        저혈청 배지에서 인간 전골수세포(HL-60)를 이용한 tPA 생산과 세포사멸기작에 관한 연구

        tPA 생산과 세포 사멸의 연구를 하기 위해서 인간 전골수세포가 배양되었다. 연속 관류 배양하에서 최대 tPA 생산량은 324 ng/ml이며 최대 세포 농도는 5.27×10 exp (6) cells/ml였다. Phorbol ester를 넣었을 때 tPA 생산량은 420 ng/ml까지 향상되었다. 형광 현미경으로 관찰했을 때, apoptosis는 세포 배양 초기부터 중반까지 우세했고 빠른 교반 속도에서도 우세했다. 그러나 necrosis는 세포배양 말기와 동결된 세포가 해동 되었을 때 우세했다. HL-60 was cultivated to produce tPA (tissue-type plasminogen activator) and study the mechanism of cell death. Maximum cell density and tPA production were obtained as 5.27×10 exp (6) cells/ml and 324 ng/ml, respectively under perfusion cultivation. tPA production was enhanced to 420 ng/ml in adding 160 nM of phorbol ester. The cells were gradually differentiated to granulocytes rather than proliferation. By Fluorescent microscope, apoptosis was prevailed except the death phase and in high agitation speed, but necrosis was prevailed in thawed cells and during the latter periods of the cultivation. It was also proved that tPA was most produced in apoptosis. To obtain higher tPA productivity, the cells must be maintained in apoptosis, not necrosis phase when the cells were dying.

        Ferromagnetic Properties in Diluted Magnetic Semiconductors (Al,Mn)N grown by PEMBE

        We present the structural, magnetic, and electrical properties in the (Al,Mn)N films with various Mn concentrations grown by plasma-enhanced molecular beam epitaxy. X-ray diffraction analyses reveal that the (Al,Mn)N films have the wurtzite structure without secondary phases. All (Al,Mn)N films showed the ferromagnetic ordering. Particularly, ($Al_{1-x}Mn_{x}$)N film with x = 0.028 exhibited the highest magnetic moment per Mn atom at room temperature. Since all the films exhibit the insulating characteristics, the origin of ferromagnetism in (Al,Mn)N might be attributed to either indirect exchange interaction caused by virtual electron excitations from Mn acceptor level to the valence band within the samples or a percolation of bound magnetic polarons arisen from exchange interaction of localized carriers with magnetic impurities in a low carrier density regime.

        <P>A design platform for a zero drive load logic inverter consisting of p-channel Si nanowire based transistors, which controlled their operating mode through an implantation into a gate dielectric layer was demonstrated.         Genetic Analysis of Carotenoids Content in Red Pepper (Capsicum annuum L.) Fruit using Complete Diallel Cross

        Regarding carotenoids content, the genetic basis, heritability and combining ability in six red pepper inbred lines were investigated using full diallel crosses. Both additive and non-additive gene actions govern inheritance of carotenoids content. The mean square of array through variance and covariance analysis (Wr-Vr) was insignificant, which suggest that inbred lines involved in diallel cross may have no epistatic effects. The Vr/Wr graph revealed the influence of partial dominant gene action towards low carotenoids content and the absence of non-allelic interaction. The H2 component was smaller than the H1 and the [H2/4H1] component was 0.187 less than 0.25, indicating unequal proportion of positive and negative alleles in the parents. The estimates of broad and narrow sense heritability for carotenoids content were 0.956 and 0.832, respectively. The variance of general combining ability (GCA) was relatively higher than that of specific combining ability (SCA), which implied that the additive gene effects were predominant as compared to both dominant and epistatic effects for the accumulation of carotenoids in this genetic population. The values of GCA of ‘62024L1’ and ‘62067L2’ were higher than those of the other parents. These 2 inbred lines, therefore, can be considered as useful breeding materials to enhance fruit carotenoids content in other red pepper varieties.

        영양 불량 입원 환자에서 영양소 섭취 변화 분석

        The purpose of this study was to examine changes in nutrient intake in patients at nutritional risk. We included106 malnourished patients who were admitted to Severance Hospital from March to September 2014. The average age was 59.0±11.6 years old and 59 patients were male (59.7%). The majority of diagnosis wascancer (94.3%). We evaluated patients` nutritional status by scored patient-generated subjective global assessment(PG-SGA), anthropometric measurement (height, weight, body composition by Bioelectrical impedanceanalysis), hand grip strength, biochemical and dietary assessment. The patients` daily intakes of energy andprotein (1,019.5±706.4 kcal, 40.4±27.7 g) during hospitalization were significantly lower than their usual intakes(1,382.0±499.8 kcal, 54.4±25.1 g, P<0.001). Serum levels of albumin, cholesterol, and total lymphocyteswere significantly reduced during hospital stay. The negatively influencing factors for reduced dietary intake wereanorexia (42.5%), abdominal distention (14.2%), pain (13.2%), and others. The results of this study could beused to establish baseline data for developing new strategies for nutritional intervention in malnourished patients.

