transfer embryos development.

        최근 4차 산업 혁명 및 코로나19로 인해 비대면 서비스 시장의 수요가 증가하고 있으며, 전 세계적으로 로봇을 이용한 다양한 서비스들이 개발되고 있는 추세이다. 본 논문에서는 사람 대신 로봇이 테이블 위에 놓인 물체들을 적절한 수거함들로 옮겨 테이블을 정리해주는 지능형 로봇 서비스를 구현함으로 목표로 한다. 효과적인 목표 서비스 개발을 위해, 본 연구에서는 딥러닝 기반의 심층 물체 자세 추정기를 이용해 물체들의 실시간 위치를 파악하고, 시멘틱 웹 기반의 지능형 로봇 체계를 활용하여 작업 환경에서 동적 상황 정보를 추론해낸다. Recently, due to the 4th Industrial Revolution and COVID-19, demand in the non-face-to-face service market has been increasing and there is a trend to develop various useful robot services around the world. In this paper, we aim to develop an intelligent robot service that the robot cleanups a table by picking and placing objects placed on the table into proper bins instead of human users. In order to develop the target service effectively, we use a deep learning-based object pose estimator to find out real-time pose of individual objects, and make use of a robot intelligence framework based on semantic web technology to infer dynamic context from the task environment.

        Purpose : In a general case, the life test could designed based on the failure mode and mechanism of components and materials or field failure cases. However, it is difficult to predict which components or parts could cause failure of an assembly in case of a new developed product or assembly that combined many different components and materials. Therefore, in this paper we studied the life test for the new developed assembly A that combined many electronic components. Methods : It is important for the life test design to choose an effective stress of the assembly A. The effective stress factor selecting is most important especially when we could not know the failure mode and mechanism. In this paper, the effects of every possible stresses were analysed using QFD(Quality Function Deployment) method. Temperature difference between ground and high altitude was found as the effective stress of the assembly A. To accelerate temperature difference we designed the life test that accelerated the life cycle. Result : The life test was designed for the new developed assembly A and the failure were found in every samples at the similar test step. Samples were three. The fault was not occurred the specific component but the connect part that connect between two circuit cards. We predicted the MCBF(Mean Cycle Between Failure) of the assembly A using every samples’ failure cycles based on a statistical analysis. Conclusion : In this paper the life test design was analysed and done for the assembly A which combined many electronic components. It is difficult to generalized the life test method for new developed assemblies. Therefore, from the beginning of various life test analyses are necessary to distinguish factors that affect the lifetime.

        NaBH4 가수분해 반응에 의한 무인항공기용 PEMFC 수소공급

        Proton Exchange Membrane Fuel Cells (PEMFC) instead of batteries is appropriate for long time flight of unmanned aero vehicles (UAV). In this work, NaBH4 hydrolysis system supplying hydrogen to PEMFC was studied. In order to decrease weight of NaBH4 hydrolysis system, enhancement of hydrogen yield, recovery of condensing water and maintenance of stable hydrogen yield were studied. The hydrogen yield of 3.4% was increased by controlling of hydrogen pressure in hydrolysis reactor. Condensing water formed during air cooling of hydrogen was recovered into storage tank of NaBH4 solution. In this process the condensing water dissolved NaBH4 powder and then addition of NaBH4 solution decreased system weight of 14%. NaBH4 hydrolysis system was stably operated with hydrogen yield of 96% by 2.0g Co-P-B catalyst for 10 hours at 2.0L/min hydrogen evolution rate.

        단호박 가루를 첨가한 호상요구르트의 품질 특성

        Sweet Pumpkin (Cucurbita maxima) is rich in minerals, vitamins, and dietary fibers and has less fat. It is thus effective for dieting and for preventing constipation. Sweet Pumpkin contains vitamin A, particularly beta-carotene, which is turned into retinol in the body and helps protect the vision. It is currently on the spotlight as a vision care, anticancer, and diet food, among others, but because it is hard to find yogurt made of Sweet Pumpkin, in this study, Sweet Pumpkin powder was added to yogurt for promotion as a new health food. Yogurt was made with 0, 0.5, 1, 2, and 3% Sweet Pumpkin powder, and the color properties (brightness, redness, and yellowness), viscosity, pH, and acidity were measured through a sensory test (color, flavor, taste, overall acceptability). The storage stability at five-day intervals for one day, five days, ten days, 15 days, etc. was also experimented on. For the color properties, the greater the amount of Sweet Pumpkin powder added, the greater the decrease in the lightness (L) value, but the redness (a) and yellowness (b) values increased (p<0.001). For the viscosity, it was decreased for five days and then increased by 47-68% for ten days. This showed a significant difference in storage stability period: one day (p<0.001), five days (p<0.01), and ten days (p<0.01) (except 15 days). Further, there was a significant difference in pH and acidity [pH: for one day (p<0.001), five days (p<0.01), and ten days (p<0.01); acidity: one day (p<0.001), ten days (p<0.05), 15 days (p<0.001)]. No significant difference in pH was shown, however, for 15 days, and in acidity for five days. The results of the sensory test are highly estimated in the order of 1%>2%.3%>0%>0.5%. The study results indicate that the addition of 1% Sweet Pumpkin powder is most suitable for manufacturing Sweet Pumpkin yogurt.

        다시마를 이용한 샐러드 드레싱 제조의 품질 특성

        This study was conducted to develop a novel salad dressing composite recipe of natural seasoning containing the dried sea tangle(Laminaria japonica) that has a high preference. Sea tangle(Laminaria japonica) is included in the vitamins and minerals, magnesium, calcium, iodine, iron content, such as high, and contained in Sea tangle alginate is not a small conference known as dietary fiber. To manufacture salad dressing with sea tangle, dressing with 0%, 3%, 6%, 9%, and 12% added sea tangle were prepared and tested for quality. The pH tended to increase with the increased sea tangle in addition but in contrast, acidity showed. The ``L`` color decreased with added sea tangle, whereas the ``a`` and ``b`` values increased. Brix measurements increase with added sea tangle. The strength texture results, 0% was the highest, lowest 9%. Bitterness and chewiness texture results, 9% was the highest, lowest 0%. According to the sensory test results, in the topic overall quality 3% was by 3.76 point the highest. But, during total nine clause, in clause six, by 6% was the highest.