        A pathologic study is made on fifty-eight cases of salivary gland tumors of Koreans, obtained during a period of twelve years, 1955^1966, at the Department of Pathology, College of Medicine, Seoul National University, and followings are the results: 1. The tumors examined consist of 38(65.2%) benign and 20(24.8%) cases of malignant tumors. 2. Incidence of histological types of these tumors is highest in benign pleomorphic adenoma (62.1 %), and thereafter malignant pleomorphic adenoma (8.6%), adenocarcinoma (8.6%) and squamous cell carcinoma (6.9%) in order. of frequency. 3. Site distribution shows major salivary glands to be involved 68.9%, in minor 20.5%, and site undeter-', mined 10.3%, respectively. Of the major ones, parotid glands are most frequently affected in 25.9%, followed ',.by submandibular 12.1% , and sublingual 6.9%. Of the minor ones, palate, lip, tongue, maxilla, and mandible are affected in order of frequency. 4. Overall age distribution shows highest peaks of benign tumors in 4th decade, and of malignant in 5th. 5, Male is more frequently affected with ratio of 1.8 : 1 to female.

        Distribution Channel and Microbial Characteristics of Pig By-products in Korea

        The distribution channel of meat by-products from the pig farm to the final consumer can include a meat processor, wholesale market, wholesaler, retailer, and butcher shop. Bacterial contamination at any of these steps remains to be a serious public health concern. The aim of this study was to evaluate the distribution channel and microbial characteristics of pig byproducts in Korea. Upon evaluation of pig by-products in cold storage, we found that the small and large intestine were significantly (p<0.05) higher in pH value compared to the heart and liver. The total plate counts were not significantly different among offals until cold storage for 7 d. The coliform count after 1 d of cold storage was significantly (p<0.05) higher in small and large intestine than in the other organs. The coliform count of heart, liver, and stomach showed a higher coliform count than small and large intestine until 7 d of cold storage. As determined by 16S rRNA sequencing, contamination of major pig by-products with Escherichia coli, Shigella spp., and other bacterial species occurred. Therefore, our results suggest that a more careful washing process is needed to maintain quality and hygiene and to ensure the safety of pig by-products, especially for small and large intestine.

        혈관 손상에 동반되는 혈관 내막 평활근의 중식에 미치는 Ramipril 의 효과

        Objective: Advanced atherosclerotic lesion consists mainly of intimal smooth muscle cell proliferation. Many factors are considered to be implicated in this process. The role of locally produced angiotensin II as a potent growth stimulus had been investigated. To evaluate the effect of ramipril, a new converting enzyme inhibitor, on the smooth muscle proliferative process this animal study was performed, Ramirpil was selected because of its unique physicochemical properties. Methods: Forty nine male New Zealand White or Chinchilla rabbits were randomly assigned to four groups: Group I (N=5), normal control; Group II (N=12), 2% cholesterol diet feeding without vascular injury; Group III (N =16), 2% cholesterol diet feeding after vascular injury; Group IV (N=16), ramipril treated group. Bilateral femoral atherosclerosis was induced by endothelial damage using desiccated nitrogen gas followed by a 4-week high cholesterol diet (2%-cholesterol & 6%-peanut oil). Ramipril, dose of 1 mg/kg/day, was given from 7 days before injury. Results: No definite atherosclerotic lesion was found in group II rabbits, But atherosclerotic lesion with intimal enlargement was induced in all group III & IV rabbits. The lesions were made of abundant cellular components, collagen fibers, & elastic fibers. The presence of intimal smooth muscle cells was confirmed by the immunohistochemical staining with muscle-specific monoclonal antibody HHF-35. Occasionally the foam cell lesions were scattered in the media as well as in the intima. There was frequent disruption of internal elastic lamina. Luminal area, neointimal area, media area, & ratio of intima to media were examined from the histologic sections of the femoral artery. There was no significant difference in all parameters between group I & group II rabbits. But between group I & group III rabbits was there significant difference in all 4 parameters. The same relationship was also found between group II & group III. So it was apparent that the hypercholesterolemic diet for 4 weeks alone could not induce atherosclerotic lesion in this animal model. The luminal area was not significantly different between group III & group IV. (0.10±0.08mm2 vs. 0.11±0.08mm2; p>0.05). The neointimal area of ramipril treated group was significantly smaller than that of group III rabbits (0.56±0.36mm2 vs. 0.17±0.12mm2; p<0.01). The media area was also smaller in ramipril treated group (0.34±0.17 mm2 vs. 0.22±0.06 mm2, p<0.05). The radio of neointima area to media area was higher in group III (1.67±0.80 vs. 0.76±0.44; p<0.01). Conclusion: This rabbit-desiccation injury-cholesterol diet model proved to be useful for the study of atherogenesis. Ramipril was considered to inhibit the intimal smooth muscle cell proliferation in this animal model.
